Due to high variation in forest communities, forest structure and the fragmentation of the forested area in Central Europe, satellite-based forest inventory methods have to meet particularly high-quality requirements. This study presents an innovative method to combine official forest inventory information at stand level with multidate satellite imagery using a spatially adaptive classification approach for producing wall-to-wall forest cover maps of important tree species and management classes across multiple ownership regions in a heterogeneous low mountain range in Germany. The classification approach was applied to a 5,200-km2 area (about 2,080?km2 of forest land, mostly mixed forests) located in the Eifel mountain range in Central Europe. In comparison with conventional classifiers, our results demonstrate a significant increase in classification accuracy in the order of 12%. The method was tested with ASTER images but holds the potential to be used for regular state forest inventories based on standard and novel earth observation data supplied for instance from the SPOT-5 and RapidEye sensors.  相似文献

The objective of the present research was to determine the effect of increasing and decreasing natural photoperiods on selected parameters of boar ejaculates. The study material consisted of 17 boars: six Polish Large White (PLW) breed, five Polish Landrace (PL) breed, and six Duroc×Pietrain (D×P) crossbreed, all aged between 8 and 12 months at the beginning of the research. Analyses were conducted on 612 ejaculates, which were collected in two experimental periods: an increasing photoperiod (IP) (January–June) and a decreasing photoperiod (DP) (July–December). A statistically proven impact of photoperiod on the volume of semen was observed in all the studied breeds (P=0.004). During the decreasing photoperiod the mean volume of semen was 261.16±75.20 ml and this was almost 17 ml higher than that for the increasing period. For boars involved in the experiment, day length also had a significant impact on the total number of motile spermatozoa (P=0.037). In the increasing photoperiod the mean number was 3.26×109 lower. A decreasing photoperiod has a positive affect on both boars and the parameters of the collected ejaculates, which were observed in the higher number of insemination doses per ejaculate. Between the different breeds, the reactions of boars to photoperiod differed and the most significant influence of photoperiod on semen parameters was noted among D×P breed boars. Least susceptible to changes in day length were PLW breed boars.  相似文献

The purposes of this study were to specify the occurrence and prevalence of Babesia canis, Borrelia burgdorferi sensu lato, and Anaplasma phagocytophilum in ticks removed from dogs in Warsaw, and to determine the Borrelia species occurring in Ixodes ricinus ticks. Among 590 collected ticks, 209 were identified as I. ricinus, and 381 as Dermacentor reticulatus. DNA of B. canis was detected in 11% of D. reticulatus ticks. We found that 6.2% of I. ricinus ticks harbored B. burgdorferi s.l. specific DNA and 2.9% harbored A. phagocytophilum DNA. In these samples sequencing of the detected Borrelia amplicon confirmed infection with Borrelia afzelii genospecies. New sequences were submitted to the GenBank database (accession no. EU152128, EU152127, EU152126). This work is the first detection of B. afzelii and A. phagocytophilum in ticks from Warsaw, and the first survey for the prevalence of B. canis, B. afzelii, and A. phagocytophilum in ticks in central Poland.  相似文献

Ultrasonographic evaluation of the spine, especially of the spinal cord, has been rarely reported in dogs. The atlanto-occipital junction provides a small acoustic window through which examination of the craniocervical transition can be performed. Normal sonographic findings of this region in 10 normal brachycephalic dogs are presented and compared with sonographic findings from 25 Cavalier King Charles Spaniels with the caudal occipital malformation syndrome. Sonographic findings were compared with magnetic resonance imaging findings to determine the extent of cerebellar herniation and syringohydromyelia. Cerebellar displacement into the foramen magnum was clearly identified sonographically; however, syringohydromyelia was not discernable due to bone overlay.  相似文献

