The Crop Protection unit of the Institute for Agricultural and Fisheries Research offers plant disease diagnostic services through its ‘Diagnostic Centre for Plants’ (ILVO‐DCP). ILVO‐DCP has requested accreditation (ISO 17025) for a number of diagnostic detection methods in bacteriology, mycology, entomology and nematology. Accreditation forms an essential part of quality control programs such as ISO17025, which can in part be realized by proving the laboratory's competence in inter‐laboratory proficiency or ring tests. In 2006, ILVO‐DCP organized such a proficiency test for the detection of Phytophthora ramorum. The protocol was developed using the standard ILAC‐G13:2000, and defined rules for participation, sample preparation and transport, communication, fraud prevention and reporting. Eight European diagnostic centres participated in the proficiency test including ILVO‐DCP. Each participant received one set of 10 coded samples, each sample consisting of either leaves or stems that were artificially inoculated with either the target or an alternative organism. Participants could use one or more methods listed in the EPPO diagnostic protocol PM 7/66. They had to report within a specific timeframe and received a detailed report of their performance. The success rate of the proficiency test was 100%. This paper lists some of the experiences gained from organising this type of proficiency test.  相似文献

BACKGROUND: In addition to their possible direct biological effects, plasma carotenoids can be used as biochemical markers of fruit and vegetable consumption for identifying diet-disease associations in epidemiological studies. Few studies have compared levels of these carotenoids between countries in Europe. OBJECTIVE: Our aim was to assess the variability of plasma carotenoid levels within the cohort of the European Prospective Investigation into Cancer and Nutrition (EPIC). METHODS: Plasma levels of six carotenoids--alpha-carotene, beta-carotene, beta-cryptoxanthin, lycopene, lutein and zeaxanthin--were measured cross-sectionally in 3043 study subjects from 16 regions in nine European countries. We investigated the relative influence of gender, season, age, body mass index (BMI), alcohol intake and smoking status on plasma levels of the carotenoids. RESULTS: Mean plasma level of the sum of the six carotenoids varied twofold between regions (1.35 micromol l(-1) for men in Malm?, Sweden vs. 2.79 micromol l(-1) for men in Ragusa/Naples, Italy; 1.61 micromol l(-1) for women in The Netherlands vs. 3.52 micromol l(-1) in Ragusa/Naples, Italy). Mean levels of individual carotenoids varied up to fourfold (alpha-carotene: 0.06 micromol l(-1) for men in Murcia, Spain vs. 0.25 micromol l(-1) for vegetarian men living in the UK). In multivariate regression analyses, region was the most important predictor of total plasma carotenoid level (partial R(2)=27.3%), followed by BMI (partial R(2)=5.2%), gender (partial R(2)=2.7%) and smoking status (partial R(2)=2.8%). Females had higher total carotenoid levels than males across Europe. CONCLUSIONS: Plasma levels of carotenoids vary substantially between 16 different regions in Italy, Greece, Spain, France, Germany, the UK, Sweden, Denmark and The Netherlands. Compared with region of residence, the other demographic and lifestyle factors and laboratory measurements have limited predictive value for plasma carotenoid levels in Europe.  相似文献

The aim of the study was to assess whether the hair of stray and domestic dogs in Egypt was contaminated with the eggs of the zoonotic parasite Toxocara canis, and also to identify risk factors for T. canis for contamination. Paired samples of hair and feces were collected from 53 stray and 47 domestic dogs, and hair samples were obtained from a further 11 stray and 9 domestic dogs. All samples were examined to identify T. canis eggs and, if eggs were found, their maturation stage. Eggs were identified in 26.6% of stray and 10.7% of domestic dog's hair samples. A significantly increased risk of embryonated T. canis eggs in hair samples was found in stray dogs (p=0.04), stray dogs had 3.18 (95% CI: 1.04-9.74) times the odds of having T. canis eggs present compared with domestic dogs. There was also a significant difference (p=0.02) between the mean quantity of eggs per gram in stray (77.6±6.54) and domestic (48.7±6.65) dog's hair. Fecal examination found a T. canis egg prevalence of 35.8% and 21.3% in stray and domestic dogs, respectively. As no domestic dogs which were positive from hair samples had negative fecal samples, this indicates that the presence of T. canis eggs in hair is probably due to self contamination. Two stray dogs had positive hair samples but negative fecal samples indicating that contamination may also be environmental. As both non-embryonated and embryonated T. canis eggs were found in the hair of domestic dogs, direct contact with dogs may be a potential risk factor for transmission of T. canis eggs to humans.  相似文献

