Effect of L-lysine Added in Diluent on the Fresh Semen Quality of Dorper Sheep

In order to explore the effect of different L-lysine levels added in semen dilution on the fresh Dorper sheep quality stored at liquid state,3 health Dorper ram were used to collect semen which were equal-packaged and added to solution containing different levels (0,0.1,0.2,0.3,0.4 and 0.5 g) L-lysine in 120 mL diluent and at 0,6,24,30,48,54,72 and 78 h,the sperm motility,membrane integrity and acrosome integrity were detected.The results showed that the sperm motility in groups added 0,0.1 and 0.2 g L-lysine were higher than other groups,and that in 0.4 and 0.5 g L-lysine groups decreased faster than other groups;The sperm motility in 0.5 g L-lysine group reduced to 0 at 30 h,and it reduced to 0 at 48 h in the group added 0.4 g L-lysine;The effective survival time and survival index in the group added 0.1 g L-lysine was higher than other groups (P< 0.05);The membrane integrity of 0.1 g group was the highest,that had no significant difference with control group from 0 to 54 h (P >0.05),while after 72 h the two groups had significant differences (P< 0.05).The membrane integrity in 0.4 and 0.5 g groups were the worst,and that was significant different with other groups (P< 0.05);The acrosome integrity of 0.1 g group was the highest which had no significant difference with control group (P >0.05),0.4 and 0.5 g groups were the worst,the difference between them was not significant (P >0.05),while was extremely significant difference with other groups except for 0 and 6 h (P< 0.01).The results suggested that dilution added a high concentration of L-lysine could inhibit sperm quality,when the count of L-lysine was more than 0.2 g,sperm quality was significantly decreased,adding 0.1 g L-lysine could improve Dorper sheep fresh sperm quality stored at liquid state.  相似文献

Effects of remifentanil on monophasic action potential and transmural dispersion of repolarization in rabbit myocardium

AIM: To study the effect of remifentanil on monophasic action potential and transmural dispersion of repolarization (TDR) in the 3-layer myocardium of isolated rabbit hearts. METHODS: Adult rabbits (n=18, 2.0 ~ 2.5 kg) were used to isolate the hearts for preparing Langendorff perfusion model. The hearts were randomly divided into 3 groups after perfusion with K-H solution for 15 min: the perfusion in control group (C group) continued for 60 min; the hearts in remifentanil group (R group) were perfused with 12 μg/L remifentanil K-H solution for 60 min; the hearts in remifentanil+aminophylline group (RA group) were given 60-min perfusion of 12 μg/L K-H remifentanil+30 mg/L aminophylline. The HR and 3 layers of myocardial monophasic action potential (MAP) in the left ventricular anterior wall were recorded at time points after balanced infusion for 15 min (T₀), and continued perfusion for 15 min (T₁), 30 min (T₂) and 60 min (T₃). The monophasic action potential duration of repolarization at 90% (MAPD₉₀) and the transmural dispersion of repolarization (TDR) were calculated. The early afterdepolarization, delay afterdepolarization and arrhythmia were also observed. RESULTS: In R group, slower HR and prolonger MAPD₉₀ and TDR at T₁~T₃ were observed as compared with those at T₀ (P<0.05). R group showed slower HR and longer MAPD₉₀ and TDR than C group and RA group (P<0.05). CONCLUSION: Remifentanil slows the HR, extends the MAPD₉₀ and increases the TDR, thus being prone to induce reentry. Aminophylline makes HR faster and MAPD₉₀ shorter, thereby reducing the TDR.  相似文献

FGF4在肺癌细胞体内和体外中表达差异的研究

FGF4已经被证明是癌基因,它涉及肿瘤的生长和转移,为了解FGF4的表达与肿瘤微环境的关系。我们利用FGF4抗体通过免疫组化对一名肺癌患者癌旁,癌组织,癌组织小鼠移植瘤,二次移植瘤以及原代培养的细胞爬片进行FGF4检测,探究其表达差异。通过对比癌组织与对照组（癌旁组织）,FGF4在癌巢中高表达;同样,移植瘤与二次移植瘤的癌巢中与癌旁组织比较,FGF4表达相对较高;但是将癌组织进行原代培养后,免疫组化检测细胞爬片FGF4,发现仅有5%±0.21%的肿瘤细胞表达FGF4,对照蛋白Cytokine作为肿瘤标记物,则在100%的肿瘤细胞中表达。研究提示免疫组化检测到FGF4在体内和体外表达不同,提示肿瘤细胞FGF4的表达与肿瘤微环境调控密切相关,肿瘤微环境对肿瘤细胞的FGF4的调控有着重要作用。  相似文献

设置5个水平的种植密度,研究了不同种植密度对甜玉米品种‘夏王'产量和主要农艺性状的影响。结果表明:在不同的种植密度下,甜玉米‘夏王'的产量和果穗性状都有明显的差异;玉米产量随着种植密度的增加而增加,增加到一定幅度后又随着密度的增加而下降;随着种植密度的增大,植株果穗长度、果穗粗度、行粒数、正品果穗率和单穗重量均逐渐下降;综合性状表现,‘夏王'在上海地区春播适宜的播种密度为4.50万—4.95万株/hm~2,从鲜穗商品性及种植效益考虑,最佳种植密度为4.50万株/hm~2。  相似文献   

鱼肠道弧菌免疫检测方法的研究

采用灭活的鱼肠道弧菌作为抗原,制备兔抗血清,建立了鱼肠道弧菌的间接荧光抗体检测技术(indirect im-munofluorescence assay test,IFAT)、Western-blotting免疫印迹检测技术。IFAT结果显示阳性信号覆盖整个菌体,明亮,清晰,荧光信号呈长弧形且两端钝圆,与鱼肠道弧菌形状一致。Western-blotting结果显示共10条蛋白带发生免疫反应,其中6条蛋白带结果较清晰,显示出较强的特异性免疫反应,阴性对照组无条带。IFAT和Western-blotting结果说明所建立检测方法能够准确、快速的检测出鱼肠道弧菌。  相似文献
