The susceptibility locus for the autoimmune disease lupus on murine chromosome 1, Sle1z/Sle1bz, and the orthologous human locus are associated with production of autoantibody to chromatin. We report that the presence of Sle1z/Sle1bz impairs B cell anergy, receptor revision, and deletion. Members of the SLAM costimulatory molecule family constitute prime candidates for Sle1bz, among which the Ly108.1 isoform of the Ly108 gene was most highly expressed in immature B cells from lupus-prone B6.Sle1z mice. The normal Ly108.2 allele, but not the lupus-associated Ly108.1 allele, was found to sensitize immature B cells to deletion and RAG reexpression. As a potential regulator of tolerance checkpoints, Ly108 may censor self-reactive B cells, hence safeguarding against autoimmunity. 相似文献

In the search for alternatives to synthetic fungicides to control postharvest disease, sulfur-containing salts were evaluated for their effects on the mycelial growth of various fungal or fungus-like pathogens and their ability to control carrot cavity spot (Pythium sulcatum) and potato dry rot (Fusarium sambucinum). Results showed that metabisulfite-containing salts provided strong inhibition of all the tested fungi. Furthermore, some sulfate-containing salts were also directly inhibitory to P. sulcatum (calcium sulfate and ammonium sulfate) and to F. sambucinum (sodium sulfate). The metabisulfite salts also provided 100% inhibition of cavity spot and dry rot at concentrations of 50 and 200 mM, respectively. Calcium sulfate and sodium sulfate also significantly reduced carrot cavity spot lesions at 50 mM and ammonium sulfate, magnesium sulfate, potassium sulfate and sodium sulfate reduced potato dry rot lesions at 200 mM. These results indicate that various sulfate and metabisulfite salts could be used to control these postharvest microorganisms. 相似文献

This study was undertaken to ascertain the antipsychotic properties of Rauwolfia tetraphylla L. leaves and to isolate and characterize the antipsychotic constituents. Among the MeOH extract and some alkaloidal fractions at different pHs, the alkaloidal CHCl(3) fraction at pH-9 (2C) showed the highest antipsychotic activity against dopaminergic (DA-D(2)) and serotonergic (5-HT(2A)) receptors in-vitro and amphetamine induced hyperactive mouse model in-vivo. The activity guided isolation of CHCl(3) fraction (2C) afforded six indole alkaloids: 10-methoxytetrahydroalstonine (1), isoreserpiline (2), an isomeric mixture of 11-demethoxyreserpiline (3) and 10-demethoxyreserpiline (4), α-yohimbine (5) and reserpiline (6). Given orally, alkaloids 3-6 showed significant antipsychotic activity in a dose dependent manner. None of the extract, alkaloidal fractions or alkaloids showed any extra pyramidal symptoms at the tested doses. It was also observed that MeOH extract was behaving similar to other clinically used novel atypical antipsychotics in having 5-HT(2A) occupancy greater than the DA-D(2) receptor at the tested doses. Further toxicity and safety evaluation studies of MeOH extracts of R. tetraphylla leaves at different doses (10, 100, 300 and 2000 mg/kg) on female Swiss albino mice showed that MeOH extract is non toxic. The susceptibility of Caspian miniature horses to hyperlipidemia was followed in a study of healthy Caspian miniature horses in which food but not water was withheld for 48 hours. Two months before the study, the horses were fed a high-energy diet. The mean of weight gain was 26.46 kg per horse during this period. All horses had a normal rectal temperature, respiratory rate, and pulse throughout the experiment. During the period of food deprivation, levels of serum triglyceride, aspartate aminotransferase, and alanine aminotransferase significantly (P < .05) increased and glucose concentration decreased. As a conclusion, Caspian miniature horses were susceptible to hyperlipidemia, but they did not show classic hyperlipemia syndrome. 相似文献

This study deals with the floral diversity and distribution patterns in plant species in relation to environmental gradients in the high altitude cold desert of Ladakh, India. An extensive literature survey and field explorations were made across the different parts of Ladakh in order to create a compilation of vascular plant species. For the quantification of vegetation, a total of eight habitat types were identified and sampled using the point intercept method. In all, 647 species of vascular plants belonging to 58 families and 243 genera were documented. Asteraceae (89 species), Poaceae (66 species), Brassicaceae (54 species), and Fabaceae (43 species) were the dominant families. Astragalus was the most dominant genus as it obtained 24 species, followed by Polygonum (18 species), Artemisia (15 species), and Potentilla (14 species). Among the habitats, riverine beds contained the highest frequency of plant species, followed by narrow gorges and tablelands. The crags and ridges had the lowest frequency of plants. Relatively high water and moisture availability may be the cause of the high frequency of plant species in riverine beds and narrow gorges. The vegetation composition was influenced by elevation; however, the intensity of change reduced at >4500-m elevation. The conservation and management of high altitude cold desert vegetation are discussed based on field observations and findings. 相似文献

The current knowledge of in vitro adherence of Staphylococcus pseudintermedius to canine corneocytes is limited to comparative analyses between strains, staphylococcal species or corneocytes collected from different breeds, body sites and hosts. However, the role played by colonization status of corneocyte donors remains unknown. The aim of this study was to evaluate the adherence properties of commensal S. pseudintermedius strains to corneocytes collected from dogs with different colonization status. For this purpose, corneocytes were collected from five dogs that were classified as persistently colonized (D1 and D2), intermittently colonized (D3 and D4) or non-colonized (D5) on the basis of the results of a previous longitudinal study. Adherence to corneocytes originating from each of the five dogs was assessed by an in vitro adhesion assay using four genetically unrelated strains isolated from the colonized dogs (S1 to S4). Irrespective of their host of origin, all strains adhered significantly better to corneocytes from D1 and D2 than to corneocytes from D3, D4 and D5 (P < 0.0001). The mean count of cells adhering to corneocytes from persistently colonized dogs was on average three times higher than the mean count using corneocytes from the other dogs. A significant difference between strains was only observed for one strain-corneocyte combination (S2-D4), indicating that S. pseudintermedius adherence to corneocytes is driven by host factors and only marginally influenced by strain factors. This finding has important implications for understanding and preventing S. pseudintermedius skin colonization and infection. 相似文献

The rice–wheat cropping system (RWCS), producing about 5–10 Mg ha–1 y–1 of grain, is the backbone of food‐crop production in South‐East Asia. However, this system shows signs of fatigue as indicated by declining yields, negative nitrogen (N) balances, and reduced responses to applied fertilizer at some research centers. The return of rice and wheat residues can recycle up to 20%–30% of the N absorbed by the crops. However, their wide C : N ratio can temporarily immobilize native and applied N. To overcome this immobilization, wheat‐straw application was supplemented with the incorporation of Sesbania green manure and mungbean residues, and their effects on productivity, agronomic N efficiency, and system's apparent N balances were studied. Combining the application of wheat straw with Sesbania green manure or mungbean residues increased cereal grain yield and agronomic N efficiency and improved the generally negative apparent N balances. The combined use of wheat straw and mungbean produced an additional 0.5–0.6 t ha–1 protein‐rich grain and thus appears to be the most promising residue‐management option for rice–wheat cropping systems in South Asia, provided that the transition cropping season between wheat harvest and rice transplanting is long enough. 相似文献