A constructed wetland composed of a pond- and a marsh-type wetland was employed to remove nitrogen (N) and phosphorus (P) from effluent of a secondary wastewater treatment plant in Korea. Nutrient concentrations in inflow water and outflow water were monitored around 50 times over a 1-year period. To simulate N and P dynamics in a pond- and a marsh-type wetland, mesocosm experiments were conducted. In the field monitoring, ammonium (NH 4 + ) decreased from 4.6 to 1.7 mg L?1, nitrate (NO 3 ? ) decreased from 6.8 to 5.3 mg L?1, total N (TN) decreased from 14.6 to 10.1 mg L?1, and total P (TP) decreased from 1.6 to 1.1 mg L?1. Average removal efficiencies (loading basis) for NO 3 ? , NH 4 + , TN, and TP were over 70%. Of the environmental variables we considered, water temperature exhibited significant positive correlations with removal rates for the nutrients except for NH 4 + . Results from mesocosm experiments indicated that NH 4 + was removed similarly in both pond- and marsh-type mesocosms within 1 day, but that NO 3 ? was removed more efficiently in marsh-type mesocosms, which required a longer retention time (2?C4 days). Phosphorus was significantly removed similarly in both pond- and marsh-type mesocosms within 1 day. Based on the results, we infer that wetland system composed of a pond- and a marsh-type wetland consecutively can enhance nutrient removal efficiency compared with mono-type wetland. The reason is that removal of NH 4 + and P can be maximized in the pond while NO 3 ? requiring longer retention time can be removed through both pond and marsh. Overall results of this study suggest that a constructed wetland composed of a pond- and a marsh-type wetland is highly effective for the removal of N and P from effluents of a secondary wastewater treatment plant.  相似文献

As little is known about the oxidant/antioxidant status in buffalo with ketosis, the present study was delineated to assess the oxidative stress level associated with clinical ketosis in water buffalo. A total of 91 parturient buffalo at smallholder farms were studied (61 suspected to be ketotic and 30 healthy). Clinical and biochemical investigations were carried out for each buffalo. Based on clinical findings and the level of beta-hydroxybutyrate (BHB), buffalo were allocated into ketotic (42), sublinical cases (19). Clinically, there was an association between clinical ketosis and anorexia (p < 0.001), constipation (p < 0.001), decreased milk yield (p < 0.001), ruminal stasis (p < 0.001), and loss of body condition (p < 0.01). Biochemically, in clinical ketosis compared with subclinical and control cases, there was a significant increase (p < 0.05) of BHB, malondialdehyde (MDA), nitric oxide (NO), aspartate aminotransferase (AST), L-alanine aminotransferase (ALT). However, there was a significant decrease of glucose, phosphorus, magnesium,total cholesterol and HDL-cholesterol. There was a positive correlation between BHB and MDA (r = 0.433), BHB and NO (r = 0.37), MDA and NO (r = 0.515), and Glucose and phosphorus(r = 0.521). However, there was a negative correlation between BHB and glucose (r = −0.341) and HDL and NO (r = −0.379). The result of the present study indicates that hyperketonemia in buffalo is associated with an increase of oxidative stress levels. Further studies need to be done on the efficacy of antioxidants as an ancillary treatment to relief the oxidative stress caused by ketosis.  相似文献

