The Mycoplasma hyopneumoniae genome contains at least 22 regions with a variable number of tandem nucleotide repeats (VNTRs) within coding DNA sequences (CDSs). In this work, the VNTR-containing CDSs were analysed in order to evaluate their degree of variation, possible correlations with antigenic properties, and their potential to be used as a basis for a strain typing PCR assay. We have analysed the VNTRs in five M. hyopneumoniae strains (J, 7448, 7422, PMS, and 232), based on published genomic sequences and on amplified and sequenced DNA segments. These VNTRs are distributed among 12 genes, most of which encode putative surface proteins, including known adhesins. The number of repeat units in any of the VNTRs is highly variable among the analysed strains, but they are, without exception, translationally in frame, and, therefore, code for a variable number of aminoacid repeats (VNTARs). These VNTARs determine putative structural, physicochemical and antigenic variations in the corresponding proteins, with potential implications for aspects associated to M. hyopneumoniae pathogenicity, such as cell adhesion and interactions with the host immune system. Considering that the characterized VNTARs are relatively stable, at least in vitro, and their sizes are strain-specific, we have developed a VNTR-based PCR assay for M. hyopneumoniae strain identification, useful for enzootic pneumonia (EP) diagnosis, strain typing, and distinction of circulating field isolates from vaccine strains in animals vaccinated against EP.  相似文献

For evaluation of the prevalence of anti-Neospora caninum antibodies and its associated risk factors, serum samples from 2109 cattle (11 beef, 50 dairy and 25 mixed farms) and 174 dogs were examined in the State of Rond?nia, Western Amazon, Brazil. An inquiry was applied in each farm. Sera were examined by the Indirect Fluorescence Antibody Test (IFAT) using cut off dilution of 1:25 for cattle and 1:50 for dogs. Statistical association between the serologic status and several variables were analyzed by linear and logistic regression. The overall herd prevalence of anti-N. caninum antibodies for 86 farms was 72% (61.3-81.2%). Prevalence values were 100, 70 and 64% in beef, dairy and mixed herds, respectively. Herd prevalence in beef herds was significantly different (P<0.05) from dairy and mixed herds. The overall animal prevalence of N. caninum in cattle was 8.8%. Prevalence values by animal were similar in different production types (P>0.05), with values of 9.5, 11.2 and 9.7% for beef, dairy or mixed cattle, respectively. Antibodies were found in 12.6% of the 174 examined dogs. Sixteen (22.8%) out of 70 farms with dogs had at least one dog with anti-N. caninum antibodies. The occurrence of antibodies in cattle was statistically associated with farms having more than 25 cows (OR 9.7, 95% IC 2.9-32.2; P=0.0002). There was no significant association between the presence of the dogs, jungle contact or reproductive variables with the occurrence of antibodies in cattle.  相似文献

Forty-eight primiparous and pregnant Holstein cows at 202 (+/-59) days in milk and average daily milk production (DMP) of 22 (+/-5.3) kg at the beginning of the trial were used to study the possibility of identifying animals that are more responsive to recombinant bovine somatotropin (rbST). Animals were housed in free-stall barns, milked three times a day and fed a total mixed ration. Animals were injected with one dose (500 mg each) of rbST every 21 days during a 105-day total period (five doses in all). Daily milk production was recorded to measure average apparent response (AAR) and peak of apparent response (PAR). Average apparent response was defined as the average increase in milk production, expressed as kilogram per cow per day, for days 0-18 after dosing, as compared to the average production calculated for days -1, -2, -3, 19, 20 and 21 after dosing. Peak of apparent response was calculated as the increase in milk production for each day from 0 to 18 days after dosing as compared to the average production for days -1, -2 and -3. There was a positive linear correlation (r = 0.608; p = 0.0001) between AAR for the first 21 days and AAR for the total 105-day period (AATR). A correlation coefficient was calculated between each individual PAR value and AATR. The highest correlation coefficient (r = 0.511; p = 0.0002) was observed for days 8, 9 and 10, and therefore these days were considered the best estimates for calculating AAR when DMP records are not available. Also, high somatic cells count had a negative impact on milk yield response to treatment (p = 0.0162).  相似文献

The location of and threats to biodiversity are distributed unevenly, so prioritization is essential to minimize biodiversity loss. To address this need, biodiversity conservation organizations have proposed nine templates of global priorities over the past decade. Here, we review the concepts, methods, results, impacts, and challenges of these prioritizations of conservation practice within the theoretical irreplaceability/vulnerability framework of systematic conservation planning. Most of the templates prioritize highly irreplaceable regions; some are reactive (prioritizing high vulnerability), and others are proactive (prioritizing low vulnerability). We hope this synthesis improves understanding of these prioritization approaches and that it results in more efficient allocation of geographically flexible conservation funding.  相似文献

Common bacterial blight (CBB) caused by Xanthomonas axonopodis pv. phaseoli (Xap) is the main bacterial disease of snap bean. The present work aimed to select snap bean genotypes that are resistant to CBB based on three components of resistance: area under the disease progress curve, latent period, and lesion diameter on pods (DL). A completely randomized two-factor factorial design with six replications was used to evaluate leaves and pods of 14 snap bean and three common bean genotypes (PI 207262, BAC-6 and A-794) with high resistance to CBB. Two Xap isolates, 1394-98 and 775-90, were used to inoculate leaves and pods. Data were analyzed using nonparametric statistics. Significant differences were observed among the evaluated genotypes for all of the components of resistance, except for DL. The snap bean UENF 1482 demonstrated good performance in two disease resistance components. For this reason, this genotype can be recommended for use in breeding programs that aim to generate snap bean genotypes resistant to Xanthomonas axonopodis pv. phaseoli.  相似文献

