This work describes the first confirmed cases of carp oedema virus disease (CEVD) in Slovakia and the Czech Republic and the phylogenetic analysis of Czech and Slovak carp oedema virus (CEV) isolates. Four cases of disease outbreak in the Czech Republic are described, the oldest dating from mid-May 2013 and one case from Slovakia dating from May 2019. In all cases, virus presence was confirmed using nested PCR. PCR products were sequenced and compared with 357-bp nucleotide sequences encoding the CEV P4a protein in GenBank. In four cases of disease outbreak (three common carp breeding facilities and one koi garden pond), CEV detected belonged to genogroup I. In one case (koi garden pond), fish were confirmed as infected with CEV from genogroup II. This work complements data on CEV occurrence in European countries and contributes to a better understanding of the pathways leading to transmission of the virus throughout Europe. 相似文献

The tetraploid wheat Triticum timopheevii Zhuk (AtAtGG) is known as a source of genes determining resistance to many diseases. An introgressive line 842, with durable resistance to leaf rust was established by crossing T. aestivum cv. ‘Saratovskaya29’ with T. timopheevii ssp. viticulosum and used for mapping leaf rust resistance genes. Molecular analysis of the line 842 with polymorphic microsatellite markers detected introgressions of T. timopheevii into the homoeologous group 2 chromosomes of common wheat. Transloca‐tion breakpoints of introgressed fragments were localized between the markers Xgwm95 and Xgwm817 on chromosome 2A, as well as Xgwm1128 and Xgwm1067 on chromosome 2B. Linkage analysis demonstrated the association of disease resistance at the seedling stage with chromosome 2A. The gene was found to be linked with marker Xgwm817 at a genetic distance of 1.5 cM. Two extracts (ethyl acetate and ethanol) and isocytisoside obtained from Aquilegia vulgaris were tested for their antioxidant and free radical scavenging activity in vitro. Inhibition both non-enzymatic (IC50: 150-219 microg/ml) and enzymatic (IC50: 23-60 microg/ml) microsomal lipid peroxidation was observed, the extracts being more active than isocytisoside. The substances tested appeared to be weak hydroxyl radical scavengers, showed very low TEAC values and moderate iron chelation ability. However, all preparations at the concentration 25 microg/ml inhibited superoxide anion formation at the range 47-68%. Despite of the lack of a potent free radical scavenging ability the substances tested demonstrated significant antioxidant activity. Relationship between this parameter and the content of phenolic groups was noticed. 相似文献

ObjectiveTo determine the minimal electrical threshold (MET) necessary to elicit muscle contraction of the pelvic limb or tail when an insulated needle is positioned outside (METout) and inside (METin) the lumbosacral epidural space in cats.Study designProspective, blinded study.AnimalsTwelve mixed-breed healthy adult cats, scheduled for a therapeutic procedure where lumbosacral epidural administration was indicated.MethodsUnder general anesthesia, an insulated needle was advanced through tissues of the lumbosacral interspace until its tip was thought to be just dorsal to the interarcuate ligament. An increasing electrical current (0.1 ms, 2 Hz) was applied through the stimulating needle in order to determine the MET necessary to obtain a muscle contraction of the pelvic limb or tail (METout), and then 0.05 mL kg?1 of iohexol was injected. The needle was further advanced until its tip was thought to be in the epidural space. The MET was determined again (METin) and 0.2 mL kg?1 of iohexol was injected. The cats were maintained in sternal position. Contrast medium spread was determined through lateral radiographic projections.ResultsThe radiographic study confirmed the correct needle placement dorsal to the interarcuate ligament in all cats. When the needle was placed ventrally to the interarcuate ligament, iohexol was injected epidurally in ten and intrathecally in two cats. The METout and METin was 1.76 ± 0.34 mA and 0.34 ± 0.07 mA, respectively (p < 0.0001).Conclusion and clinical relevanceNerve stimulation can be employed as a tool to determine penetration of the interarcuate ligament but not the piercing of the dura mater at the lumbosacral space in cats. 相似文献

Allelochemical effects were observed when Tribolium castaneum (Herbst) adults were treated with Baccharis salicifolia (Ruiz & Pavon) Pers essential oil. The main biological activities were toxicity and repellence. Terpenes present in the essential oil were identified by GC-MS, and some authentic samples were tested to assess their activity individually. The most acutely toxic compounds after 3 days were beta-pinene and pulegone. Most of the monoterpenes elicited symptoms indicative of neurotoxicity. The most repellent compound was alpha-terpineol. Toxic and repellent effects of chemical derivatives of the major sesquiterpene present in B salicifolia essential oil, as well as a series of monoterpenes, were evaluated in order to investigate structure-activity relationships. The reduced derivatives of the monoterpenes and sesquiterpenes were more repellent that their carbonyl analogues. Paenibacillus larvae is the causative agent of American Foulbrood (AFB), a severe disease that affects larvae of the honeybees. Due to the serious effects associated with AFB and the problems related to the use of antibiotics, it is necessary to develop alternative strategies for the control of the disease. The aim of the present work was to evaluate the effect of a propolis ethanolic extract (PEE) against P. larvae and its potential for the control of AFB. In vitro activity of PEE against P. larvae isolates was evaluated by the disk diffusion method and the minimum inhibitory concentration (MIC) was determined. Toxicity for honeybees was evaluated by oral administration of PEE and its lethal concentration was assessed. Lastly, colonies from an apiary with episodes of AFB on previous years were divided into different groups and treated with sugar syrup supplemented with PEE by aspersion (group one), sugar syrup by aspersion (group two), fed with sugar syrup supplemented with PEE (group three) and fed with sugar syrup only (group four). All isolates were sensitive to PEE and the MIC median was 0.52% (range 0.32-0.64). PEE was not toxic for bees at least at 50%. Field assays showed that 21 and 42 days after the application of the treatments, the number of P. larvae spores/g of honey was significantly lower in colonies treated with PEE compared to the colonies that were not treated with PEE. To our knowledge, this is the first report about the use of propolis for the treatment of beehives affected with P. larvae spores. 相似文献
