Field evaluation of melatonin implants to advance the breeding season in 1-year-old farmed red deer hinds

The efficacy of controlled-release melatonin implants to advance the onset of the breeding season was assessed in 1-year-old red deer hinds on five commercial deer farms in various localities in the North Island of New Zealand. Between 44 and 60 hinds in each of six herds were equally divided among treatment and control groups at each site. Melatonin treatment commenced between 27 November and 16 December and was achieved by the subcutaneous administration of two 18 mg melatonin implants. Three doses were given at about 30 day intervals. Two adult stags for each hind group were treated with three 18 mg melatonin implants concurrently on either two or three occasions. On each property, treated and control hinds were joined as one herd to treated stags commencing 30 January-10 February and concluding 15 May-2 June. The hinds in the four experimental herds underwent rectal ultrasound examination May-June to estimate conception rate and foetal age. Calving dates, hind and calf mortalities, weaning weights, and the antler growth cycle and harvesting data were recorded. Overall, treatment with melatonin resulted in an average advance of the median calving date of 22 days (range 12-36 days) when compared with untreated controls in the same herds. Pregnancy rates were 91.3-100% in treated hinds and 63.6-100% in untreated hinds. There were no differences in calf mortality or calf sex ratio between treated and untreated groups. No hind deaths could be attributed to melatonin treatment. The weaning weights of calves were 5.68 kg and 4.43 kg heavier for the male and female offspring of treated hinds respectively, compared with those of control hinds. Treated stags commenced rutting behaviour earlier than normal and the antler casting and growth cycle was advanced. Treatment resulted in advancement of the seasonal pattern of coat changes in hinds and stags, but no untoward side effects of the melatonin treatments were observed.  相似文献

Action of deltamethrin on N-type (Ca_v2.2) voltage-sensitive calcium channels in rat brain

Isolated presynaptic nerve terminals prepared from whole rat brain were used to evaluate the action of deltamethrin on voltage-sensitive calcium channels by measuring calcium influx and endogenous glutamate release. Deltamethrin-enhanced K⁺-stimulated calcium influx and subsequent Ca²⁺-dependent glutamate release. The effect of deltamethrin was concentration-dependent, stereospecific, blocked by ω-conotoxin MVIIC but unaltered in the presence of tetrodotoxin. These results suggest that N-type voltage-sensitive calcium channels are a site of action at the presynaptic nerve terminal. Electrophysiological studies were carried out using rat brain Ca_v2.2 and β₃ subunits coexpressed in Xenopus oocytes to validate such action. Deltamethrin reduced barium peak current in a concentraion-dependent and stereospecific manner, increased the rate of activation, and prolonged the inactivation rate of this channel. These experiments support the conclusion that N-type voltage-sensitive calcium channel operation is altered by deltamethrin.  相似文献

Effect of recent cropping history and herbicide use on the degradation rates of isoproturon in soils

Five soil samples were taken from each of five fields with different crop management histories. Three of the fields were in an arable rotation, the fourth field was temporary grassland, and the final field was under permanent grass. Of the three arable fields, two had been cropped with winter wheat in three of the preceding 6 years, and the third had last been cropped with winter wheat once only, 6 years previously. With one exception, the winter wheat had been sprayed with the herbicide isoproturon. The rate of isoproturon degradation in laboratory incubations was strongly related to the previous management practices. In the five soils from the field that had been treated most regularly with isoproturon in recent years, <2.5% of the initial dose remained after 14 days, indicating considerable enhancement of degradation. In the soils from the field with two applications of the herbicide in the past 6 years, residues after 27 days varied from 5% to 37% of the amount applied. In soils from the other three sites, residue levels were less variable, and were inversely related to microbial biomass. In studies with selected soils from the field that had received three applications of isoproturon in the previous 6 years, kinetics of degradation were not first‐order but were indicative of microbial adaptation, and the average time to 50% loss of the herbicide (DT₅₀) was 7.5 days. In selected soils from the field that had received just one application of isoproturon, degradation followed first‐order kinetics, indicative of cometabolism. Pre‐incubation of isoproturon in soil from the five fields led to significant enhancement of degradation only in the samples from the two fields that had a recent history of isoproturon application.  相似文献
