Our aim was to evaluate the ability of different strains of Bifidobacteria to cross the gut wall and reach the liver of weaning pigs, and to study the influence of different doses of a probiotic on this parameter. The ability of bacteria to pass from the intestinal lumen to mesenteric lymph nodes and other tissues is called translocation. Several factors promote bacterial translocation, including intestinal bacterial overgrowth, deficiencies in host immune response, and gut barrier damage. Translocation can also be used as a parameter to evaluate a candidate probiotic. Few data are available on the probiotic translocation in weaning pig. We analysed the liver samples from two trials performed with 32 and 64 pigs, respectively, reared from 21 to 35 days of age, fed seven different diets: control diet (C), or C supplemented with one of six different Bifidobacteria strains (1010/CFU/d) (trial 1); or supplemented with four different levels of B. animalis (0, 107, 109, 1011) crossed with 0% or 2% sugar beet fructo-oligosaccharides (trial 2). We found Bifidobacterium spp. genus-specific DNA in liver of subjects from all dietary treatments (control fed pigs included), and no relationship between oral dose and strain appeared. On average 53.1% (trial 1) and 48.4% (trial 2) of the pigs showed the presence of Bifidobacterium spp. genus-specific DNA. We conclude that both endogen and exogenous bacteria can translocate to the liver.  相似文献

Not nocuous bacteria are important for the maturation and the modulating activity of the gut immune system. However, the humoral immune response against commensal and probiotic bacteria is less documented, particularly in farmhouse animals. Blood serum and saliva were collected in two trials where probiotics, Lactobacillus rhamnosus GG (LbR), well-defined human isolate (Trial A), and a novel and abundant porcine commensal, Lactobacillus sobrius strain 001T (LbS) (Trial B), were supplemented to weaning pigs. Anti-LbR IgA were present in serum of pigs before treatment with LbR, but also after 1 or 2 weeks in control pigs, notwithstanding the absence of DNA from LbR in colon. Pigs fed or not LbS for 1 or 2 weeks had LbS-specific IgA, in saliva and in serum. Colon contents of control subjects were positive for DNA from LbS. We hypothesized that part of this IgA strain-specific activity is partially related to immune cross-reactivity between different Lactobacillus-species. With the procedure of Shu et al. [Shu, Q., Bird, S.H., Gill, H.S., Rowe, J.B., 1999. Immunological cross-reactivity between the vaccine and other isolates of Streptococcus bovis and Lactobacillus. FEMS Immunol. Med. Microbiol. 26, 153–158], after ELISA test on blood serum or saliva pre-incubated with LbR or LbS, each strain blocked a relevant part of IgA specific for the other. So bacteria with different affinity for the pig present reciprocal crossed immune activity. When probiotics are supplied to weaning pigs, the possible action of already present multi-effective IgA should be considered. The mechanism of IgA induction by certain probiotics needs to be addressed in further studies.  相似文献

Immunohistochemical studies were performed on male and female bladder and urethra collected from 4 adults dogs and 10 foetal specimens with crown-rump length from 53 to 155 mm (medium-sized breeds, presumptive 38 days of gestation to term). A panel of antisera was tested, including PGP 9.5 to describe the general intramural innervation, ChAT and TH to depict the cholinergic and nor-adrenergic components and NOS1, CGRP, SP, NPY, VIP, SOM, GAL, 5-HT to investigate the possible nitrergic, peptidergic and aminergic ones. A rich cholinergic innervation was present in adult bladder and urethra, along with a lesser number of adrenergic nerves and a small number of nitrergic ones. Either bladder or urethra received numerous CGRP-, SP-, NPY-, VIP-containing nerve fibres which were distributed throughout the muscle layers. All over the lower urinary tract strong to weak ChAT-, CGRP-, SP- and NPY-immunoreactivity was detected in intramural ganglia, in peripheral nerve bundles and around blood vessels. 5-HT-immunoreactive endocrine cells were present in the urethral epithelium. Early foetal organs were supplied only by cholinergic nerve fibres. Few NOS-, CGRP- and SP-ergic components appeared at the end of pregnancy. It can be guessed that sensory mediators such as CGRP and SP increase in postnatal ages while other neuropeptides, such as NPY and VIP, appear only after birth, as the urinary reflex consolidates.  相似文献

