Putative genetic markers have been associated with ETEC F4 (Mucine 4 [MUC4]; MUC4GG;CG as susceptible; MUC4CC as resistant) and F18 (Fucosyltransferase 1 [FUT1]; FUT1GG;AG as susceptible; FUT1AA as resistant) resistances respectively. In this study, 71 post‐weaning pigs were followed from d0 (35 days old) to d42 (77 days of age) to investigate the effect of MUC4 or FUT1 genotypes on the mid‐jejunal microbiota composition, pigs expression of genes related to inflammation (IL8, GPX2, REG3G, TFF3, CCL20 and LBPI) and glycomic binding pattern profile (Ulex europaeus agglutinin I [UEA] fucose‐binding lectin and peanut agglutinin [PNA] galactose‐specific), and on blood plasma targeted metabolomics profile, faecal score and performance parameters of growing healthy pigs. The MUC4 and FUT1 resistant genotypes improved the pigs’ growth performance and had firmed faecal score susceptible genotypes in d0–d21 period. Pigs with MUC4GG genotype had a higher jejunal expression of genes relate to immune function (CCL20 and REG3G) than MUC4CG and MUC4CC pigs (p < 0.05). MUC4CG pigs had higher expression of TFF3 (implicated in mucosal integrity) than MUC4GG and MUC4CC (p < 0.05). FUT1 influenced the alpha‐ and beta‐jejunal microbial indices. The FUT1AA group had a higher number of operational taxonomic units (OTUs) belonging to Lactobacillus genus, while FUT1GG group had a higher number of OTUs belonging to Veillonella genus. MUC4CC pigs had lower scores for UEA on brush borders and goblet cells in villi than MUC4GG (p < 0.05). FUT1AA pigs had lower UEA positivity and higher PNA positivity on brush borders and goblet cells than FUT1AG and FUT1GG (p < 0.05). Both FUT1 and MUC4 influenced the metabolic profile of healthy pigs. Results highlight the role of MUC4 and FUT1 on pig intestinal homoeostasis and improved the knowledge regarding the potential interaction between host genetics, gut microbiota composition and host metabolism in a healthy status.  相似文献

The aim of this paper is to review the most relevant findings obtained over the last four years on the physiology, microbiology and immunology of the gastrointestinal tract of pigs as influenced by weaning and nutrition in the post-weaning period through the action of feed components or alternative substances to in-feed antibiotics. In recent years, some light has been shed on the physiology of the stomach, including motility, gastric emptying and acid secretion. This organ is important in the control of pathogen entry into the intestines. Recent studies on basic intestinal physiology around weaning have been scarce. Most investigations have been devoted to nutritional approaches on the effects of diet components (e.g. protein, fibre, etc.) and alternative substances (e.g. organic acids, zinc, prebiotics, probiotics, other substances of animal or plant origins) on pig performance and various aspects of gastrointestinal tract physiology, microbiology and immunology. Among these, beneficial effects of some organic acids have been confirmed. Additional data with zinc sources include possible mechanisms of protection. Overall it appears that manipulating carbohydrate (prebiotic) composition of the weaning diet may be the most promising way to improve gut health in weaned piglets. In that respect, consistent results have been obtained with lactose supplementation, and interactions with other feed constituents have been studied. Significant positive results have also been produced with probiotics (bacteria, yeasts) fed to the piglets or to the sows. The major parts of the responses appear to be mediated through changes in gastrointestinal microbiology, including enhanced numbers of favourable bacteria (lactobacilli, bifidobacteria) and/or decreased numbers of potentially pathogenic bacteria (Escherichia coli, Clostridia, etc.) together with more favourable profiles of fermentation products along the intestines. In contrast, measurable effects of these dietary factors on intestinal physiology and mucosal immunology have been limited or difficult to interpret in many cases. Finally, investigations have been conducted with varying diet composition (other than carbohydrates: protein source or level, amino acids, etc) or supplementing with different substances of animal or plant origin. However, positive effects on the gut have often been more variable and sometimes inconsistent.  相似文献

A histopathological and immunohistochemical study on the intestines of 45 specimens of farmed rainbow trout, Oncorhynchus mykiss (Walbaum), from Loch Awe, Scotland, revealed a number of cellular deviations in individuals naturally infected with the pseudophyllidean cestode Eubothrium crassum (Bloch, 1779). Twenty-five individuals (55.5%) were infected with an average worm burden of 18.84 +/- 4.06 (mean +/- SE) cestodes per host (range, 2-80 worms; total 471 worms). The cestodes, measuring an average 8.23 +/- 1.10 cm (mean +/- SE; range, 5.3-13.0 cm) in length, were found attached by their scolices to the mucosal lining of the distal portion of the pyloric caeca. Within the caeca, the strobila evoked a mild catarrhal enteritis, namely an enhanced mucus production with epithelial cellular desquamation, a leucocytic infiltration of the lamina propria-submucosa and vacuolization of the intestinal epithelial cells. Eosinophilic granular cells of the stratum granulosum exhibited granular depletion, while within the catarrh, the presence of a high number of rodlet cells was noticed. Immunohistochemically, the occurrence of E. crassum caused a significant reduction in the number of bombesin-, gastrin-releasing peptide and glucagon-like immunoreactive endocrine cells, but an increase in the relative densities of endocrine cells containing cholecystokinin-8- and gastrin-like substances. There were, however, no significant differences in the number of endocrine cells that were immunoreactive to secretin, neuropeptide Y and peptide histidine-isoleucine antisera in the digestive tracts of either the infected or non-infected O. mykiss.  相似文献

