The aim of this work is to explore the suitability of the complementary use of mass spectra and the corresponding statistical analysis (principal components-Pareto analysis (PCA) and discriminant analysis (DA)) of these spectra to differentiate diverse humic samples as a function of their structural and conformational features. To this end, the mass spectra of humic samples belonging to the main humic fraction types (gray humic acid, brown humic acid, and fulvic acid) were obtained by electrospray ionization mass spectrometry (ESI-MS). The results obtained showed that the application of PCA yielded a clear separation between blanks and humic samples. However, a clear differentiation among the humic fraction types was not achieved. The DA of PCA data, however, yielded a clear separation among the humic substances (HS) samples belonging to each HS fraction type considered: gray humic acids, brown humic acids, and fulvic acids. These results showed that the mass spectra of each humic sample include characteristic mass/charge (m/z) distribution values that can be considered as a "fingerprint" representative of its specific structural features. Our results also indicate that, although the m/z values principally corresponded to single-charged ions, we cannot identify these molecular weight distributions with those of humic samples, since sample molecular fragmentation, as well as partial molecular ionization, cannot be ruled out under our experimental and instrumental conditions.  相似文献

Our previous studies of the native rhizobial population associated with peanut nodules in the Córdoba soils of Argentina revealed that this population is highly diverse and includes slow- and fast-growing isolates. The native fast-growing isolates NCHA22 and NET30 were selected on the basis of their plant growth promoting properties and their chromosomal genotypes were determined by 16S rDNA sequencing. NCHA22 and NET30 16S rDNA alleles were found to cluster with those of Rhizobium tropici group IIB and Rhizobium giardinii bv. giardinii strain H152, respectively. We have now characterized these isolates by analyzing the glnA and nifH genes to clarify their taxonomic position. These studies confirmed that fast-growing isolates belonging to species earlier described as bean symbionts were obtained from nodules of a leguminous plant that has been described as efficiently nodulated exclusively by slow-growing rhizobial strains.  相似文献

Current approaches for rapid assessment of carbon source utilization by whole soil communities (i.e., community-level physiological profiling or CLPP) provide a limited, biased view of microbial communities with little connection to in situ activities. We developed an alternative CLPP approach based upon fluorometric detection of dissolved oxygen consumption in a microtiter platform which offers flexible manipulation of experimental factors. In the attempt to reduce oxygen re-dissolution, the wells were filled with liquid to very near the top and sealed. We found that filling the wells with 240 vs. 150 μl of sample improved the sensitivity of the system to discern both the response to a substrate amendment as low as 10 mg l−1 and un-amended, endogenous respiration. The preparation of a soil slurry facilitates inoculation into the microplate. Disruption of soil samples had a limited effect on the endogenous respiration in comparison to intact soil microbags in a 24-well microplate. Storage time (up to 33 days) reduced the level of activity in intact soil microbags but not in disrupted samples. A microcosm fertilization experiment was set to study the effects of N availability on the respiratory response in the plates. The use of soil organic carbon (SOC) and amended C-substrates (50 mg l−1) was increased by the addition of nitrogen (N) in the plate, and appeared N-limited shortly after microcosm fertilization. The addition of the eukaryotic inhibitor cycloheximide delayed the initial increase in fluorescence (time to minimum response) of several C sources (casein, acetate, asparagine, coumaric acid), varying among soils, which could be explained by the fungal use of these compounds. However, the extent of the inhibition caused by cycloheximide did not increase at higher fungal to bacteria ratios as estimated by PLFA analysis, indicating that the direct estimation of the fungal biomass from cycloheximide addition is not feasible. This paper provides an optimized, standardized protocol for soil analysis, and sets the basis for further validation studies that will continue to define the underlying capabilities/biases of this approach.  相似文献

The aims of our study were to compare the effectiveness of poultry manure (PM) and banana waste (BW), with regard to their use as inoculant carriers of a bacterial consortium constituted by strains of Azospirillum, Azotobacter and P-solubiliser bacteria and to establish the most efficient dose of biofertilizer for a soil cultivated with banana (Musa paradisiaca AAA Simmonds), with respect to improving plant performance and soil physical and microbiological properties. Six months after planting, plant growth had increased with increase in dose of the biofertilizers applied. The biofertilizer prepared on BW enhanced the density of P-solubiliser bacteria, the concentrations of available P and foliar P to a greater extent than with the biofertilizer prepared on PM. The increases produced by the biofertilizer prepared on PM for the soil aggregate stability, enzymatic activities and the labile carbon fractions were highly correlated to the dose applied. Both biofertilizers can be considered potentially useful as inoculant carriers of PGPR but the usefulness of BW appears to be restricted to moderate doses of application (≤3%).  相似文献