Wood density is defined as the ratio of mass to volume and therefore in principle it should be possible to calculate a unique partial least squares regression (PLS-R) model for several species. PLS-R models for wood density based on X-ray microdensity data were calculated for each species Pinus pinaster and Larix × eurolepis and for both species together. After cross-validation and test set validation the data sets were combined and final models were calculated. The common model gave a residual prediction deviation (RPD) of 3.1, a range error ratio (RER) of 11.7, and a SEP/SEC of 1.06. The single models for Pinus pinaster and Larix?×?eurolepis gave RPD’s of 3.5 and 3.2, RER’s of 13 and 11, and a SEP/SEC of 1.2. To the best knowledge of the authors all obtained PLS-R models are the first ones that fulfil the requirements according to AACC Method 39-00 (AACC in AACC Method, 39-00:15, 1999) to be used at least for screening (RPD?≥?2.5). Although this method and the defined limits were developed for the analysis of grains they can be used as a rough rule of thumb until limits for wood are available. The improvement of the PLS-R models, compared to published results, might be due to three facts (1) the higher number of scans collected for a single spectrum, (2) that the samples were better represented by the NIR spectra and X-ray microdensity values, and (3) that the sites for the measurement of NIR spectra and X-ray microdensity were coincided as strictly as possibly.  相似文献

Oligoryzomys (Cricetidae, Sigmodontinae) is a common rodent genus from South America that includes a couple of very similar species. Related species have been used as experimental model for understanding several diseases for which these species are reservoirs. In order to provide a better understanding of the embryological aspects of this group, herein we showed data on the embryonic and fetal development in Oligoryzomys sp. Eight specimens of different stages of gestation were obtained from the Collection of the Zoology Museum of University of Sao Paulo, Brazil. Gestational ages were estimated by crown-rump-length according to Evans and Sack (1973). To address our analysis after examining the gross morphology, tissues from several organs were processed for light and scanning electron microscopy. Morphological data on the systems (nervous system, cardiorespiratory system, intestinal tract and urogenital system) were described in detail. Finally, the findings were compared with what is known about embryological aspects in other rodent species in order to establish similarities and differences during the organogenesis in different species.  相似文献

The effects of epidural and intravenous (IV) methadone (0.5mg/kg) on the minimum alveolar concentration of isoflurane (ISO(MAC)) were compared in dogs. Six dogs (16.5 ± 2.5 kg bodyweight) received three treatments in random order during isoflurane anaesthesia, with a 7 day washout interval between each study. Methadone was injected via a lumbosacral epidural catheter introduced 10 cm cranially into the epidural canal and the electrical stimulation for ISO(MAC) determination was applied either to the thoracic (EP(T) treatment) or to the pelvic limb (EP(P) treatment) during separate study days. In the IV treatment, ISO(MAC) was determined via electrical stimulation of the pelvic limb. Variables were recorded before (baseline), 2.5 and 5h after drug injection. The ISO(MAC) decreased significantly (P<0.05) from baseline at 2.5 and 5h after methadone in all treatments. At 2.5h, the magnitude of ISO(MAC) reduction did not differ between treatments (mean decreases from baseline: 30-33%). The ISO(MAC) reduction lasted longer following epidural methadone in the thoracic limb (decreases from baseline: 30% at 5h in the EP(T) treatment vs. 19% and 16% in the EP(P) and IV treatments, respectively). Although the isoflurane sparing effect provided by epidural methadone was not significantly greater than IV methadone during the initial stage (2.5h), it was more prolonged than the IV route in specific dermatomes (5h in the thoracic limb) with the epidural technique employed. Methadone may therefore provide a greater isoflurane sparing effect when administered epidurally, compared to IV, when noxious stimulation occurs in specific dermatomes.  相似文献

Over a period of 1 year, the production of verotoxin was investigated in 1127 Escherichia coli isolated from 243 dairy cattle from 60 small farms in southern Brazil. Vero cell assay was used to detect toxins in culture supernatants from E. coli isolated from bovine feces. Shiga toxin-producing E. coli (STEC) detection rates were 95% (57 of 60) for farms and 49% (119 of 243) for cattle. Prevalence of STEC-positive cattle in the farms ranged from 0 to 100%. Ninety-six percent (315 of 327) of the STEC isolates did not react in the panel of sera used for typing. Twelve isolates, all non-motile, belonged to serogroups previously associated with human diseases, and 67% (8 of 12) were of only two serotypes (O91:H- and sorbitol-fermenting O157:H-). These results indicate that dairy cattle from the region surveyed may be a source of STEC potentially pathogenic for humans.  相似文献

Pastures are a major soil cover in Central Brazil, especially in rich soils previously occupied by dry forests. We simulated a scenario in which the wooden fences in Paranã Valley are replaced by live fences and isolated trees are left in the pasture fields, and we verified changing in tree cover by adding trees and avoiding logging for wooden fences. The simulation involved the analysis of a 20-year historic series of LANDSAT satellite images to determine the average time of pasture renewal. The average amount of wooden fences produced per hectare of local forest was estimated based on the literature and field data. The high spatial-resolution satellite images available in the Google Earth? program were analyzed to estimate the total length of the fences and the average and maximum number of isolated trees per hectare found in the pastures of the region. The results showed that pasture renewal happens every 8.1 years. It is possible to produce an average of 1,472 stakes per hectare of forest. In the study area, we estimated the existence of an average of 842 km of wooden fences and 3.9 isolated trees per hectare of pasture (maximum = 48 isolated trees). The results of the simulation showed that the adoption of live fences can increase the crown coverage up to 7.5 % or even up to 14.3 % if all of the pasturelands are managed to have live fences and farmers begin to adopt cover-development practices, such as keeping an average of 48 isolated trees per hectare of pasture.  相似文献
