The aim of the present study was to evaluate the effect of high pre‐slaughter environmental temperature on meat (Longissimus lumborum muscle) quality traits of Italian autochthonous pigs Suino Nero Lucano (SNL). High pre‐slaughter environmental temperature influenced the meat quality of autochthonous pigs. In particular, the meat from SNL pigs exposed to heat stress (H) showed significantly lower pH values at 24 and 48 hr post mortem, haem iron content and redness (a*) value, and a significantly higher shear force, drip loss and lightness (L*) values, compared to meat from SNL pigs reared under comfortable environmental temperature (C). Moreover, H meat showed a significant reduction of vitamin A, vitamin E, creatine and carnosine content, compared to C meat. Despite this, no pale‐soft‐exudative pork was found in our study, and no significant difference was detected between two studied groups for intramuscular fat and protein content of meat. In addition, despite the meat from SNL pigs exposed to heat stress has suffered a qualitative decrease, it showed a good endogenous antioxidants content.  相似文献

Objective

To study the effect of alternating the order of midazolam and alfaxalone administration on the incidence of behavioural changes, alfaxalone induction dose and some cardiorespiratory variables in healthy dogs.

Study design

Prospective, randomized, controlled, clinical trial.

Animals

A total of 33 client-owned dogs undergoing elective procedures.

Methods

Following intramuscular acepromazine (0.02 mg kg?1) and morphine (0.4 mg kg?1) premedication, anaesthesia was induced intravenously (IV) with a co-induction of either midazolam (0.25 mg kg?1) prior to alfaxalone (0.5 mg kg?1; group MA), or alfaxalone followed by midazolam at identical doses (group AM). The control group (CA) was administered normal saline IV prior to alfaxalone administration. Additional alfaxalone (0.25 mg kg?1 increments) was administered as required in all groups until orotracheal intubation was possible. Changes in behaviour, quality of induction, ease of intubation and incidence of adverse events at induction were recorded. Heart rate (HR), respiratory rate (fR) and systolic arterial blood pressure (SAP) were measured before treatments (baseline values), 30 minutes after premedication and at 0, 2, 5 and 10 minutes postintubation.

Results

The incidence of excitement was higher in group MA compared with groups CA (p = 0.005) and AM (p = 0.013). The mean induction dose of alfaxalone was lower in group AM compared with group CA (p = 0.003). Quality of induction and ease of intubation were similar among groups. Mean HR values decreased after premedication and increased after alfaxalone administration in all groups. Mean SAP values were similar between groups. The number of animals that required manual ventilation was higher in the MA group.

Conclusions and clinical relevance

Despite a lower occurrence of adverse events at induction in group AM compared with group MA and a reduction of alfaxalone dose requirement in group AM compared with group CA, the use of an alfaxalone–midazolam co-induction does not seem to produce any cardiovascular or respiratory benefits in healthy dogs.  相似文献

The objectives of this research study were to isolate, evaluate, and compare the fine structures of starch fractions obtained from a wild‐type (normal) corn starch and amylose‐extender25, dull39, sugary2, and sugary1 corn mutants in the same genetic background using three different fractionation procedures based on gel‐permeation chromatography or alcohol‐precipitation methods. Starch fractions obtained from each of the three methods were enzymatically debranched and analyzed using high‐performance anion‐exchange chromatography with a postcolumn amyloglucosidase reactor and a pulsed amperometric detector. The separations were performed by fractionation on a GPC column, by precipitation with 1‐butanol, and by preferential precipitation with 1‐butanol and isoamyl alcohol. Using any of these methods, no apparent differences in the molecular weight distributions of amylopectin or of amylose among the different starches were observed. The proportions of branch chain lengths of the starch components obtained by the various fractionation methods were very similar among methods for each of the starch types analyzed, such as the predominance of long branch chains in ae25 corn and that of the short branch chains in su2 corn. Overall, the effect of the corn mutations was more important to the differences observed among the starch types than was the method of fractionation used.  相似文献

This study reports the changes in fatty acids, taste, and microstructure of cooked southern king crab meat (Lithodes santolla) during storage at 0°C for 10 days and at ?20°C for 90 days. At the end of both storage times, the iodine value decreased by 16.5%, while 83.5% of the initial fatty acid quality remained unchanged. The polyene ratio decreased by 32% at 0°C and 35.9% at ?20°C, whereas the atherogenic and thrombotic indices remained at values that do not represent any risk to human health. Free amino acids that contribute to taste (taste activity value, TAV > 1) were: glycine and alanine (sweetness), arginine (bittersweetness), and histidine (bitterness). The bittersweet taste imparted by arginine (initial TAV = 16.4) was prevalent even at the end of frozen storage (TAV = 7.9). The umami taste was elicited by disodium 5′-adenosine monophosphate (AMP) nucleotide. The equivalent umami concentration in g MSG/100 g meat changed from 0.031 to 0.045 in refrigerated samples and to 1.6 in frozen samples. A loss of the original fibrous structure of the meat was evidenced during both treatments. Refrigerated samples presented a disintegrated and homogeneous texture at 10 days, while freezing formed a spongy tissue at 90 days.  相似文献

