The aim of the present research is to study the effect of pH values on the sperm rheotaxis properties. Semen collected from bulls was diluted with SOF medium (1:10). pH of the medium was adjusted using a digital pH meter to the following pH values: 6.0, 6.2, 6.4, 6.4, 6.8, 7.0. All kinetic parameters of sperm (n = 3,385) were determined through a computer‐assisted sperm analysis (CASA) system using microfluidic devices with controlled flow velocity. The following parameters were determined: total motility (TM%), positive rheotaxis (PR%), straightline velocity (VSL, μm/s), average path velocity (VAP, μm/s), linearity (LIN, as VSL/VCL, %), beat cross‐frequency (BCF, Hz) and curvilinear velocity (VCL, μm/s). Nitric oxide, calcium and potassium were estimated in semen at different pH values. To confirm the effect of nitric oxide and K+, we used sodium nitroprusside (an NO donor) and KCL as (a K+ donor) to see their effect on sperm PR%. The results showed no difference in TM% at pH (6–7). The PR% was the lowest at pH 6 and 7. The best parameters for the PR% were at pH 6.4–6.6. The concentration of Ca+2 did not change at different pH values. The mean NO values decreased with the increase of pH; however, the mean values of K+ increased with the increase of pH. Addition of high concentration of NO and K+ to the semen media at fixed pH level had a negative effect on TM% and PR%. In conclusion, the bull sperm had the best rheotaxis properties at pH 6.4–6.6 and sensitive to the change of seminal NO and K+. 相似文献

Photosynthesis is an important process for plants in which plants utilize the energy from sunlight to manufacture and produce all their own food. Although UV-C damage is not physiologically relevant for plants growing in the sun, short-wavelength (UV-C) radiation from germicidal lamps has been used to study cell damage in animals as well as in plants. This study provides a review of the effects of a methanolic extract of spinach (SE) as a natural powerful antioxidants used at two concentrations (25 and 50?ppm) on fenugreek seedlings grown under UV-C stress conditions. The results revealed that exposure of fenugreek seedlings to low UV-C period (30?min) increased growth criteria of the produced seedlings. In addition, the contents of chlorophyll, total pigments, total protein, free amino acids, non-enzymatic (carotenoids, total phenol, flavonoids, α?tocopherol & ascorbic acid contents) and enzymatic antioxidants were increased at this period. By increasing exposure periods of UV-C radiation, the growth criteria and chemical constitutes of the seedlings were decreased. Application of lower concentration of SE (25?ppm) improve the growth of the seedlings at 30-min UV-C and was able to protect seedlings from high periods of UV-C irradiation by increasing all measured parameters except CAT. These increases might be an adaptive mechanism to minimize the adverse effects of longer exposure of UV-C radiation. Moreover, treating the irradiated seedlings with spinach extract led to an obvious alteration in gene expression including synthesis of some proteins bands and disappearance of other protein sets of the protein profile. 相似文献

The present study aimed to determine floristic composition, the structure of the vegetation, and species distribution at 16 sample plots in Wadi Fatimah, Mecca, Saudi Arabia, highlighting the ecological factors that affect species distribution. The study revealed 100 plant species belonging to 34 families. Fabaceae and Poaceae were the dominant families, and chamaephytes and therophytes were predominant life forms, which demonstrate a typical desert life-form spectrum. Altitude showed a highly significant association with species frequency distribution. The highest species richness value of 23 species per plot was recorded in the coastal plain (Vegetation groups II & III). The lowest species richness value of 9 species per plot was recorded in the littoral salt marsh (Vegetation group I). Chorological analysis exhibited the Saharao-Arabian–Sudano-Zambezian chorotypes, 47% of the studied species were bi-regional. Using TWINSPAN, DCA, and CCA multivariate analysis, four major vegetation groups were classified into three main ecosystems (I) Halopeplis perfoliata, Suaeda fruticosa, Acacia tortilis, and Suaeda fruticosa, representing the littoral salt marsh; (II) Rhazya stricta and Haloxylon salicornicum, characterized by the coastal plain; and (III) Senna italica, Glinus lotoides, and Tribulus macropterus were the leading groups in the inland mountainous country. The CCA ordination showed that the separation of Vegetation group III along the axis was affected by Mg, K, and P concentration, and VG IV was significantly associated with silt, clay, and fine sand content. Furthermore, the Vegetation group I was significantly correlated with Na and Ca concentration, and VG II group was significantly correlated with heavy metal concentrations of elements as Fe, Pb, Zn, Ni, and Cu. 相似文献

