Some of the advantages of soilless production systems over conventional systems are higher yield, better pest control management and more efficient labor use. Among berries, strawberry responds quite well to soilless production systems. Major yield-affecting factors for strawberries in soilless systems may be listed as; growing medium, source and types of plantlets, density of plants and efficiency of fertigation system. The present research was conducted with strawberry plants (Fragaria x ananassa ‘Camarosa’) grown in four different media; (1) Cocopeat (C), (2) Peat (P) and (3 and 4) 1:1 mixtures with Pumice (Pu), to evaluate the effects of soilless media on marketable yield and root-shoot morpho-physiological parameters. Correlation measurements were made to assess if there is any interaction between yield and any tested morpho-physiological feature. The experiment was conducted in greenhouse, using frigo plantlets (11.6 plantlet/m2) planted in July and through 1 June of following year. Results showed that growth medium had an effect on yield, numbers of developed leaves, shoot-root dry mass, number and length of roots. Yield was correlated with these morphological features. Growth medium also had an influence on plant nutrient accumulations i.?e., shoot accumulation of nitrogen (N) and potassium (K) and root accumulation of phosphorus (P) and all the micro-nutrients that with exception of boron (B). There were some interactions between yield and plant nutrient content of tissues determined by correlation analyses. The most striking were negative correlations with yield and shoot N–K amounts and positive correlation with yield and root P amount. Some positive correlations were also found between yield and copper (Cu), manganese (Mn) and zinc (Zn) contents of both shoot and root tissues. Boron (B) contents was correlated only with the root tissues. The medium which had the best overall performance for soilless strawberry production was C?+?Pu and should be recommended to be used with frigo plantlets of ‘Camarosa’.  相似文献

This experiment was carried out to determine the percentage of internal connection between rootstock and scion in graft union with nondestructive and noninvasive MRI (Magnetic Resonance Imaging) method and to follow performance status according to internal connection ratios of the grafted rooted vines after planting to vineyard field. Research was established in a factorial randomized block design and carried out with cvs. Merlot and Syrah grafted onto 110?R rootstock and 4 different internal (MRI) connection levels in 3 replications. The percentages of grafted rooted vine internal determination by MRI by four sides (13.75%) in graft union were found to be very low than others at the pre-planting stage. An increase in the internal connection ratio in the majority of the grafted rooted vines was determined after the vegetative growth phase. Therefore, the internal connection ratios of the graft union of rooted vines of cvs. Merlot and Syrah varieties showed a tendency to increase during the second year of development. Again in both cultivars, loss rate of grafted rooted vines showed a decreasing tendency depending on increase of internal connection ratio of graft union. As a result, in case of decrease of MRI costs, it is thought that grafted rooted vine producers may have the opportunity to supply better quality seedlings to vine growers using MRI techniques.  相似文献

The objective of the joint project between the two universities of Bonn and Sarajevo was to study the effects of climate change associated with warmer winter and spring temperatures on fruit tree phenology. Changes in flowering date of apple and cherry were correlated with local weather data obtained concomitantly on-site at four locations in the Balkan countries Slovenia, Serbia, B & H and at Bonn, Germany.Phenological data of flower opening (F1; BBCH 61) of apple cv. ‘Golden Delicious’ originated from 55 years at Cacak (44?°N, Serbia) and at Klein-Altendorf, University of Bonn (50?°N), cv. ‘Bobovec’ from the University of Ljubljana (46?°N, Slovenia) and cherry cv. ‘Burlat’ (35 years at Klein-Altendorf, 50?°N) and 20–30 years at Tuzla (44?°N, B & H) and Mostar (43?°N, Bosnia & Herzegovina; B & H).The annual precipitation showed a negligible change in the last 55 years of ±5% in the Balkan countries (e.?g. Cacak, 699?mm), but only ±1.8% at Klein-Altendorf (605?mm) with a slight shift to more winter rainfall.Regression analysis showed an increase in mean temperature of 1.7?°C (annual) and 1.45?°C for the vegetation period at Klein-Altendorf (last 55 years), Germany. In the evaluated Balkan stations, the greatest increase in annual air temperature between two climate phases (until 1987 versus from 1988 until now) was at Ljubljana (1.4?°C) and Cacak (1.2?°C) in contrast to the least increase (0.3?°C) at Tuzla (annual average 10?°C) and an intermediate value (0.9?°C) in the warmer Mostar (annual average 14.5?°C).This recent temperature increase advanced flower opening by 10–14 days for apple such as cv. ‘Golden Delicious’ at Cacak and 7–10 days (last 55 years) at Bonn and 4–5 days (last 35 years) for the early cherry cv. ‘Burlat’ at Klein-Altendorf/Bonn; at Mostar and Tuzla, the effect of climate change was greater with the early cherry cv. ‘Burlat’ (26 days) compared with the difference of the late cherry cultivar (16 days). Before recent climate change (1973–1987), cherry flower was advanced by 10 days in the warmer med climate at Mostar but delayed by 16 days in the cooler Tuzla. The results, i.?e. the flower advancement in the Continental climate but flower delay in the med climate, are discussed with respect to the risk of late frost and countermeasures to combat climate change effects.  相似文献

