The intensification of agriculture has led to a loss of biodiversity and subsequently to a decrease in ecosystem services, including regulation of pests by natural enemies. Biological regulation of pests is a complex process affected by both landscape configuration and agricultural practices. Although modeling tools are needed to design innovative integrated pest management strategies that consider tritrophic interactions at the landscape scale, landscape models that consider agricultural practices as levers to enhance biological regulation are lacking. To begin filling this gap, we developed a grid-based lattice model called Mosaic-Pest that simulates the spatio-temporal dynamics of Meligethes aeneus, a major pest of oilseed rape, and its parasitoid, Tersilochus heterocerus through a landscape that changes through time according to agricultural practices. The following agricultural practices were assumed to influence the tritrophic system and were included in the model: crop allocation in time and space, ploughing, and trap crop planting. To test the effect of agricultural practices on biological regulation across landscape configurations, we used a complete factorial design with the variables described below and ran long-term simulations using Mosaic-Pest. The model showed that crop rotation and the use of trap crop greatly affected pollen beetle densities and parasitism rates while ploughing had only a small effect. The use of Mosaic-Pest as a tool to select the combination of agricultural practices that best limit the pest population is discussed.  相似文献

Tick salivary proteins are promising targets for the development of anti-tick vaccines. Recently, we described two paralogous anti-complement proteins, called Ixodes ricinus anti-complement (IRAC) proteins I and II, that are co-expressed in tick I. ricinus salivary glands. However, our previous attempts to immunize rabbits against IRAC via infection with recombinant Bovine herpesvirus 4 (BoHV-4) vectors invariably failed although both recombinants expressed high levels of functional IRAC proteins in vitro. As IRAC are soluble monovalent antigens, one of the possible explanations is that monovalent ligation of the B-cell receptor induces receptor activation but fails to promote antigen presentation, a phenomenon that is thought to induce a state of B-cell tolerance. In the present study, we tried to increase IRAC immunogenicity by expressing them as oligovalent antigens. To this end, IRAC were fused to membrane anchors and BoHV-4 vectors expressing these recombinant forms were produced. The immunization potentials of recombinant viruses expressing either secreted or transmembrane IRAC proteins were then compared. While the former did not induce a detectable immune response against IRAC, the latter led to high titres of anti-IRAC antibodies that only marginally affected tick blood feeding. All together, the data presented in this study demonstrate that the immunogenicity of a soluble antigen can be greatly improved by anchoring it in membrane.  相似文献

The parasite Trypanosoma brucei possesses a large family of transmembrane receptor-like adenylate cyclases. Activation of these enzymes requires the dimerization of the catalytic domain and typically occurs under stress. Using a dominant-negative strategy, we found that reducing adenylate cyclase activity by about 50% allowed trypanosome growth but reduced the parasite's ability to control the early innate immune defense of the host. Specifically, activation of trypanosome adenylate cyclase resulting from parasite phagocytosis by liver myeloid cells inhibited the synthesis of the trypanosome-controlling cytokine tumor necrosis factor-α through activation of protein kinase A in these cells. Thus, adenylate cyclase activity of lyzed trypanosomes favors early host colonization by live parasites. The role of adenylate cyclases at the host-parasite interface could explain the expansion and polymorphism of this gene family.  相似文献

Pterostilbene, a natural methoxylated analogue of resveratrol, was evaluated for antioxidative potential. The peroxyl-radical scavenging activity of pterostilbene was the same as that of resveratrol, having total reactive antioxidant potentials of 237 +/- 58 and 253 +/- 53 microM, respectively. Both compounds were found to be more effective than Trolox as free radical scavengers. Using a plant system, pterostilbene also was shown to be as effective as resveratrol in inhibiting electrolyte leakage caused by herbicide-induced oxidative damage, and both compounds had the same activity as alpha-tocopherol. Pterostilbene showed moderate inhibition (IC50 = 19.8 microM) of cyclooxygenase (COX)-1, and was weakly active (IC50 = 83.9 microM) against COX-2, whereas resveratrol strongly inhibited both isoforms of the enzyme with IC50 values of approximately 1 microM. Using a mouse mammary organ culture model, carcinogen-induced preneoplastic lesions were, similarly to resveratrol, significantly inhibited by pterostilbene (ED50 = 4.8 microM), suggesting antioxidant activity plays an important role in this process.  相似文献