Background: Colostrum is the first secretion produced by mammary glands during the hours immediately preceding and succeeding parturition. This secretion differs from milk and represents an essential vehicle of passive immunity,prebiotic compounds and growth factors involved in intestinal development. Most of the literature concerning colostrum composition refers mainly to human and cow; and little is known about pig colostrum metabolome and how it varies between pig breeds and different farrowing parity. Thus, the aim of the present research is to provide new information about pig colostrum composition and the associations between metabolites, the sows' breed and the survival and growth rates of their litters.Results: Colostrum samples were gathered from 58 parturitions of sows belonging to three different breeds chosen for their importance in Italian heavy pig production: 31 Large White, 15 Landrace and 12 Duroc respectively. The defatted and ultrafiltered colostrum samples were analysed using1 H–NMR spectroscopy. Principal Components Analysis(PCA) was assessed on the obtained spectra. In addition, using a Stepwise Regression and a Linear Regression analyses the metabolites named after the signals assignment were tested for their associations with piglets' performances.Twenty-five metabolites were identified, comprehending monosaccharides, disaccharides(such as lactose), organic acids(lactate, citrate, acetate and formate), nitrogenous organic acids(such as creatine) and other compounds,including nucleotides. PCA results evidence a clustering due to breed and season effects. Lactose was the main compound determining the assignment of the samples into different clusters according to the sow breed. Furthermore, some metabolites showed to be associated with piglets' performance and survival traits: acetate and taurine were positively related to litter weight gain and piglets' survival rate, respectively, while dimethylamine and cis-aconitate were linked to new-borns' impaired ability to survive.Conclusions: The results obtained suggest that colostrum composition is affected by breed, which, together with environmental conditions, may cause changes in colostrum metabolites content with possible consequences on piglets' performances. Among the identified metabolites, acetate, taurine, dimethylamine and cis-aconitate showed consistent associations with piglets' survival rate and litter weight gain, implying that these compounds may affect new-borns' ability to survive.  相似文献

The aim of this study was to determine the effects of 3 antibiotics used for pulmonary pathologies added in the feed of weaned pigs on growth performance, commensal microbiota, and immune response. At weaning, a total of 72 pigs were randomly assigned by BW and litter to 1 of the following diets: control (typical weaning diet), control + 400 mg of tilmicosin/kg, control + 600 mg of amoxicillin/kg, and control + 300 mg of doxycycline/kg. Individually penned pigs were slaughtered after 3 wk (12 pigs/treatment) or 4 wk (6 pigs/treatment). During the fourth week, all pigs received the control diet to test the residual effect of the antimicrobial supplementation. The antibiotic supplementation increased growth and feed intake during the first week (P < 0.01) and over the first 3 wk combined (P < 0.05). Gain-to-feed ratio tended to improve during the first week (P = 0.076) by the antibiotics compared with the control. Among the antibiotic treatments, no difference was observed in ADG and feed intake, which were also unchanged by the diet in the fourth week. The fecal enterobacteria counts were increased by amoxicillin on d 14 and 21 (P < 0.05 and 0.01, respectively) and were decreased by tilmicosin (P < 0.001) compared with the control. Amoxicillin decreased lactic acid bacteria (P < 0.01) counts compared with the control. The antibiotic supplementation tended to decrease total bacteria variability in the jejunum (Shannon index, P = 0.091) compared with the control. The antibiotic treatment decreased the mean total serum IgM concentration (P = 0.016) after 3 wk and did not change the mucosal histomorphometry of the small intestine. For tilmicosin, the observed positive action on piglet performance and feed intake can originate by the decreased costs of immune activation determined by the action on intestinal microbiota. For amoxicillin and doxycycline, the observation on intestinal and fecal microbiota seems to be not sufficient to explain their growth-promoting effect.  相似文献