The aim of this study was to develop an anatomical model of the feline hip joint for low‐field magnetic resonance imaging (LF‐MRI) based on high‐field magnetic resonance imaging (HF‐MRI). The study was performed on six adult clinically healthy European shorthair cats, aged 1–3 years, with body weight of 2.8–4.4 kg. The animals were examined with the use of the Vet‐MRI Grande Esaote LF (0.25 T) scanner and high‐field Siemens Magnetom TRIO (3 T) MRI scanner. In the LF‐MRI, most satisfactory results in T1‐weighted images were obtained when TE was 26 ms in all three planes and when TR was 350–950 ms in the transverse plane, 950–1150 ms in the sagittal plane and 520–750 ms in the dorsal plane. In T2‐weighted images, TE was 90 ms in the transverse and dorsal plane and 120 ms in the sagittal plane. The results were presented as images acquired with LF‐MRI scanners in three planes. The slice thickness was 3 mm for each plane. In LF‐MRI, muscles in the hip joint region and round ligament were well visualized. Unlike in LF‐MRI, the cross section of the femoral nerve was identified in HF‐MRI scans. In examinations of the feline hip joint, the main limitations of LF‐MRI were a lack of reliable contrast between articular cartilage and synovial fluid as well as longer scan time. Despite the above, LF‐MRI images were characterized by good contrast between bones and the surrounding soft tissues.  相似文献

South African Proteaceae are adapted to the low soil phosphorus (P) concentrations of the Cape Floristic Region. The efficient P uptake by Proteaceae means that these plants experience phosphorus (P) toxicity at lower rhizosphere [P] than crop plants. This is only problematic when cultivating Proteaceae (and many plants from this region) on previously agricultural land with high residual soil [P]. In this study we hypothesize that P toxicity will result in element imbalances in leaves of Proteaceae and information from this study aims to facilitate ameliorative treatments. Phosphorus toxicity was induced on-farm in Leucadendron ‘Safari Sunset’ (Proteaceae) with subsequent mapping of element distribution in non-necrotic leaf tissue using micro particle-induced X-ray emission spectrometry. Phosphate supply up to 0.01 mM in a fertigation solution resulted in increased stem length of Leucadendron ‘Safari Sunset’ while P concentrations in excess of this resulted in decreased stem length, increased leaf [P] up to 0.25% (w/w) and, between 1 mM and 5 mM P supply, typical P toxicity symptoms were observed. High P supply (5 mM P) resulted in increased leaf [P] in most leaf tissues including the epidermis, where calculations from an equilibrium speciation model indicated that there was 30% more dissolved PO43− in the epidermis compared to leaves at low P supply (0 mM added P on soil with 34 mg P kg−1). Concomitantly, bundle sheath and epidermal [Ca] were reduced and 10% more Ca was predicted to be adsorbed and precipitated as hydrapatite at high P supply. High P supply resulted in increased leaf [Cl] and [Mn] in all tissues studied; decreased total leaf [Fe], bundle sheath, xylem, phloem and epidermal [Fe] and decreased total leaf [Zn] and xylem and phloem [Zn]. The observed symptoms of P toxicity in Leucadendron ‘Safari Sunset’ (necrosis in some plants, chlorosis and leaf rosetting) co-occurred with (1) excess PO43−, which may bind Ca in the epidermis (leading eventually to necrosis); (2) reduced [Fe] and increased [Mn] (leading to chlorosis) and (3) reduced total and vascular [Zn] (leading to leaf rosetting).  相似文献

Background

Genome editing of monocot plants can be accomplished by using the components of the CRISPR/Cas9 (clustered regularly interspaced short palindromic repeat/CRISPR associated Cas9) technology specifically optimized for these types of plants. Here, we present the development of RNA-guided Cas9 system for simplex and multiplex genome editing in barley.

