Three regions of the classical swine fever virus (CSFV) genome that have been widely sequenced were compared with respect to their ability to discriminate between isolates and to segregate viruses into genetic groups. Sequence data-sets were assembled for 55 CSFVs comprising 150 nucleotides of the 5' non-translated region, 190 nucleotides of the E2 envelope glycoprotein gene and 409 nucleotides of the NS5B polymerase gene. Phylogenetic analysis of each data-set revealed similar groups and subgroups. For closely related viruses, the more variable or larger data-sets gave better discrimination, and the most reliable classification was obtained with sequence data from the NS5B region. No evidence was found for intertypic recombination between CSFVs. A larger data-set was also analysed comprising 190 nucleotides of E2 sequence from 100 CSFVs from different parts of the world, in order to assess the extent and global distribution of CSFV diversity. Additional groups of CSFV are evident from Asia and the nomenclature of Lowings et al. (1996) [Lowings, P., Ibata, G., Needham, J., Paton, D., 1996. J. Gen. Virol. 77, 1311-1321] needs to be updated to accommodate these. A tentative assignment, adapting rather than overturning the previous nomenclature divides CSF viruses into three groups with three or four subgroups: 1.1, 1.2, 1.3; 2.1, 2.2, 2.3; 3.1, 3.2, 3.3, 3.4. The expanding data-base of CSFV sequences should improve the prospects of disease tracing in the future, and provide a basis for a standardised approach to ensure that results from different laboratories are comparable. 相似文献

The reference strains of the 12 serotypes of Actinobacillus pleuropneumoniae express one or two of three different RTX exotoxins designated Apx I, Apx II and Apx III. The toxins are important virulence factors. In the present study, ELISAs with purified Apx I, Apx II and Apx III, respectively, as antigen were evaluated as candidates for serological diagnosis of Actinobacillus pleuropneumoniae infection in pigs. The pigs were inoculated with biotype 1, serotypes 1-12, and biotype 2, serotype 14, respectively. A strong humoral antibody response was seen to all the three antigens in most pigs irrespective of the serotype used for inoculation. However, titers to the exotoxins secreted by the serotype used for inoculation were generally highest. The results show that toxin proteins of Actinobacillus pleuropneumoniae are antigenically related and that a correlation between serotype and secretion of exotoxin is not revealed serologically in the ELISA test. 相似文献

A migration assay was used to separate a pyrantel-susceptible and -resistant isolate of the pig nematode, Oesophagostomum dentatum. The experiment had three steps. In the first step, LD(50) values for pyrantel of the two isolates in the assay were established. In the second step, susceptible and resistant worms were mixed in various proportions prior to exposure to a fixed concentration of pyrantel and thereafter assayed. The inhibition of migration showed to be linearly correlated with the proportion of resistant worms in a sample. In step three, this line was used as a standard curve to predict the number of resistant worms in samples from pigs infected with mixed samples of susceptible and resistant larvae. 相似文献

Using specific ELISAs, antibody levels of four different isotypes to bovine respiratory syncytial virus (BRSV) were determined in calves, following experimental BRSV infection.Most calves experienced an increase in the specific IgM and IgG1 titres about 6-10 days after infection with BRSV. The IgM titre was transient showing positive titres for only 5-10 days, while specific IgG1 was present for a longer time. IgA was detected concomitantly with IgM but at a lower level. Production of IgG2 anti-BRSV antibodies was detected from 3 weeks after infection.In two closed herds, repeated blood samplings were performed on young stock to analyse maternal immunity. The passively transferred antibodies were mainly of the IgG1 isotype and the half-life of IgG1 to BRSV was estimated to be 26.6 days. One of the herds had an outbreak of enzootic pneumonia, diagnosed to be caused by BRSV. Furthermore, another herd with acute BRSV was followed by weekly blood samples in six calves; in both herds IgM and IgG1 was detected shortly after the appearance of clinical signs. Serum samples from 50 Danish dairy herds (453 samples) were tested for immunoglobulins of the isotypes IgG1, IgG2 and IgM. The presence of antibodies to BRSV was widespread and more than 54% of the samples had BRSV antibodies of both the IgG1 and IgG2 isotypes indicating a high herd prevalence to BRSV. The prevalence of degenerative joint disease (DJD) in the distal tarsal joints and the relation between radiographic and clinical signs compatible with the disease were estimated in a population of Icelandic horses used for riding. The material consisted of 614 horses age 6-12 years (mean age = 7.9 years). Radiographs with 3 projections of each tarsus were made and a clinical examination, including palpation of the medial aspect of the distal tarsus and motion evaluation of the hindlimbs before and after a flexion test of the tarsus, was performed. Radiographic signs of DJD in the distal tarsal joints were found in 30.3% of the horses and the prevalence was strongly correlated with age. Hindlimb lameness before and after flexion test and palpation abnormalities were significantly associated with the radiographic findings. The lameness was usually mild and, in most cases, detectable only after the flexion test. The prevalence of lameness was not significantly correlated with age. Lameness could not be predicted by details of the radiographic findings. 相似文献

