Four experiments involving 265, 410, 894, and 554 sows (Exp. 1 to 4, respectively) were conducted to determine the effect of spray-dried plasma (SDP) at 0 or 0.25% (Exp. 1 and 2) and 0 or 0.50% (Exp. 3 and 4) in lactation diets on average daily feed disappearance (FD), sum of sow BW, fetal and placental loss from d 110 gestation to weaning (SWL), litter size at weaning, litter weight at weaning, and average days from weaning to first estrus (WEI). Experiments 1, 3, and 4 were conducted during summer months, and Exp. 2 was conducted during fall to winter months. Experiment 1 used only parity 1 and parity 2 sows and Exp. 4 used only mature (>2 parities) sows, whereas Exp. 2 and 3 used all parity groups. Sows fed SDP in Exp. 1 had increased (P < 0.01) FD and a tendency for reduced (P = 0.06) SWL and WEI (P = 0.06). Sows fed SDP in Exp. 2 had a tendency for increased (P = 0.09) sow BW at weaning and reduced (P = 0.09) SWL, whereas other variables were not different between diets. Parity 1 and 2 sows fed SDP in Exp. 3 had increased (P < 0.01) FD, but mature sows fed SDP had reduced (P = 0.02) FD. Pig survival and litter size at weaning for all parity groups was not different between diets. The WEI for parity 1 sows fed SDP was reduced (P = 0.02) and tended to be reduced (P = 0.10) for mature sows fed SDP, but was not different between diets for parity 2 sows. More parity 1 sows fed SDP were detected (P = 0.01) in estrus 4 to 6 d after weaning, and fewer were detected (P < 0.01) in estrus 6 d after weaning compared with control parity 1 sows. In Exp. 4, FD was reduced (P < 0.01) for mature sows fed SDP; however, litter weight and average pig BW at weaning was increased (P < 0.01) with more (P < 0.01) marketable pigs (pig BW > 3.6 kg) weaned per litter. Relatively low dietary levels of SDP (0.25 to 0.50%) fed to parity 1 sows farrowed during summer months increased lactation FD and reduced WEI. Mature sows fed SDP during summer months consumed less lactation feed without compromising WEI, but had an increased litter weight, average pig BW, and number of marketable pigs at weaning.  相似文献

The American Veterinary Medical Association (AVMA) Council on Education (COE) has challenged veterinary schools to improve self-assessment of curricular outcomes. One way to assess the quality of education is to gather feedback from alumni. To successfully gather feedback using a questionnaire, questions must be pertinent to veterinary education and include quantifiable responses. Several principles must be applied in questionnaire development to ensure that the questions address the intended issues, that questions are interpreted correctly and consistently, and that responses are quantifiable. The objectives of the questionnaire for alumni of Mississippi State University's College of Veterinary Medicine (MSU-CVM) were twofold: (1) to determine whether graduates were comparable to their US peers in terms of work opportunities and salary, and (2) to evaluate how well the CVM curriculum prepared students to begin their veterinary careers. Demographic categories used by the AVMA and published knowledge, skills, attitudes, and aptitudes of veterinary graduates were used in developing the questions. College-specific questions, such as those relating to student activities and impressions of college resources, were also incorporated. Questionnaires were mailed to participants, who could respond via the World Wide Web. Questionnaire results allowed leaders within the college to determine which aspects of alumni's experiences were exceptionally positive, which needed immediate response, and which might require further study. This article describes the application of principles in developing, administering, and analyzing responses to a questionnaire regarding veterinary education.  相似文献