The ovaries and uterus were collected after ovariohysterectomy from a 16-month-old Labrador bitch in diestrus that never mated. Discrete swellings were found in the uterine horns, with the macroscopic appearance of normal early pregnancy. At histologic examination, the endometrium, devoid of any conceptus and chorion, showed a marked proliferation, on the basis of which a diagnosis of deciduoma was made. A remarkable population of stromal eosinophilic granular lymphocytes was present, especially in the axis of the endometrial folds. Periodic acid-Schiff and Dolichos biflorus-lectin histochemical reaction and a panel of 10 immunohistochemical markers were used to characterize eosinophilic granular cells. Our findings allowed us to compare these granular cells with the granulated decidual cells, whose presence was until now described only in primates, rodents, or a few other epitheliochorial species. On the basis of our results, the importance of eosinophilic granular cells in a decidualization process is hypothesized to occur also in the bitch.  相似文献

Medetomidine (0.02-0.06 mg/kg) in combination with zolazepam-tiletamine (0.8-2.3 mg/kg) were evaluated for reversible anesthesia in four species of Southeast Asian primates: Bornean orangutan (Pongo pygmaeus pygmaeus), Bornean gibbon (Hylobates muelleri), long-tailed macaque (Macaca fascicularis), and pig-tailed macaque (Macaca nemestrina). Twenty-three anesthetic procedures of captive-held and free-ranging primates were studied in Sabah, Malaysia. The induction was smooth and rapid. Respiratory and heart rates were stable throughout anesthesia, whereas body temperature and systolic arterial blood pressure decreased significantly. Atipamezole at five times the medetomidine dose effectively reversed anesthesia, with first signs of recovery within 3-27 min.  相似文献

Ingested nitrate is absorbed in the small intestine, recirculated into the saliva and reduced to nitrite by oral bacteria. In pigs receiving a moderate dietary addition of nitrate, the recirculation into the saliva is modest, so we aimed to assess the effect of higher nitrate doses to find out how the animal reacts to this new situation and to evaluate if a higher nitrate level could enhance the nitrate reduction process, improving the nitrite production Trial 1. Six piglets received 100 g of a commercial diet with 2.45% KNO3. In relation to baseline values, nitrate in blood serum and saliva increased 15 times, and declined after 6 h vs. 2 h. Salivary nitrite increased seven times after the addition and declined after 6 h vs. 2 h. Trial 2. Six piglets were fed a diet with or without 1.22% KNO3 for 2 weeks. Salivary nitrate and nitrite increased with the addition of KNO3: nitrate increased from d0 to the end of the trial, nitrite increased 15 times after 1 week, but decreased after 2 weeks to 4.5‐fold the control. After 2 weeks, nitrate reduced Shan diversity index of salivary microbiota. The present results indicate that the long exposure to high quantities of nitrates impairs the oral reduction of nitrate to nitrite and engenders a reduction of the mouth’s microbiota diversity.  相似文献

The histopathology, immunohistochemistry and ultrastructure of the alimentary canal of chub, Leuciscus cephalus (L.), from the River Brenta, naturally infected with the acanthocephalan Pomphorhynchus laevis Müller, 1776, was studied and described. Of 62 chub examined, 54 (87%) were infected with P. laevis ; the intensity of infection ranged from five to 130 parasites per host, and a density of 8 P. laevis per cm2 was common. Examination of histological material of infected chub revealed that both male and female acanthocephalans deeply penetrated all layers of the gut wall by means of their slender neck, bulb and proboscis. As a result, a capsule was formed around the bulb and proboscis on the external surface of the host intestine. In parasitized chub, four main types of reaction against the body of the acanthocephalan were recognized. Pomphorhynchus laevis caused local damage to the intestinal wall, eliciting catarrhal-erosive enteritis in the lumen and a fibroblastic-collagenous and fibro-epithelioid encapsulation in its thickness with tissue zonation according to the depth of parasite penetration. Furthermore, eosinophilic granular cells (EGC) within the inflammatory tissue were identified by immunohistochemical methods and transmission electron microscopy.  相似文献

Healthy weaned pigs susceptible to enterotoxigenic Escherichia coli F4 (ETEC) require more tryptophan (Trp) to maximize their performance. This may be related to an effect on intestinal microbiota. We studied the intestinal bacterial diversity of healthy pigs with different susceptibility to ETEC and fed different Trp levels. Thirty-six littermate weaned pigs were selected to obtain a set potentially formed of 50% ETEC-susceptible and 50% non-susceptible pigs, based on a Mucin 4 gene polymorphism. Pigs were fed a diet with 0.17 (TrpL) or 0.22 (TrpH) standardized ileal digestible Trp:Lys ratio for 21 days. Slaughtered pigs were classified into non-susceptible, mildly susceptible, and susceptible, by testing ETEC adhesion to intestinal villi. Bacterial diversity in jejunum content was assessed by the 16S rRNA gene-targeted denaturing gradient gel electrophoresis (DGGE) fingerprinting analysis and expressed by the Shannon index. Susceptible pigs had a reduced bacterial diversity, particularly with TrpL diet (p = 0.003). The ETEC adhesion class affected the quantification of enterobacteria DNA (p = 0.027). One DGGE band, which referred to Clostridium bartlettii, was not evidenced in all the susceptible pigs; less DNA from this microbe was quantified by RT-PCR in the jejunum from TrpH susceptible pigs (p = 0.025) compared to TrpL. The gene expression for β-galactoside α-2,3-sialyltransferase 1 was higher in jejunal tissue of ETEC-susceptible pigs (p = 0.019). In studies on pig gut microbiota, the presence of intestinal receptors for ETEC should be considered because of their contribution to a reduced bacterial diversity. This effect could be partially reversed by dietary Trp addition.  相似文献
