It is important to couple phenotypic analysis with genetic diversity for germplasm conservation in gene bank collections. The use of molecular markers supports the study of genetic marker-trait associations of biological and agronomic interest on diverse genetic material. In this report, 19 Greek traditional sweet cherry cultivars and two international cultivars, which were used as controls, were grown in Greece and characterized for 17 morpho-physiological traits, 15 simple sequence repeat (SSR) loci and 10 inter simple sequence repeat (ISSR) markers. To our knowledge, this is the first report on molecular genetic diversity studies in sweet cherry in Greece. Principal component analysis (PCA) of nine qualitative and eight quantitative morphological parameters explain over 77.33% of total variability in the first five axes. The SSR markers yielded a combined matching probability ratio (MPR) of 9.569 × e−12. The 15 SSR loci produced a total of 92 alleles. Ten ISSR primers generated 91 bands, with an average of 9.1 bands per primer. Expected heterozygosity (gene diversity) values of 15 SSR loci and 10 ISSR markers averaged at 0.683 and 0.369, respectively. Based on stepwise multiple regression analysis (MRA), SSR alleles were found associated with harvest time and fruit polar diameter. Furthermore, three ISSR markers were correlated with fruit harvest and soluble solids and four ISSR markers were correlated with fruit skin color. Stepwise MRA identified six SSR alleles associated with harvest time with a high correlation (P < 0.001), with linear associations with high F values. Hence, data analyzed by the use of MRA could be useful in marker-assisted breeding programs when no other genetic information is available.  相似文献

Sewage sludge treated with 15% bentonite, vermiculite or biochar was evaluated as a soil amendment in comparison to limed and untreated sludge. Seven treatments were established to two soils, an acid and an alkaline, in three replications, i.e. 2% addition of sludge treated with bentonite, vermiculite, biochar and lime and application of 2% untreated sludge, inorganic fertilization and no sludge or inorganic fertilizers (control). Then, the soil treatments were used in a pot experiment with perennial ryegrass (Lolium perenne L.) as a test plant. Sludge treated with the clay minerals or biochar improved pH of the acid soil and significantly increased organic matter and available nutrients of both soils compared to control. Although no salinity or sodicity hazard was evidenced, the initial salinity of acid and alkaline soil increased by four-eight and two-three times, respectively, upon addition of all sludge treatments, especially that of untreated sludge. Moreover, soil available zinc (Zn) increased by four-eight times. Soil application of sludge treated with the clay minerals or biochar increased the total aboveground biomass yield of ryegrass in the acid and alkaline soil by 133%–171% and 72%–88%, respectively, compared to control and enhanced nutrient uptake by plants. Furthermore the microbial metabolic quotient indicated lack of low pH and heavy metal stress with addition of sludge to the acid soil. After three harvests of ryegrass, the residual effect of sludge on pH of acid soil and salinity, available phosphorus (P), Zn and boron (B) of both soils still persisted. Thus sewage sludge treated with 15% bentonite, vermiculite or biochar could be applied to soils at a rate of 2% (≈80 Mg ha−1) to serve as soil amendment and fertilizer for grasses and pasture species; however, caution is needed regarding possible P build-up, Zn phytotoxicity and salinization risks.  相似文献

The presence of organotin (OT) compounds was investigated in coastal areas (10 stations), wastewater treatment systems (six stations), rivers (19 stations) and lakes (12 stations) throughout Greece, in three sampling campaigns organized between October 1998 and September 1999. A screening method for the determination of toluene extractable organotins (TEOTs) by Electrothermal Atomic Absorption Spectrometry (ETAAS) was used during the first two sampling campaigns. TEOTs (sum of tributyltin, TBT, dibutyltin, DBT and triphenyltin, TPhT) were detected in most seawater and wastewater samples at concentrations up to 19.4 and 89.9 ng l?1, respectively, while they were detected occasionally in surface water. During the third sampling campaign, OT compounds were extracted in selected stations using direct derivatization with NaBEt4 in acidic medium and analysed by Gas Chromatography–Mass Spectrometry (GC–MS). In seawater, the levels of TBT, DBT and monobutyltin (MBT) were varied between < 2 and 70, 159 and 19 ng l?1, respectively and tend to decrease with increasing distance from the coastline. TPhT was not detected in any of the samples. Significant concentrations of OT species were detected in influent wastewater, up to 384, 76.2 and 67.8 ng l?1 for TBT, DBT and MBT, respectively. The application of a procedure based on XAD-4 adsorption resin permitted the obtainment of an enriched polyphenolic extract from Sesamum indicum seeds. Chemical analysis of the obtained extract led to the identification of 12 lignans. Among them, 2 lignans, (+)-sesamolinol-4'-O-β-D-glucoside and disaminyl ether, are reported for the first time as natural compounds. Their structure has been determined by spectroscopic methods, mainly by the application of one-dimensional (1D) and two-dimensional (2D) nuclear magnetic resonance (NMR) techniques [heteronuclear multiple-quantum coherence (HMQC), heteronuclear multiple-bond correlation (HMBC), and nuclear Overhauser effect spectrometry (NOESY)] and mass spectroscopy. The isolated compounds were evaluated for their antimutagenic activity. Among the tested lignans, the most active lignan was found to be sesamolin, followed by sesamolinol and samin, against H(2)O(2). Additionally, some of the tested lignans showed desmutagenic activity against benzo[a]pyrene (BaP).  相似文献