A radical scavenging guided phytochemical study on the stem bark of Tecoma mollis afforded seven active phenylpropanoid glycosides (17), including a new one (4), and one iridoid (8). The structures of the isolated compounds were elucidated on the basis of spectroscopic evidences and correlated with known compounds. Compounds (17) displayed promising antioxidant activity (DPPH assay) in relation to ascorbic acid (positive control). The antimicrobial activity for compounds (18) was evaluated against five bacterial and five fungal strains. The isolated compounds exhibited nonselective weak to moderate antimicrobial activity. The highest antileishmanial activity against Leishmania donovani was observed for compound (7) with an IC50 value of 6.71 μg/ml, using pentamidine and amphotericin B as drug controls. Compound (5) exhibited moderate antimalarial activity (45% inhibition) against chloroquine sensitive (D6) clones of Plasmodium falciparum.  相似文献

Background

Pinnipeds have a thick blubber layer and may have difficulty maintaining their body temperature during hot weather when on land. The skin is the main thermoregulatory conduit which emits excessive body heat.

Methods

Thorough evaluation of the skin histology in three pinniped species; the California sea lion-Zalophus californianus, the Pacific harbor seal-Phoca vitulina richardsi, and the Northern elephant seal-Mirounga angustirostris, was conducted to identify the presence, location and distribution of skin structures which contribute to thermoregulation. These structures included hair, adipose tissue, sweat glands, vasculature, and arteriovenous anastomoses (AVA). Thermal imaging was performed on live animals of the same species to correlate histological findings with thermal emission of the skin.

Results

The presence and distribution of skin structures directly relates to emissivity of the skin in all three species. Emissivity of skin in phocids (Pacific harbor and Northern elephant seals) follows a different pattern than skin in otariids (California sea lions). The flipper skin in phocids tends to be the most emissive region during hot weather and least emissive during cold weather. On the contrary in otariids, skin of the entire body has a tendency to be emissive during both hot and cold weather.

Conclusion

Heat dissipation of the skin directly relates to the presence and distribution of skin structures in all three species. Different skin thermal dissipation patterns were observed in phocid versus otariid seals. Observed thermal patterns can be used for proper understanding of optimum thermal needs of seals housed in research facilities, rescue centers and zoo exhibits.  相似文献

Field-applied salicylic acid (SA) could provide a potential protection against drought stress in onion large-scale production. Two-season field experiments were consecutively conducted in 2013/2014 and 2014/2015 to study the effect of 1 and 2 mM SA on growth, yield, plant water relations, chlorophyll a fluorescence, osmoprotectants, and water-use efficiency (WUE) in onion plants under four levels of irrigation (I120 = 120%, I100 = 100%, I80 = 80%, and I60 = 60% of crop evapotranspiration). Foliar application of SA enhanced drought stress tolerance in onion plants by improving photosynthetic efficiency and plant water status as evaluated by membrane stability index and relative water content. These results were positively reflected by improving plant growth, productivity, and WUE under drought stress conditions. Therefore, SA application may, in future, find application as a potential growth regulator for improving plant growth and yield under deficit irrigation by 20–40%.  相似文献

The Tataouine province in southern Tunisia is well known for its severe pedoclimatic conditions. Using solid phase microextraction (SPME) and gas chromatography (GC), coupled to flame ionization and mass spectrometer detectors, we characterized virgin olive oils from Chemlali Tataouine, Fakhari Douirat, Zarrazi Douirat, and Dhokar Douirat varieties, which grow in the harsh arid region of Tataouine. Significant differences in the proportions of volatile constituents from oils of different varieties were detected. The results showed that lipoxygenase products were generally the major metabolites of the volatile fraction, and (E)-Hex-2-enal was the principal compound characterizing the olive oil headspace for most samples, though the absolute levels varied greatly, never exceeding 76.45 and 32.16%, respectively. The C5 compounds were unusually abundant, comprising 42.97% of the total lipoxygenase products and a remarkably high level of penten dimers. Each autochthonous variety could thus be differentiated according to the percentage of each metabolite.  相似文献