This paper presents transformations of saturated hydrocarbons of petroleum type pollutants during ex situ bioremediation of soil on the pilot heap (halde), during a period of 6 months, within the grounds of Petroleum Refinery Pan?evo (Serbia). Samples for analysis were taken in time intervals of 2 weeks (P1–P12 samples). Organic substance was extracted by Soxhlet’s method and quantified. Isoprenoid aliphatics, in particular pristane and phytane, and polycyclic aliphatics of sterane and triterpane types in saturated hydrocarbon fractions were analysed by GC-MS (SIM method). Significant amounts of n-alkanes have not been detected. The MS-chromatogram revealed only marginal amounts of pristane and phytane in sample P1. Pristane and phytane occurred in sample P8, and in even higher quantities in the final sample P12. The proceeding bioremediation process was accompanied by the decrease of the relative amounts of pentacyclic terpanes of hopane type, compared to tri- and tetracyclic terpanes. In the initial sample P1 the distribution of steranes and hopanes follows a pattern, which is characteristic for crude oils. However, their identification by SIM method was not possible in samples P8 and P12 because of the reduced concentration. The observed changes in the alkane fractions’ compositions may be considered as atypical, referring to the fact that during oil biodegradation under natural conditions, decomposition of isoprenoids occurs much easier and faster than decomposition of polycyclic alkanes of tri-, tetra- and pentacyclic terpane, sterane and diasterane types, after the decomposition of n-alkanes has been almost completed.  相似文献

In order to investigate the Urias Coastal Lagoon (UCL) hydrodynamics, a vertically integrated semi-implicit, non-linear, finite difference model, has been applied. The flow dynamics in this model has been described by the depth integrated shallow water equations and has been forced by prescribed free surface elevations at the open boundary in the inlet of the lagoon. The predicted instantaneous tidal elevation and the vector field of tidal velocities, reflect reasonably well the flood and ebb conditions in the coastal lagoon. Maximum tidal velocities of 0.6 m/s at the navigation channel of the lagoon and tidal ranges of 1.2 m were predicted for spring tides. Residual current of 0.01–0.06 m/s have also been predicted. The advection-diffusion process of a hypothetical pollutant released at two discrete points in the UCL depended on the intensity of water circulation; sites with slow instantaneous tidal velocities and residual currents of small magnitude presented slow advection and diffusion of the pollutant and may be considered vulnerable to the contamination, specifically the head of the lagoon where the pollutant was difficult to be removed by the tidal currents. The main channel, where the tidal currents exceed 0.6 m/s and the residual currents reached 0.06 m/s, behaved as a natural conduct for the pollutant motion. The forces involved in water circulation within the channel would be the best driving mechanism to flush contaminants from the UCL into the Ocean.  相似文献

Coke Wastewater Treatment by a Three-Step Activated Sludge System   总被引:1,自引:0,他引:1  
The treatment of industrial coke wastewater was studied in a laboratory-scale activated sludge system. The concentrations of the main pollutants in the wastewater ranged between 800 and 1870 mg COD/l, 100–221 mg phenols/l, 198–427 mg SCN/l, 133–348 mg ${\text{NH}}_4^ + - {{\text{N}} \mathord{\left/ {\vphantom {{\text{N}} {\text{l}}}} \right. \kern-0em} {\text{l}}}$ and 11–41 mg CN?/l. To avoid inhibition phenomena resulting from the high concentrations of thiocyanate, ammonium nitrogen and cyanide a three-step process was implemented. The first step was anoxic for the removal of nitrates, followed by an oxic step during which biodegradation of phenols and thiocyanates took place, and by a second oxic step to oxidize ammonium nitrogen to nitrate. The dilution effect due to the recirculation of the final effluent to the head of the process and also, the separation of the nitrification step from the biodegradation of thiocyanate led to much higher efficiencies than when the process was carried out simultaneously. Very high removals were obtained (99% phenols, 97% SCN?, 63% COD, 98% ${\text{NH}}_4^ + - {\text{N}}$ , 90% total-N and 99% cyanide) employing hydraulic residence times of 15.4 h for denitrification, 98 h for phenol and thiocyanate biodegradation and 86 h for nitrification.  相似文献