Cropping systems that integrate cover crops into crop rotations, reduce tillage intensity and frequency, and maintain residue cover have the potential to improve agricultural sustainability in drylands. However, there is much yet to learn about the benefits of cover crops in sustainable dryland farming in the southern Great Plains (SGP). We reviewed the literature on the effects of cover crops on soil organic carbon (SOC), nitrogen, soil water conservation, and crop yields in dryland cropping systems of the US Great Plains (GP), and analyzed the opportunities and challenges for integrating cover crops into dryland crop-fallow systems of the SGP. Majority of the studies in the central Great Plains (CGP) and the northern Great Plains (NGP) of the United States suggest that cover cropping improves sustainability of cropping systems through their positive effects on SOC accumulation, nutrient cycling, soil erosion control, weed suppression, and soil health improvement. However, integrating cover crops into dryland cropping systems of the SGP faces challenges because of low quantity of soil-water availability. More research on the tradeoff between water use and other agroecosystem benefits of cover cropping is required to successfully integrate cover crops into dryland cropping systems in the SGP. 相似文献

The present study aimed to evaluate the effect of organic and inorganic selenium (Se) supplementation on semen quality and blood serum profiles of buffalo bulls. Nine mature buffalo bulls were divided into three groups: control (non‐supplemented); organic Se (10 mg Sel‐Plex®/head twice weekly) and inorganic Se (10 mg sodium selenite/head twice weekly). Semen was collected twice a week for 3 months during Se supplementation. Semen properties were evaluated from fresh ejaculate. Moreover, fructose concentration, aspartate and alanine transaminase (AST and ALT) activities, total protein and total cholesterol were assayed in seminal plasma. Additionally AST, ALT, testosterone and Se levels were determined in the blood serum. Results showed that Se supplementation significantly (P < 0.05) influences the semen parameters during 3 months of treatment. Organic Se significantly (P < 0.05) increased the percentage of viable sperms compared to inorganic Se and the control group. Fructose concentration was significantly higher (P < 0.05) in the seminal plasma of organic Se‐treated bulls. Serum testosterone and Se concentrations were significantly (P < 0.05) increased in the Se supplemented groups than the control group. In conclusion, Se supplementation improved the parameters of buffalo bull semen and more precisely, organic Se was more effective for the improvement of semen quality and some blood components than inorganic Se. 相似文献

Viral isolation, polymerase chain reaction (PCR), dot blot hybridization (DBH), and indirect enzyme-linked immunosorbent assay
(iELISA) were used for the diagnosis of lumpy skin disease in clinically infected, fevered, and apparently normal dairy cows.
Lumpy skin disease virus (LSDV) was isolated from skin biopsies and blood samples collected from clinically infected cows
in percentages of 72% and 20%, respectively. The virus recovered from blood samples collected from fevered cows in percentage
of 33.3%. Both PCR and DBH detected viral DNA in 100% of skin biopsies collected from clinically infected cows whereas the
detection rates in blood samples collected from clinically infected animals were 100% and 84% using PCR and DBH, respectively.
Viral DNA was detected in blood samples collected from fevered cows using PCR and DBH in percentages of 77.8% and 66.6%, respectively.
Only 19.1% of blood samples collected from in-contact cows was positive for both of PCR and DBH. Detection rates of antibodies
against LSDV using iELISA in serum samples collected from clinically infected and fevered cows were 56% and 11.1%, respectively,
whereas all in-contact cows had no antibodies against the virus. 相似文献

