A field experiment with separately tile-drained plots was used to study the ability of oilseed radish (Rhaphanus sativus L.), as a cover crop sown after harvest of a main crop of cereals or peas, to reduce nitrogen (N) and phosphorus (P) leaching losses from a clay loam in southern Sweden over 6 years. In addition to oilseed radish in pure stand, two cover crop mixtures (hairy vetch (Vicia villosa) and rye (Secale cereale) for 3 years and oilseed radish in mixture with buckwheat (Fagopyrum esculentum) for 2 years) were tested. The cover crop plots (three replicates per treatment) were compared with unplanted plots as a control. Plots cropped with oilseed radish during autumn (August–November) had significantly smaller yearly mean N concentration in drainage water over 5 of 6 years compared with unplanted controls. Mineral N content in the soil profile in autumn was significantly less in oilseed radish plots than for control plots in all years. The cover crop mixtures of hairy vetch and rye or buckwheat and oilseed radish also showed the potential to reduce soil mineral N in autumn and N concentration in drainage water, compared with unplanted controls. The cover crops had no impact on P leaching. In conclusion, oilseed radish has the ability to reduce leaching losses of N, without increasing the risk of P leaching.  相似文献

To investigate the incidence of co-colonization of different strains of Campylobacter species present in canine and feline stool samples, isolates were recovered by culture from 40 samples from dogs (n=34) and cats (n=6). Animals were of different ages, with diarrhoea or without clinical signs. Three isolation procedures were used: two selective agars and a filtration method. In each stool sample, multiple colonies were identified to the species level by PCR, subsequently genotyped by Amplified Fragment Length Polymorphism (AFLP) and pattern similarities (451 isolates) were calculated to investigate their phylogenetic relationships. Genetic heterogeneity of strains in individual stool samples was detected within the species Campylobacter jejuni, C. upsaliensis and C. helveticus, though to a different degree in dogs and cats. In 3 of the 34 (9%) canine samples, more than one genotype of the same Campylobacter species was present, while strain variation was detected in four of the six feline samples. The results show that preferably, multiple colonies should be analyzed in molecular epidemiological and aetiological studies.  相似文献

Objectives To study in horses (1) the relationship between cardiovascular variables and muscle perfusion during propofol–ketamine anaesthesia, (2) the physiological effects of a single intravenous (IV) detomidine injection, (3) the metabolic response of muscle to anaesthesia, and (4) the effects of propofol–ketamine infusion on respiratory function. Study design Prospective experimental study. Animals Seven standardbred trotters, 5–12 years old, 416–581 kg. Methods Anaesthesia was induced with intravenous (IV) guaifenesin and propofol (2 mg kg?1) and maintained with a continuous IV infusion of propofol (0.15 mg kg?1 minute?1) and ketamine (0.05 mg kg?1 minute?1) with horses positioned in left lateral recumbency. After 1 hour, detomidine (0.01 mg kg?1) was administered IV and 40–50 minutes later anaesthesia was discontinued. Cardiovascular and respiratory variables (heart rate, cardiac output, systemic and pulmonary artery blood pressures, respiratory rate, tidal volume, and inspiratory and expiratory O2 and CO2) and muscle temperature were measured at pre‐determined times. Peripheral perfusion was measured continuously in the gluteal muscles and skin using laser Doppler flowmetry (LDF). Muscle biopsy samples from the left and right gluteal muscles were analysed for glycogen, creatine phosphate, creatine, adenine nucleotides, inosine monophosphate and lactate. Arterial blood was analysed for PO2, PCO2, pH, oxygen saturation and HCO3. Mixed venous blood was analysed for PO2, PCO2, pH, oxygen saturation, HCO3, cortisol, lactate, uric acid, hypoxanthine, xanthine, creatine kinase, creatinine, aspartate aminotransferase, electrolytes, total protein, haemoglobin, haematocrit and white blood cell count. Results Circulatory function was preserved during propofol–ketamine anaesthesia. Detomidine caused profound hypertension and bradycardia and decreased cardiac output and muscle perfusion. Ten minutes after detomidine injection muscle perfusion had recovered to pre‐injection levels, although heart rate and cardiac output had not. No difference in indices of muscle metabolism was found between dependent and independent muscles. Anaerobic muscle metabolism, indicated by decreased muscle and creatine phosphate levels was evident after anaesthesia. Conclusion Muscle perfusion was closely related to cardiac output but not arterial blood pressure. Total intravenous anaesthesia with propofol–ketamine deserves further study despite its respiratory depression effects, as the combination preserves cardiovascular function. Decreases in high‐energy phosphate stores during recovery show that muscle is vulnerable after anaesthesia. Continued research is required to clarify the course of muscle metabolic events during recovery.  相似文献

