The primary objective of this study was to characterize Fusarium spp. associated with the economically devastating mango malformation disease (MMD) in Mexico. In all, 142 Fusarium strains were isolated from symptomatic mango inflorescences and vegetative tissues in eight geographically diverse Mexican states from 2002 through 2007. Initially, all the Mexican isolates were screened for genetic diversity using appolymerase chain reaction and random amplified polymorphic DNA markers and were grouped into seven distinct genotypes. Based on results of these analyses, evolutionary relationships and species limits of the genetically diverse MMD-associated Fusarium spp. were investigated using multilocus DNA sequence data and phylogenetic species recognition. Maximum parsimony analyses of a five-locus data set comprising 5.8 kb of aligned DNA sequence data indicated that at least nine phylogenetically distinct Fusarium spp. within the Gibberella fujikuroi species complex are associated with MMD, including one species within the African clade (Fusarium pseudocircinatum), two species within the Asian clade (F. mangiferae and F. proliferatum), and at least six species within the American clade (F. sterilihyphosum and five undescribed Fusarium spp.). Molecular phylogenetic analyses indicate that a novel genealogically exclusive lineage within the American clade was the predominant MMD associate in Mexico. This new Fusarium sp. caused MMD and could be distinguished from all other known species morphologically by the production of mostly sterile, coiled hyphae which are typically associated with sporodochial conidiophores together with unbranched or sparsely branched aerial conidiophores. Koch's postulates were completed for isolates of the new species on nucellar seedlings of mango cv. Ataulfo. This pathogen is formally described herein as F. mexicanum.  相似文献

BACKGROUND: Cardamom, an important spice crop often attacked by many insect pests, is controlled mainly using synthetic insecticides. As honey bees play a vital role in pollination in cardamom, the impact of insecticides on honey bees needs to be explored to assess its safety. RESULTS: Risk assessment based on contact toxicity revealed diafenthiuron to be a non‐selective insecticide to bees with a low selectivity ratio (the ratio between the LD50 for beneficial and pest species). A dose of diafenthiuron that killed 90% of cardamom borer, Conogethes punctiferalis Guenee, was found to kill 100% of Indian bees. Based on the hazard ratio (the ratio between the field‐recommended dose and the LD50 for the beneficial), diafenthiuron was found to be slightly to moderately toxic to bees. Diafenthiuron, even at low concentrations of LC1 (the concentration that killed 1% of bees), was found to affect the foraging and homing behaviour of Indian bees. Of bees fed with 30 µg mL?1 of diafenthiuron, 40% were found missing on the third day after exposure. However, diafenthiuron did not affect bee visits to the cardamom fields. CONCLUSION: Diafenthiuron is more highly toxic to Apis cerana indica F. than to C. punctiferalis by contact, using selectivity ratio and probit substitution methods of risk assessment, but the hazard ratio revealed diafenthiuron to be a slightly to moderately toxic chemical. Diafenthiuron was found to affect the foraging and homing behaviour of bees at sublethal concentrations. Thus, sublethal effects are more relevant in risk assessment than lethal and acute effects. This case series describes clinical, radiographic, and histopathological features of mandibular swellings in 5 immature, large breed dogs. The dogs originated from different regions of the United States. In each case, intraoral dental radiography of the jaw swelling revealed a two-layered (double) ventral mandibular cortex. Biopsy was performed in 4 of the 5 puppies, revealing periosteal new bone formation. Resolution of the mandibular swelling was spontaneous in the 4 dogs available for follow-up examination. The authors postulate that the clinical, radiographic, and histopathological presentation of mandibular swelling in these 5 dogs is a distinct pathological entity consistent with an inflammatory condition of the maturing human mandible known as periostitis ossificans.  相似文献

Laminin‐332 (laminin‐5) is a basement membrane heterotrimeric protein composed of alpha‐3, beta‐3 and gamma‐2 laminin chains. Laminin‐332 polypeptides are targeted by auto‐antibodies in human patients with mucous membrane (cicatricial) pemphigoid or, more rarely, subepidermal vesicular diseases that resemble epidermolysis bullosa acquisita (EBA) or bullous pemphigoid (BP). The objectives of this report were to characterize the clinical, histopathological and immunological characteristics of nine dogs with auto‐antibodies targeting laminin‐332. Immunological investigations consisted of direct immunofluorescence (IF), indirect IF with intact and salt‐split canine gingival, and salt‐split normal or laminin‐332‐deficient human skin, immunoblotting with purified human laminin‐332 and immunoblotting with recombinant NC1 domain of human collagen VII. All dogs exhibited varying degrees of skin blistering and ulceration associated with microscopic subepidermal vesiculation with or without inflammatory cells. Indirect IF established that circulating IgG auto‐antibodies bound the dermal side of salt‐split canine lip and human skin. In five dogs, IgG variably recognized the basement membrane of laminin‐332‐deficient human skin (three dogs negative, two dogs positive). In all nine dogs, IgG auto‐antibodies detected purified human laminin‐332 by immunoblotting. In two dogs, additional targeting of collagen VII‐NC1 was present. These observations establish laminin‐332 as a novel basement membrane antigen in dogs with autoimmune blistering diseases with variable clinical phenotypes. The names ‘acquired junctional epidermolysis bullosa’, ‘anti‐laminin‐332 mucous membrane pemphigoid (MMP)’ and ‘mixed auto‐immune subepidermal blistering dermatosis’ are proposed for dogs with clinical signs reminiscent of EBA, MMP or BP respectively.  相似文献

