Germanium(Ge) exhibited a strong inhibition on the growth of rice plants and necrotic spots appeared on leaf bladea which had absorbed 5 ppm Ge via their roots for 4 days. The translocating form of Ge in the exudate was found to be 68GeO2 which was the form given to the roots. Nearly equal radioactivities of the 68Ge in the shoots were extracted from the water-extractable, 0.25 N HCl-extractable and residual fractions. The radioactivities in the water extractable fraction were fractionated into 2 peaks by a molecular sieving effect using a Sephadex G-25 column. Some of the 68Ge in the higher molecular fraction was found to be associated with proteins. On the other hand, the 68Ge in the smaller molecular fraction was proved to be an unchanged form of the 68GeO2 which had been given to the roots.  相似文献

Although there have been some attempts to obtain intergeneric hybrids between Japanese pear (Pyrus pyrifolia Nakai) and apple (Malus × domestica Borkh.) mature hybrids have not been generated due to hybrid lethality. Shoots from immature hybrid embryos were gamma irradiated and cultured at normal temperature conditions to obtain viable intergeneric plants. Gamma irradiation doses of 50 and 100 Gy cultured at 25 °C killed all shoots but there was one survivor at 150 Gy and 25 °C. Hybrid status of the shoot was confirmed by using morphology, flow cytometry and SSR markers. The intergeneric plant had similar leaf serrations to apple and similar petioles to Japanese pear. Flow cytometric analysis revealed that the intergeneric shoot had a DNA content intermediate between the parents. SSR analysis showed one allele from each parent for all 16 SSR loci investigated. These data strongly suggest that the obtained shoot is a true hybrid between Japanese pear and apple.  相似文献

To examine the efficacy of juvenile salmon research as a tool for forecasting adult returns, the results from a study on the early marine life stage of juvenile chum salmon, conducted in the Nemuro Strait during 1999–2002 (i.e., 1998–2001 brood years), were compared with the return rates of adult salmon. Among the four brood years, the 2000 brood year (i.e., salmon migrating to the sea in 2001) was previously reported as showing higher abundance, higher growth rate and better somatic condition during the coastal residency period. Consequently, we expected it to have the highest return rate, under a hypothesis that juvenile survival in coastal residency regulates brood-year strength. Contrary to this expectation, the 2000 brood year had almost the lowest return rate. Alternatively, a statistical model in which sea surface temperature during the first year of marine life and size at release were utilized as explanatory variables reconstructed the actual variability in return rates more accurately than that based on the early marine life stage. Possible reasons for the discrepancy between the results of the juvenile salmon research and adult returns are discussed, and we suggest improvements for future research on juvenile salmon.  相似文献

We have discovered multiple acidic cysteine protease inhibitors, in addition to the known Eel-CPI-1, in the skin mucus extract of the Japanese eel Anguilla japonica by using the two-dimensional gel system of gelatin reverse zymography. Two of the acidic inhibitors, which we have named Eel-CPI-2 and Eel-CPI-3, were purified to homogeneity by anion exchange chromatography on a column of DEAE-Sepharose CL-6B, followed by fast protein liquid chromatography on Superdex 75 10/300 GL and HiTrap Q HP columns. The amino acid compositions of Eel-CPI-2 and Eel-CPI-3 were found to be almost identical and closely similar to that of the eel galectin AJL1. The molecular masses of Eel-CPI-2 and Eel-CPI-3 were elucidated to be 16,089.080 and 16,089.137 Da, respectively, by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. The apparent dissociation constant of Eel-CPI-2 and Eel-CPI-3 for cysteine protease papain was determined to be 1.79 × 10−7 and 1.05 × 10−7 M, respectively, by a quartz crystal microbalance technique.  相似文献