Late season production of raspberries and blackberries has become more common due to increased consumer demand and new production techniques, but information is scarce regarding the effect on nutritional quality, especially during storage. Antioxidant levels in berries can be influenced by pre-harvest climate conditions (light intensity, day length and temperature) and post-harvest storage time. This study analysed the effect of these factors on the content of different antioxidants (vitamin C, ellagic acid, anthocyanins and total phenolics) in raspberries (Rubus idaeus L. cv. Polka) and blackberries (Rubus fructicosus L. cv. Loch Ness) during storage. The analyses were performed using high performance liquid chromatography (HPLC) with photodiode array detection. Ellagic acid content during storage ranged between 104 and 114 mg/100 g fresh weight in raspberries and 172 and 182 mg/100 g fresh weight in blackberries. Anthocyanin content increased during storage from 64 to 77 mg/100 g fresh weight in raspberries, but decreased from 199 to 162 mg/100 g fresh weight in blackberries. Vitamin C content in raspberries ranged during storage between 22 and 28 mg/100 g fresh weight, but in blackberries values were lower (10–12 mg/100 g fresh weight). Significant changes in total phenolics and sugars occurred during different storage periods (0–9 or 10 days). The results suggest that late season raspberries and blackberries maintain high quality in terms of antioxidant content.  相似文献

Postmortem changes in pork muscle protein phosphorylation in relation to the RN(-) genotype were investigated using one-dimensional gel electrophoresis and a phosphor specific staining. The phosphorylation levels of several protein bands were found to be affected by the RN(-) genotype and to change during postmortem development. Glycogen phosphorylase, phosphofructokinase, and pyruvate kinase were found in protein bands affected by the RN(-) genotype, and the phosphorylation profile indicates that part of the increased rate and extended pH decline of the RN(-) genotype could be a consequence of phosphorylation of these key enzymes during the postmortem metabolism. The results illustrate that the protein phosphorylation level of the muscle proteins could be interpreted as a global metabolic fingerprint containing information about the activity status of the enzymes in the postmortem metabolism.  相似文献

An homologous radioimmunoassay for brown trout vitellogenin (VTG) was developed. Intact VTG, isolated from juvenile brown trout by selective precipitation and anion exchange chromatography was labelled with Na125I, with 1,3,4,6-tetrachloro-3,6-diphenylglycoluril (Iodogen) as the oxidizing agent. Incorporation of Na125I into VTG was higher than 75% and there was little degradation of the labelled protein. Labelled VTG eluted at the same position as unlabelled, purified brown trout VTG when analyzed by gel filtration on Sepharose 6B. Antisera with high titers, i.e. 1250 000, against brown trout VTG were raised in rabbits. The sensitivity of the assay was 5 ng VTG/ml and the standard curve was linear between 10 and 100 ng VTG/ml. Plasma from maturing female brown trout, as well as estradiol-treated and untreated juvenile brown trout diluted parallel to the standard curve, while plasma from maturing female rainbow trout and estradiol-treated arctic charr diluted non-parallel to the standard curve for brown trout VTG. Purified rainbow trout VTG and plasma from maturing female rainbow trout diluted parallel to each other, but with lower sensitivity than for brown trout VTG. Determinations of protein-bound phosphorus in the plasma of estradiol-treated juvenile brown trout correlated well with the RIA determinations of VTG. Repeated freezing and thawing of plasma samples yielded up to a hundred-fold increase in the apparent VTG level, while storage of a plasma sample for one year at –20°C did not affect the VTG level as measured by RIA.  相似文献

The present study demonstrates that acceleratedphotoperiod advances ovulation in Atlantic salmon, and that exposure to cold water prior to spawning further advances and synchronizes this process while improving egg-survival. High water temperature inhibited both sperm release and ovulation, whereas a GnRHa treatment overrode this temperature effect in most individuals. A decrease in water temperature seemed to accelerate both ovulation and sperm release, and water temperature modulated the plasma 17,20βP profiles around ovulation and sperm release. The GnRHa treatment markedly increased the volume of strippable milt and the plasma 17,20βP levels in males.  相似文献
