The aim of this study was to classify the histopathological features of pigs infected with Mycobacterium avium complex (MAC). We used slaughtered pig organs systemically infected with MAC. The results showed granulomatous lesions which were observed predominantly in the digestive organs and regional lymph nodes rather than respiratory organs. The histological picture showed a wide range of granulomatous stages from exudative to fibrotic reactions to the MAC infection. Eosinophils and giant cells were characteristically observed in the exudative reactions. The histopathological type in primary focus tended to be maintained in the respective organs. Most strains with the same genotype showed pathogenicity for guinea pigs irrespective of the type of granuloma. Although these findings suggest that different stages of a granulomatous lesion originating from the same causative agent might influence histological patterns, other possibilities such as the hereditary background of the host, or the effects of viral infections should be considered. 相似文献

Arthropathy in dogs induced by ofloxacin, a quinolone antimicrobial agent, was pathophysiologically investigated. In the in vivo studies, ofloxacin was administered orally once or twice at 20 mg/kg/day to male juvenile (3-month-old, n=3) or adult (36-month-old, n=2) dogs, and the humeral and femoral heads were examined pathologically. Unlike adult dogs, fluid-filled vesicles were macroscopically observed on the articular surfaces of one juvenile dog 24 hours after a single treatment with ofloxacin. These lesions were seen in all juvenile dogs by twice dosing. Microscopically, fissures or cavity formations in the middle zone of the articular cartilage were noted only in juvenile dogs. Furthermore, the cartilage matrix from the abnormal area to the articular surface showed a decreased safranin-O staining intensity, suggesting proteoglycan depletion. Ultrastructurally, chondrocytes in the middle zone of juvenile dogs displayed dilatation of the cisternae in the rough endoplasmic reticulum as an initial hallmark. In the in vitro studies, chondrocytes isolated from the articular cartilage of naive juvenile dogs were exposed to ofloxacin at 6.3-100 microg/ml for 24 hours. Although no changes were noted in the deoxyribonucleic acid synthesis, protein synthesis, or proteoglycan release at concentrations of up to 100 microg/ml, the proteoglycan synthesis was evidently decreased in a dose-dependent manner from 12.5 microg/ml. The results obtained suggest that the inhibitory action of ofloxacin on proteoglycan syntheses in the chondrocytes may largely contribute to the early morphologic features in the articular cartilage of the juvenile dog. 相似文献

Analysis of cellular components at multiple levels of biological information can provide valuable functional insights. We performed multiple high-throughput measurements to study the response of Escherichia coli cells to genetic and environmental perturbations. Analysis of metabolic enzyme gene disruptants revealed unexpectedly small changes in messenger RNA and proteins for most disruptants. Overall, metabolite levels were also stable, reflecting the rerouting of fluxes in the metabolic network. In contrast, E. coli actively regulated enzyme levels to maintain a stable metabolic state in response to changes in growth rate. E. coli thus seems to use complementary strategies that result in a metabolic network robust against perturbations. 相似文献

In the rich and long-standing literature on the flow-induced formation of oriented precursors to polymer crystallization, it is often asserted that the longest, most extended chains are the dominant molecular species in the "shish" of the "shish-kebab" formation. We performed a critical examination of this widely held view, using deuterium labeling to distinguish different chain lengths within an overall distribution. Small-angle neutron-scattering patterns of the differently labeled materials showed that long chains are not overrepresented in the shish relative to their concentration in the material as a whole. We observed that the longest chains play a catalytic role, recruiting other chains adjacent to them into formation of the shish. 相似文献

