The recently developed “second generation” of Roundup Ready® soybean (RR2) cultivars commercially available for farmers in 2008 were promoted as higher yielding relative to the “first generation” RR cultivars (RR1). Previous studies showed that glyphosate reduced such yield components as photosynthesis, water absorption, nutrient uptake and symbiotic N2 fixation in RR soybean cultivars; however, no data are available regarding the glyphosate effects on these physiological factors in RR2 soybean. Thus, the objective of this research was to evaluate the nutrient accumulation and nodulation of both generations of RR soybeans at different rates of glyphosate applied at various growth stages. In general, increased glyphosate rates and late applications decreased the nutrient accumulation, nodulation, and shoot and root biomass in both RR1 and RR2. All macro- and micronutrients, with exception of N and K, accumulated more in RR1 than RR2. Although this result may be an individual cultivar characteristic, it suggests that the RR2 cultivar was also inefficient in nutrient uptake and translocation or was unable to rapidly recover from potential chelating effects of glyphosate. These studies suggest that applying glyphosate at early growth stages using the lowest glyphosate rate might have less damage on growth and productivity of RR soybeans.  相似文献

Recent reports of weed‐control failures after the use of glyphosate led to suspicion about the selection of resistant biotypes of Conyza at locations in west and north Paraná, Brazil. Plants were collected, identified as Conyza sumatrensis and subsequently evaluated for possible resistance to glyphosate in four stages of weed development. The experiments were carried out in a greenhouse by combining biotypes, stages of development and a range of glyphosate doses. All the suspected biotypes were collected from locations in Cascavel, Toledo, Assis Chateaubriand, Tupãssi and Campo Mourão with a history of glyphosate use in burndown and in glyphosate‐resistant soybean for at least the four previous years and were compared to a susceptible biotype (São Jorge do Ivaí) with no previous history of herbicide use. The doses of glyphosate ranged from 0 to 5760 g ae ha?1. The biotypes were considered as resistant if two combined criteria were present (resistance factor > 1 and the rate required to achieve 80% control is >720 g ha?1). The results provided evidence that there is a marked difference in the level of control of older plants and also confirmed the presence of some resistant biotypes. For applications at the first stage of development, two biotypes that were resistant to glyphosate were identified (Cascavel‐1 and Tupãssi‐6). For applications in the second stage of development, beyond the biotypes that were found in the first stage, three other biotypes were considered as resistant: Toledo‐5, Assis Chateaubriand‐7 and Floresta‐10. However, for applications at the third and fourth stages, all the biotypes were considered as resistant.  相似文献

Bacterial leaf blight of aroids is caused by a heterogeneous group of xanthomonads listed as Xanthomonas axonopodis pv. dieffenbachiae (Xad) on the EPPO A2 quarantine list. Recently, Xad strains were shown not to belong to X. axonopodis but to the species X. citri, X. phaseoli and X. euvesicatoria. Here, to verify the pathovar designation, 11 representative strains were tested for pathogenicity on six aroid genera. They had overlapping host ranges and only the strain isolated from Syngonium showed host specificity. The X. citri strains, isolated from various hosts, showed dissimilarity in virulence to the tested aroid genera. The X. phaseoli strains, isolated from Anthurium and Syngonium, were generally more virulent and, additionally, induced systemic infections. The X. euvesicatoria strains, isolated from Philodendron, were scored as not pathogenic on the tested aroids. Four representative strains were genome sequenced and showed a variable virulence‐associated gene content. Pathogenicity to aroids was correlated with the presence of three specific T3 effector genes and with a T6SS gene sequence. Together, the phylogenetic and pathogenic differentiation among Xad strains justifies the installation of three pathovar epithets for the pathogens on aroids: X. phaseoli pv. dieffenbachiae comb. nov. for the strains isolated from Anthurium; X. phaseoli pv. syngonii comb. nov. for the strain isolated from Syngonium; and X. citri pv. aracearum comb. nov. for the strains isolated from Aglaonema, Xanthosoma and Dieffenbachia. It is proposed that phytosanitary regulations for xanthomonads on aroids are restricted to these three pathovars.  相似文献

Genomic selection is a promising breeding methodology that could increase selection accuracy and intensity and reduce generation interval. As the cost of genotyping decreases, it will be important to optimize training populations for costly phenotypic experiments for many complex traits. The aim of this research was to evaluate different optimization strategies, by using historical data from the Norwegian oat breeding programme at Graminor. In this paper, we focus on the optimization criteria: genetic diversity, phenotypic variance and genetic similarity between the training and testing populations. The four training population strategies—prediction core, diversity core, phenotypic selection and random selection—were applied to an oat candidate population of 1124 lines. An independent testing population was used to calculate the mean prediction abilities for the traits days to heading and plant height. Moreover, the strategies were tested in three independent wheat populations. The results showed that prediction core was the most promising strategy to select training populations with high genetic similarity to the testing set, high genetic diversity, and high phenotypic variance, which resulted in higher prediction ability across population sizes and traits.  相似文献

