Objective

To compare the effects of intravenous (IV) medetomidine-morphine and medetomidine-methadone on preoperative sedation, isoflurane requirements and postoperative analgesia in dogs undergoing laparoscopic surgery.

Study design

Randomized, crossover trial.

Animals

Twelve adult Beagle dogs weighing 15.1 ± 4.1 kg.

Methods

Dogs were administered medetomidine (2.5 μg kg?1) IV 5 minutes before either methadone (MET) or morphine (MOR) (0.3 mg kg?1) IV. Anaesthesia was induced with propofol, maintained with isoflurane in oxygen, and depth was clinically assessed and adjusted by an anaesthetist blinded to the treatment. Animals underwent laparoscopic abdominal biopsies. Sedation and nausea scores, pulse rate (PR), respiratory rate (fR), noninvasive systolic arterial blood pressure (SAP), rectal temperature (RT) and pain scores were recorded before drug administration, 5 minutes after medetomidine injection and 10 minutes after opioid administration. Propofol dose, PR, fR, SAP, oesophageal temperature (TOES), end-tidal carbon dioxide and end-tidal isoflurane concentration (Fe′Iso) were recorded intraoperatively. Pain scores, PR, fR, SAP and RT were recorded 10 minutes after extubation, every hour for 6 hours, then at 8, 18 and 24 hours. The experiment was repeated with the other drug 1 month later.

Results

Nine dogs completed the study. After opioid administration and intraoperatively, PR, but not SAP, was significantly lower in MET. Fe′Iso was significantly lower in MET. Temperature decreased in both treatments. Pain scores were significantly higher in MOR at 3 hours after extubation, but not at other time points. Two dogs required rescue analgesia; one with both treatments and one in MOR.

Conclusion and clinical relevance

At the dose used, sedation produced by both drugs when combined with medetomidine was equivalent, while volatile anaesthetic requirements and PR perioperatively were lower with methadone. Postoperative analgesia was deemed to be adequate for laparoscopy with either protocol, although methadone provided better analgesia 3 hours after surgery.  相似文献

Objective

To determine the dose of cis-atracurium needed to produce a moderate neuromuscular blockade (NMB) in pigs.

Study design

Prospective experimental study.

Animals

Seven pigs [five females and two males; median (range) body weight: 47 (36–64) kg].

Methods

Pigs were premedicated with intramuscular midazolam (0.3 mg kg?1) and ketamine (7 mg kg?1). Anaesthesia was induced with intravenous (IV) propofol 3 (1–4) mg kg?1 and maintained with isoflurane in oxygen. Based on a preliminary study, the subjects were administered 0.3 mg kg?1 cis-atracurium followed by 0.48 mg kg?1 hour?1 constant rate infusion (CRI) IV. A moderate NMB was defined as a train-of-four (TOF) count of ≤2 by acceleromyography. When the TOF count was >2, 0.1 mg kg?1 cis-atracurium was administered and the CRI was increased. The cis-atracurium CRI was decreased when the TOF count was under 2 for more than 15 minutes. The total dose of cis-atracurium required to maintain a moderate NMB was calculated as the total amount of cis-atracurium used (both CRI and supplementary boluses) divided by the administration time.

Results

The cis-atracurium CRI lasted for 87 (76–151) minutes. To induce and maintain a moderate neuromuscular blockade, the initial dose of cis-atracurium was 0.3 (0.3– 0.5) mg kg?1 and the CRI was 0.71 (0.37–0.98) mg kg?1 hour?1.

Conclusions and clinical relevance

The doses described in our study may help researchers obtain a moderate NMB using cis-atracurium in pigs.  相似文献

Insect vector-borne plant diseases, particularly those whose causative agents are viral, or phloem- and xylem-restricted bacteria, greatly impact crop losses. Since plants are immobile, the epidemiology of vector-borne diseases greatly depends on insect vectors, which are the only means of dissemination for many pathogens. The effectiveness of a vector-borne pathogen relies upon the vectorial capacity, which is affected by vector density, feeding activity on hosts, longevity before and after pathogen ingestion, duration of the incubation period, and vector competence. During the last decade, research on human vector-borne epidemics has stimulated interest in novel control strategies targeting different parts of the vector cycle, and our purpose here is to draw parallels between this field of research and agronomy. We review the literature on insect vectors of crop diseases and their symbiotic microorganisms with the aim of suggesting future integrated management techniques based on current research on insect-vectored human diseases. Vector transmission is a complex process and different modes of transmission are encountered irrespective of the pathogen. Facultative symbionts have varied effects on life history traits that could be used for vector population control. Symbiont selection, transformation, and their manner of dissemination are important when developing an integrated vector management system based on symbiont manipulation. In the short term, progress on our knowledge of the microflora of insect vectors of plant diseases must be made. In the long term, symbiont manipulation, which has been successfully demonstrated against human insect-vectored diseases, could be adapted to insect-borne plant diseases to increase sustainable crop production.  相似文献

