In the present study four methicillin-resistant Staphylococcus pseudintermedius (MRSP) strains isolated from a dog (n = 3) and the anterior nares of the dog owner (n = 1) were investigated by conventional and molecular methods. The species identity of the four S. pseudintermedius strains was confirmed by conventional methods, by PCR mediated amplification of S. intermedius/S. pseudintermedius specific segments of thermonuclease encoding gene nuc and by restriction fragment length polymorphism analysis of phosphoacetyltransferase encoding gene pta. Investigation of the four S. pseudintermedius for toxinogenic potential revealed that all four strains were positive for the exfoliative toxin encoding gene siet and the leukotoxin encoding genes lukS, lukF. The oxacillin and penicillin resistance of the four S. pseudintermedius strains could be determined by cultivation of the strains on oxacillin resistant screening agar base, ChromID MRSA Agar and Brilliance MRSA Agar and by multiplex PCR detecting the resistance genes mecA and blaZ. The genetic relatedness of the strains was studied by macrorestriction analysis of their chromosomal DNA using pulsed field gel electrophoresis (PFGE). According to PFGE all four S. pseudintermedius strains represent an identical bacterial clone indicating a cross transmission between the dog and the dog owner.  相似文献

Rain-cracking of post-veraison grape berries (Vitis vinifera L.) reduces yield and quality of must and wine. Under vineyard conditions, cracking is often associated with bunch rots. The objective of the present study was to identify the role of Botrytis cinerea Pers.: Fr. as the dominant bunch rot fungus in cracking of Riesling berries. For these experiments grape berries with characteristic symptoms of Botrytis infections were selected in the vineyard. Berries infected by Botrytis always cracked in those regions of the berry where symptoms of the infections (brownish purple colour of the fruit skin) were present. In 55 % of these berries, cracks were orientated perpendicularly to the pedicel/stylar scar axis. Microscopic cracks referred to as microcracks were often present in the stylar scar region and occasionally in the vicinity of lenticels. These cracks allowed penetration of the fluorescent tracer acridine orange. The area infiltrated by the dye was positively related to crack length (r2?=#x003D;?0.43*). Water uptake of detached berries increased with time. Rates of uptake were highly variable and higher for infected berries as compared to berries without infections. Rates of water uptake into infected berries were positively related to cumulative crack length (number of cracks multiplied with average crack length) per berry (r2?=#x003D;?0.45*). Exposing the outer surface of the strained grape berry exocarp to water increased the frequency of microcracks. The increase was larger in infected berries than in those without symptoms. Our data demonstrate that Botrytis infections increase the cracking susceptibility of Riesling grape berries by weakening the grape berry exocarp, increasing crack length per berry which in turn resulted in increased water uptake.  相似文献

Background: Previous studies have suggested an association between equine immune-mediated hemolytic anemia and clostridial infections or neck abscesses.
Objective: The purpose of this report was to describe and characterize the hematologic abnormalities in a horse with Clostridium -associated immune-mediated hemolytic anemia. We also retrospectively evaluated hematologic abnormalities in 8 horses with clostridial myositis or subcutaneous emphysema.
Methods: A 7-year-old Foreign Warm-Blood gelding was evaluated for anemia and a cervical abscess. CBCs and reticulocyte counts were obtained using an Advia 120 analyzer and evaluation of Wright's-stained smears. All cases of equine Clostridium spp. myositis or subcutaneous emphysema over a 7-year period were identified in a retrospective search of the University of Minnesota Veterinary Teaching Hospital database. Clinical, hematologic, and microbiological findings were recorded.
Results: Clostridium perfringens genotype A was isolated from the neck wound of the gelding. The CBC was characterized by severe regenerative anemia, intravascular hemolysis, and RBC agglutination. A direct Coombs' test was positive. Moderate numbers of spheroechinocytes were observed. The total automated reticulocyte count was 4.5% (56,700/μL), with most reticulocytes having low absorbance (mature reticulocytes). The anemia responded to penicillin and steroidal and nonsteroidal immunosuppressive drugs. Of 8 horses with myositis, all of which involved the cervical region, 5 were anemic, 1 had a positive direct Coombs' test, and 2 had increased numbers of type III echinocytes and spheroechinocytes.
Conclusions: The presence of type III echinocytes or spheroechinocytes may be helpful in diagnosing immune-mediated hemolytic anemia associated with clostridial infections in horses. Automated reticulocyte counts may detect very low levels of reticulocytosis in hemolytic anemia in horses.  相似文献

Background: The ADVIA 120 is a laser-based hematology analyzer with software applications for animal species. Accurate reference values would be useful for the assessment of new hematologic parameters and for interlaboratory comparisons.

