Purpose

Characterization of tannery sludge (TS) for its plausible use in amelioration and phytoremediation of heavy metal rich TS treated soil by growing economically important plants (Ricinus communis, Brassica juncea and Nerium oleander).

Materials and methods

Treatments were prepared by amalgamation of TS (0, 5, 10, 20, 30, 50, 75, 100 %) with garden soil (GS). All treatments were analysed for chemical properties, total and DTPA (Diethylene triamine pentaacetic acid) extractable heavy metals (Cr, Pb, Cu, and Mn). Seed germination experiment was conducted; unvarying saplings were selected and planted in concerned pots and allowed to grow for 90 days in green house. At harvest, plant samples were washed with distilled water and used for determination of growth parameters (biomass of root, shoot and total biomass on dry weight basis) and metal accumulation in different parts of the plant. Translocation factor (TF) and bioconcentration factor (BCF) have been calculated to check the phytostabilization capability of studied plants.

Results and discussion

Application of TS in fixed quantity as an amendment resulted in significant improvements of GS characteristics (alkaline pH with high electrical conductivity, organic carbon, available NPK and heavy metals) and in treatments. DTPA extractable heavy metal concentrations were found very low and total heavy metal concentrations were also found under allowable range in control and treated soil (T-I to T-VI). The maximum seed germination percentage, plant growth, biomass production for all plant species were observed in T-III treatment (20 % TS + 80 % GS) with majority of the metal accumulation in underground part (BCF >1) and meagre translocation in aerial part (TF <1). From T-IV to T-VII treatment, accumulation of heavy metals in plant parts has generally increased; however, biomass has been tremendously decreased.

Conclusions

TS was found rich in NPK content with significant concentration of heavy metals. Pot growth experiment suggested amelioration of GS with specific quantity of 20 % TS can tremendously enhance the plant growth, help in the utilization of TS and can act as a substitute of synthetic fertilizer. Majority of the metals was accumulated in root part (BCF >1) and meagre translocation (TF <1) in aerial part, concludes R. communis and B. juncea could be suitable plant species to be grown in heavy metal rich TS treated soil, vis-à-vis for phytostabilization of heavy metals. In addition, these oil yielding and medicinal plants can also be used for phytoremediation of moderately contaminated tannery soils.

In cattle production systems, an intense selection pressure for production traits has resulted in the decline of fertility traits. To optimize an efficient reproduction system, the inclusion of both male and female fertility traits in the selection process is very much essential. RAPD (Random Amplified Polymorphic DNA) was developed as a molecular biology tool and has been extensively used, to study intra- and interspecific genetic diversity. The present study was undertaken to utilize RAPD primers to investigate the association between DNA markers and semen quality traits viz. Sperm concentration, total sperm count ejaculate and initial sperm motility and thereby to identify good/poor semen producers. DNA isolated from the blood samples of healthy bulls was subjected to RAPD-PCR. The multiple regression analysis followed by independent t test was carried out to identify suitable markers. Based on the results, only 12 bands were identified as marker suitable for any of the quality trait. This includes, OPA2 ~ 760, OPA2 ~ 700, OPA6 ~ 1,200, OPA9 ~ 400, OPA9 ~ 380, OPA12 ~ 970, OPA14 ~ 715, OPA14 ~ 605, OPA16 ~ 485, OPA17 ~ 860 and OPA18 ~ 480. Multiple regression analysis selected, OPA2 ~ 760 and OPA2 ~ 1,750 for sperm concentration and OPA2 ~ 760, OPA2 ~ 700, OPA9 ~ 620, OPA4 ~ 670 and OPA18 ~ 1,015 for total sperm count/ejaculate. But the t test revealed a significant association between OPA2 ~ 760 and total sperm count. Further, discriminant function analysis also identified this marker in the first step itself. The results of the present study can be exploited as a low-cost alternative strategy for identification of good /poor semen producers in crossbred bulls at an early age.  相似文献

High population density and unscientific land use practices have induced severe land degradation in the fragile Lower Himalayan ecosystem of India. Land cover management has been an effective strategy in managing land degradation through the reduction of water, soil and nutrient losses and improvement in soil fertility and quality. Acacia nilotica (Acacia)-based silvipastoral systems with five intercrops, viz., Eulaliopsis binata (bhabbar), Saccharum munja (munj), Vetiveria zizanioides (vetiver), natural grasses and no grass, were evaluated in a long-term study in degraded bouldery lands in Haryana state of India. All grasses resulted in a reduction of soil, water and nutrient losses and improved microbial properties. However, their association adversely affected the growth of Acacia, and the decline varied with grass species. After 11 years of establishment, sole Acacia plantation had the maximum height (7.58 m), diameter at breast height (dbh) (21.32 cm) and crown spread (7.41 m). Munj produced the highest biomass under Acacia, but most adversely affected its growth, resulting in minimum survival (48 %), height (7.07 m), dbh (16.23 cm) and crown spread (6.57 m). Yield of all the grasses increased during the initial 5–6 years, but declined sharply thereafter, with the maximum decline in bhabbar. Detailed investigations established that the sharp decline in survival and growth of bhabbar was due to shade and not because of nutrient competition or allelopathy. Acacia + V. zizanioides proved the most effective silvipastoral system for resource conservation and biomass production. It also provided the highest NPV (Rs 1.88 lakhs ha?1), B:C (2.37) and IRR (24.70 %) as compared to Rs 6,998 ha?1, 1.05 and 8.76 % under pure Acacia plantation, respectively.  相似文献

beta-Defensins are small antimicrobial peptides of the innate immune system produced in response to microbial infection of mucosal tissue and skin. We demonstrate that murine beta-defensin 2 (mDF2beta) acts directly on immature dendritic cells as an endogenous ligand for Toll-like receptor 4 (TLR-4), inducing up-regulation of costimulatory molecules and dendritic cell maturation. These events, in turn, trigger robust, type 1 polarized adaptive immune responses in vivo, suggesting that mDF2beta may play an important role in immunosurveillance against pathogens and, possibly, self antigens or tumor antigens.  相似文献