Fruits of European plum (Prunus domestica L.) show high rates of transpiration. Consequently, they are highly perishable fruit. In the present study, the potential of an edible coating (Versasheen®-based with sorbitol as plasticizer) to improve storage life and keeping quality was evaluated on fruit of the plum cultivars ‘Jojo’ and ‘Tophit plus’. Plums were picked at commercial harvest date and stored at 2?±?0.5?°C (90?±?2% rh) for up to 28?d plus 2?d at 20?°C to simulate shelf life. Every seventh day, plums were analysed for transpiration, colour changes and laser light backscattering imaging (LLBI) at a wavelength of 785?nm to non-destructively investigate variations in quality properties of plums. Finally, fruit flesh firmness, soluble solids content, and titratable acidity were determined destructively. Results indicated that coating of fruit of both cultivars increased their surface resistance against water vapour transfer resulting in lower transpiration rates and thus lower mass losses compared to controls. Consequently, coating of fruit considerably delayed the decrease in flesh firmness. Moreover, edible coating retarded the increase of FWHM785 in coated plums. In this context, FWHM785 proved as a rapid and non-destructive tool to monitor changes in mechanical properties in heavily coloured plums. Results highlight that edible coatings successfully extend post-harvest life of plums.  相似文献

The aim of the present research was to investigate the fruit quality of twenty different local pear varieties (Pyrus communis L.), namely Ahlat, Ankara, Arpa, B?ld?rc?n, Çermai, Cinci, Gelin Bo?an, Hac? Hamza, Hah?r, Kabak, K?z?l, Kokulu, Mehrani, Menendi, Sulu, ?algam, Tokat Sultan?, Tur?u, Yaz, and Yaz Meyri?i, grown in Gumushane province in terms of pomological and morphological characteristics and chemical compositions. The fruit mass, fruit width and length, fruit stem thickness and length, fruit kernel width and length, hardness of pulp, number of seeds, leaf width and length, leaf stem length and thickness, and water soluble dry matter (WSDM) of the pear fruits have been determined as pomological and morphological characteristics. On the other hand, the chemical compositions of the pear varieties have been evaluated in terms of protein, ash, sucrose, fructose, glucose, total sugar, titratable acidity, moisture, and mineral element levels.Both pomological and morphological results demonstrated that the local pear varieties are important in terms of rehabilitation studies and detailed selection studies on these local varieties should be performed. The chemical analyses result of the pear varieties revealed that there is no component that may be harmful to human health when consumed, and also these varieties contains the necessary amount of mineral elements.  相似文献

