根据病毒的理化特性，从猫三联活疫苗中分离出猫鼻气管炎、猫西洋太和猫泛白细胞减少症病毒；筛选出每种病毒敏感的细胞，并将该3种病毒分别分别在其中传代与扩增；通过电镜观察此3种病毒的形态学特征及其在宿主细胞内增殖部位、分布情况等，并以此对猫3种病毒进行了初步鉴定。  相似文献   

Genotype evaluation,muscle histopathology and ultrastructure in a Duchenne muscular dystrophy animal model

AIM: To evaluate the genotype , muscle histopathology and ultrastructure in dko mice. METHODS: Dystrophin/Utrophin-deficient double knockouts (dko) mice were obtained from university of Oxford, UK. Genotype of filial generation of heterozygote was evaluated by PCR-SSP. HE staining and fluorescent immunohistochemistry by SABC-Cy3 were used to detect striated muscle of dko mouse, and the muscle ultrastructure was observed by transmission electron microscope(TEM). RESULTS: In 112 filial generation mice, there were 28 mdx (25.0%), 26 dko (23.2%) and 58 heterozygote (51.8%), which coincided with the law of Mendelian inheritance. HE staining showed that the myocytes were not very uniform, there were phenomenon of round outline, centrally nucleated fibers, widening interspace, inflammatory cell infiltration and connective tissue proliferation in dko mice. There were no any immunofluorescent expression of dystrophin and utrophin in sarcolemma in dko mice. TEM showed sarcolemma breakage, separation and edema, and loose myofibril texture, inflammatory cell infiltration and connective tissue proliferation in dko mice. CONCLUSION: PCR-SSP is a very quick and accurate way for genotype evaluation of filial generation. The pathophysiology of dko mouse was very similar to Duchenne muscular dystrophy (DMD), and dko mouse is an ideal animal model for study of DMD clinical therapy.  相似文献

利用表达 H 5亚型禽流感病毒 (AIV)血凝素基因的重组鸡痘病毒 (r FPV- HA)以不同剂量免疫 1日龄 SPF鸡、有或无母源抗体 (FPV、AIV H5)的商品鸡 ,并于免疫后 2 1d利用同亚型 AIV通过肌肉注射进行致死性攻击 ,通过检测免疫后 HI抗体应答、比较攻毒后发病率和死亡率评价免疫剂量和母源抗体对 r FPV- HA免疫效力的影响。结果发现 ,免疫后 2 1d,15 %～ 2 0 %的 SPF鸡和无母源抗体商品鸡可检出 HI抗体 ,而含母源抗体商品鸡检测不到 HI抗体。利用H5亚型 AIV致死性攻击后 ,10 3～ 10 6 PFU的 r FPV- HA可保护 95 %～ 10 0 %的 SPF鸡和无母源抗体商品鸡抵御强毒攻击 ,使之免于发病和死亡 ;而不同剂量 r FPV- HA接种的含母源抗体商品鸡有 80 %～ 90 %发病和死亡。结果表明 ,在较宽的免疫剂量范围内 ,r FPV- HA对 SPF鸡和无母源抗体商品鸡可提供良好的保护 ,显示出一定的应用前景 ;母源抗体影响 r FPV- HA诱导的免疫应答 ,且提高免疫剂量亦不能克服其干扰作用 ,这提示在实际应用中需优化免疫程序 ,避免母源抗体干扰。  相似文献

Mid-storage and pre-sowing seed treatments for lettuce and carrot

Mid-storage hydration-dehydration treatments with medium-vigour lettuce seed very effectively reduced physiological deterioration in storage and ultimately showed better field emergence and final yield. Soaking-drying treatment of high-vigour lettuce seed was not effective. Pre-sowing treatment of medium-vigour lettuce seed, though less effective than mid-storage treatment, gave better field performance, but high-vigour seed did not benefit from the treatment. For carrot, both mid-storage and pre-sowing seed treatments proved beneficial, with the former showing better performance than the latter. Unlike lettuce, well-preserved carrot seed responded positively to the pre-sowing treatment.  相似文献   

土地利用景观格局动态分析——以阜康荒漠绿洲为例

阜康荒漠绿洲土地利用景观格局受自然分异和水土资源开发利用程度的制约 ,通过划分景观样带和选取景观空间格局指标 ,本文重点揭示该区域景观格局的变化及其影响因子。结果表明 :( 1 ) 1 987年至 1 998年间阜康土地利用景观格局变化主要表现为耕地面积在洪积扇上增加 ,在冲积平原上下降 ;荒地则刚好相反。林地面积在低山丘陵带和洪积扇带下降 ,在冲积平原上没有变化。牧草地面积在低山丘陵带上升 ,其他各带均下降。城镇面积在各样带上都有所增加 ,水域各带基本不变。 ( 2 )低山丘陵带和洪积扇带景观多样性和均匀度下降 ,优势度上升 ;而冲积平原则多样性和均匀度上升 ,优势度下降。景观破碎度有增加的趋势 ,表明了人为影响的不断增强。  相似文献   

Effect of bFGF on glucose regulated protein 78 expression and apoptosis in human ovarian cancer CAOV3 cells

AIM: To evaluate the effect of bFGF on the apoptosis and the expression of glucose regulated protein 78 (GRP78) in ovarian cancer CAOV3 cells.METHODS: Starvation induced cell apoptosis was conducted. After treatment with bFGF, the cell cycle and apoptosis were determined by FACS analysis and Annexin V/PI staining, respectively. The expression of protein kinase B (PKB) and GRP78 were detected by Western blotting and RT-PCR. RESULTS: Compared to starvation group, the cells treated with bFGF were still viable and increased activation of PKB and high expression of GRP78 were observed, which were prevented by PKB inhibitor wortmannin effectively (P<0.01). CONCLUSION: bFGF plays a critical role in anti-apoptosis and proliferation in human ovarian cancer through PKB signal transduction pathway, partly by upregulating the expression of GRP78.  相似文献   

氮吸附法分析生物质焦孔隙结构

采用氮吸附法对4种生物质焦(稻壳、树叶、玉米秆、棉花秆)的孔隙结构进行测量,结果表明,不同种类焦样的比表面积和孔径分布有明显差别,树叶的比表面积最大,为242.21 m2·g-1,玉米秆的比表面积最小,为0.81 m2·g-1.850℃时,稻壳、树叶、玉米秆焦样的孔径分布曲线在微孔和中孔范围各有一个分布峰,而棉花秆焦样的孔径分布曲线只在中孔范围内出现一个分布峰.热解温度是影响孔隙结构的一个重要因素,在高温条件下,同步热解得到的焦样的比表面积较大,微孔较多.在本研究中,600℃、850℃的稻壳焦样和850℃的树叶焦样具有较大的比表面积,比较适合做吸附剂.  相似文献