Rainbow trout (Oncorhynchus mykiss) maintained in crowded (100 kg m− 3) and uncrowded (20 kg m− 3) conditions were fed 42 days with five experimental diets having different levels of vitamin E (25.6 and 275.6 mg kg diet− 1), C (0 and 1000 mg kg diet− 1) and HUFA (highly unsaturated fatty acids, 12.5 and 320.5 g kg diet− 1): −E−HUFA, −E+HUFA, +E−HUFA, +E+HUFA, −C+E+HUFA. Cortisol, plasma metabolites, tissue glycogen, fish composition, and tissue fatty-acid profile were evaluated at the end of the experimental period. In general, no changes in cortisol levels were associated with crowding, although +E+HUFA and −C+E+HUFA fish showed higher levels (mean ± SE, 55.5 ± 11.1 and 78.0 ± 11.3 ng ml− 1) as a consequence of a possible interaction between chronic crowding and diet composition. Protein and glucose con-centration in plasma displayed no effect of crowding, but liver glycogen showed a general tendency to decrease in −E−HUFA, −E+HUFA, +E−HUFA, +E+HUFA, −C+E+HUFA crowded groups (70.2 ± 2.1, 52.1 ± 2.5, 73.4 ± 7.4, 91.7 ± 3.3, 74.2 ± 8.4 mg g− 1 tissue, respectively) compared to uncrowded groups (108.9 ± 14.2, 82.7 ± 8.8, 92.4 ± 10.7, 99.1 ± 10.0, 103.5 ± 15.6 mg g− 1 tissue, respectively), thus proving significant in −E+HUFA fish. Variations in total lipids, triglycerides, total cholesterol and HDL as well as LDL cholesterol in plasma were manifested under crowding conditions, displaying a certain influence of vitamin E and HUFA dietary content. Final body composition, in general, showed no change attributable to fish density, but some differences associated with diet composition were found in lipid and moisture percentages of crowded fish. Liver and muscle fatty-acid profile revealed a clear effect of the dietary lipid source that was more evident in muscle than in liver at normal fish density, and in some cases this effect was modulated by dietary vitamin E and C content and fish-culture conditions.  相似文献

Degradation of litter strongly depends on its chemical composition, which in turn affects the associated microorganisms. In the alpine region, the abandonment of pastures leads to a rigorous change in the composition of the litter layer, shifting from grass to highly recalcitrant dwarf shrub litter, thus affecting the food web and decomposition processes. Three species belonging to different functional groups (grasses, herbs, dwarf shrubs) and indigenous on managed and/or abandoned alpine pastureland were selected for this study, the annual grass Dactylis glomerata, characteristic for managed sites, Trollius europaeus as representative of a herb common on both managed and abandoned areas, and Vaccinium vitis-idea as typical dwarf shrub arising on abandoned alpine pastures. Litter bags containing litter material from either one of the three plant types were incubated on the soil surface for 9 weeks. DNA was extracted from the substrate, amplified and analysed using PCR–DGGE. Fingerprinting analyses of bacterial and fungal communities showed that the microbiota attached to the litter differed considerably depending on the plant type. Nonetheless, specific bacterial bands were present in the fingerprinting patterns of all three litter types. Identifying these organisms applying the 16S rRNA clone libraries revealed the dominance of bacteria from the class Bacteroidetes, representing two thirds of all identified band positions.  相似文献

This study was conducted to describe the ultrasonographic appearance and technique for the evaluation of sciatic nerve (ScN) in cats. An anatomical study was carried out using six feline cadavers to determine anatomic landmarks. An ultrasonographic 'in vitro' study was performed using eight pelvic limbs from four fresh feline cadavers to assess the ScN in three regions (glutea, femoralis and popliteal) using a 4-13MHz linear transducer. Five healthy adult experimental cats were employed for an 'in vivo' ultrasonographic study of the ScN using the same protocol described for the 'in vitro' study. The ultrasonographic images of the ScN were well correlated with those obtained in the anatomical study. The ScN was easily identified in all the approaches. The ScN was observed as a hypoechoic structure with internal echoes, outlined by hyperechoic lines. This study shows the usefulness of ultrasound to evaluate the entire pathology of the ScN in the cat.  相似文献

The effects of handling on haematocrit, haemoglobin, plasma glucose, lactate and total amino acids, liver glycogen and hepatic activity of fructose 1,6‐bisphosphatase (FBPase) and alanine aminotransferase (AAT) were investigated in common dentex (Dentex dentex Linnaeus, 1758). A total of 42 fish (50‐g weight) were subjected to handling (netting and shaking in the air for 45 s). Six fish were sampled at 0, 0.5, 1, 2, 4, 8 and 12 h after handling. Six unstressed fish were also sacrificed at each time and used as controls. Handling resulted in a rise in plasma glucose and lactate levels, as well as in enhanced hepatic FBPase and AAT activities. In most cases values returned to control levels 8 h after handling. There were no significant changes in haematocrit, haemoglobin, plasma total amino acids and liver glycogen levels as a consequence of handling. The results suggest that dentex is rather tolerant to handling; apparently, sensitivity to handling is not the main factor responsible for the low survival exhibited by this species in aquaculture.  相似文献