The effects of aquarium background colour and feed colour on survival, growth rates and feed utilization efficiency of thinlip mullet (Liza ramada) larvae (0.035 g) were investigated in two experiments. In the aquarium background colour trial, 50 larvae were stocked in duplicates in 120 L glass aquaria filled with dechlorinated tap water. The outside walls and bottoms of each pair of the aquaria were covered with coloured paper sheets to achieve one of six colours (white, black, red, green, yellow and blue), while noncoloured aquaria served as a control. The fish were fed an experimental diet (35% crude protein) at a daily rate of 5% of their body weight (BW), twice a day for 8 weeks. The best growth rates, feed efficiency and survival were achieved in larvae reared in light‐coloured aquaria (white, noncoloured and yellow). Fish performance was significantly retarded in larvae reared in dark‐coloured aquaria (red, green, black and blue). Body composition was not significantly affected by aquarium colour. In a feed colour trial, duplicate groups of larvae (0.035 g) were stocked at 50 fish per 120 L aquarium and fed a test diet (35% crude protein) with six different colours [dark blue, red, yellow, light brown (control), light green and dark brown] at a daily rate of 5% BW, twice a day for 8 weeks. The best performance and survival were achieved in fish fed on dark‐coloured diets (red, dark blue and dark brown). Light‐coloured diets (yellow, light green and light brown) resulted in inferior performance. Body composition was not significantly affected by feed colour. These results suggest that light‐coloured tanks should be used for rearing thinlip mullet, L. ramada larvae, while dark‐coloured diets are more preferable to light‐coloured diets. 相似文献

Due to the fact that the persistence ofBacillus thuringiensis (B.t.) is very short in the field as affected by exposure to ultraviolet radiation, some biochemical approaches have been adopted
in an attempt to increase potentiation of the pathogen against the two lepidopterous corn borers,Chilo agamemnon andOstrinia nubilalis. These approaches were based on the incorporation of some selected non-toxic chemical compounds with different modes of action
with the endotoxin ofB.t. fed to the larvae and thus resulting in its potentiation.
Among the compounds tested, some representatives of inorganic salts, organic acids, protein and lipid solubilizing agents,
amino acids and amides showed an obvious potentiation to the endotoxin activity against the target insects. Inorganic salts,
such as, calcium oxide, calcium carbonate, zinc sulphate and potassium carbonate at 0.1% potentiated the activity of the product
Dipel 2X (B.t. var.kurstaki) against the two tested species in varying degrees. With regard to protein solubilzing agents, urea, sodium thioglycollate
and EDTA enhanced the potency ofB.t. againstO. nubilalis with a fold increase of 1.4–2.3. The lipid emulsifying agent Tween 80 (0.5%), caused 1.3 fold increase in the potency ofB.t. With respect toC. agamemnon, sodium thioglycollate and EDTA (0.1%) were effective in potentiating the activity ofB.t. with fold increase of 3.1 and 1.2, respectively, while urea caused a decrease in the potency ofB.t as compared with the control. The lipid emulsifying agent Tween 80 (0.5%) caused 1.3 fold increase in the potency ofB.t. The potentiating effect of aromatic compounds is not obvious with respect to the tested insect species. With amino acids
and amides, it appears that some of the tested compounds enhanced the potency ofB.t. against the tested insect species but in varying degrees. The mode of action of the tested compounds has been discussed in

The physiochemical characteristics of β‐glucan in oat and barley foods can affect human physiological response. A method for continuous measurement of β‐glucan viscosity with a Rapid Visco Analyzer (RVA) was developed to overcome the complexity of the common protocols based on in vitro digestion methods. The effects of several parameters on viscosity and solubility were considered. Oat cereal foods showed different RVA viscosity profiles depending on their physiochemical characteristics. Products high in starch exhibited a high initial viscosity that was reduced by α‐amylase action, whereas products with low amounts of starch exhibited a slow increase in viscosity. The viscosity of all samples reached a plateau in the viscosity curve after 1–2 hr, which is the key for obtaining reproducible results. Optimum digestion condition was achieved using sodium phosphate buffer (pH 6.9) and 1% β‐glucan dispersion at 37°C and 160 rpm. A particle size of <0.6 mm gave more consistent viscosities than did larger particles without affecting the solubility of β‐glucan. Pancreatin and α‐amylase concentrations affected the viscosity profile by influencing the digestion rate of protein and starch in the samples, but pepsin had limited influence at pH 6.9. Highly significant Pearson correlation between the in vitro digestibility protocol and RVA methods was achieved, indicting that the developed method could be used as an effective alternative for measurement of β‐glucan viscosity. 相似文献