A male crossbred calf developed a limp and pain upon deep pressure on the right hind limb and the right forelimb. The radiographic findings of affected limbs and pathological findings of bone biopsy were similar to those observed in canine panosteitis. To clarify the lowering mechanism of loss tangen (tan) of sitka spruce (Picea sitchensis Carr.) wood impregnated with extractives of pernambuco (Guilandina echinata Spreng. synCaesalpinia echinata Lam.), we examined the vibrational properties of the impregnated wood in relation to the adsorbed water. The results obtained were as follows: (1) The equilibrium moisture content (EMC) of impregnated sitka spruce decreased to some extent compared with untreated wood. (2) Frequency dependencies of tan a about 400–8000Hz showed that impregnated wood has much lower tan than untreated wood at around 9% mois ture content (MC), except for the high-frequency region. At high relative humidity, impregnated wood has a minimum tan (at around 4000Hz); and at other frequency ranges the tan of impregnated wood did not differ considerably from that of untreated wood. (3) The apparent activation energy of the mechanical relaxation process (E) concerned with adsorbed water molecules was higher for impregnated specimens than for untreated ones at moderately high relative humidity, whereas at high relative humidity the difference was not observed. Based on these results, it is thought that the tan of impregnated wood decreased at low rela tive humidity because of the formation of direct hydrogen bonds between impregnated extractives and wood components. Five wood species were acetylated with acetic anhydride (AA) solution of glucose pentaacetate (GPA) at 120°C for 8h, and the effect of GPA on the dimensional stability of the acetylated wood was investigated. Some GPA was introduced into the wood cell wall during acetylation. The GPA remaining in the cell lumen penetrated the cell wall effectively after heating to more than 140°C for 10min. The bulking effects of GPA resulted in a 10%–30% increase in the anti-swelling efficiency of the acetylated wood with 20% GPA/AA solution in place of AA. Hydrophobic GPA did not deliquesce under highly humid conditions and it remained in the cell wall after boiling in water.Part of this paper was presented at the 51st Annual Meeting of the Japan Wood Research Society, Tokyo, April 1988  相似文献

Leukemia inhibitory factor (LIF) is essential for embryo implantation in mice and plays an important role in other mammals including humans. Intraperitoneal (i.p.) injections with anti-LIF antibody (7.5 μg/g body weight, 3 times) between D3 (D1 = day of vaginal plug detection) and D4 effectively blocked embryo implantation; complete inhibition was achieved in C57BL/6J mice, and implantation was dramatically reduced in ICR mice (reduced to 27%). Normal rabbit IgG used as the control did not disturb embryo implantation. Anti-LIF antibody was localized not only in the stroma, but also in the luminal epithelium and the glandular lumen after i.p. injections. Growth-arrested blastocysts were recovered from the uterus without any implantation sites in both strains. Blastocysts made contact with the LE on the antimesometrial side; however, uterine stromal cells did not undergo secondary decidual reaction, and the uterine lumen was open, even at D7. Several regions of decidualization in ICR mice treated with anti-LIF antibody were smaller than those of the control, and development of blastocysts was delayed. The expression of LIF-regulated genes, such as immune-responsive gene-1 and insulin-like growth factor binding protein-3, was significantly decreased in C57BL/6J mice treated with anti-LIF antibody compared with the control, but not in ICR mice. The present study demonstrated that simple ip injections of an antibody are sufficient to block one of the important factors involved in embryo implantation in mice, and this method should also be easily applicable to the investigation of other factors involved in implantation.  相似文献

The distribution and composition of extracellular matrix (ECM) of the spleen in two species of fruit bats, namely Cynopterus titthaecheilus and Rousettus leschenaultii, were examined by histochemistry and immunohistochemistry. Reticular fibres accompanied by laminin were identified to make up the splenic stromal network. Types I and III collagen were identified in various spleen compartments with varying intensities. Thin and short elastin fibres were scattered in several parts of the spleen. Visualization of the ECM of the spleen can better demonstrate spleen compartmentalization. The alleged vascular space structure in the fruit bats spleen was the sinus structure that was strengthened by the presence of reticular fibres that limit the sinus basement membrane. The present study identified periellipsoidal lymphoid sheath (PELS)-like structure in fruit bats spleen that had never been identified in mammals before. In addition to describing the structure, this study highlighted the variations in ECM composition of the spleen between species that can provide new insight into the phylogenetic study of spleen morphology.  相似文献