Neoparamoeba pemaquidensis is an ubiquitous amphizoic marine protozoan and has been implicated as the causative agent for several diseases in marine organisms, most notably amoebic gill disease (AGD) in Atlantic salmon. Despite several reports on the pathology of AGD, relatively little is known about the protozoan and its relationship to host cells. In this study, an in vitro approach using monolayers of a rainbow trout gill cell line (RTgill-W1, ATCC CRL-2523) was used to rapidly grow large numbers of N. pemaquidensis (ATCC 50172) and investigate cell-pathogen interactions. Established cell lines derived from other tissues of rainbow trout and other fish species were also evaluated for amoeba growth support. The amoebae showed preference and highest yield when grown with RTgill-W1 over nine other tested fish cell lines. Amoeba yields could reach as high as 5 x 10(5) cells mL(-1) within 3 days of growth on the gill cell monolayers. The amoebae caused visible focal lesions in RTgill-W1 monolayers within 24 h of exposure and rapidly proliferated and spread with cytopathic effects destroying the neighbouring pavement-like cells within 48-72 h after initial exposure in media above 700 mOsm kg(-1). Disruption of the integrity of the gill cell monolayers could be noted within 30 min of exposure to the amoeba suspensions by changes in transepithelial resistance (TER) compared with control cell monolayers maintained in the exposure media. This was significantly different by 2 h (P < 0.05) compared with control cells and remained significantly different (P < 0.01) for the remaining 72 h that the TER was monitored. The RTgill-W1 cell line is thus a convenient model for growing N. pemaquidensis and for studying host-pathogen interactions in AGD.  相似文献

Levels of reproductively-related steroids were determined in captive male sand tiger sharks, Carcharias taurus, maintained at two institutions: SeaWorld Adventure Park Orlando and the National Aquarium in Baltimore. Sexual conflicts were absent at the former, but were documented at the latter. Serum titers of 17β-estradiol, progesterone, testosterone, and 5α-dihydrotestosterone were determined via radioimmunoassay in adult male sharks from 1988 to 2000. Sampling overlap between the two institutions occurred for 3 months of the year, but steroid concentrations were compared only for April due to the occurrence of sexual conflicts in the sharks at the National Aquarium in Baltimore in that month. For April, testosterone and dihydrotestosterone were significantly higher in the SeaWorld males, and progesterone was significantly higher in the National Aquarium in Baltimore males, while estradiol was not significantly different. Steroid levels were also determined from serial samples taken monthly over 17 months from three male sharks and one female shark at the National Aquarium in Baltimore in 2001–2002 and were compared with corresponding observed sexual conflicts. The steroid levels obtained showed distinct annual hormonal cycles in the male sharks and corroborated a biennial cycle for the single serially-sampled female shark. Furthermore, the steroid levels for individual males correlated with sexual conflicts as well as their position within the male dominance hierarchy. As this species is depleted in some regions globally, insight into the steroid profile of mature sand tiger sharks is important for a greater understanding of the relationship between their reproductive physiology and behavior, and may aid in captive management and reproduction. The amino acid composition and the physicochemical and functional properties of quinoa protein isolates were evaluated. Protein isolates were prepared from quinoa seed by alkaline solubilization (at pH 9, called Q9, and at pH 11, called Q11) followed by isoelectric precipitation and spray drying. Q9 and Q11 had high levels of essential amino acids, with high levels of lysine. Both isolates showed similar patterns in native/SDS-PAGE and SEM. The pH effect on fluorescence measurements showed decreasing fluorescence intensity and a shift in the maximum of emission of both isolates. Q9 showed an endotherm with a denaturation temperature of 98.1 degrees C and a denaturation enthalpy of 12.7 J/g, while Q11 showed no endotherm. The protein solubility of Q11 was lower than that of Q9 at pH above 5.0 but similar at the pH range 3.0-4.0. The water holding capacity (WHC) was similar in both isolates and was not affected by pH. The water imbibing capacity (WIC) was double for Q11 (3.5 mL of water/g isolate). Analysis of DSC, fluorescence, and solubility data suggests that there is apparently denaturation due to pH. Some differences were found that could be attributed to the extreme pH treatments in protein isolates and the nature of quinoa proteins. Q9 and Q11 can be used as a valuable source of nutrition for infants and children. Q9 may be used as an ingredient in nutritive beverages, and Q11 may be used as an ingredient in sauces, sausages, and soups.  相似文献

Anoxia occurs annually in deeper waters of the central portion of the Chesapeake Bay and presently extends from Baltimore to the mouth of the Potomac estuary. This condition, which encompasses some 5 billion cubic meters of water and lasts from May to September, is the result of increased stratification of the water column in early spring, with consequent curtailment of reoxygenation of the bottom waters across the halocline, and benthic decay of organic detritus accumulated from plankton blooms of the previous summer and fall. The Chesapeake Bay anoxia appears to have had significant ecological effects on many marine species, including several of economic importance.  相似文献