The computer‐assisted sperm analysis (CASA) has become a standard laboratory tool. Although it contributes a lot to the objective sperm motility assessment, its measurements may be affected by many factors. The aim of the study was to evaluate the effect of chamber on boar semen CASA results. Totally, 100 extended (30 × 106 sperm/ml) boar semen samples were analysed by CASA. Each sample was evaluated using Makler, Leja 4 chamber 20 μm and conventional glass slide/coverslip chambers (MC, LC and GSC, respectively). The differences in values between MC and LC and between MC and GSC were significantly positive (higher values for MC compared with LC and GSC) for total motility, progressive, rapid movement, VCL, VSL, VAP, STR and hyperactive, thus indicating a systematic effect. Between LC and GSC, the differences in many parameters (non‐progressive, progressive, slow, LIN, STR, hyperactive) were evenly distributed around zero, while in all other parameters the differences were significantly positive (higher values for LC compared with GSC), except for medium movement. Based on the estimated intraclass correlation coefficients, the method agreement between MC and LC and between LC and GSC was overall moderate to good, depending on the parameter; nonetheless, it was poor between MC and GSC. The limits of agreement between methods can vary considerably depending on the parameter and should be considered when comparisons between CASA measurements of different andrology laboratories or studies have to be performed.  相似文献

Bacillus anthracis infecting cattle is usually identified based on the typical symptom: sudden death. Bacillus anthracis causing atypical symptoms may remain undiagnosed and represent a potential occupational health hazard for, that is veterinarians and producers, butchers and tanners. In the year 2004, one case of sudden death in a dairy farm in southern Finland was diagnosed as bovine anthrax. Four years later 2008, an atypical case of anthrax was diagnosed in the same holding. The bull was taken to the Production Animal Hospital of the Faculty of Veterinary Medicine, University of Helsinki because of fever, loss of appetite and a symmetrically swollen scrotal sac. Penicillin treatment cured the fever but not the swollen scrotum. Before the intended therapeutic castration, a punctuate consisting of 10 ml fluid collected into a syringe from the scrotal sac was cultivated on blood agar at 37°C. After 24 hr, an almost pure culture of a completely non‐hemolytic Bacillus cereus‐like bacteria was obtained. The strain was identified as B. anthracis using Ba‐specific primers by the Finnish Food Safety Authority (RUOKAVIRASTO). After the diagnosis, the bull was euthanized and destroyed, the personnel were treated with prophylactic antibiotics and the clinic was disinfected. In this particular case, treatment with water, Virkon S and lime seemed to be effective to eliminate endospores and vegetative cells since no relapses of anthrax have occurred in 10 years. This case is the last reported anthrax case in Finland.  相似文献

Over the last four decades, plastination has been one of the best processes of preservation for organic tissue. In this process, water and lipids in biological tissues are replaced by polymers (silicone, epoxy, polyester) which are hardened, resulting in dry, odourless and durable specimens. Nowadays, after more than 40 years of its development, plastination is applied in more than 400 departments of anatomy, pathology, forensic sciences and biology all over the world. The most known polymers used in plastination are silicone (S10), epoxy (E12) and polyester (P40). The key element in plastination is the impregnation stage, and therefore depending on the polymer that is used, the optical quality of specimens differs. The S10 silicone technique is the most common technique used in plastination. Specimens can be used, especially in teaching, as they are easy to handle and display a realistic topography. Plastinated silicone specimens are used for displaying whole bodies, or body parts for exhibition. Transparent tissue sections, with a thickness between 1 and 4 mm, are usually produced by using epoxy (E12) or polyester (P40) polymer. These sections can be used to study both macroscopic and microscopic structures. Compared with the usual methods of dissection or corrosion, plastinated slices have the advantage of not destroying or altering the spatial relationships of structures. Plastination can be used as a teaching and research tool. Besides the teaching and scientific sector, plastination becomes a common resource for exhibitions, as worldwide more and more exhibitions use plastinated specimens.  相似文献

Previous greenhouse studies have demonstrated that photosynthesis in some cultivars of first‐ (GR1) and second‐generation (GR2) glyphosate‐resistant soybean was reduced by glyphosate. The reduction in photosynthesis that resulted from glyphosate might affect nutrient uptake and lead to lower plant biomass production and ultimately reduced grain yield. Therefore, a field study was conducted to determine if glyphosate‐induced damage to soybean (Glycine max L. Merr. cv. Asgrow AG3539) plants observed under controlled greenhouse conditions might occur in the field environment. The present study evaluated photosynthetic rate, nutrient accumulation, nodulation, and biomass production of GR2 soybean receiving different rates of glyphosate (0, 800, 1200, 2400 g a.e. ha–1) applied at V2, V4, and V6 growth stages. In general, plant damage observed in the field study was similar to that in previous greenhouse studies. Increasing glyphosate rates and applications at later growth stages decreased nutrient accumulation, nodulation, leaf area, and shoot biomass production. Thus, to reduce potential undesirable effects of glyphosate on plant growth, application of the lowest glyphosate rate for weed‐control efficacy at early growth stages (V2 to V4) is suggested as an advantageous practice within current weed control in GR soybean for optimal crop productivity.  相似文献

The susceptibility of eleven Prunus rootstocks to iron (Fe) deficiency was studied in hydroponics by growing them with 20 μM Fe, 0 μM Fe or 3 μM Fe+10 mM sodium bicarbonate. Based on the intensity of leaf chlorosis, the peach-almonds PR 204/84, Stylianidis K and KID2, produced at the Pomology Institute of Naoussa (Greece), showed the same or even greater tolerance than GF 677, the Greek peach-almond Retsou x Nemaguard, the plum-almond Myrandier 617 and the peaches GF 305, IDS 37, Greek wild peach seedling the greatest susceptibility whereas the plums St. Julien GF655/2 and Myrobalan 29C intermediate. Rootstocks without Fe presented significantly lower nitrogen and Fe whereas with bicarbonate significantly lower nitrogen, phosphorus, Fe and zinc. Root ferric chelate reductase activity was significantly increased in ?Fe rootstocks but negatively correlated with their tolerance; physiological and morphological changes were observed along a zone of a few centimeters length, 1–2 mm behind the root tip.  相似文献
