Wheat landraces carry abundant genetic variation in heading and flowering times. Here, we studied flowering-related traits of two Nepalese varieties, KU-4770 and KU-180 and a Japanese wheat cultivar, Shiroganekomugi (SGK). These three wheat varieties showed similar flowering time in a common garden experiment. In total, five significant quantitative trait loci (QTLs) for three examined traits, the heading, flowering and maturation times, were detected using an F2 population of SGK/KU-4770. The QTLs were found at the Ppd-1 loci on chromosomes 2B and 2D and the 2B QTL was also confirmed in another F2 population of SGK/KU-180. The Ppd-D1 allele from SGK and the Ppd-B1 alleles from the two Nepalese varieties might be causal for early-flowering phenotype. The SGK Ppd-D1 allele contained a 2-kb deletion in the 5′ upstream region, indicating a photoperiod-insensitive Ppd-D1a allele. Real-time PCR analysis estimating the Ppd-B1 copy number revealed that the two Nepalese varieties included two intact Ppd-B1 copies, putatively resulting in photoperiod insensitivity and an early-flowering phenotype. The two photoperiod-insensitive Ppd-1 homoeoalleles could independently contribute to segregation of early-flowering individuals in the two F2 populations. This study examined the immunological responses of pregnant cattle and their foetuses following an experimental challenge with live Neospora caninum tachyzoites at day 210 of gestation. Animals were bled prior to and weekly throughout the experiment and sacrificed at 14, 28, 42 and 56 days post inoculation (dpi). At post mortem examination, samples of lymph nodes and spleen were collected from both dam and foetus for immunological analysis. Subcutaneous (sc) inoculation over the left prefemoral (LPF) lymph node of pregnant cattle at day 210 of gestation, led to the vertical transmission of parasites by 14 dpi, however no foetal deaths were observed in the infected animals. Foetuses from infected dams mounted Neospora-specific humoral and cell-mediated immune (CMI) responses by 14 dpi. These responses involved anti-Neospora IgG, antigen-specific lymphocyte proliferation, and the production of the cytokines IFN–γ, interleukin (IL)-4 and IL-10. There was also evidence of innate immunity during the response against Neospora from infected dams, with statistically significant (p < 0.05) increases in mean expression of toll like receptors (TLR)-2 on 56 dpi in maternal spleen, LPF, right prefemoral (RPF), left uterine (LUL) and right uterine (RUL) lymph nodes and TLR-9 in retropharyngeal (RLN), LPF and RPF lymph nodes from 28 dpi. Statistically significant (p < 0.05) increases in mean TLR-9 were detected in spleen samples from foetuses of infected dams, compared to the foetuses from control animals. Our results show that vertical transmission of the parasite occurred in all infected dams, with their foetuses showing effective Neospora-specific cell mediated, humoral and innate immune responses. 相似文献

Despite Neospora caninum being a major cause of bovine abortion worldwide, its pathogenesis is not completely understood. Neospora infection stimulates host cell-mediated immune responses, which may be responsible for the placental damage leading to abortion. The aim of the current study was to characterize the placental immune response following an experimental inoculation of pregnant cattle with N. caninum tachyzoites at day 210 of gestation. Cows were culled at 14, 28, 42 and 56 days post inoculation (dpi). Placentomes were examined by immunohistochemistry using antibodies against macrophages, T-cell subsets (CD4, CD8 and γδ), NK cells and B cells. Macrophages were detected mainly at 14 days post inoculation. Inflammation was generally mild and mainly characterized by CD3+, CD4+ and γδ T-cells; whereas CD8+ and NK cells were less numerous. The immune cell repertoire observed in this study was similar to those seen in pregnant cattle challenged with N. caninum at early gestation. However, cellular infiltrates were less severe than those seen during first trimester Neospora infections. This may explain the milder clinical outcome observed when animals are infected late in gestation. 相似文献

