Methoxyfenozide (RH-2485) was found to be 7.5-fold less toxic in terms of LD50 values against last-instar larvae of a greenhouse-selected strain of the beet armyworm Spodoptera exigua (Hübner) that was collected in July 2001 in an experimental greenhouse for resistance at Murcia in southern Spain, in comparison with a laboratory susceptible strain. To date, the compound is the newest member of this new group of moulting hormone accelerating IGRs to reach the marketplace against Lepidoptera. To understand this different potency in the greenhouse-selected S exigua, oxidative metabolism and acetylcholinesterase activities were measured in last-instar larvae and adults. In addition, we determined, by the use of 14C-labelled methoxyfenozide, the pattern of absorption in body tissues and excretion via faeces in last-instar larvae of the greenhouse-selected strain and compared the results with those from the laboratory susceptible strain. It was striking that the rate of excretion was about twice as high in the greenhouse-selected strain, resulting in a more rapid clearance of insecticide amounts from the insect body. Data are discussed in relation to mechanisms of lower toxicities for this new group of IGRs.  相似文献

The effect of irrigation time on the dispersion of 1,2-dibromo-3-chloroproparie (DBCP) in soil after a shallow injection treatment was determined for Yolo fine sandy loam. When injections equivalent to 23.5 litres of DBCP/ha at a depth of 7 to 8 cm were subsequently followed by 15 cm of irrigation water, adequate distribution of DBCP was obtained if the soil was irrigated within 5 days. However, the most rapid and deepest penetration of fumigant was obtained by irrigating 2 to 5 days after application. There was insufficient downward movement of DBCP in non-irrigated soil.  相似文献

Deep-rooting trees in agroforestry systems may promote distribution of spores of arbuscular mycorrhizal fungi (AMF) at deeper soil levels. We investigated the vertical distribution of AMF spores in Oxisols under agroforestry and monocultural (unshaded) coffee systems in on-farm experiments (Brazil). The number of AMF spores was considered as an indicator of mycorrhiza incidence in soil. Spores were extracted from 0–1, 2–3, 5–7.5, 10–15, 20–30, 40–60 cm soil-depths in agroforestry and monocultural coffee systems, of three different age groups (young, medium-aged and old), using centrifugation methods, and counted. Fine roots were collected and dry-weighed from 0–30 cm in young and old systems and from several depths in medium-aged systems. Soils were characterised with respect to texture, pH, organic matter, calcium, magnesium, phosphorus and potassium. Agroforestry had a higher percentage of spores (12–21% of the total number of spores) and roots (on average 1.5 g L–1 soil) in deeper layers (20–60 cm), and a lower percentage (79–88%) closer to the surface (0–15 cm) than the monocultural fields (respectively 3–12%, 0.6 g L–1 soil and 88–97%). Greater numbers of spores in the deeper soil layers may be explained by greater amounts of roots and may be an indicator of greater incidence of mycorrhiza in agroforestry than in monocultural coffee systems. Greater mycorrhizal incidence at deeper soil layers in the agroforestry system may change the dynamics of phosphorus cycling in soil, making this nutrient more available to plants.This revised version was published online in November 2005 with corrections to the Cover Date.  相似文献

Phosphorus (P) is a primary limiting nutrient for crop production in weathered tropical soils. The deficiency is mainly caused by sorption of phosphate onto Al- and Fe- (hydr)oxides. We hypothesise that the distribution of soil P among various pools is influenced by land use. Our objective was to characterise the soil inorganic (Pi) and organic P (Po) pools and to compare the various pools at different depths in agroforestry (shaded) and monocultural (unshaded) coffee cultivation systems. The study was carried out in the Atlantic Coastal Rainforest domain, Brazil, with Oxisols as the dominant soil type. Soils were collected from four farmers' coffee (Coffea arabica L.) fields, two agroforestry and two monocultural systems. Three profiles were sampled per field, at depths of 2–3, 10–15 and 40–60 cm. A simplified sequential P fractionation was carried out, using resin, 0.5 M NaHCO3, 0.1 M NaOH, 1 M HCl and concentrated HCl as extractants. Sum-P (resin, NaHCO3 NaOH, 1 M HCl and concentrated HCl) ranged from 370 to 830 mg kg–1. Concentrated HCl extracted the largest portion (74%), followed by NaOH (22.5%). Labile (sum of resin, NaHCO3 and NaOH) P ranged from 13 to 40% of Sum-P. The major part (62%) of the labile fraction was Po. In the agroforestry fields, the amount of Po decreased less with depth and the percentage of Po in labile pools was higher than in monocultural fields. This suggests that agroforestry maintains larger fractions of P available to agricultural crops by influencing the dynamics of P through the conversion of part of the Pi into Po, thereby reducing P losses to the unavailable pools.This revised version was published online in November 2005 with corrections to the Cover Date.  相似文献

Shoot multiplication using seedling materials was achieved by subculture on Murashige and Skoog salts with Gamborg's B5 vitamins (MSB5) medium containing a combination of 5 M 6-benzyladenine (BA), 5 M thidiazuron (TDZ), and 1 M indole-3-butyric acid (IBA) with three green ash (Fraxinus pennsylvanica) clones, SD1009 (South Dakota origin), SD2002 (South Dakota origin), and KA2018 (Kansas origin). Shoots were rooted using in vitro and ex vitro methods. For in vitro rooting studies, elongated shoots were transferred to rooting plugs supplied with liquid MSB5 medium containing a 3×3 factorial arrangement of two different auxins, -naphthaleneacetic acid (NAA) and IBA, at three concentrations (0, 5, and 10 M). The most effective treatment for in vitro root number, root length, and shoot height was 5 M IBA. The three clones also were tested for ex vitro rooting using a quick dip in 1 mM NAA, 1 mM IBA, or control (no auxin). The maximal ex vitro rooting response occurred when shoot explants of the three clones were dipped in 1 mM IBA. Significant clonal differences were noted in response to in vitro and ex vitro rooting treatments. Rooted plantlets were acclimated to the greenhouse.  相似文献

