The incorporation of Se to fungi has been studied, focusing on element distribution among different cellular compartments and, in particular, polysaccharide structures contained in cell walls. Se-enriched mycelia of Pleurotus ostreatus were obtained in submerged cultures. The incorporation of selenium from the growth medium to mycelia was observed with the relative distribution between cytosol plus cell membranes fraction (CCM) and cell walls fraction (CW) of about 44 and 56%, respectively. CCM fractions were analyzed by size exclusion chromatography with on-line UV (280 nm) and ICP-MS detection (80Se). The results obtained showed selenium binding to components of different molecular masses (about 24% of total selenium coeluted with the compounds of molecular mass > 10 kDa). A polysaccharide-containing fraction of mycelia was treated alternatively with Tris-HCl at pH 7.5 or with chitinase. Better solubility and increased contribution of low molecular mass compounds were observed in chitinase extracts (UV detection), confirming the degradation of polysacharides by the enzyme. The total area under the ICP-MS chromatogram of chitinase extract was 2 times higher with respect to the area for Tris-HCl extract. Furthermore, the relative contribution of selenium in the low molecular mass fraction (molecular mass < 1 kDa) in chitinase extract was 72% as compared to 45% in Tris-HCl extract (based on peak area measurements with respect to total area under the chromatogram). The results obtained suggest selenium binding to chitin-containing polysaccharide structures in fungi cell walls.  相似文献

Garlic (Allium sativum L.) is an essential part of Polish, Ukrainian, and Israeli cuisine. The aim of this investigation was to compare the changes in bioactive compounds, proteins, and antioxidant potentials in fresh Polish, Ukrainian, and Israeli garlic samples after subjection to cooking temperature. Dietary fiber and essential trace elements were comparable. The antioxidant potentials were determined by four scavenging methods using beta-carotene, 1,1-diphenyl-2-picrylhydrazyl (DPPH), nitric oxide (NO), and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS(*)(+)) radical cation with K(2)S(2)O(8) or MnO(2) assays. Polyphenols, tocopherols, proteins, and antioxidant potentials were higher in Polish garlic, but not significantly (P > 0.05). The SDS- and native-PAGE electrophoretic patterns of all three fresh garlic samples were without significant differences. Most of the proteins were in the molecular mass range of 24-97 kDa, and the more intensive major bands were concentrated at 50 and 12 kDa. The 50 kDa protein nearly disappears and the intensity of the 12 kDa lectin bands slightly decreases during cooking. It was observed that the bioactive compounds, antioxidant potential, and proteins in garlic decrease significantly after 20 min of cooking at 100 degrees C (P < 0.05). In conclusion, (a) the bioactive compounds, electrophoretic patterns, and antioxidant potential of fresh Polish, Ukrainian, and Israeli garlic samples are comparable; (b) garlic samples subjected to 100 degrees C during 20 min preserve their bioactive compounds, antioxidant potential, and protein profile and are comparable with fresh garlic; and (c) fresh garlic should be added to dishes cooked at 100 degrees C in the last 20 min of the cooking process.  相似文献

Examining the actual state of canopy gaps in forests is critically important for evaluating forest community structure and dynamics, and for comparing gap disturbance regimes in different forests. We, therefore, re-examined our own previously published data from three studies of three different gap survey methods - the Belt transect method, the canopy coverage census method, and an aerial photography method - in order to ascertain how gap parameters (area, size and density of gaps, and related gap characteristics) obtained from these methods differ from each other. We found that the Belt transect method is easy to apply and practical in the field, and the method used for gap measurement is simple. Mean gap size and size distribution obtained using the Belt transect method were not different statistically from those of the other gap survey methods, and quantified gap parameters were in the range recorded for other forests of the world. For preliminary studies of gap dynamics in different forest types, an easy, speedy and workable gap survey method is needed. In addition, surveying a large number of gaps and a large area improve the accuracy of the method. We conclude that the Belt transect method is most suitable method for preliminary gap surveys in the field.  相似文献

Boulder is a round white selection with medium specific gravity that can be used in both the tablestock and chip-processing markets. The tubers will chip process out-of-the-field and from 10 C storage. The tubers of Boulder are large in size with a low incidence of internal defects. Boulder was tested in Michigan State University trials, the North Central Regional trials, on-farm trials in Michigan and other out-of-state replicated agronomic trials. Under irrigated conditions in Michigan, the yield is similar to or better than Atlantic, but specific gravity is less. Boulder has a full-season vine maturity that is similar to Snowden, but the tubers size early. Based upon our trials, an in-row spacing of less than 25.4 cm to reduce the percentage of oversize tubers is recommended. In simulated bruise tests, Boulder had less blackspot bruising than Atlantic, but was more blackspot susceptible than Onaway. Based upon five years of trials, Boulder has resistance to scab that is greater than Atlantic, but less than Onaway.  相似文献

Liberator is a round white chip-processing selection with medium-high specific gravity and resistance to scab (Streptomyces scabies Thaxter). The tubers will chip process out of the field and from 10 C storage. The tuber appearance is similar to Norchip. Post-harvest tuber tests indicate a tolerance to fusarium dry rot. Liberator was tested in the North Central Regional Trials and the National Snack Food Association Trials. Under irrigated conditions the yield and specific gravity are similar to Snowden and Atlantic. Liberator has a full-season vine maturity that is similar to Snowden and tuber dormancy equal to Atlantic.  相似文献