Abstract This work investigated eight species of Lepidoptera associated with Miconia calvescens DC. (Myrtales: Melastomataceae) in Brazil, including six defoliators, Salbia lotanalis Druce (Lepidoptera: Pyralidae), Druentia inscita Schaus (Mimallonidae), Antiblemma leucocyma Hampson (Noctuidae), three Limacodidae species, a fruit borer Carposina cardinata Meyrick (Carposinidae), and a damager of flowers Pleuroprucha rudimentaria Guenée (Geometridae). Based on host specificity and the damage caused to plants, S. lotanalis and D. inscita are the most promising species for biological control of M. calvescens. Sustainability of agricultural systems has become an important issue all over the world. The activity of enzymes is potentially an important quality bioindicator in soils. The aim of the present study was to develop a simple and convenient assay to determine the activity of arylsulphatase (AS), acid (ACP) and alkaline phosphatase (ALP) in agricultural soil. The activities of these enzymes were detected using a non-electroactive substrate, which produces an electroactive product. To this end, p-aminophenyl phosphate (pAPP) was used as a substrate which is converted to p-aminophenol (pAP) after enzymatic dephosphorylation; and 4-nitrocatechol sulphate (4-NCS) was used as a substrate for AS activity based on its catalytic effect on the hydrolysis of 4-NCS into 4-nitrocatechol (4-NC). The products of both enzymatic reactions were quantified on carbon-based screen-printed electrodes (SPCEs) modified with carbon nanotubes (CNTs), using Osteryoung square-wave voltammetry (OSWV). The determination of the reaction products allowed more sensitive determination of ALP, ACP and AS activities in soil than that obtained with a spectrophotometric method. This assay also diminishes the generation of waste, which is desirable in green analytical chemistry. The optimization of the analytical procedure in terms of the nature of electrode type, applied potential, pH of solution, and precision of measurements is reported. 相似文献

This study aimed to verify the preference and infestation level of Anastrepha fraterculus (Diptera: Tephritidae) [South American fruit fly] in fruits of guava cultivars and to correlate them to variables such as peel coloration, soluble solids and pH of fruit cultivars. The following cultivars were used: Pedro Sato, Paluma, Cascão and Século XXI. The infestation was evaluated in cages, considering two scenarios: no-choice and multiple choice. In both tests, evaluations of the fruit attraction to insects were conducted for a period of 1′, 3′, 5′, 10′, 20′, 30′, 1 h, 2 h, 6 h, 12 h and 24 h. The visit of A. fraterculus on the assayed cultivars in relation to the time was studied by logistic regression. After 10 days, the number of larvae in each fruit was recorded. In the multiple choice test, the visit proportions were significantly higher in the fruits of cvs. Século XXI and Pedro Sato than in those of cvs. Paluma and Cascão. In the no-choice test, the visit proportions were significantly lower in the Paluma fruits. In both tests, the rate of fruit infestation by A. fraterculus did not differ among cvs. Pedro Sato, Paluma and Cascão, whereas the fruits of cv. Século XXI were more infested. The indexes of pH did not interfere with the infestation of A. fraterculus, whereas a high rate of soluble solids and low color angle appear to be crucial for discriminating the fruits of the most susceptible cultivars. Infestation rate of Século XXI fruits displayed significant correlations with: °Brix (r= 0.7078) and color angle (h) (r= –0.9499) of guava fruits under the multiple choice conditions. 相似文献

The assay of angiotensin-converting enzyme (ACE) inhibition by food-derived peptides is usually carried out by using soluble ACE in a batch process. The purification of this enzyme from tissues is not an easy task, and the resulting preparation loses activity very fast. In addition, ACE commercial preparations are very expensive. In this work the immobilization of ACE, through lysine amino groups, to 4% beads cross-linked (4 BCL) glyoxyl-agarose is described. The amount of immobilized enzyme increased with increasing concentrations of enzyme and with incubation time until a saturation point was reached at 50 mg protein/mL gel and 3.5 hours, respectively. The IC50 values for a noncompetitive sunflower peptide inhibitor were similar for the soluble (30.56 microM) and immobilized (32.7 microM) enzymes. An immobilized derivative was obtained that was 60 times more stable than the soluble enzyme at 60 degrees C. This procedure yields a derivative that can be reused and has increased thermal stability compared to that of the soluble enzyme. Thus, ACE immobilization is a good alternative to using soluble freshly prepared or commercial preparations because of economical and practical reasons. 相似文献