Slope correction for LAI estimation from gap fraction measurements   总被引:1,自引:0,他引:1  
Digital hemispherical photography poses specific problems when deriving leaf area index (LAI) over sloping terrain. This study proposes a method to correct from the slope effect. It is based on simple geometrical considerations to account for the path length variation within the canopy for cameras pointing vertically. Simulations over sloping terrain show that gap fraction increases up-slope while decreasing down-slope. As a consequence of this balance between up- and down-slope effects, effective LAI estimates derived from inversion of the Poisson model are marginally affected for low to medium slopes (<25°) and LAI (LAI < 2). However, for larger slopes and LAI values, estimated LAI values may be strongly underestimated. The proposed correction was evaluated over four forested sites located over sloping terrain. Results indicate that in these conditions (LAI between 0.6 up to 3.0, clumped canopies with relatively erectophile leaf distribution), the effect of the slope (between 25° and 36°) was moderate as compared to other potential sources of problems when deriving LAI from gap fraction measurements, including clumping, leaf angle inclination and spatial sampling.  相似文献

A gnamma or weathering pit is a small-sized cavity sculpted into solid rock which refills with rainwater. This study describes the main hydrological processes in gnammas and the underlying chemical interactions with the environment. Four gnammas developed in a granite tor from central Spain were chosen for monitoring and studying the dynamics of the gnamma–water system. Parameters monitored were air and water temperature, pH, ORP (redox potential), and conductivity. These measurements describe the system dynamics, without the need for special sampling or other potential interferences under low water volumes. Additionally, chemical analyses were carried out to contrast the chemical properties with the chemical composition. As a consequence of water volumes less than 100 L, daily fluctuations tended to cause the largest amplitude changes. Photosynthesis and respiration/putrefaction processes in the gnamma are responsible for daily pH fluctuations up to 4 units, with pH values often greater than 9. ORP generally alternates from positive to negative values as the solution oscillates diurnally between basic and acidic conditions. Conductivity values ∼ 10 μS/cm occur most of the year, with only minor daily oscillations. During dry periods, conductivity increases significantly with maximum values > 400 μS/cm. These high conductivity values reflect atmospheric inputs by summer rainstorms after prolonged dry periods. Therefore, conductivity does not relate directly with chemical species dissolved from the host rock. Silica was the main weathering product of granite dissolution during the year, with highest concentrations (> 200 ppm) in the rainy season. Low Si values measured during the dry season are in accordance with observed precipitation of amorphous silica within the gnamma. Seasonal variations in concentration are also present in Al and Fe as a response to ORP variations and evaporative dynamics, causing mineral dissolution and new mineral precipitation during the year. Relations among the measured parameters are influenced by several factors, including biological processes, temperature effects and water level variations. Factors such as large pH oscillations, basic pH values, acids from biological activity, and alkali cations all increase silicate solubility, thus favoring granite dissolution. Overflow of water during the winter, when the water is chemically enriched, is an effective mechanism to remove weathering products.  相似文献

The maize of Latin America, with its enormous diversity, has played an important role in the development of modern maize cultivars of the American continent. Peruvian highland maize shows a high degree of variation stemming from its history of cultivation by Andean farmers. Multivariate statistical methods for classifying accessions have become powerful tools for classifying genetic resources conservation and the formation of core subsets. This study has two objectives: (1) to use a numerical classification strategy for classifying eight Peruvian highland races of maize based on six vegetative traits evaluated in two years and (2) to compare this classification with the existing racial classification. The numerical classification maintained the main structure of the eight races, but reclassified parts of the races into new groups (Gi). The new groups are more separated and well defined with a decreasing accession within group × environment interaction. Most of the accessions from G1 are from Cusco Gigante, all of the accessions from G3 (except one) are from Confite Morocho, and all of the accessions from G7 are from Chullpi. Group G2 has four accessions from Huayleño and four accessions from Paro, whereas G4 has four accessions from Huayleño and five accessions from San Geronimo. Group G5 has accessions from four races, and G6 and G8 formed small groups with two and one accession each, respectively. These groups can be used for forming core subsets for the purpose of germplasm enhancement and assembling gene pools for further breeding.  相似文献
