Sodium channels have been a major target for the development of insecticides such as synthetic pyrethroids. However, insecticides currently available induce resistance and present limited selectivity to insect pests. Molecular and biochemical studies, as well as binding experiments using radiolabelled neurotoxins, have shown that sodium channels expressed in various insect orders must be structurally and pharmacologically different. At least three groups of peptide neurotoxins derived from scorpion venom are highly active on insects and very weakly or practically inactive on mammals. It is proposed that various insecticides are examined for possible cooperative interactions with the peptide toxins highly active on insects, and pairs of ligands are identified that will increase the selectivity not only between mammals and insects but also between different pest and non-pest insects. This is feasible on the basis of the differential allosteric modulations observed between LqhαIT, an α-toxin highly active on insects, and brevetoxin on locust versus cockroach and rat brain sodium channels. Moreover, combination of LqhαIT with the pyrethroid deltamethrin increased the binding of [125I] LqhαIT by more than 1.8-fold, and the combined presence of brevetoxin further increased the binding. Such allosteric modulation may provide a new approach to increase the selective activity of pesticides on target organisms by simultaneous application of allosterically interacting drugs, designed on the basis of the selective peptide toxins.  相似文献

Novel aryl chain isosters and analogues of 7-keto-8-aminopelargonic acid (KAPA) and 7,8-diaminopelargonic acid (DAPA), the vitamer intermediates involved in the biosynthetic pathway of biotin, possessing chain lengths of eight carbon atoms, were prepared and evaluated as potential herbicides. In the greenhouse test the most active compounds were the fluorinated derivative 9d and the selenophenyl/furan mixture 17m/17p, which were most active against Foxtail millet. In the more sensitive Arabidopsis test the most active substances were 9a and 17m, which displayed GR(50) (concentration of active compound causing 50% growth inhibition) values of 0.2 and 0.5 mg kg(-1) respectively (values of < 50 mg kg(-1) are considered herbicidal).  相似文献

The neuronal elements of the vomeronasal organ (VNO) of camel were investigated immunohistochemically. PGP 9.5 labeled the receptor cells in the vomeronasal sensory epithelium, but not the supporting or basal cells. OMP stained some receptor cells, but no immunoreactive signals for OMP were detected in the non-sensory epithelium. PLCβ2 labeled scattered cells in the sensory epithelium and a larger number of cells in the non-sensory epithelium. Double labeling immunohistochemistry revealed that the PLCβ2-positive cells were surrounded by substance P-positive nerve fibers. Collectively, these data suggest that the camel VNO bears, in addition to the mature vomeronasal receptor cells, trigeminally-innervated solitary chemosensory cells which are expected to play a substantial role in the control of stimulus access to the VNO.  相似文献

During 2008–2011 model field experiments were carried out at the Joni?k?lis Experimental Station of Lithuanian Research Centre for Agriculture and Forestry on a clay loam Endocalcaric Endogleyic Cambisol. The study was aimed to establish the comparison of various postharvest practices (mineral nitrogen fertilizer alone or together with a bioactivator Penergetic k, livestock slurry, red clover biomass, and straw incorporation in the soil by a stubble cultivator at a 10 cm depth) on the acceleration of the initial (nine-month period) decomposition of winter wheat straw. During this period straw mass decomposition intensity (DIM) was 20.7–29.1%, carbon (C) concentration decreased by 6.5– 22.8%, while an increase occurred in nitrogen (N) 1.1–2.2 times. The highest straw decomposition rate was recorded when after straw incorporation autumn was warm and humid. That year straw mass C to N ratio (C/N) was 38–46. Under less favourable autumn conditions, the highest decomposition of straw was achieved having applied mineral N (with and without Penergetic) and livestock slurry and having incorporated the straw in the soil (C/N = 40–55). A slower decomposition rate was observed for the straw spread on the soil surface with mineral N addition or on undersown red clover.  相似文献

