To assess the effects of a single supraphysiological postnatal administration of a progestogen on uterine glands in dogs, 10 females were randomly assigned to a medroxyprogesterone acetate 35 mg (MPA; n = 6) or placebo (n = 4) group within the first 24 h of birth. The safety of the treatment was also evaluated. A transient mild clitoris enlargement appeared in MPA-treated females. Microscopic postpubertal uterine assessment revealed the presence of uterine glands in all cases without significant differences in the area occupied by the glands per µm2 of endometrium nor in the height of the uterine epithelium. 相似文献

This study evaluated the bioenergetic map of mitochondria metabolism in cryopreserved bovine sperm. The detected oligomycin-sensitive basal respiration supported ATP production; frozen–thawed spermatozoa were found to have a coupling efficiency higher than 0.80. Cell respiration, however, was not stimulated by the protonophoric action of FCCP, as its titration with 1, 2, 4 and 6 μM did not stimulate the uncoupling activity on oxidative phosphorylation as highlighted by unresponsive oxygen consumption. The unusual effect on the stimulation of maximal respiration was not related to fibronectin- or PDL-coated plates used for cellular metabolism analysis. Conversely, irradiation of frozen–thawed bovine sperm with the red light improved mitochondrial parameters. In effect, the maximal respiration of red-light-stimulated sperm in PDL-coated plates was higher than the non-irradiated. In spite of this, red-light irradiation had no impact on membrane integrity and mitochondrial activity evaluated by epifluorescence microscopy. 相似文献

Maternal periconceptional undernutrition is associated with altered development and increased risks of adverse outcomes in the offspring. The aim of this work was to determine the effect of periconceptional undernutrition on behavioural and reproductive aspects of the offspring. Fifty ewes were synchronized in oestrus (day 0) and allocated to two groups (n = 25) to be fed diets that provided 1.5 (C) or 0.5 (L) times the requirements for maintenance until day 15. Ewes were mated and fed the control diet from day 16 until lambing. Two months after lambing, 26 lambs were exposed to tests to determine their cognitive/emotional responses. Six ewe lambs were euthanized and in vitro oocyte maturation and fertilization procedures performed. The experimental diets produced no changes of mean live weight (LW) of C ewes, L ewes presenting a reduction in their initial LW with significant differences at day 15, in comparison with C ewes (p < 0.05). L ewes experienced a significant reduction in their body condition (BC) in comparison with C ewes (p < 0.05). Fourteen days after the onset of the experimental diets, mean LW and BC of L ewes was significantly lower than those of C ewes (p < 0.05). Undernourished ewes presented a trend to a reduction of prolificacy and fecundity (p < 0.10) in comparison with C ewes. Emotional and cognitive test revealed a similar response between groups. Ewe lambs from the undernourished ewes presented a population of oocytes 1.7 times higher than ovaries from control ewe lambs (66.0 ± 0.73 vs. 113.7 ± 15.6 oocytes; p < 0.05) and had more oocytes in the ‘good’ (p < 0.05) and ‘healthy’ (p < 0.05) categories. In conclusion, a low plane of nutrition around conception significantly increases quantity and quality of the oocyte population of 60‐day‐old female descendants. Modifications of the cognitive and emotional responses of the progeny have not been evidenced. 相似文献

The Périgord black truffle (Tuber melanosporum Vittad.) can mycorrhize with rockroses (Cistus L.) in Mediterranean forests. These shrubs may be of interest for truffle cultivation as they produce early carpophores,
collaborate in tree mycorrhization and in the survival of Tuber melanosporum mycelium. However, there are very few quantitative studies on truffle production associated with Cistus. For this reason, we have carried out a study on the natural production of Tuber melanosporum associated with Cistus laurifolius L. in central Spain. An Anova test confirmed the existence of significant differences in carpophore production in four habitats,
with and without the presence of C. laurifolius. Twenty-one burns associated with C. laurifolius showed less significant production than the rest. We also confirmed that in 122 burns, Tuber melanosporum was always unequivocally associated with the root base of Quercus L. or Corylus L. trees. Similarly, five truffle-collectors confirmed that they had never found a single burn with carpophore production
associated exclusively with rockroses. It has also been statistically verified that many Cistus die in Tuber melanosporum burns, whereas in old burns with minimum carpophore production, a new colonization of young Cistus has been observed. This study also confirms that C. laurifolius mycorrhizes easily with Tuber melanosporum. We propose that Cistus laurifolius may act as transmitters of Tuber melanosporum, although they do not induce fruiting. Consequently, the commercial cultivation of Cistus laurifolius mycorrhized with Tuber melanosporum for production is not recommended in truffle culture. The results reported substantiate the negative observations in cultivations