The chemoprophylactic effects of paromomycin sulfate against natural cryptosporidiosis in young kids were investigated. Two studies were carried out using two groups of 18 and 12 animals in two pens. In each pen, kids were allocated to treated or control groups. The treatment consisted of oral paromomycin given at 100 mg kg−1 body weight day−1 for 11 consecutive days from 2 days of age. All kids were weighed at 2, 6 and 10 days of age. Infection was monitored by collecting fecal samples and staining fecal smears every 3–4 days from days 4 or 5 to days 15 or 19. The results clearly showed the efficacy of paromomycin in reducing cryptosporidial oocyst output. Moreover, paromomycin prevented clinical signs and mortality.  相似文献

The effect of recovery from training has not been studied in horses. Therefore, the effect of recovery was examined with exercise of known effect within a conditioning period (CP). A standardized exercise test was performed at the beginning of CP to determine v4, v10, and v180 (horse’s speed, which produced a blood lactate concentration of 4 and 10 mmol/L and a heart rate of 180 beats/min). Six horses were conditioned for three periods of 2 weeks, 5 times per fortnight at their individual v10 for two bouts of 5 minutes on a treadmill. Every 2 weeks of conditioning was followed by 1 week with reduced workload. Standardized exercise test was repeated after each 2 weeks of conditioning and 2 weeks after finishing CP. Exercise speed was individually adapted to the new v10 for every 2 weeks of conditioning. In addition, peak oxygen consumption before, after 3 weeks of conditioning, and at the end of the CP was measured. The mean v4 increased steadily during CP. v180 did not change, whereas peak oxygen consumption increased between the beginning and after 3 weeks of conditioning and leveled off thereafter. In conclusion, reducing the workload for 1 week after 2 weeks of conditioning 5 times per fortnight at v10 for two bouts of 5 minutes allowed for a continuous increase of v4, but the extent of the increase was smaller than in another study with a similar conditioning program but for the recovery week. Water efficiency is a key concept to solve water-shortage problems in semiarid areas. Deficit irrigation (DI) in many crops has frequently proved to be an efficient tool to optimise water-use efficiency. Three different DI strategies were studied for commercial orchards of mature sweet orange (Citrus sinensis L. Osbeck, cv. Salustiana and cv. Navelina) from 2006 to 2008: sustained deficit irrigation (SDI), regulated deficit irrigation (RDI), and low-frequency deficit irrigation (LFDI) all defined physiologically with stem-water potential thresholds. The experimental research plots were located in the Guadalquivir river basin, SW Spain. The effects of DI treatments on the fruit yield and on the crop-water status, by the integrated stem-water potential (Ψint) were analysed. Also, the benefits of DI in terms of agricultural water-use efficiency (WUEagr) and financial water-use efficiency (WUEf) were estimated for each irrigation strategy. Different relationships were estimated between these parameters and irrigation and total water applied, in order to establish the best irrigation strategy for different irrigation regimes. Yield and Ψint showed significant differences consistent with the water amounts applied, although the crop response was influenced by other parameters such as crop variety and irrigation strategy. In this sense, treatments with similar irrigation rates and Ψint resulted in different yield values, evidencing the importance of these factors. Regarding the crop variety, the results showed that cv. Salustiana responded better than cv. Navelina to DI, from the physiological and agricultural perspectives. In terms of water savings, the RDI and LFDI reduced water use by between 1000 and 1250 m3 ha−1, respectively, with similar yields in comparison to the fully irrigated treatment, significantly improving the WUE. Consequently, the WUEf, and WUEagr were more strongly affected by deficit-irrigation strategy rather than the total water supplied. Thus, the amount of irrigation water would have a relative importance but other variables such as the irrigation strategy, would decidedly influence prudent water management in semiarid areas.  相似文献

Eukaryotic cells contain nontranslating messenger RNA concentrated in P-bodies, which are sites where the mRNA can be decapped and degraded. We present evidence that mRNA molecules within yeast P-bodies can also return to translation. First, inhibiting delivery of new mRNAs to P-bodies leads to their disassembly independent of mRNA decay. Second, P-bodies decline in a translation initiation-dependent manner during stress recovery. Third, reporter mRNAs concentrate in P-bodies when translation initiation is blocked and resume translation and exit P-bodies when translation is restored. Fourth, stationary phase yeast have large P-bodies containing mRNAs that reenter translation when growth resumes. The reciprocal movement of mRNAs between polysomes and P-bodies is likely to be important in the control of mRNA translation and degradation. Moreover, the presence of related proteins in P-bodies and maternal mRNA storage granules suggests this mechanism is widely adapted for mRNA storage.  相似文献