Seasonal variation in chemical composition of essential oils obtained from Satureja parnassica subsp. parnassica Heldr. & Sart ex Boiss. and Satureja thymbra L. was determined by GC and GC-MS analysis. The in vitro antibacterial activities of these essential oils were evaluated at two different initial levels against two common foodborne bacteria. Their minimum inhibitory concentration (MIC) and noninhibitory concentration (NIC) values were determined via an innovative automated technique, which was applied on a Malthus apparatus combining the conductance measurements with the common dilution method. The essential oil concentration fluctuated greatly during the period examined and established that the Satureja oils obtained during the flowering period were the most potent, exhibiting the lowest MIC values and retaining remarkable bactericidal properties. Thus, they represent an inexpensive source of natural antibacterial substances that exhibit potential for use in food systems to prevent the growth of foodborne bacteria and to extend the shelf life of the processed food.  相似文献

Fresh white asparagus spears were subjected to peeling or left unpeeled before storage at 10 °C for 7 days. CO2 and ethylene production rates were determined during storage, while color, toughness and DPPH radical scavenging activity as well as soluble solids, total soluble phenols and nitrate content were determined before and after storage in both peeled and unpeeled spears. Peel CO2 and ethylene production rates were also determined during storage. Spears subjected to peeling were more tender, and had lower lightness and chroma and higher hue angle values compared to the unpeeled ones. However, both peeled and unpeeled spears had similar DPPH radical scavenging activity, soluble solids, total soluble phenols and nitrate contents. Peeling resulted in increased CO2 and C2H4 production rates, but peel contributed to total increase of CO2 and C2H4 production. As the measured metabolic activity of the peeled asparagus spears was similar to that of the unpeeled, peeling did not adversely affect quality during storage.  相似文献

An increasing demand for cherry production (Prunus avium L.) in Greece led to the development of new high quality sweet cherry cultivars. Self-incompatibility in cherry is one of the most challenging issues for the species’ cultivation and top breeding priority. Τhe present study focuses on the development of new hybrids with improved traits such as productivity, fruit size, organoleptic characteristics and self-compatibility. For this purpose, thirty different cultivars were crossed and produced hybrids that were evaluated according to 34 morpho-physiological characteristics. The results were analyzed using the XLSTAT (version 2014.1) software and a dendrogram was constructed using the agglomerative hierarchical clustering method. Optimal hybrid clustering was achieved when characteristics of great economic importance such as fruit shape and size, growth habit and days to blooming were included in the analysis. Based on the results, new sweet cherry hybrids with the special character of self-compatibility were developed. Our findings provide crucial new information for sweet cherry future breeding programs and cultivation.  相似文献

Osteoarthritis belongs to the most common joint diseases in humans and animals and shows increased incidence in older patients. The bioactivities of collagen hydrolysates, sulfated glucosamine and a special fatty acid enriched dog-food were tested in a dog patient study of 52 dogs as potential therapeutic treatment options in early osteoarthritis. Biophysical, biochemical, cell biological and molecular modeling methods support that these well-defined substances may act as effective nutraceuticals. Importantly, the applied collagen hydrolysates as well as sulfated glucosamine residues from marine organisms were strongly supported by both an animal model and molecular modeling of intermolecular interactions. Molecular modeling of predicted interaction dynamics was evaluated for the receptor proteins MMP-3 and ADAMTS-5. These proteins play a prominent role in the maintenance of cartilage health as well as innate and adapted immunity. Nutraceutical data were generated in a veterinary clinical study focusing on mobility and agility. Specifically, key clinical parameter (MMP-3 and TIMP-1) were obtained from blood probes of German shepherd dogs with early osteoarthritis symptoms fed with collagen hydrolysates. Collagen hydrolysate, a chondroprotective food supplement was examined by high resolution NMR experiments. Molecular modeling simulations were used to further characterize the interaction potency of collagen fragments and glucosamines with protein receptor structures. Potential beneficial effects of collagen hydrolysates, sulfated glycans (i.e., sulfated glucosamine from crabs and mussels) and lipids, especially, eicosapentaenoic acid (extracted from fish oil) on biochemical and physiological processes are discussed here in the context of human and veterinary medicine.  相似文献
