The phylogenetic relationships among Erwinia amylovora biovar 4 (the pathogen of bacterial shoot blight of pear in Japan), other biovars of E. amylovora, and Erwinia pyrifoliae were investigated using the sequences of 16S rRNA, gyrB, and rpoD genes. The tested isolates formed two distinct monophyletic groups in the phylogenetic trees constructed based on the gyrB gene, rpoD gene, or a combination of the three genes: group 1 contained E. amylovora biovars 1, 2, and 3; group 2 contained E. amylovora bv. 4 and E. pyrifoliae. This phylogenetic analysis showed that E. amylovora bv. 4 was more closely related to E. pyrifoliae than to other biovars of E. amylovora.

A preemergence damping-off of tobacco (Nicotiana tabacum L.) was found in Fukuoka, Japan, in 2009. A fungus repeatedly isolated from the black-rotted seeds reproduced the symptoms in tobacco seeds, and the preemergence damping-off developed after flower and soil inoculation with the fungus. Brown spots and blight developed on leaves of tobacco seedlings after inoculation with the isolate. The fungus was reisolated from the rotted seeds and blighted leaves. The causal fungus was identified as Stemphylium lycopersici based on morphology and phylogenetic analysis. This is the first report to confirm the pathogenicity of S. lycopersici in tobacco in the world. 相似文献

To determine the effects of artificial headlands (groins with an anchor-shaped head portion) on the surf-zone fish assemblage structure of a sandy beach with strong wave action, three study sites (headland site sheltered behind the head portion, pocket-beach site between two adjacent headlands and exposed reference site without headlands) were sampled on the Kashimanada Coast, central Japan, in summer and autumn in 2012 and 2013. Distinct differences in physical conditions were recognized between the headland site and the other two sites throughout the study period, the former being characterized by lower wave height and turbidity, finer sediment particles and richer detritus. The fish assemblage structures also differed among the sites, the headland site supporting greater species and individual numbers. Cluster analysis showed clear differences in species composition of fish assemblages between the headland site and the other two sites. In addition, small-sized fishes tended to be more abundant at the headland site compared with the other two sites, although no differences were apparent in fish assemblage structure between the pocket-beach and reference sites. These results suggested that the presence of headlands had marked effects on the surf-zone fish assemblage structure in the study area. 相似文献

In 2006, stem rot and blue-green crusty lesions were found on the stems of tomato plants in Chiba Prefecture, Japan. Penicillium oxalicum was isolated repeatedly from the diseased plants. The causal fungus reproduced natural symptoms after artificial inoculation
of tomato plants and was re-isolated from symptomatic plant tissue. P. oxalicum is a new pathogen that causes blue mold on tomato plants in Japan. 相似文献

BACKGROUND: Grey mould caused by the fungus Botrytis cinerea Pers. ex Fr. is one of the major diseases in grapes. The use of fungicides is a simple strategy to protect grapes against B. cinerea disease. However, phenotypes exhibiting resistance to fungicides have been detected in B. cinerea populations. The variation of fungicide‐resistant B. cinerea isolates renders B. cinerea disease control difficult in grapevine fields. RESULTS: The authors have developed a nested polymerase chain reaction–restriction fragment length polymorphism (PCR‐RFLP) method to detect fungicide‐resistant B. cinerea isolates at an early growth stage of grapes in grapevine fields. The nested PCR‐RFLP method was carried out to detect benzimidazole‐, phenylcarbamate‐ and/or dicarboximide‐resistant B. cinerea isolates from grape berries and leaves at Eichorn–Lorenz growth stage 25 to 29. This method successfully detected fungicide‐resistant B. cinerea isolates at an early growth stage of grapes. In addition, only 8 h was required from tissue sampling to phenotyping of fungicide resistance of the isolates. CONCLUSION: It is proposed that the early diagnosis of fungicide‐resistant B. cinerea isolates would contribute to further improvement of integrated pest management against B. cinerea in grapevine fields, and that the nested PCR‐RFLP method is a high‐speed, sensitive and reliable tool for this purpose. Electroretinography (ERG) is an effective method for the diagnosis of retinal disease. In the dog, dependable ERG recording is difficult without the use of an expensive device like a Ganzfeld full-field stimulator. The International Society for Clinical Electrophysiology of Vision has defined the standard flash stimulus condition (SF) and evaluation of the retina using the b/a ratio in humans. In dogs, evaluation using the b/a ratio has not been reported, whereas the intensity of SF has been defined. In this study, we performed a convenient ERG recording method using a contact lens electrode with a built-in light source (LED-electrode), and confirmed SF as reported previously. ERG recordings were performed on 15 healthy beagle dogs under sedation. We performed bilateral ERG at 12 different intensities after 30 min dark adaptation. After 10 min light adaptation, we recorded single flash cone and flicker cone response using the SF determined in this study. In this study, SF of 3.0 cd/m(2)/sec (6,000 cd/m(2), 0.5 msec) resulted in b/a=2. The intensity for rod response that recorded only the b-wave was 0.0096 cd/m(2)/sec (80 cd/m(2), 0.12 msec). We could achieve ERG for each response easily and smoothly under sedation, and without general anesthesia. Using an LED-electrode, we could perform more quantitative and reproducible ERG examinations than with traditional methods. We propose that the b/a ratio is the most useful parameter in ERG reporting for evaluating retinal function. 相似文献