Mycoplasma pneumonia of swine (MPS) is caused by Mycoplasma hyopneumoniae (M.hp) and is a common chronic respiratory disease of pigs. Recently, a genetically selected variant of the Landrace pig (Miyagino L2) has a lower incidence of pulmonary MPS lesions. We investigated the pathological and immunological characteristics of MPS resistance in these pigs (n = 24) by comparing with the normal landrace pig (control: n = 24). The pathological MPS lung lesion score in MPS‐selected landrace pigs was significantly lower than in the control. The gene expression of interleukin (IL)‐12p40, which acts as a chemoattractant and a component of the bioactive cytokines IL‐12 and IL‐23, was significantly higher at the hilar lymph nodes, lung, and spleen in MPS‐selected landrace pigs than in control landrace pigs, and these were negatively correlated with the macroscopic MPS lung lesion score. In summary, we demonstrate that resistance against MPS in Miyagino L2 pigs is associated with IL‐12p40 up‐regulation, in comparison with normal landrace pigs without the MPS vaccine. In addition, a comparative study of macroscopic MPS lung lesions and IL‐12p40 gene expression in lung and hilar lymph nodes may lead to beneficial selection traits for the genetic selection for MPS resistance in pigs. 相似文献

Osmoregulatory actions of growth hormone (GH) and its mode of action in salmonids are reviewed. We present evidence suggesting
that insulin-like growth factor I (IGF-I) mediates some of the actions of GH on seawater acclimation. Plasma concentration
and turnover of GH rise following exposure to seawater. Exogenous GH (in vivo) increases gill Na+,K+-ATPase activity and the number of gill chloride cells, and inhibits an increase in plasma osmolarity and ions following transfer
of fish to seawater. A single class of high affinity GH receptors is present in the liver, gill, intestine, and kidney. The
levels of IGF-I mRNA in the liver, gill and kidney increased after GH-injection. After transfer to seawater, IGF-I mRNA increased
in the gill and kidney following the rise in plasma GH, although no significant change was seen in the liver. Injection of
IGF-I improved the ability of the fish to maintain plasma sodium levels after transfer to seawater. GH treatment also sensitizes
the interrenal to adrenocorticotropin (ACTH), increasing cortisol secretion. Both cortisol and IGF-I may be involved in mediating
the action of GH in seawater adaptation, although studies on the effect of GH on osmoregulatory physiology of non-salmonid
species are limited. An integrated model of the osmoregulatory actions of GH is presented, and areas in need of research are
outlined.
Résumé Cet article est une revue des effets osmorégulateurs de l'hormone de croissance et de son mode d'action. Nous présentons des résultats qui suggèrent que le facteur de croissance de type insuline (IGF-I) est un médiateur de certaines des actions de la GH sur l'adaptation à l'eau de mer. Les concentrations plasmatiques et le renouvellement de la GH augmentent après transfert en eau de mer. La GH exogène stimule (in vivo) l'activité Na+,K+-ATPase et le nombre de cellules à chlorure branchialeset inhibe les augmentations de l'osmolarité et des concentrations ioniques du plasma observées après transfert en eau de mer. Une seule classe de récepteurs à haute affinité pour la GH est présent dans le foie, les branchies, l'intestin et le rein. Les niveaux d'ARNm d'IGF dans le foie, les branchies et le rein augmentent après injection de la GH. Après transfert en eau de mer, les ARNm de l'IGF augmentent dans les branchies et dans le rein en suivant l'augmentation de GH plasmatique, bien qu'aucune modification ne soit observée au niveau du foie. L'injection d'IGF augmente la capacité du poisson à maintenir ses niveaux de sodium plasmatique après transfert en eau de mer. Le traitement à la GH augmente la sensibilité à l'adrenocorticotropine (ACTH) et stimule donc les niveaux de cortisol. A la fois le cortisol et l'IGF-I semblent impliqués comme médiateurs des effets de la GH dans l'adaptation à l'eau de mer, bien que les études sur les effets de la GH sur la physiologie de l'osmorégulation chez les espèces non-salmonidés restent encore limitées. Un modèle intégré des actions de la GH sur l'osmorégulation est présenté et les domaines de recherche à développer sont soulignés.相似文献