Objective:


Objective: The goal of this study was to establish reference intervals for CBC results and new parameters for RBC morphology, reticulocytes, and platelets in healthy dogs and cats using the ADVIA 120 hematology system.

Methods:


Methods: The ADVIA 120, with multispecies software (version 1.107-MS), was used to analyze whole blood samples from clinically healthy dogs (n=46) and cats (n=61). Data distribution was determined and reference intervals were calculated as 2.5 to 97.5 percentiles and 25 to 75 percentiles.

Results:


Results: Most data showed Gaussian or log-normal distribution. The numbers of RBCs falling outside the normocyticnormochromic range were slightly higher in cats than in dogs. Both dogs and cats had reticulocytes with low, medium, and high absorbance. Mean numbers of large platelets and platelet clumps were higher in cats compared with dogs.

Conclusions:


Conclusions: Reference intervals obtained on the ADVIA 120 provide valuable baseline information for assessing new hematologic parameters and for interlaboratory comparisons. Differences compared with previously published reference values can be attributed largely to differences in methodology.  相似文献

An endemic area of bovine tuberculosis (TB) (Mycobacterium bovis) currently affecting wild white-tailed deer (Odocoileus virginianus) in northern lower Michigan, USA, constitutes the first self-sustaining outbreak of the infection in free-ranging North American cervids. Given this precedent, epidemiologic insights gained from the outbreak afford the opportunity to guide not only current surveillance and intervention but also control efforts for future outbreaks involving wildlife reservoirs. Our specific objectives were to evaluate retrospective data from field surveillance conducted from 1995 to 2000 to determine apparent prevalence, trends in apparent prevalence, and the effects of various factors on the odds of being M. bovis positive.Data were gathered from post-mortem examinations of 62,560 wild deer collected from all 83 Michigan counties. Records of survey method, sex, age, geographic area and infection status as determined by mycobacterial culture were subjected to trend analysis and multivariable logistic regression. Apparent prevalence for the period was 0.54% (336/62,560) statewide. Prevalence varied widely with geographic area, but significantly decreased since 1995 in the core area of the outbreak-which coincided with implementation of control strategies. Significant risk factors were geographic area, sex, age, and the sex-by-age interaction. The survey method by which deer were obtained for testing was not a predictor of infection.Our results to date suggest an outbreak characterized by broad areas of very low prevalence surrounding focal areas where prevalence is sometimes orders-of-magnitude higher (e.g., deer originating from the core area were up to 147 times more likely to be TB positive than deer from other areas). Our results also identify older male deer as most likely to be M. bovis positive (OR=11.3, 95% CI 3.2, 40.3 for bucks > or =5 years vs. does < or =1.5 years)-an observation consistent with the biology and behavior of the species. Synthesizing these results with those of other ongoing investigations, we hypothesize a two-stage model of disease transmission where TB is maintained at very low prevalence in matriarchal groups, with primary dissemination of the disease attributable to the dispersal and movements of bucks (as well as to the large aggregations of animals created by human activities).  相似文献

BACKGROUND: Besides flow cytometric detection of cellular hemoglobin (HGB) concentration, the ADVIA 2120 uses a novel cyanide-free colorimetric method to determine extracellular total HGB concentration. In human samples, the results are equivalent to those of the cyanmethemoglobin method on the ADVIA 120. Cyanide-free HGB measurement has not been evaluated in animal samples. OBJECTIVES: The aim of this prospective study was to compare the 3 methods of HGB analysis on the ADVIA 2120 and ADVIA 120 in blood samples from dogs, cats, and horses. METHODS: Consecutive fresh K(3)EDTA blood samples from 119 dogs, 113 cats, and 151 horses submitted to the Central Laboratory, Department of Veterinary Clinical Sciences, Justus-Liebig University Giessen, were included. A CBC was performed on each sample using the ADVIA 2120 and ADVIA 120. Colorimetric and cellular HGB concentrations and all calculated variables based on HGB measurement were compared using linear regression, Passing Bablok regression, and Bland Altman plots, using the ADVIA 120 as the reference method. RESULTS: In samples from all species, an excellent correlation was found for colorimetric HGB results (r=0.99). HGB measured with the cyanide-free method was overestimated on the ADVIA 2120 compared with the cyanide-based method on the ADVIA 120, with a mean proportional bias of -21.0% (dog), -22.0% (cat), and -19.4% (horse). The correlation of cellular HGB concentration between analyzers was excellent (r=0.99); however, imprecision was higher than for colorimetric methods. Excellent to fair agreement was found for all calculated variables. CONCLUSION: The cyanide-free method of HGB determination is appropriate for use in blood samples from animals, provided the proportional bias is considered.  相似文献