A new speciation model developed and implemented in Polymath was found to be successful in predicting struvite precipitation in soils. Struvite (NH4MgPO4) has been identified as a mineral for the recovery of nitrogen (N) and phosphorus (P). Predicting struvite precipitation potential in soil is important for optimal quantification of nutrient species. Polymath and Visual Minteq models were used for prediction of several solid phases in the soil. One approach to immobilize P for solid-phase formation is by co-blending. Immobilization was achieved through the blending of an Al-based water treatment residual (Al-WTR) and with Ca?CMg-based materials [slag and magnesium oxide (MgO)]. The results suggest that Polymath model revealed solid Phases of dicalcium phosphate pentahydrate (DCPP), magnesium hydroxide (MHO), magnesium orthophosphate (v) docosahydrate (MP22), magnesium orthophosphate (v) octahydrate (MP8), and struvite, which were lacking in the modeling from Visual Minteq. Residual leachate from the co-blended amendments; Soil+WTR+Slag, Soil+WTR+MgO, Soil+MgO, Soil+Slag, Soil+WTR, and the control (without amendment) had struvite of 353, 199, 119, 90, 37, and 12?mg?l-1, respectively. This implies that struvite, a phosphate mineral can be precipitated in the soil and could be released as nutrients for plant uptake. Struvite precipitation in soil and for reuse may reduce cost and may be a safe practice for sustainable environmental nutrient management.  相似文献

A study was conducted to establish a growth rate response of potato plant (Solanum tuberosum L.) tops to exposure to 72-hr periods of six constant temperature treatments (10, 15, 20, 25, 30, 35°C) and to determine a thermal optimum for vegetative potato growth rates. The plants were maintained under “no stress” soil moisture conditions throughout the study. The temperature treatments were accomplished by inserting the plants into a growth chamber for 72 hours at selected temperatures. Maximum growth (as measured by percent change in leaf area) occurred at 25°C with temperature growth differences significant at the 5% level. Plant height measurements (percent change occurring during treatment time) showed maximum stem elongation at 30°C during an early growth stage and 25°C during stage 2 (4 weeks later). Predictive models developed from the data indicate maximum rates of leaf area increase in both growth stages occur at 24.7°C and that maximum rates of stem elongation occur at 31.3 and 27.4°C for growth stages 1 and 2, respectively. Leaf water potential and stomatal resistance measurements taken at the end of each treatment period indicated that no water stress occurred.  相似文献

Potato chip color is an important trait in determining quality and acceptability of cultivars for processing. Therefore, the genetics of potato chip color is important in varietal breeding. The use of 2xS. tuberosum Group Tuberosum haploid × 2x wild species hybrids provides disomic inheritance for genetic analysis of chip color. Two hundred forty haploid-species hybrids representing crosses between 15 haploids and 10 species were evaluated for potato chip color after 21 days reconditioning (18 to 20C) following 6 months at 4C. Phenotypic variation for chip color was found among the haploid-species hybrids. Crosses between good, medium, and poor performing clones were made in all combinations and 136 families were obtained. Progeny were evaluated for reversion resistance (chipping direct from 10C storage) and reconditioning (chipping from 4C after tuber warming) chip color. Variation for chip color was found among the progeny for each trait. Significant correlations between reversion resistance and reconditioning were found. Yet, the use of at least two chip tests (one for reversion resistance and one for reconditioning) is more informative than the use of one test and subsequent predictions made, regarding the other trait. A three locus hypothesis for both reversion resistance and reconditioning is proposed for the inheritance of each trait. One dominant allele at each of the three loci is necessary for good chipping. One or two loci may be common to both characteristics. Good chipping 2x hybrids that produce 2n pollen can be used to transfer this trait to the 4x level through use of 4x × 2x crosses.  相似文献

In humans with atopic dermatitis and in mouse models of IgE-mediated allergic diseases, evidence is mounting that the stratum corneum (SC) provides an important barrier against environmental allergens. At this time, it is not known whether the SC has a similar role in dogs, especially in those with atopic dermatitis. The objectives of this pilot study were to determine whether SC removal led to earlier and stronger sensitization of atopic dogs to Dermatophagoides farinae (Df) house dust mites. Five Maltese-beagle atopic (MBA) dogs were sensitized epicutaneously after the SC was removed with ten tape strips (TS group), while sensitization was done without tape strips in five other MBA dogs (nontape stripping; NTS group). During this 16 week study, sensitization was assessed with allergen-specific IgE serology, intradermal testing with Df allergens and determination of stimulation indices of blood mononuclear cells cultured with Df and stained for CD4 and the activation markers CD25 or CD30. Compared with dogs from the NTS group, those of the TS group exhibited earlier rises in Df-specific IgE serum levels, usually had higher allergen-specific IgE titres, showed higher intradermal test reactivity and had earlier increases and higher percentages of CD25- or CD30-positive activated allergen-specific peripheral CD4-positive T lymphocytes. These observations implicate a role of the SC as a barrier limiting sensitization to exogenous allergens in this experimental atopic dog model.  相似文献