The pear (Pyrus spp.) is one of the most important fruits consumed in daily life. The aim of this study was to determine the total phenolic and ascorbic acid contents, as well as the antimicrobial, antioxidant, and antimutagenic activities, of various pear cultivars. The fruits of five pear (Pyrus communis L.) cultivars (‘Deveci’, ‘Kizil’, ‘Egirsah’, ‘Gugum’, and ‘Banda’) were used in this study. It was determined that the peel and pulp of the ‘Kizil’ pear had the highest total phenolic content (402.5?mg/100?g and 215.2?mg/100?g, respectively), while those of the ‘Banda’ pear had the lowest total phenolic content (326?mg/100?g and 126.1?mg/100?g, respectively). Additionally, the ‘Kizil’ pear showed the highest antioxidant capacity in the 2,2’-azino-bis(3-ethylbenzothiazoline-6-6-sulphonic acid) (ABTS) and ferric-reducing ability assay (FRAP) (1.72 μmol TE/g FW and 161.25?μmol Fe II/g FW, respectively) and the highest ascorbic acid content (16.02?mg/100?g). The ‘Banda’ pear showed the highest antibacterial activity against the test bacteria. However, none of the pear extracts displayed antifungal activity. While all doses of the ‘Kizil’, ‘Gugum’, and ‘Banda’ pear extracts used in this study, except 80?μL/plate, exhibited antimutagenic activities, only the lowest dose (10?μL/plate) of the ‘Deveci’ pear extract showed the antimutagenic activity against induced mutagenesis in the Salmonella typhimurium TA 98 strain. Consequently, the five Turkish pear cultivars used in this study exhibited different levels of antimicrobial, antioxidant, and antimutagenic activities.  相似文献

Background

Genome editing of monocot plants can be accomplished by using the components of the CRISPR/Cas9 (clustered regularly interspaced short palindromic repeat/CRISPR associated Cas9) technology specifically optimized for these types of plants. Here, we present the development of RNA-guided Cas9 system for simplex and multiplex genome editing in barley.

Results

We developed a set of customizable RNA-guided Cas9 binary vectors and sgRNA modules for simplex and multiplex editing in barley. To facilitate the design of RNA-guided Cas9 constructs, the pBract derived binary vectors were adapted to Gateway cloning and only one restriction enzyme was required for construction of the sgRNA. We designed a synthetic, codon optimized Cas9 gene containing the N terminal SV40 nuclear localization signal and the UBQ10 Arabidopsis 1st intron. Two different sgRNAs were constructed for simplex editing and one polycistronic tRNA-gRNA construct (PTG) for multiplex editing using an endogenous tRNA processing system. The RNA-guided Cas9 constructs were validated in transgenic barley plants produced by Agrobacterium-mediated transformation. The highest mutation rate was observed in simplex editing of the cytokinin oxidase/dehydrogenase HvCKX1 gene, where mutations at the hvckx1 locus were detected in 88% of the screened T0 plants. We also proved the efficacy of the PTG construct in the multiplex editing of two CKX genes by obtaining 9 plants (21% of all edited plants) with mutations induced in both HvCKX1 and HvCKX3. Analysis of the T1 lines revealed that mutations in the HvCKX1 gene were transmitted to the next generation of plants. Among 220 screened T1 plants we identified 85 heterozygous and 28 homozygous mutants, most of them bearing frameshift mutations in the HvCKX1 gene. We also observed independent segregation of mutations and the Cas9-sgRNA T-DNA insert in several T1 plants. Moreover, the knockout mutations of the Nud gene generated phenotype mutants with naked grains, and the phenotypic changes were identifiable in T0 plants.

Conclusions

We demonstrated the effectiveness of an optimized RNA-guided Cas9 system that can be used for generating homozygous knockout mutants in the progeny of transgenic barely plants. This is also the first report of successful multiplex editing in barley using a tRNA processing system.

Background

To investigate plant hybrid sterility, we studied interspecific hybrids of two cultivated rice species, Asian rice (Oryza sativa) and African rice (O. glaberrima). Male gametes of these hybrids display complete sterility owing to a dozen of hybrid sterility loci, termed HS loci, but this complicated genetic system remains poorly understood.

Results

Microspores from these interspecific hybrids form sterile pollen but are viable at the immature stage. Application of the anther culture (AC) method caused these immature microspores to induce callus. The segregation distortion of 11 among 13 known HS loci was assessed in the callus population. Using many individual calli, fine mapping of the HS loci was attempted based on heterozygotes produced from chromosome segment substitution lines (CSSLs). Transmission ratio distortion (TRD) from microspores was detected at 6 of 11 HS loci in the callus population. The fine mapping of S1 and S19 loci using CSSLs revealed precise distances of markers from the positions of HS loci exhibiting excessive TRD.

Conclusions

We demonstrated that AC to generate callus populations derived from immature microspores is a useful methodology for genetic study. The callus population facilitated detection of TRD at multiple HS loci and dramatically shortened the process for mapping hybrid sterility genes.