In Mediterranean climates trees may go through two periods of dormancy, resulting in special anatomical features such as false rings and other intra-annual density fluctuations (IADFs). In this paper, ring growth and the presence of IADFs were studied in Pinus pinea L. growing in the coastal and inland regions of Alentejo (southern Portugal). In order to identify the triggering factors associated with the IADFs, a new classification was proposed for the IADFs in P. pinea: Type E (latewoodlike cells within earlywood); Type E+ (transition cells between earlywood and latewood); Type L (earlywoodlike cells within latewood) and Type L+ (earlywoodlike cells between latewood and earlywood of the next tree ring). Response function analyses showed that radial growth of P. pinea was strongly correlated with precipitation in southern Portugal. The climatic response of P. pinea was higher in the inland area where the summer drought is more severe, the winter temperatures are lower and the soils have low water-holding capacity, in comparison with the coastal area. IADFs were frequent in P. pinea and most of the IADFs were observed in latewood. The presence of IADFs was correlated with fluctuations in climate parameters during the growing season. The IADF type E+ was linked to precipitation events early in summer. The IADF type L and L+ were associated with above-average precipitation in early autumn. 相似文献

Summary The antinutritional factors found in soybean, lectin (SBL) and Kunitz trypsin inhibitor (SKTI), are usually inactivated by heat treatment. However, residual activity of these factors can be found in several types of soy-derived products. Heat treatment does not completely eliminate these factors, and in addition it may considerably decrease protein solubility. The genetic elimination of these antinutritional factors could be an alternative to the heat treatment. This study aimed to develop soybean lines devoid of SKTI and SBL in their seeds. The population under study was obtained by crossing the normal cv. Monarca with a soybean line lacking SKTI and SBL. Specific DNA primers were designed for the identification of the recessive alleles that condition the absence of SKTI and SBL. F2 seeds presenting the DNA markers that identify the recessive alleles were selected and the corresponding F2 plants were backcrossed with the recurrent parent (‘Monarca’), producing the BC1F1 generation. The F2 generation was analyzed by SDS-PAGE in order to confirm the genotypes of the F2 selected. The segregation tests confirmed that these traits are governed by two genes that segregate independently. 相似文献

Summary Frost tolerance of 30 barley (Hordeum vulgare L.) cultivars have been field evaluated in North Italy during the 1990/1991 winter season that was characterized by exceptionally low temperatures without snow cover. The results showed a significant correlation between cold injury and grain yield loss (r=0.61**). Five cultivars chosen for their varying degree of frost tolerance were further evaluated using laboratory tests. Measurements of survival rate and membrane damage were used to assess the influence of hardening on frost resistance. The reliability of the tests is shown by the high correlation to the field data. For both the laboratory temperature regimes and field conditions, the tested cultivars showed the same order of classification. The effect of a rise in temperature at the end of the hardening treatment on frost tolerance is also reported. The laboratory tests here proposed can be integrated in a breeding programme for improving frost tolerance in barley. 相似文献

Summary Molecular adaptation to cold and drought involves a series of biochemical and molecular changes leading plants to improve their winter hardiness or drought resistance.We are interested to study the molecular basis of cold acclimation and drought response of barley to survive under stress. Several genes regulated by low temperatures and sometimes by drought have been isolated from the barley genome. In this review the most significant results of our recent work will be presented and discussed.The protein encoded by cDNA clone pt59 and induced in barley by cold was over-expressed in E. coli to produce the matching antibody, which in vivo recognizes a cold-induced protein of 14 kDa (COR14). The COR14 is stored in amounts only slightly greater in the cold resistant Onice than in the susceptible Gitane, although the former has a higher induction-temperature threshold of COR14 than the latter. This fact is an evolutionary advantage enabling the resistant varieties in the field to prepare for the cold well ahead of the susceptible ones.Two other cDNA clones, paf93 and cdr29, are regulated by low temperature and drought stress but not by exogenous ABA treatment. Indeed during the early stage of dehydration, the mRNAs are expressed before the induction of known ABA regulated genes such as dehydrins and when only a small increase occurs in ABA content. The sequence analysis revealed that paf93 encodes for a protein homologous to the cold-regulated protein COR47 of Arabidopsis, whereas cdr29 represents a plant gene homologous to yeast and mammalian sequences coding for acyl-Coenzyme A oxidase.Abbreviations ABA
