The formation of heat-induced aggregates of kappa-casein and denatured whey proteins was investigated in milk-based dairy mixtures containing casein micelles and serum proteins in different ratios. Both soluble and micelle-bound aggregates were isolated from the mixtures heated at 95 degrees C for 10 min, using size exclusion chromatography. Quantitative analysis of the protein composition of the aggregates by reverse phase high-performance liquid chromatography strongly suggested that primary aggregates of beta-lactoglobulin and alpha-lactalbumin in a 3 to 1 ratio were involved as well as kappa-casein, and alpha(s2)-casein in micellar aggregates. The results gave evidence that heat-induced dissociation of micellar kappa-casein was implicated in the formation of the soluble aggregates and indicated that a significant amount of kappa-casein was left unreacted after heating. The average size of the aggregates was 3.5-5.5 million Da, depending on the available kappa-casein or the casein:whey protein ratio in the mixtures. The size and density of these aggregates relative to those of casein micelles were discussed.  相似文献

The formation of electrostatic complexes of whey protein (WP) and a nongelling carrageenan (CG) was investigated as a function of pH, ionic strength, temperature, and protein-to-polysaccharide (Pr:Ps) ratio. On lowering the pH, the formation of soluble WP/CG complexes was initiated at pH(c) and insoluble complexes at pH(phi), below which precipitation occurred. The values of the transition pH varied as a function of the ionic strength. It was shown that at [NaCl] = 45 mM, the value of pH(phi) was the highest, showing that the presence of monovalent ions was favorable to the formation of complexes by screening the residual negative charges of the CG. When CaCl(2) was added to the mixtures, complexes of WP/CG were formed up to pH 8 via calcium bridging. The electrostatic nature of the primary interaction was confirmed from the slight effect of temperature on the pH(phi). Increasing the Pr:Ps ratio led to an increase of the pH(phi) until a ratio of 30:1 (w/w), at which saturation of the CG chain seemed to be reached. The behavior of WP/CG complexes was investigated at a low Pr:Ps ratio, when the biopolymers were mixed directly at low pH. It resulted in an increase of the pH of the mixture, as compared to the initial pH of the separate WP and CG solutions. The pH increase was accompanied by a decrease in conductivity. The trapping of protons inside the complex probably resulted from a residual negative charge on the CG. If NaCl was present in the mixture, the complex took up the Na(+) ions instead of the H(+) ions.  相似文献

The enzymatic properties of Plumeria rubra latex have been evaluated for the first time, showing a high activity in both hydrolysis and synthesis reactions, and compared to the biocatalytic behavior of babaco (Vasconcellea x Heilbornii cv.) latex. Both biocatalysts have been optimized by studying the various parameters that influence reaction kinetics. The optimum temperatures for hydrolysis reactions were 50 and 55 degrees C for babaco and Plumeria, respectively. The optimum pH for babaco latex was 7, whereas for Plumeria latex, two optimal pH values (4 and 7) were observed. With regard to esterification and acyl transfer reactions such as alcoholysis and interesterification, the influence of thermodynamic water activity on reaction yields was determined and correlated with water sorption and desorption isotherms. When babaco latex is used as a biocatalyst, optimal synthesis reaction yields are obtained when the enzymatic extract is stabilized at a water activity value of 0.38, which corresponds to a water content of 5.7%. This optimal level of hydration is located on the linear portion of the biocatalyst's sorption isotherm, where the water molecules exhibit high-energy interactions with the protein network. In synthesis reactions (esterification, alcoholysis, and interesterification) biocatalyzed by Plumeria latex, correlation between best reaction yields and water activity cannot be done. Indeed, the sorption isotherm plot has an atypical shape, indicating that water might be trapped in the latex matrix and, consequently, that the water content of the biocatalyst is highly dependent on the hydration history of the latex.  相似文献

Changes in the acid gelation properties of skim milk as a result of variations in the micelle/serum distribution of the heat-induced whey protein/kappa-casein aggregates, induced by the combination of heat treatment and limited renneting, were investigated. No dramatic change in the zeta potential or the isoelectric point of the casein micelles was suggested, whether the aggregates were all attached to the casein micelle or not. Fluorescence intensity measurement using 8-anilino-1-naphthalenesulfonic acid (ANS) showed that the heat-induced aggregates were highly hydrophobic. Dynamic oscillation viscosimetry showed that acid gelation using glucono-delta-lactone (GDL) started at a higher pH value in prerenneted milk. However, no change in the gelation profile of skim milk could be related to the proportion of aggregates bound to the surface of the casein micelles. The results support the idea of an early interaction between the serum aggregates and the casein micelles on acidification.  相似文献