The study addresses the first two weeks of piglet weaning and is aimed at i) identifying new probiotic strains of Bifidobacterium spp. and non-digestible oligosaccharides (NDO) able to increase the level of autochthonous bifidobacteria in the gut, and ii) finding the best probiotic doses, and probiotic-NDO combinations for piglet growth and gut health.Initially, on a total of 60 pigs we administered 12 strains of 4 bifidobacteria species taken from our collection of strains of different ecological origin. We found the most promising probiotic candidates to be two strains of B. animalis subsp. lactis (M 354 and Ra 18) and one strain of B. choerinum (Su 891), as these led to an increase in the number of viable bifidobacteria in the caecum. In a second trial, a different set of 64 piglets were fed a diet containing a galacto-oligosaccharide (1%) from milk whey, and two fructo-oligosaccharides (4%) from chicory inulin and sugar beet (SbFOS). There was virtually no effect on the hindgut microbiota, except for SbFOS at 4% which tended to increase the endogenous bifidobacteria. Then, in a third trial employing 128 weaning pigs, the probiotic strains Ra 18 and Su 891 were tested individually, at different doses (0; 107; 109; 1011 per pig per day) and combined with 0% or 2% SbFOS supplementation. It was found that Ra 18 improved the live weight growth linearly, while the Su 891 did not. The FOS supplementation showed no interaction with the Ra 18 or Su 891 probiotics for growth performance and no effect on this response criterion.Our selection procedure has highlighted that B. animalis subsp. lactis Ra 18, at 1011 CFU per pig per day, appears to be the best probiotic choice for improved growth performance in weaning piglets.  相似文献

Specimens of testis, excurrent duct including the male accessory glands and urethra, were studied in boars, bulls, horses and donkeys, in order to localize endocrine/paracrine cells. Silver impregnation methods were used to test the argentaffinity and/or argyrophilia of cells. Immunoreactivities to chromogranin A, 5-hydroxytryptamine, somatostatin, [met]- and [leu]- enkephalins, gastrin-releasing peptide, calcitonin gene-related peptide, neuropeptide Y, substance P, vasoactive intestinal peptide, beta-endorphin antisera were tested by a streptavidin-biotin method. In the testis, epididymis, ductus deferens and vesicular gland no endocrine cells were found in any of the animals studied. Chromogranin-A, serotonin, somatostatin and enkephalins were present in endocrine/paracrine cells in the surface or glandular epithelia, whereas all other antisera gave negative results. In the prostatic complex and the urethral epithelium, the most consistent number of endocrine cells was serotonin-immunoreactive. Few cells were also argentaffin and a very limited number of them showed argyrophily and chromogranin-A immunoreactivity. Somatostatin-and enkephalin-immunoreactive cells were rare in the bull and boar, absent in stallions. This comparative study carried out on different species of domestic ungulates has shown deeply different immunophenotypes, even comparing species that are in a very close zoological relationship with one another, such as the horse and the donkey.  相似文献

The presence of Aquaporins 1 (AQP1) and 9 (AQP9), integral membrane water channels that facilitate rapid passive movement of water and solutes, was immunohistochemically detected in the excurrent ducts collected from sexually mature buffalo bulls of proven fertility during the mating (late autumn–winter) and non‐mating (late spring to the beginning of autumn) seasons. Furthermore, the research was performed also on the epididymal cauda of a senile buffalo bull with inactive testis. Aquaporins 1 and 9 were immunolocalized at distinct levels. In the efferent ducts, AQP1 immunoreactivity was strongly evidenced at the apical surface of the non‐ciliated cells and weakly along the basal membrane of the epithelial cells. The latter reactivity disappeared during the non‐mating season. No AQP1 immunoreactivity was detected in the epithelium of epididymis and vas deferens, whereas AQP1 was expressed in the smooth muscle layer of the vas deferens. Aquaporin 1 was present in the blood vessels and in small nerve bundles all along the genital tract. The supranuclear zone of the epididymal principal cells was AQP9 immunoreactive, limited to the corpus and cauda regions, and vas deferens. The samples collected in the two reproductive seasons showed a weaker AQP9 immunoreactivity during the non‐mating season. A typical AQP9 immunoreactivity was noticed in the old buffalo examined. The tested AQP molecules showed a different expression pattern in comparison with laboratory mammals, primates, equine, dog and cat. In addition, seasonal differences were noticed which are possibly useful in regard to the comprehension of the morphophysiology of reproduction in the bubaline species, which are still a matter of debate.  相似文献
