Prions are infectious pathogens essentially composed of PrP(Sc), an abnormally folded form of the host-encoded prion protein PrP(C). Constrained steric interactions between PrP(Sc) and PrP(C) are thought to provide prions with species specificity and to control cross-species transmission into other host populations, including humans. We compared the ability of brain and lymphoid tissues from ovine and human PrP transgenic mice to replicate foreign, inefficiently transmitted prions. Lymphoid tissue was consistently more permissive than the brain to prions such as those causing chronic wasting disease and bovine spongiform encephalopathy. Furthermore, when the transmission barrier was overcome through strain shifting in the brain, a distinct agent propagated in the spleen, which retained the ability to infect the original host. Thus, prion cross-species transmission efficacy can exhibit a marked tissue dependence.  相似文献

The stratum corneum (SC) forms the main part of the permeability barrier of the skin. In mice and in humans, cutaneous barrier disruption can be generated by removing the SC with tape stripping (TS) and the skin barrier function can be assessed by measurement of transepidermal water loss (TEWL). The aim of the present study was to characterize the skin barrier restoration in the dog following mechanical disruption and to analyse the correlation between the skin barrier recovery and TEWL measurement. Thirty sequential TS were performed on 12 sites on four healthy beagle dogs. The number of TS was chosen to ensure a sufficient barrier disruption with a slow recovery. Skin repair was assessed for 72 h by clinical and histological examinations, and TEWL measurements. The results showed that performing 30 TS was adequate to disrupt the skin barrier in the dog. The homeostatic repair response, initiated in the skin, was rapid and characterized by complete restoration of the SC within 72 h, accompanied by greater basal cell proliferation, and dermal eosinophilic inflammation. TEWL was significantly increased by complete removal of the SC but recovered along with restoration of the SC (Scheffe test, P ≤ 0.05). Characterization of a canine model of barrier disruption and restoration and assessment of the skin barrier function by TEWL measurements could help better understand the events implied in skin barrier function. Development of this canine model is also necessary for future studies on the effects of treatments aimed at restoring the skin barrier.  相似文献

A spot-on formulation combining permethrin, dinotefuran and pyriproxyfen (Vectra 3D? spot-on solution for dogs - one 10-25kg pipette contains 196mg dinotefuran, 1429mg permethrin and 17mg pyriproxyfen) was evaluated in adult Beagle dogs in a study designed to measure its efficacy to control Aedes aegypti (anti-feeding effect and mortality effect). The trial was performed according to Animal Welfare and Good Clinical Practice. Twelve dogs (five males and seven female, >3 years old, weighing 8.8-13.0kg) were randomly allocated to treatment groups on pre-treatment mosquito counts: six dogs served as untreated controls, and six dogs were treated with the test formulation. Treatment consisted of applying a combination formulation to deliver at least 46.6mgkg(-1) permethrin, 6.40mgkg(-1) dinotefuran and 0.57mgkg(-1) pyriproxyfen. The combination is designed to control fleas, ticks, sand flies and mosquitoes. Each dog was infested with approximately 100 adult unfed A. aegypti once before treatment (day 6) then at 1, 7, 14, 21 and 28 days post-treatment. Counts and engorgement determination of dead and live mosquitoes were performed after 1h exposure period. In the treated group (group A), the repellency effect of the product based on engorgement status (anti-feeding effect), was 91.5%, 94%, 94.7%, 94% and 87% at 1, 7, 14, 21 and 28 days post-treatment. Mortality effect or insecticidal efficacy calculated at the end of the 1-h exposure was almost identical when calculated 24h after the 1-h exposure and remained above 93% until the end of the in-life phase. No adverse events were observed following treatment, including observations conducted 2, 4 and 24h after the last dog was treated.  相似文献

Ornithobacterium rhinotracheale (ORT) is an emerging respiratory pathogen of poultry in North America that is causing millions of dollars in economic losses to the poultry industry. Ornithobacterium rhinotracheale is associated with airsacculitis, pleuritis, pneumonia, and consolidation of lungs. Little is known about the molecular mechanisms of infection. In this study, the mechanism of iron acquisition by O. rhinotracheale was explored. O. rhinotracheale strains grown under iron deprivation in media containing 200 microM 2,2'-dipyridyl did not secrete siderophores as measured by the chrome azurol S (CAS) agar and CAS solution assays. Filter disks impregnated with various protein-bound iron compounds and inorganic iron salts of Fe(III) and Fe(II) placed on iron-restricted agar inoculated with a lawn of O. rhinotracheale supported growth from sheep and porcine hemoglobins, ovotransferrin, Fe(III), and Fe(II), but they did not support growth from bovine transferrin, bovine apo-transferrin, bovine lactoferrin, and hemin. However, both bovine hemoglobin and transferrin supported growth of O. rhinotracheale serotype C. Four immunoreactive proteins involved in iron acquisition were identified in an O. rhinotracheale membrane extract by using mass spectrometry. Furthermore, O. rhinotracheale field strains showed differential sensitivity to 2,2'-dipyridyl. Of the 72 field strains tested, 22 strains were resistant to the iron chelator at concentrations of 50 microM and 100 microM, suggesting this attribute may be related to disease-producing potential of these strains. This is the first report on the identification of the iron acquisition mechanism of O. rhinotracheale.  相似文献