Though the ovine stifle is commonly used to study osteoarthritis, there is limited information about the age‐related morphometric changes of the tidemark. The objective of this study was to document the number of tidemarks in the stifle of research sheep without clinical signs of osteoarthritis and of various ages (n = 80). Articular cartilage of the medial and lateral tibial condyles and of the medial and lateral femoral condyles was assessed by histology: (a) to count the number of tidemark; and (b) to assess the OARSI (Osteoarthritis Research Society International) score for structural changes of cartilage. The number of tidemarks varied between anatomical regions, respectively, from 4.2 in the medial femoral condyle to 5.0 in the lateral tibial condyle. The axial part showed a significant higher number of tidemarks than the abaxial part, for all regions except the medial tibial condyle. Whilst the tidemark count strongly correlated with age (Spearman's correlation coefficient = 0.70; 95% confidence interval (95% CI): 0.67–0.73; p < 0.0001), the OARSI score was weakly correlated with age in our cohort of sheep (Spearman's correlation coefficient = 0.25; 95% CI: 0.19–0.30; p < 0.0001). Interestingly, no tidemark was seen in the three animals aged 6 months. Our data indicate that the number of tidemarks increases with age and vary with anatomical region. The regional variation also revealed a higher number of tidemarks in the tibia than in the femur. This could be attributed to the local variation in cartilage response to strain and to the difference in chondrocyte biology and density.  相似文献

A collection of 191 durum wheat accessions representing Mediterranean Basin genetic diversity was grown in nine different environments in four countries, with productivities ranging from 0.99 to 6.78 t ha−1. The population breeding structure comprised eight genetic subpopulations (GSPs) using data derived from 97 evenly distributed SSR markers. The phenotypic structure was assessed: (i) from the mean values of six agronomic traits across environments (multivariate), and (ii) from data representing each trait in each environment (univariate). Mean daily maximum temperature from emergence to heading was significantly (P < 0.05) and negatively associated to yield, accounting for 59% of yield variations. Significant but weak relationships were obtained between the genetic similarities among accessions and their overall agronomic performance (r = 0.15, P < 0.001), plant height (r = 0.12, P < 0.001), spike–peduncle length (r = 0.06, P < 0.01) and thousand kernel weight (r = 0.03, P < 0.05), suggesting a very low possibility of prediction of the agronomic performance based on random SSR markers. The percentage of variability (measured by sum of squares) explained by the environment varied between 76.3 and 98.5% depending on the trait, while that explained by genotypes ranged between 0.4 and 12.6%, and that explained by the GE interaction ranged from 1.1 to 12.5%. The clustering of the accessions based on multivariate phenotypic data offered the best explanation of genotypic differences, accounting for 30.3% (for yield) to 75.1% (for kernel weight) of the observed variation. The genotype × environment interaction was best explained by the phenotypic univariate clustering procedure, which explained from 28.5% (for kernel weight) to 74.9% (for days to heading) of variation. The only accessions that clustered both in the genetic dissimilarities tree and the tree obtained using Euclidean distances based on standardized phenotypic data across environments were those closely related to the CIMMYT hallmark founder ‘Altar 84’, the ICARDA accessions adapted to continental-dryland areas, and the landraces, suggesting that genetic proximity corresponded to agronomic performance in only a few cases.  相似文献

Nowadays, biofouling is responsible for enormous economic losses in the maritime sector, and its treatment with conventional antifouling paints is causing significant problems to the environment. Biomimetism and green chemistry approaches are very promising research strategies for the discovery of new antifouling compounds. This study focused on the red alga Sphaerococcus coronopifolius, which is known as a producer of bioactive secondary metabolites. Fifteen compounds, including bromosphaerol (1), were tested against key marine biofoulers (five marine bacteria and three microalgae) and two enzymes associated with the adhesion process in macroalgae and invertebrates. Each metabolite presented antifouling activity against at least one organism/enzyme. This investigation also revealed that two compounds, sphaerococcinol A (4) and 14R-hydroxy-13,14-dihydro-sphaerococcinol A (5), were the most potent compounds without toxicity towards oyster larvae used as non-target organisms. These compounds are of high potential as they are active towards key biofoulers and could be produced by a cultivable alga, a fact that is important from the green chemistry point of view.  相似文献
