ABSTRACT: The effects of the different catching methods (gillnet, longline, harpoon) on sensory, chemical (pH, total volatile base nitrogen, K -value) and microbiological (total viable count [TVC]) changes in pike perch Sander lucioperca stored in ice were investigated. The same soaking time was used for both gillnet and longline fishing. The catching method had considerable influence on the freshness quality of pike perch. The acceptable shelf life was 15 days for pike perch caught by gillnet, and 22 days for longline and harpoon. The initial concentrations of inosine monophosphate (2.4 μmol/g) in pike perch caught by gillnet were significantly lower ( P < 0.05) than longline (4.1 μmol/g), and especially by harpoon (16.7 μmol/g). However, the initial K -values for fish caught by harpoon were significantly ( P < 0.05) lower (24.36%) than fish caught by longline and gillnet (57.69%, 64.41%, respectively). The average K , Ki, G and H -values at rejection day in terms of sensory assessment were approximately 90, 98, 156 and 40%, respectively, for all catching methods during ice storage. However, TVC reached 7.0 log cfu/g after approximately 11 days of storage for fish caught by gillnet, 19 days for fish caught by longline and 8 days for fish caught by harpoon. The result of this study suggests that the best catching method for preserving the freshness of pike perch is longline, based on the data obtained from the sensory and microbiological analysis. 相似文献

ABSTRACT: Post-release movement and diel activity patterns of hatchery-reared and wild black-spot tuskfish were examined using ultrasonic telemetry. Five hatchery-reared and four wild fish were released in the sandy bottom of Urasoko Bay, Ishigaki Island, Okinawa, Japan, and monitored using automated monitoring receivers from November 2005 to February 2006. Both hatchery-reared and wild fish tended to stay near the release site for over two weeks, before leaving the release site. Both hatchery-reared and wild tuskfish showed diurnal rhythm intermittently; signals were recorded more frequently in the daytime and less frequently in the nighttime, suggesting that the fish of both origins were active during the day and inactive during the night. These findings indicate that the one-year-old hatchery-reared tuskfish have some consistent behavioral characteristics with those of the wild. 相似文献

ABSTRACT: This study was undertaken to describe and quantitatively analyze the pattern of development of gametes in the Japanese scallop Patinopecten yessoensis . Incorporation of 5-bromo-2'-deoxyuridine (BrdU) into gonial cells was used to quantitatively detect mitotically active gonial cells. Oogonia increased in number from November to December and decreased rapidly to March. A small number of oocytes was detected in November. Oocytes steadily increased in number and size up to March. The number of spermatogonia slightly increased from November to December, and increased markedly from January to March. Both ratios of BrdU-immunopositive gonial cells in the ovary and testis to gonial cells moderately increased from September to December. The ratio of BrdU-immunopositive spermatogonia to gonial cells drastically increased from January to February and kept an elevated level in March, whereas the oogonia started to disappear in January. The results suggest that the pattern of proliferation of gonial cells can be divided into two phases: (i) phase I, oogonia and spermatogonia slowly proliferate through the growing stage; and (ii) phase II, oogonia develop into oocytes and spermatogonia begin to proliferate rapidly through the mature and spawning stages. 本文研究了腌制鱼的水分活度(Aw值)变化及其与其他有关参数的关系。以鲳鱼作为试验原料,用混合腌渍法对鱼品进行不同时间的腌制,然后观察鱼体中水分活度、盐分含量和水分含量的变化情况,并进一步探索这些参数之间相互的变化关系。实验结果表明,鱼体中水分活度和水分含量的降低,盐分的增加均在开始腌制至18小时时为最快;18小时后变化缓慢;34小时后基本趋向平衡。实验表明,水分活度,盐分含量与水分含量之间存在简单的线性关系,因此在腌制过程中,测定出水分和盐分含量就可以计算出水分活度Aw值,从而达到控制鱼品质量的目的。此方法在提高鱼品的保藏效果上具有一定的实用价值。 相似文献

Epitope-based vaccines are always questioned for their cross-protection against the antigenically variable foot-and-mouth disease virus (FMDV). In this study, we proved the cross-protection effect of a multi-epitope vaccine incorporated with poly(I:C) against three topotypes of O type FMDV. A total of 45 naïve pigs were vaccinated with different doses of multi-epitope protein vaccine incorporated with poly(I:C). At 28 days post-vaccination, 45 vaccinated and 6 unvaccinated control pigs (two pigs for each group) were challenged with three topotypes of virulent O type FMDV, namely, O/Mya/98 (Southeast Asia topotype), O/HN/CHA/93 (Cathay topotype) and O/Tibet/CHA/99 (PanAsia topotype) strains. All unvaccinated pigs developed generalised FMD clinical signs. Results showed that all pigs (n = 15) conferred complete protection against the O/Mya/98 and O/HN/CHA/93 FMDV strains, 11 of which were protected against the O/Tibet/CHA/99 FMDV strain. The 50% protective dose values of the vaccine against the O/Mya/98, O/HN/CHA/93 and O/Tibet/CHA/99 FMDV strains were 15.59, 15.59 and 7.05, respectively. Contact challenge experiment showed that transmission occurred from the donors to the unvaccinated but not to vaccinated pigs. These results showed that vaccination with multi-epitope protein vaccine incorporated with poly(I:C) can efficiently prevent FMD in pigs. 相似文献

选用嗜水气单胞菌、副溶血弧菌、荧光假单胞菌三种常见水产病原菌,用于测定柠檬酸的抑菌效果及最小抑菌浓度,并研究了柠檬酸抑菌作用的稳定性。三种菌均为革兰氏阴性菌。试验结果表明,柠檬酸对三种菌都有不同程度的抑菌作用,并得到最小抑菌浓度,依次是:0.0011g/mL,0.0008g/mL,0.00125g/mL;在最小浓度下抑菌作用可达6d之久,柠檬酸抑菌效果有一定的稳定性。将Nisin溶于0.00125g/mL,得到复合液的抑菌效果强于柠檬裴单独作用,且稳定性增强。 相似文献