The potential negative environmental impact of manure and biological waste (BW) can be minimised at a profit by recycling plant nutrients in the food chain. Current large-scale livestock production, epizootic diseases and increasing globalisation increase the need for biosecurity, to minimise the risk of disease transmission to the food chain. Arable use of manure and BW can inadvertently spread infectious diseases; opinion differs concerning the risk levels. To obtain general acceptance for arable use, a hygienically safe end-product is needed. This paper provides a detailed discussion of treatment alternatives for co-treatment, i.e., mixture before treatment of manure and BW. Composting, anaerobic digestion and ammonia treatment are the three options given. A decision support tool is also presented and discussed. Suitable treatment methods must combine biosecurity aspects with environmental, economic and nutrient recycling aspects to create a beneficial whole-farm approach. 相似文献

Disease resistance mediated by the resistance gene Xa21 is developmentally controlled in rice. We examined the relationship between Pathogenesis Related (PR) defense gene expression and Xa21-mediated developmental disease resistance induced by Xanthomonas oryzae pv. oryzae (Xoo). OsPR1a, OsPR1b, and OsPR1c genes were cloned and their induction was analyzed, in addition to the OsPR10a gene, at the juvenile and adult stages in response to a wildtype Xoo strain that induces a resistance response (incompatible interaction) and an isogenic mutant Xoo strain that does not (compatible interaction). We found that the adult stage leaves are more competent to express these OsPR1 genes and that the Xa21 locus is required for the highest levels of induction. 相似文献

ABSTRACT Avirulent isolates of Pyricularia oryzae and isolates of Bipolaris sorokiniana, a nonrice pathogen, were used to suppress rice blast caused by P. oryzae. In greenhouse experiments, both fungi substantially reduced leaf blast when applied 24 h or more before the pathogen. B. sorokiniana, but not avirulent isolates of P. oryzae, systemically reduced disease in leaf 5 when applied to whole plants at the four-leaf stage. In field experiments, both fungi were able to reduce neck blast significantly. No increase in grain yield was obtained by using avirulent isolates of P. oryzae, whereas five sprays with B. sorokiniana from seedling to heading stages increased the grain yield in two of three experiments conducted at two locations in Nepal. The significant increase in yield was observed under high inoculum pressure of P. oryzae. Induced resistance is suggested to be involved in the suppression of disease. 相似文献

To investigate the variations of active compounds between species, varieties and individuals of Valeriana cultivated under the same environmental condition, the contents of valerenic acid derivatives and valepotriates in rhizomes and roots of different plant material were analysed by a HPLC method. Different species or varieties of Valeriana yielded 11.65-0.15 mg/g of valerenic acid derivatives, and 1.81-0.03 mg/g of valepotriates. The variation between individuals of one commercial cultivar of V. officinalis ranged from 12.34 to 3.01 mg/g of valerenic acid derivatives, and 3.67-0.92 mg/g of valepotriates. Individuals from self-pollinated mother plants, normal or regenerated, showed a similar variation. The variation of micropropagated plants was much less than the seed propagated plants. The ratio of mean values between valerenic acid derivatives and valepotriates was similar in all groups (3 < ratio < 8) except one group of micropropagated plants (ratio > 20). 相似文献

Urinations of ruminants on grazed pastures increase the risk of nitrate leaching. The study investigated the effect of reducing the length of the grazing season on nitrate leaching from a coarse sandy, irrigated soil during 2006–2007 and 2007–2008. In both years, precipitation was above the long‐term mean. The experiment was initiated in a 4‐yr‐old grass‐clover sward in south Denmark. Three treatments were as follows grazing only (G), spring cut followed by grazing (CG) and both spring and autumn cuts with summer grazing (CGC). Nitrate leaching was calculated by extracting water isolates from 80 cm depth using ceramic suction cups. Because of considerable variation in measured nitrate concentrations, the 32 installed suction cups per treatment were insufficient to reveal differences between treatments. However, weighted nitrate leaching estimations for G, CG and CGC showed estimated mean nitrate N concentrations of 23, 19 and 13 mg/L for an estimated proportion area occupied by urine patches of 0.33, 0.26 and 0.16, respectively. Thus, N concentrations in G and CG exceeded the EU limit of 11.3 mg N/L. Under the prevailing conditions, the time of urination did not appear important. The estimated background leaching calculated from suction cups presumably not situated under urine patches resulted in mean nitrate N concentrations of 2.6 mg/L. 相似文献