Deliberate and malevolent use of plant pathogens, i.e. agroterrorism lato sensu (anticrop bioterrorism and use of bioweapons against the agricultural sector), may represent a non-negligible threat for crops and forests in Europe. In order to assess this risk objectively, a methodology based on a critical review of existing Pest Risk Analysis schemes has been elaborated and is described in the present paper. In this methodology, three interdependent steps are suggested: (i) definition of an a priori list of potentially dangerous plant pathogens for Europe, (ii) detailed elaboration and analysis of theoretical scenarios of possible acts of agroterrorism, and (iii) elaboration and use of a risk assessment scheme adapted to agroterrorism.  相似文献

Our research illustrates how a landscape mosaic changes in association with a mixed natural-anthropogenic disturbance history. Our study area is the Northwest Wisconsin (USA) Sand Plain (NWSP), a region with a rich disturbance history including fire, insects and clearcut forestry. We integrated historic airphotos from 1938, 1960, 1980 and 1998 within a GIS to describe change among four landcover classes describing a canopy-closure gradient: closed forests, woodlands, savannas and “open barrens”. Our work addresses two literature needs: empirical studies of mixed-disturbance landscapes, and nonforest habitats within a forest matrix. Our analysis shows that: the area of open barrens fluctuated, woodlands and savannas declined severely and closed forests increased through time. Falling median patch sizes and other landscape metrics suggest that the woodlands are becoming more fragmented. The landcover transitions driving this change vary according to time and place. The dominant transitions are toward closed forests from all classes, and transitions toward open barrens are also consistently important. The woodlands, savannas and open barrens habitats are mostly comprised of transient patches, persisting for less than 20 years. This contrasts with closed forests that often persist for 40 plus years. These changes are consistent with the disturbance regime that is shifting from fire- to forestry-dominance. Our results show a trend towards landscape simplification, manifest as losses of intermediate-density habitats (woodland and savanna) and shrinking patch sizes. The transient nature of the nonforest habitats shows that disturbance resulting in total or partial canopy removal will be vital for their conservation at a landscape scale.  相似文献

Theory predicts that local mate competition (LMC) favors the evolution of female-biased sex ratios. Empirical support of this prediction is indirect and comes from comparative studies or from studies showing that individuals can adjust their offspring sex ratio in response to varying LMC intensities. Replicate lines from a population of the spider mite Tetranychus urticae were selected under three LMC intensities for up to 54 generations. Within each selection regime, the final sex ratio matched theoretical predictions. Furthermore, the ability of individuals to adjust their offspring sex ratio diminished in females evolving under strict LMC, but not in females evolving under relaxed LMC levels. These results provide direct experimental evidence for the evolutionary process by which LMC modifies sex-allocation strategies and suggest that evolution under strict and constant LMC may lead to a loss of phenotypic plasticity.  相似文献

The generation of pluripotent stem cells from an individual patient would enable the large-scale production of the cell types affected by that patient's disease. These cells could in turn be used for disease modeling, drug discovery, and eventually autologous cell replacement therapies. Although recent studies have demonstrated the reprogramming of human fibroblasts to a pluripotent state, it remains unclear whether these induced pluripotent stem (iPS) cells can be produced directly from elderly patients with chronic disease. We have generated iPS cells from an 82-year-old woman diagnosed with a familial form of amyotrophic lateral sclerosis (ALS). These patient-specific iPS cells possess properties of embryonic stem cells and were successfully directed to differentiate into motor neurons, the cell type destroyed in ALS.  相似文献

Micrometer-sized bubbles are unstable and therefore difficult to make and store for substantial lengths of time. Short-term stabilization is achieved by the addition of amphiphilic molecules, which reduce the driving force for dissolution. When these molecules crystallize on the air/liquid interface, the lifetime of individual bubbles may extend over a few months. We demonstrated low gas-fraction dispersions with mean bubble radii of less than 1 micrometer and stability lasting more than a year. An insoluble, self-assembled surfactant layer covers the surface of the microbubbles, which can result in nanometer-scale hexagonal patterning that we explain with thermodynamic and molecular models. The elastic response of the interface arrests the shrinkage of the bubbles. Our study identifies a route to fabricate highly stable dispersions of microbubbles.  相似文献