We propose that from approximately 3 to 1 million years ago, ice volume changes occurred in both the Northern and Southern Hemispheres, each controlled by local summer insolation. Because Earth's orbital precession is out of phase between hemispheres, 23,000-year changes in ice volume in each hemisphere cancel out in globally integrated proxies such as ocean delta18O or sea level, leaving the in-phase obliquity (41,000 years) component of insolation to dominate those records. Only a modest ice mass change in Antarctica is required to effectively cancel out a much larger northern ice volume signal. At the mid-Pleistocene transition, we propose that marine-based ice sheet margins replaced terrestrial ice margins around the perimeter of East Antarctica, resulting in a shift to in-phase behavior of northern and southern ice sheets as well as the strengthening of 23,000-year cyclicity in the marine delta18O record.  相似文献

[目的]研究草酸胁迫对甘蓝型油菜抗性相关蛋白含量和酶活性的影响,初步揭示草酸毒素对甘蓝型油菜致病性的生理生化机制,为油菜抗菌核病资源的初筛提供一种更方便、快速的方法.[方法]以草酸溶液对苗期甘蓝型油菜材料GH01进行浸根处理,设置3个浓度(清水对照、1和2 mmol/L草酸溶液)、7个胁迫处理时间点(0、6、12、24、36、48和60h),分别测定油菜叶片中可溶性物质含量和抗性相关蛋白酶酶活性.[结果]草酸溶液胁迫下,随着胁迫时间增加,油菜叶片中可溶性蛋白和可溶性总糖含量不断增加,而抗性相关酶均呈先升后降趋势,酶活力曲线呈现“峰状”.其中,2 mmol/L草酸溶液胁迫下,SOD、PPO、POD和MDA含量分别在48、24、60和36 h达最大值,分别为359.66 U/g、257.50 U/g·min、44.94 U/g· min和72.86 μmol/L;在1 mmol/L草酸溶液胁迫下,SOD、PPO、POD和MDA含量分别在48、24、36和24 h达最大值,分别为302.17 U/g、215.00 U/g·min、24.01 U/g· min和75.75 μmol/L.[结论]可利用草酸对甘蓝型油菜抗菌核病性进行初步鉴定,草酸溶液浓度以2 mmol/L为宜,其SOD、PPO、POD活性和MDA含量分别在处理48、24、60和36 h时达最高水平.  相似文献

OBJECTIVE: To determine whether severity of leukocytosis correlates with severity of postmortem lesions in dogs with immune-mediated hemolytic anemia (IMHA). DESIGN: Retrospective study. ANIMALS: 34 dogs with IMHA that had CBC performed within 48 hours prior to death and complete necropsy examinations. PROCEDURE: Dogs were independently assigned to 4 leukocytosis groups (within reference range; mild leukocytosis, moderate leukocytosis, marked leukocytosis) and 3 lesion severity groups (mild lesions, moderate lesions, severe lesions). RESULTS: Moderate to marked leukocytosis correlated with moderate to severe postmortem lesions. Ischemic necrosis within liver, kidney, heart, lung, and spleen attributable to thromboembolic disease or anemic hypoxia were the most common important lesions found at necropsy. None of the dogs with mild lesions had moderate or marked leukocytosis. Four of 14 severely affected dogs had WBC counts within reference range, but all 4 had neutrophilic left shifts. Three of these 4 dogs had toxic change in neutrophils. CONCLUSION AND CLINICAL RELEVANCE: Moderate to marked leukocytosis, neutrophilic left shift, and toxic change in neutrophils in dogs with IMHA should alert clinicians to the potential for moderate to severe tissue injury, which could complicate treatment and worsen prognosis. Lesions appear to be secondary to anemic hypoxia, thromboembolic disease, or both; therefore, treatment objectives should focus on improving blood oxygen-carrying capacity and monitoring for thromboembolic disease.  相似文献