Eight Hevea brasiliensis Muell. Arg. clones (GT1, YUNYAN77-4, IRCA707, IRCA317, PB217, PB260, PR107 and RRIM600) were compared for their tolerance towards chilling stress. Net photosynthesis (Pn), stomatal conductance (Gs), optimal and effective photochemical efficiencies (F(v)/F(m) and ), non-photochemical quenching, cellular lysis and leaf necrosis were measured on trees chilled at 10?°C for 96?h, as well as upon recovery at 28?°C. In addition, ascorbate peroxidase, catalase, dehydroascorbate reductase, glutathione reductase, monodehydroascorbate reductase and superoxide dismutase activities were monitored. Clone RRIM600 appeared to be the most tolerant, because it showed no cellular lysis or leaf necrosis and the best recovery as revealed by Pn, Gs, F(v)/F(m) and . Its ability to sustain chilling stress seemed related in part to the fast closure of stomata, suggesting an 'avoidance strategy' for this clone. IRCA707, GT1 and YUNYAN77-4 were also tolerant to the cold treatment as only a few leaf injuries were observed. However, YUNYAN77-4 showed a particular behaviour with a large stomata opening during the first hour of chilling, some photosynthetic activity after 96?h at 10?°C, but the slowest recovery in Pn. The greatest cell or leaf damage was observed on PB260, IRCA317, PR107 and PB217 clones, thus classified as sensitive to chilling. These clones showed the strongest decrease in Pn, F(v)/F(m) and and the slowest recovery for F(v)/F(m) and , indicating a high sensitivity of photosystem II to cold temperatures. Punctual increases of various enzymatic activities were observed for all clones during chilling kinetics. During recovery, the strongest increases in enzymatic activity were observed for the most tolerant clones, suggesting that efficient reactive oxygen species elimination is a crucial step for determining chilling tolerance in Hevea although the enzymes implicated varied from one tolerant clone to another. This study points out contrasted strategies of the Hevea clones in copping with chilling stress and recovery.  相似文献

This study examined the genetic variability and genotype × diet interactions during early growth (initial mean body weight 1.2 g) among seven heterozygous clones of rainbow trout, Oncorhynchus mykiss. The clones were hand-fed a diet containing either fishmeal or plant proteins during a 49-day trial divided into two periods (P1, 26 days, and P2, 23 days). Weight, variation of weight within clone, feed intake, feed efficiency and mortality were calculated for both periods.There was a highly significant effect of diet and of clone for all traits at both periods, except for feed efficiency and mortality at P1. Highly significant interactions between diet and clone were also recorded for all these traits, except for mortality at P1. The occurrence of genotype × diet interactions when feeding juvenile rainbow trout with an all plant-protein diet indicates that a highly performing genotype on a fishmeal diet may perform poorly when fed a plant-protein diet. Interactions were found for the two major determinants of growth, i.e. feed intake and feed efficiency, showing that the dietary response differs according to the genotype. Monitoring of the within-clone variability of weight showed that a plant-based diet is likely to enhance the overall phenotypic variance in a population, whatever its initial genetic variability.  相似文献

Background

Plant root systems play a major role in anchoring and in water and nutrient uptake from the soil. The root cone angle is an important parameter of the root system architecture because, combined with root depth, it helps to determine the volume of soil explored by the plant. Two genes, DRO1 and SOR1, and several QTLs for root cone angle have been discovered in the last 5 years.

Results

To find other QTLs linked to root cone angle, a genome-wide association mapping study was conducted on two panels of 162 indica and 169 japonica rice accessions genotyped with two sets of SNP markers (genotyping-by-sequencing set with approximately 16,000 markers and high-density-rice-array set with approximately 300,000 markers). The root cone angle of all accessions was measured using a screen protractor on images taken after 1 month of plant growth in the Rhizoscope phenotyping system. The distribution of the root cone angle in the indica panel was Gaussian, but several accessions of the japonica panel (all the bulus from Indonesia and three temperate japonicas from Nepal or India) appeared as outliers with a very wide root cone angle. The data were submitted to association mapping using a mixed model with control of structure and kinship. A total of 15 QTLs for the indica panel and 40 QTLs for the japonica panel were detected. Genes underlying these QTLs (+/?50 kb from the significant markers) were analyzed. We focused our analysis on auxin-related genes, kinases, and genes involved in root developmental processes and identified 8 particularly interesting genes.

Conclusions

The present study identifies new sources of wide root cone angle in rice, proposes ways to bypass some drawbacks of association mapping to further understand the genetics of the trait and identifies candidate genes deserving further investigation.

In the semi-arid Brazilian Northeast, the exploitation of alluvial aquifers for irrigation and domestic supply to rural communities over the last 10 years has upset the traditional mechanisms of water resources management. In the Forquilha watershed (221 km2; 5°17″S, 39°30″W), the two main water resources are reservoirs (with a capacity exceeding 0.9-6.7 × 106 m3), used for domestic water supply only, and an alluvial aquifer (2.3 × 106 m3), used for irrigation and domestic water supply. From 1998 to 2006, the irrigated area with alluvial groundwater increased from 0 to 75 ha, and the fraction of population supplied through domestic water networks, using reservoirs and the aquifer, increased from 1% to 70%. Based on physical and socioeconomic issues, three main water territories have been defined (“Aquifer”, “Reservoirs”, and “Disperse Habitat”). Considering the next 30 years with a realistic population growth, three hypotheses regarding irrigated area (i.e., 0, 75, or 150 ha), and several possible water-management scenarios, hydrological balance models were built and used to simulate the different impacts on water resource availability and salinity. Simulation results showed that, in all cases, releases from the upstream main reservoir are necessary to keep reservoir salinity below 0.7 g L−1 and for guaranteeing domestic needs in the whole watershed. As a consequence, a management approach that takes into account the interrelations among the three territories is necessary. Moreover, the simulations showed that the area of irrigated fields cannot exceed the current extent (75 ha), or serious restrictions on water availability and salinity will take place. Moreover, important socioeconomic problems are expected, including a high cost of palliative water supply with tank trucks from external sources.  相似文献
