A cell-based high-throughput screen that assessed the cellular stability of a tumor suppressor protein PDCD4 (Programmed cell death 4) was used to identify a new guanidine-containing marine alkaloid mirabilin K (3), as well as the known compounds mirabilin G (1) and netamine M (2). The structures of these tricyclic guanidine alkaloids were established from extensive spectroscopic analyses. Compounds 1 and 2 inhibited cellular degradation of PDCD4 with EC50 values of 1.8 μg/mL and 2.8 μg/mL, respectively. Mirabilin G (1) and netamine M (2) are the first marine natural products reported to stabilize PDCD4 under tumor promoting conditions.  相似文献

The effects of specific immunization against 5α-androstenone have been examined in large, genetically homologous groups of boars reared either to bacon weight (90–95 kg live weight) or to heavy manufacturing weight (115–120 kg live weight). At the lighter weight, immunization significantly (P < 0.05) reduced the concentration of androstenone in the adipose tissue from a mean value of 1.77 (S.E. 0.2) μg g?1 fat in untreated boars (n=39) to 1.10 (S.E. 0.18) μg g?1 for animals (n=19) treated with 5α-androstene-3-BSA. In contrast, boars (n=20) treated with 5α-androstenone-11-BSA as immunogen accumulated androstenone to a level of 1.99 μg g?1 fat (S.E. 0.38). At the heavier weight, immunization reduced the accumulation of androstenone in adipose tissue from a mean value of 1.81 μg g?1 fat (S.E. 0.22) in untreated boars (n=76) to 1.17 μg g?1 (S.E. 0.19) for animals (n=22) treated with androstene-3-BSA as immunogen. In contrast, boars (n=21) treated with androstenone-11-BSA as immunogen accumulated androstenone to a mean level of 1.74 μg g?1 fat (S.E. 0.46). No detrimental side-effects were observed in the immunized animals and the advantages of male-type performance and carcass composition were fully preserved.  相似文献

Reproductive traits were evaluated in Bos taurus and Bos indicus crossbred heifers that were fed different diets during the postweaning period. The study was designed in a 2 x 2 x 2 factorial arrangement of treatments. Angus x Hereford (AH; n = 148) and Brahman x Hereford (BH; n = 148) heifers were sorted after weaning by body weight into light (LW) and heavy (HW) weight blocks. Heifers in each weight block were assigned to diets calculated to reach a target weight of 55% (LE) or 65% (HE) of their projected mature weights by the start of the breeding season. Puberty was determined after a 160-d observation period and characterized by the following criteria: 1) behavioral estrus, 2) presence of a palpable corpus luteum (d 6 to 10; estrus = d 0), and 3) rise in serum progesterone above 1 ng/ml (d 6 to 10). A higher (P = .01) proportion of AH heifers than of BH heifers reached puberty by the breeding season (93% vs 67%). Interactions of breed x weight block and energy level x weight block also contributed to this difference. Weight at puberty was heavier (P = .001) among HE than among LE heifers and greater for heifers in HW than for those in LW blocks (P = .02). Differences in prebreeding weight, body condition, average daily gain, hip height, and pelvic area were influenced selectively by breed, energy level, or weight block. Pregnancy rates were higher (P = .01) among AH than among BH heifers. Incidence and severity of dystocia was influenced by the breed x energy level interaction (P = .01). Brahman x Hereford heifers had less (P = .01) dystocia than AH heifers, HE heifers had less (P less than .02) dystocia than LE heifers, and HE-AH heifers had less (P less than .01) dystocia than LE-AH contemporaries. Subsequent duration of the postpartum interval to estrus was shorter (P = .002) among AH than among BH females. Pregnancy rates at the end of the 2nd yr were higher (P = .02) among LW than among HW females and weights were heavier (P = .001) at weaning among calves weaned from BH dams.  相似文献

Herbicides released through agricultural activities to surface waters and drinking water systems represent a risk to human and environmental health, as well as a cost to municipalities for removal. This study focuses on the viability of glyphosate tolerant cropping systems as an alternative to atrazine-based systems, and the impact of tilling historically no-till ground on the runoff pollution potential of these systems. Variable intensity field rainfall simulations were performed on 2 m long × 1 m wide plots within a field in first-year disk and harrow following no-till (CT), and within a long-term no-tilled (NT) field, both treated with atrazine and glyphosate according to label. Rainfall sequence was: 50 mm h−1 for 50 min followed by 75 mm h−1 for 15 min, 25 mm h−1 for 15 min, and 100 mm h−1 for 15 min. Runoff was collected at regular time intervals during two simulated rainfall events and analyzed for herbicide concentration, sediment content, and volume. Maximum glyphosate concentration in runoff was 233 μg L−1 for NT and 180 μg L−1 for CT (approximately 33% and 26% of the maximum contaminant limit (MCL) for glyphosate (700 μg L−1), respectively, while maximum atrazine concentrations in runoff was 303 μg L−1 for NT and 79 μg L−1 for CT (approximately 100 times and 26 times the atrazine MCL (3 μg L−1)). Atrazine concentration and loading were significantly higher in runoff from NT plots than from CT plots, whereas glyphosate concentration and loading were impacted by tillage treatment to a much lesser degree. Results suggest that glyphosate-based weed management may represent a lower drinking water risk than atrazine-based weed management, especially in NT systems.  相似文献

The objectives of this study were to simplify sample preparation and validate mercury detection in soil and plant samples using inductively coupled plasma atomic emission spectroscopy (ICP-AES). A set of mercury contaminated and mercury free soil and plant samples were digested and analyzed by ICP-AES, inductively coupled plasma mass spectrometry (ICP-MS), and cold vapor atomic absorption spectroscopy (CVAAS). Results show that mercury measurements in soil and plant samples using ICP-AES were in agreement with those analyzed using ICP-MS and CVAAS. The concentrations of mercury in soils and plant tissues determined by ICP-AES were 92.2% and 90.5% of those determined by CVAAS and ICP-MS, respectively. Digestion of soil samples with 4 M HNO3 and direct measurement by ICP-AES without reduction of Hg2+ to Hg0 gave a reasonable and acceptable recovery (92%) for determining Hg in soils. We conclude that ICP-AES with optimized conditions (addition of gold chloride, extension of washing time, linear working range, and selection of wavelength – 194 nm) resulted in reliable detection of mercury in environmental samples.  相似文献

The first photographs ever returned from the surface of Mars were obtained by two facsimile cameras aboard the Viking 1 lander, including black-and-white and color, 0.12 degrees and 0.04 degrees resolution, and monoscopic and stereoscopic images. The surface, on the western slopes of Chtyse Planitia, is a boulder-strewn deeply reddish desert, with distant eminences-some of which may be the rims of impact craters-surmounted by a pink sky. Both impact and aeolian processes are evident. After dissipation of a small dust cloud stirred by the landing maneuvers, no subsequent signs of movement were detected on the landscape, and nothing has been observed that is indicative of macroscopic biology at this time and place.  相似文献