  1. Camouflage grouper, Epinephelus polyphekadion (Bleeker, 1849), and squaretail coralgrouper, Plectropomus areolatus (Rüppell, 1830), are commercially important medium-bodied groupers that aggregate at specific sites and times to spawn and are highly vulnerable to fishing during these events. Populations of both species are in decline globally, such that management specifically targeting these species is warranted.
  2. A 12-month fish market survey in Chuuk, Federated States of Micronesia, provided an opportunity to examine age-based reproductive life history of these two groupers and assess whether current management acts to conserve them. Life history characteristics of both E. polyphekadion and P. areolatus include a functionally gonochoristic sexual pattern and rapid growth particularly during early life history.
  3. P. areolatus demonstrated early maturity (2.8 years) and a relatively brief lifespan (10 years), while delayed maturity (4.5 years) and higher longevity (25 years) was shown for E. polyphekadion. The spawning seasons for E. polyphekadion and P. areolatus were 2 and 3 months, respectively, which fall entirely within the January–April grouper sales, catch, and export ban period for Chuuk. Marketed catch included 22% E. polyphekadion and 15% P. areolatus juveniles, suggesting that size limits may aid in the conservation of these species.
  4. Findings from this and recent grouper life history studies suggest that the current 4-month ban in Chuuk be applied only to grouper species known to reproduce during these months to minimize economic impacts to fishers and market owners, and prevent shifts in fishing pressure to more vulnerable species, such as those with low population turnover times, slow growth, or late maturity. Size limits for catch, sale and export are also warranted as an additional management option.

Costimulatory ligands, B7.1 and B7.2, have been incorporated into viral and DNA vectors as potential nonchemical adjuvants to enhance CTL and humoral immune responses against viral pathogens. In addition, soluble B7 proteins, minus their transmembrane and cytoplasmic domains, have been shown to block the down regulation of T-cell activation through blockade of B7/CTLA-4 interactions in mouse tumor models. Recently, we developed swinepox virus (SPV) vectors for delivery of feline leukemia antigens for vaccine use in cats [Winslow, B.J., Cochran, M.D., Holzenburg, A., Sun, J., Junker, D.E., Collisson, E.W., 2003. Replication and expression of a swinepox virus vector delivering feline leukemia virus Gag and Env to cell lines of swine and feline origin. Virus Res. 98, 1-15]. To explore the use of feline B7.1 and B7.2 ligands as nonchemical adjuvants, SPV vectors containing full-length feline B7.1 and B7.2 ligands were constructed and analyzed. Full-length feline B7.1 and B7.2 produced from SPV vectors were natively processed and costimulated Jurkat cells to produce IL-2, in vitro. In addition, we explored the feasibility of utilizing SPV as a novel expression vector to produce soluble forms of feline B7.1 (sB7.1) and B7.2 (sB7.2) in tissue culture. The transmembrane and cytoplasmic regions of the B7.1 and B7.2 genes were replaced with a poly-histidine tag and purified via a two-step chromatography procedure. Receptor binding and costimulation activity was measured. Although feline sB7.1-his and sB7.2-his proteins bound to the human homolog receptors, CTLA-4 and CD28, both soluble ligands possessed greater affinity for CTLA-4, compared to CD28. However, both retained the ability to partially block CD28-mediated costimulation in vitro. Results from these studies establish the use of SPV as a mammalian expression vector and suggest that full-length-vectored and purified soluble feline B7 ligands may be valuable, nonchemical immune-modulators.  相似文献

This paper offers a forest operations perspective in environmentally sensitive areas in Europe and the United States. The objective is to help inform key sustainability knowledge gaps in forest operations to reduce impacts on the environment and the equipment operators in these increasingly targeted and difficult to operate in areas. Under existing guidelines and best management practices, the protection of ecosystem services linked to soil, water, wildlife and biodiversity have rightly received the highest considerations. However, the connection between protecting these ecological values, safe operational practices and technological advancements is rarely well articulated. One means to address the operational and technological gap is the selection of equipment and tailor the work conditions to terrain type, forest features and management objectives. To that effect, this paper discusses the environmental, planning, equipment selection and operator considerations and proposes possible solutions and guidelines to contain both the operators’ safety and environmental risks in forest operations.  相似文献

Multi-factor crop management field experiments were conducted in Vezaiciai, in southwestern Lithuania, from 2004 to 2006 to assess the influence of tillage and organic fertilizer management on stem base disease development. The treatments included 4-year crop rotations of continuous grass, winter triticale, winter oilseed rape and spring barley in combination with three tillage systems (shallow (8–10 cm) non-ploughing tillage, shallow (10–12 cm) tillage and deep (20–25 cm) ploughing). Eyespot prevailed both in winter triticale and spring barley crops. Winter triticale was more susceptible than spring barley. Shallow tillage decreased eyespot incidence and severity relative to deep tillage. Crop residue management exerted a weak but statistically significant effect on the disease development on triticale; in general, slightly higher numbers of affected stems were recorded in triticale plots where the straw from the previous crop had been removed prior to planting. However, complete straw removal from plots significantly decreased eyespot severity in comparison with crop residue or manure management treatments in spring barley. The highest number of affected triticale and barley plants was found in shallow (10–12 cm) tillage plots. The effects of residue management on minimising eyespot severity can be expected only after a delay of a few years.  相似文献