F4 enterotoxigenic Escherichia coli (ETEC) cause diarrhoea and mortality in piglets leading to severe economic losses. Oral immunization of piglets with F4 fimbriae induces a protective intestinal immune response evidenced by an F4-specific serum and intestinal IgA response. However, successful oral immunization of pigs with F4 fimbriae in the presence of maternal immunity has not been demonstrated yet. In the present study we aimed to evaluate the effect of maternal immunity on the induction of a systemic immune response upon oral immunization of piglets. Whereas F4-specific IgG and IgA could be induced by oral immunization of pigs without maternal antibodies and by intramuscular immunization of pigs with maternal antibodies, no such response was seen in the orally immunized animals with maternal antibodies. Since maternal antibodies can mask an antibody response, we also looked by ELIspot assays for circulating F4-specific antibody secreting cells (ASCs). Enumerating the F4-specific ASCs within the circulating peripheral blood mononuclear cells, and the number of F4-specific IgA ASCs within the circulating IgA+ B-cells revealed an F4-specific immune response in the orally immunized animals with maternal antibodies. Interestingly, results suggest a more robust IgA booster response by oral immunization of pigs with than without maternal antibodies. These results demonstrate that oral immunization of piglets with F4-specific maternal antibodies is feasible and that these maternal antibodies seem to enhance the secondary systemic immune response. Furthermore, our ELIspot assay on enriched IgA+ B-cells could be used as a screening procedure to optimize mucosal immunization protocols in pigs with maternal immunity.  相似文献

OBJECTIVE: To characterize an outbreak of valine-associated scrapie, assess the relative risk of scrapie infection in relation to allele frequency at codon 136, and investigate lateral transmission of infection in a sheep flock within the United States. ANIMALS: 1,006 sheep. PROCEDURE: To determine genotypes, blood or semen samples were assessed via commercial testing; in 190 slaughtered sheep, scrapie status was determined via immunohistochemical evaluation of tissues. Scrapie-positive sheep born to scrapie-negative dams and sheep infected after 1 year of age were identified to assess lateral transmission. RESULTS: Genotypes were determined for codon 171 (164 sheep) or codons 136 and 171 (842 sheep). Forty-four of 160 slaughtered sheep of known genotype were scrapie positive. In these sheep, the presence of at least 1 valine allele at codon 136 (V136) was highly correlated with scrapie-positive status. Lateral transmission was the probable source of infection for 4 scrapie-positive sheep born to scrapie-negative dams and for 11 sheep in which scrapie was diagnosed at > 50 months of age. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that the outbreak of scrapie was associated with a relatively high frequency of V136 in the flock, introduction of a valine-dependent scrapie strain, and the occurrence of lateral transmission. Genotyping of sheep may assist management decisions following diagnosis of scrapie in a sheep with at least 1 V136. It may be prudent to remove sheep of the diploid genotype AVQR (at codons 136 and 171) from infected flocks.  相似文献

OBJECTIVE: To describe the epidemiologic features of Campylobacter infection among cats in the Minneapolis-Saint Paul metropolitan area. DESIGN: Prevalence survey. ANIMALS: 152 cats examined at 3 private veterinary clinics and an animal humane society. PROCEDURES: Fecal samples were submitted for bacterial culture for Campylobacter spp. To determine the duration of Campylobacter carriage, follow-up fecal samples were collected from cats with positive Campylobacter culture results. RESULTS: Campylobacter organisms were cultured from 37 of the 152 (24%) fecal samples. Campylobacter isolates were identified as Campylobacter upsaliensis (29 cats), Campylobacter jejuni (2), and Campylobacter coli (1); species of the remaining 5 isolates could not be determined. Campylobacter organisms were isolated from 36 of the 122 (30%) cats that were < or = 1 year old but from only 1 of the 30 (3%) cats that were > 1 year old, and shedding was more common during the summer and fall months. No association between Campylobacter shedding and clinical signs of disease was identified. For 4 of 13 cats from which follow-up fecal samples were obtained, duration of Campylobacter carriage could not be determined because Campylobacter organisms were isolated from all follow-up samples. For the remaining 9 cats, median duration of Campylobacter carriage was 44 days. CONCLUSIONS AND CLINICAL RELEVANCE: C. upsaliensis can commonly be isolated from the feces of overtly healthy kittens in the Midwest United States. Because carriage may be prolonged, veterinarians should encourage good hand hygiene among owners of cats, especially among owners with new kittens in their household.  相似文献

During breadmaking, a continuous protein network is formed which confers visco-elasticity to dough. The properties of this protein network are highly dependent on the characteristics of the gluten proteins of the wheat flour. A good quality (highly elastic) gluten network retains the carbon dioxide that is produced by the yeast, giving dough and bread with optimal properties. However, the properties of the gluten proteins can differ substantially between wheat flours and are highly dependent on genetic, environmental and post-harvest conditions. Deficiencies in wheat quality for breadmaking can be overcome by incorporating exogenous components which alter the functionality of the gluten proteins during breadmaking. These include additives (e.g. potassium bromate, iodate, chlorine dioxide and chlorine, azodicarbonamide, ascorbic acid and peroxides) and enzymes affecting protein crosslinking. Transglutaminase, glucose oxidase, hexose oxidase and laccase all promote the formation of covalent bonds between gluten proteins and, hence, can serve as alternatives to chemical bread improvers.  相似文献