Proper management of soil organic matter is an important issue in the context of sustainable agriculture. The intensification of production and the loss of organic carbon associated with agriculture reduce the efficiency of production and the quality of the environment, especially in relation to areas exposed to erosion. The aim of this study was to determine the impact of specific tillage systems and plant cover on the organic carbon losses, as well as on runoff and soil losses, over a 6-year study period following the introduction of no-till. The first factor in the experiment was the tillage system: conventional tillage (CT) and no-till (NT). The second factor was plant cover: horse bean, spring wheat and winter oilseed rape. The results showed that runoff was 4.3 ± 0.6% higher under NT than under CT, while soil loss was 66.8 ± 2.7% lower under NT than under CT. Compared to CT, NT limited the total organic carbon losses by an average of 46.0 ± 2.9% and organic carbon bound with sediment losses by 53.2 ± 0.7%, whereas for dissolved organic carbon, there were no significant differences for the tillage systems. The anti-erosion effectiveness of NT was lower in the first year, but it increased in subsequent years after the introduction of this tillage system. Plant cover also had a significant impact on organic carbon losses and soil protection. The crops were ranked according to runoff, soil losses and organic carbon losses in the following order from lower to higher losses: winter oilseed rape > spring wheat > horse bean.  相似文献

The effect of olive oils on lipid metabolism and antioxidant activity was investigated on 60 male Wistar rats adapted to cholesterol-free or 1% cholesterol diets. The rats were divided into six diet groups of 10. The control group (control) consumed the basal diet (BD) only, which contained wheat starch, casein, cellulose, and mineral and vitamin mixtures. To the BD were added 10 g/100 g virgin (virg group) or Lampante (Lamp group) oils, 1 g/100 g cholesterol (chol group), or both (chol/virg group) and (chol/Lamp group). The experiment lasted 4 weeks. Plasma total cholesterol (TC), LDL-cholesterol (LDL-C), HDL-cholesterol (HDL-C), triglycerides (TG), total phospholipids (TPH), HDL-phospholipids (HDL-PH), total radical-trapping antioxidative potential (TRAP), malondialdehyde lipid peroxidation (MDA), and liver TC were measured. Groups did not differ before the experiment. In the chol/virg and chol/Lamp vs chol group, the oil-supplemented diets significantly (P < 0.05) lessened the increase in plasma lipids due to dietary cholesterol as follows: TC (25.1 and 23.6%), LDL-C (39.3 and 34.7%), TG (19.3 and 17.0%), and TC in liver (36.0 and 35.1%) for the chol/virg and chol/Lamp group, respectively. The chol/virg and chol/Lamp diets significantly decreased the levels of TPH (24.7 and 21.2%; p < 0.05 in both cases) and HDL-PH (22.9 and 18.0%; p < 0.05 in both cases) for the chol/virg and chol/Lamp group, respectively. Virgin and Lampante oils in rats fed basal diet without cholesterol did not affect the lipid variables measured. Virgin, and to a lesser degree Lampante, oils have increased the plasma antioxidant activity in rats fed BD without cholesterol (an increase in TRAP, 20.6 and 18.5%; and a decrease in MDA, 23.2 and 11.3%, respectively). In the rats of chol/virg and chol/Lamp vs Chol diet groups the added oils significantly hindered the decrease in the plasma antioxidant activity (TRAP, 21.2 and 16.7%; and MDA, 27.0 and 22.3%, respectively). These results demonstrate that virgin, and to less degree Lampante, oils possess hypolipidemic and antioxidant properties. It is more evident when these oils are added to the diets of rats fed cholesterol. These positive properties are attributed mostly to the phenolic compounds of the studied oils.  相似文献

As is known from various animal models, the spread of agents causing transmissible spongiform encephalopathies (TSE) after peripheral infection affects peripheral nerves before reaching the central nervous system (CNS) and leading to a fatal end of the disease. The lack of therapeutic approaches for TSE is partially due to the limited amount of information available on the involvement of host biological compartments and processes in the propagation of the infectious agent. The in vivo model presented here can provide information on the spread of the scrapie agent via the peripheral nerves of hamsters under normal and altered axonal conditions. Syrian hamsters were unilaterally footpad (f.p.) infected with scrapie. The results of the spatiotemporal ultrasensitive immunoblot-detection of scrapie-associated prion protein (PrP(Sc)) in serial nerve segments of both distal sciatic nerves could be interpreted as a centripetal and subsequent centrifugal neural spread of PrP(Sc) for this route of infection. In order to determine whether this propagation is dependent on main components in the axonal cytoskeleton (e.g. neurofilaments, also relevant for the component ;a' of slow axonal transport mechanisms), hamsters were treated -in an additional experiment- with the neurotoxin beta,beta-iminodiproprionitrile (IDPN) around the beginning of the scrapie infection. A comparison of the Western blot signals of PrP(Sc) in the ipsilateral and in the subsequently affected contralateral sciatic nerve segments with the results revealed from IDPN-untreated animals at preclinical and clinical stages of the TSE disease, indicated similar amounts of PrP(Sc). Furthermore, the mean survival time was unchanged in both groups. This in vivo model, therefore, suggests that the propagation of PrP(Sc) along peripheral nerves is not dependent on an intact neurofilament component of the axonal cytoskeleton. Additionally, the model indicates that the spread of PrP(Sc) is not mediated by the slow component ;a' of the axonal transport mechanism.  相似文献