This article used ethnographic methods to examine how Ohio hunters’ knowledge, attitudes, and practices affect risk exposure to infectious disease. Semi-structured interviews were conducted with hunters from Southeast Ohio, and an online survey was distributed to a random sample of licensed Ohio hunters. Data analyses indicated that Ohio hunters learn about wildlife disease through word-of-mouth, hunting publications, and online sources. They perceived low-to-no risk of exposure to infectious diseases. Although hunters were generally knowledgeable about infectious wildlife diseases, they were more concerned about the impact on wildlife populations than their own health. The results contribute to a better understanding of the role of hunter behavior in response to disease events, the identification of future interventions that would most effectively inform hunters about wildlife diseases, and how to minimize their risk of exposure.  相似文献

Several experiments were conducted to evaluate the protein quality of various raw and rendered animal by-product meals commonly used in companion animal diets. Six freeze-dried raw animal meals (beef lungs, pork lungs, sheep lungs, pork livers, oceanfish, chicken necks) and 3 rendered animal meals (lamb meal, regular ash poultry by-product meal, and low ash poultry by-product meal) were fed in chick assays to determine Lys and TSAA bioavailability, protein efficiency ratio (PER), and net protein ratio (NPR). Each experimental diet was offered to 4 replicates of 5 chicks per pen in all growth assays. Furthermore, each animal by-product meal was fed to mature White Leghorn roosters for determination of true AA digestibility. All freeze-dried, raw animal meals were offered to 5 replicate roosters, and all rendered animal meals were offered to 4 replicate roosters. Most raw animal meals exhibited moderate to high protein quality. Lysine bio-availabilities ranged from 86 to 107% and 70 to 99% for raw and rendered animal meals, respectively. Bio-availability of TSAA ranged from 64 to 99% and 61 to 78% for raw and rendered animal meals, respectively. The PER values ranged from 2.83 to 4.03 and 2.01 to 3.34 for raw and rendered animal meals, respectively. The NPR values ranged from 3.83 to 4.8 and 3.05 to 4.12 for raw and rendered animal meals, respectively. Despite a numeric increase in NPR vs. PER values, the overall ranking of animal meals remained similar. Lamb meal had the poorest PER and NPR values, and pork lungs had the greatest values. Total essential AA digestibility and total AA digestibility ranged from 93.6 to 96.7 and 90.3 to 95.5%, respectively, for raw animal meals and 84.0 to 87.7 and 79.2 to 84.8%, respectively, for rendered animal meals. Rendered animal meals generally had lower protein quality than raw animal meals, with lamb meal consistently having the poorest protein quality and pork livers having the greatest protein quality.  相似文献

Effects of droplet size and carrier volume on foliar uptake and transport of daminozide were investigated. A constant dose of daminozide (100 µg per leaf) was applied to both primary leaves of 10‐day‐old Phaseolus vulgaris (cv Nerina) in droplet sizes of 1–10 µl and carrier volumes of 10 to 200 µl per leaf. Decreasing droplet size or increasing carrier volume decreased daminozide penetration, but increased translocation. Plotting the logarithm of the leaf surface/droplet interface area vs daminozide uptake yielded a negative linear relationship, but for translocation an optimum quadratic type relationship was obtained. Some phytotoxicity occurred at low carrier volumes and large droplet sizes. The degree of phytotoxicity was positively related to the amount of daminozide deposited per unit wetted area above 0.7 µg daminozide mm−2. Below this threshold, there was no visual evidence of phytotoxicity. At the breakpoint, the deposit covered an area of 276 mm2 on both primary bean leaf surfaces. Since the maximum in the relationship of translocation with interface area was in close agreement with the threshold amount of deposit above which phytotoxicity occurred, the inverse relationship between daminozide uptake and translocation at low interface areas was attributed to phytotoxicity. © 2000 Society of Chemical Industry  相似文献
