The present study sought to quantitatively examine mucosal inflammatory and immune responses in dogs with gastritis and the relationship of these responses to infection with Helicobacter. Gastric biopsies from 30 dogs were evaluated for B- and T-lymphocytes, neutrophils, eosinophils, macrophages, and mast cells. Mucosal atrophy, fibrosis, cellularity, and severity of gastritis were graded qualitatively. Messenger-RNA (mRNA) for actin, interleukin-1beta (IL-1beta), IL-4, IL-8, and IL-10, transforming growth factor beta (TGF-beta), and interferon gamma (IFN-gamma) was quantified by polymerase chain reaction (PCR). The presence of Helicobacter spp. was determined by urease activity, histology, PCR, and enzyme-linked immunosorbent assay. mRNA for IL-1beta, IL-8, IL-10, TGF-beta, and IFN-gamma was detected in most dogs. IL-4 mRNA was detected in only 1 dog. Correlations were observed for IL-1beta versus IL-8 and IL-10; IL-8 versus IL-10, IFN-gamma, and TGF-beta; and IL-10 versus IFN-y. Mucosal pathology was related to cytokine mRNA expression (neutrophils to IL-8 and IFN-gamma, macrophages and lymphocytes to IFN-gamma, and fibrosis to IL-1beta). Gastritis was categorized as lymphoplasmacytic in all dogs, and its histologic severity correlated with atrophy, infiltration with lymphocytes and macrophages, and expression of IL-10 and IFN-gamma. Of the dogs examined, 76.7% were infected with Helicobacter spp. Infection was associated with increased expression of TGF-beta and fibrosis. Circulating anti-Helicobacter immunoglobulin G titers were higher in uninfected than infected dogs. We conclude that lymphoplasmacytic gastritis in dogs is characterized by concurrent activation of proinflammatory and immunomodulatory cytokines, with increased mRNA expression related to mucosal pathology. No significant associations between Helicobacter infection and proinflammatory cytokine expression, severity of gastritis, or differences in the pathogenicity of different Helicobacter spp. were found.  相似文献

Interleukin (IL)-8-encoding regions of five avian species were cloned, sequenced and characterized. Each IL-8-encoding region is 312 nucleotides long and encodes IL-8 which is 103 amino acids. Pairwise sequence analysis showed that sequence identities of IL-8-encoding regions ranged from 87% to 100%. The IL-8 protein identities varied from 84% to 100%. Phylogenetic analysis indicated that IL-8-encoding regions and encoded proteins of chicken, duck, goose and turkey clustered together and evolved into a distinct phylogenetic lineage from that of pigeon which evolved into a second lineage. The results from binding reactivities of antiserum against each recombinant IL-8 (rIL-8) protein to homologous or heterologous rIL-8 proteins, chemotactic activities of each rIL-8 protein or reduction levels of the chemotactic activity of rIL-8 protein which was pretreated with homologous or heterlogous antiserum have suggested that all five IL-8 proteins were functionally active, and shared structural and functional identity with each other.  相似文献

We present a brief introduction to FT‐Raman spectroscopy and examples of its use in cereal science for readers not familiar with Raman spectroscopy. We illustrate the use of FT‐Raman spectroscopy as an analytical tool to measure the degree of chemical modification of chemically modified starches and as a technique to determine conformational and structural changes in plant proteins under different environmental conditions. Results from the neutral mass spectrometer carried on the aeroshell of Viking 1 show evidence for NO in the upper atmosphere of Mars and indicate that the isotopic composition of carbon and oxygen is similar to that of Earth. Mars is enriched in (15)N relative to Earth by about 75 percent, a consequence of escape that implies an initial abundance of nitrogen equivalent to a partial pressure of at least 2 millibars. The initial abundance of oxygen present either as CO(2) or H(2)O must be equivalent to an exchangeable atmospheric pressure of at least 2 bars in order to inhibit escape-related enrichment of (18)O.  相似文献

Comparison of two types of dried scallops, Chlamys farreri and Patinopecten yessoensis, and effects of preparation methods (boiling and steaming) on the composition of their volatile components were carried out by simultaneous steam distillation and extraction and with analysis by gas chromatography-mass spectrometry. One hundred and seventy-two compounds were identified, three tentatively. Qualitatively, both scallops had similar components. Quantitatively, C. farreri contained more components with higher levels in aldehydes, alkanes, naphthalenes, esters, furans, miscellaneous compounds, alcohols, and ketones, whereas P. yessoensis had more components with higher levels in aromatics, pyrazines, pyridines, sulfur-containing compounds, and terpenes from both boiling and steaming methods. Comparison between methods for the same scallop showed that similar qualities of components were detected. Overall, more compounds with higher levels were detected from the boiling method.  相似文献

The effects of different thinning and pruning methods on the bending strength and dynamic modulus of elasticity (DMOE) of young Taiwania (Taiwania cryptomerioides Hay) were investigated. The average DMOE, modulus of elasticity (MOE), and modulus of rupture (MOR) in the thinning treatments showed the following trend: no thinning > medium thinning > heavy thinning. This indicates that thinning reduces average bending properties. The average DMOE, MOE, and MOR in the pruning treatments showed the following trend: medium pruning > no pruning > heavy pruning. According to this tendency, better average qualities of lumber and specimens were from wood subjected to no-thinning and medium-pruning treatments according to an ultrasonic wave technique and static bending tests. However, most results showed no statistically significant differences among thinning, pruning, and thinning and pruning treatments. The average values of DMOE, MOE, and MOR of visually graded construction-grade lumber were significantly greater than those of below-grade lumber. Moreover, there were very significant positive relationships between density, ultrasonic velocity, DMOE, MOE, and MOR, although the determination coefficients were small.  相似文献

Studies are reported on two small lakes at Sudbury, Ontario located close to a nickel-copper smelter which closed in 1972. At that stage, Baby Lake had a pH of 4.0–4.2 while the adjacent Alice Lake had a pH 5.9–6.3. Both lakes were almost entirely devoid of algae and had neither Zooplankton nor fish. Soon after the closure of the smelter, with its large airborne volume of sulphur dioxide and of copper and nickel containing particulates, the chemistry of the lakes began to change. By 1985, Baby Lake had changed from pH 4.0 to 6.8 and is now at pH 7.2. The pH of Alice Lake increased from a low of 5.9 in the early 1970s to 6.9–7.4 in the mid 1980s and is now at 7.3. Copper and nickel concentrations also decreased in both lakes during this period. The first biota found in the lakes in the post-smelter stage in the early 1980s were benthic red chironomids, planktonic rotifers, and a limited number of phytoplankton species, of which Rhizosolenia was the most common. By the 1990s, 13 phytoplankton species were present in each lake, with a substantial Zooplankton fauna (14 species) of rotifers, copepods, and cladocerans. There are now numerous insect larvae in the sediment and some small fish in both lakes. The biological recovery, which followed substantial reductions in acidity and in soluble nickel and copper concentrations in the waters, is a slower process than chemical recovery and is initially characterized by the dominance of a few species.  相似文献