Results

We developed a set of customizable RNA-guided Cas9 binary vectors and sgRNA modules for simplex and multiplex editing in barley. To facilitate the design of RNA-guided Cas9 constructs, the pBract derived binary vectors were adapted to Gateway cloning and only one restriction enzyme was required for construction of the sgRNA. We designed a synthetic, codon optimized Cas9 gene containing the N terminal SV40 nuclear localization signal and the UBQ10 Arabidopsis 1st intron. Two different sgRNAs were constructed for simplex editing and one polycistronic tRNA-gRNA construct (PTG) for multiplex editing using an endogenous tRNA processing system. The RNA-guided Cas9 constructs were validated in transgenic barley plants produced by Agrobacterium-mediated transformation. The highest mutation rate was observed in simplex editing of the cytokinin oxidase/dehydrogenase HvCKX1 gene, where mutations at the hvckx1 locus were detected in 88% of the screened T0 plants. We also proved the efficacy of the PTG construct in the multiplex editing of two CKX genes by obtaining 9 plants (21% of all edited plants) with mutations induced in both HvCKX1 and HvCKX3. Analysis of the T1 lines revealed that mutations in the HvCKX1 gene were transmitted to the next generation of plants. Among 220 screened T1 plants we identified 85 heterozygous and 28 homozygous mutants, most of them bearing frameshift mutations in the HvCKX1 gene. We also observed independent segregation of mutations and the Cas9-sgRNA T-DNA insert in several T1 plants. Moreover, the knockout mutations of the Nud gene generated phenotype mutants with naked grains, and the phenotypic changes were identifiable in T0 plants.

Conclusions

We demonstrated the effectiveness of an optimized RNA-guided Cas9 system that can be used for generating homozygous knockout mutants in the progeny of transgenic barely plants. This is also the first report of successful multiplex editing in barley using a tRNA processing system.

Yersinia ruckeri (Y. ruckeri) can cause mortalities that are contributing to substantial economic losses in the rainbow trout (Oncorhynchus mykiss) aquaculture sector. Because of its most characteristic clinical signs, the disease in rainbow trout caused by this pathogen is called enteric redmouth disease. Although it is considered to affect mainly salmonids, there are reports in the available literature of isolating this bacterium from other fish species, both clinically healthy and diseased. The aim of this study was to analyse the available data concerning yersiniosis in non‐salmonid fish. The analysed data indicate that Y. ruckeri is a threat not only to rainbow trout. Some of the affected species have high commercial importance and mortalities may contribute to high economic losses. The disease symptoms may not be specific and can be different from those characteristic for enteric redmouth in trout, which may lead to misdiagnosis. Collected information indicates that infection with Y. ruckeri should be taken into account in the diagnostic procedures not only in salmonids.  相似文献

1. The effect of riparian ecotone functional complexity and stream hydraulics on an upland river ecosystem has been analysed. 2. The amount of nutrients retained by bottom sediment was lowest on a sandy substrate (range: 26–104 mg m−2 P-PO4) and highest in wetland bays (range: 558–5368 mg m−2 P-PO4). A stream bed covered by Berula erecta had about three times higher retentive nutrient capacity ( = 584 mg m−2 day−1) than did a sandy substrate( = 205 mg m−2 day−1). 3. The amount of allochthonous organic matter (CPOM) deposited on the stream bed decreased with current velocity. The trophic potential of CPOM, measured as total protein, was significantly correlated with the amount of deposited CPOM (r = 0.863; p<0.00001) and depended on stream order. 4. Both invertebrate and fish biomass in the upland river were significantly correlated with calcium/bicarbonate (benthos: r = 0.858; p<0.006; fish: r = 0.918; p<0.001). 5. Fish biomass, diversity and species richness were highest in pools, lower in riffles and lowest in the run/transition zone. 6. Macroinvertebrate biomass was highest at an intermediate riparian ecotone complexity with an adequate supply of organic matter and incident light. Fish biomass followed the same trend, being lowest in heavily shaded areas and in open channels without riparian vegetation (range: 1–4.5 g m2), but highest in ecotones of intermediate complexity (range: 1.6–92.8 g m2). The ‘cascading effect’ of invertebrate density depletion, which was inversely related to fish biomass, was observed seasonally. 7. The above results indicate that riparian ecotone structure and the heterogeneity of the stream channel may regulate biodiversity, productivity and nutrient retention in the fluvial corridor.
