Insect pests are a major constraint to cowpea production in Africa. Therefore the aim of this study was to identify cowpea material that exhibit greater pest resistance and grain yield for evaluation by farmers. About 45 cowpea genotypes were assayed for flavonoids and anthocyanins with the objective of quantifying the levels of these phenolics in seed extracts as markers for effective plant defence. The results revealed significant differences in the concentration of flavonoids and anthocyanins in seed extract. Farmer varieties such as Sanzie, Bensogla and Omondaw exhibited much higher levels of phenolics compared with improved genotypes like ITH98-46, TVu1509 and IT93K-452-1. When planted in the field in Ghana and Tanzania, the genotypes that had high concentrations of flavonoids and anthocyanins in seed extracts (e.g. Bensogla, Omondaw and IT86D-2075) showed relatively lower infestation by thrips, pod-sucking bugs, aphids and alcidodes. Providing minimum protection with insecticide spray further showed that, genotypes with low infestation by thrips and pod-sucking bugs produced more grain yield without spraying, an indication of their natural resistance to these insect pests. Furthermore, farmer-selected varieties such as Sanzie, Bensogla, and Omondaw produced more grain yield without protection than their improved counterparts. These results showed that the higher the concentration of flavonoids and anthocyanins in cowpea seed extracts, the lower the insect pest incidence on seedlings raised from those seeds. Correlation analysis further confirmed a direct relationship between high flavonoids/anthocyanins in seed extracts and enhanced insect pest.  相似文献

Even among forest specialists, species-specific responses to anthropogenic forest fragmentation may vary considerably. Some appear to be confined to forest interiors, and perceive a fragmented landscape as a mosaic of suitable fragments and hostile matrix. Others, however, are able to make use of matrix habitats and perceive the landscape in shades of grey rather than black-and-white. We analysed data of 42 Chiroxiphia caudata (Blue Manakin), 10 Pyriglena leucoptera (White-shouldered Fire-eye) and 19 Sclerurus scansor (Rufous-breasted Leaftosser) radio-tracked in the Atlantic Rainforest of Brazil between 2003 and 2005. We illustrate how habitat preferences may determine how species respond to or perceive the landscape structure. We compared available with used habitat to develop a species-specific preference index for each of six habitat classes. All three species preferred old forest, but relative use of other classes differed significantly. S. scansor perceived great contrast between old forest and matrix, whereas the other two species perceived greater habitat continuity. For conservation planning, our study offers three important messages: (1) some forest specialist species are able to persist in highly fragmented landscapes; (2) some forest species may be able to make use of different anthropogenic habitat types to various degrees; whereas (3) others are restricted to the remaining forest fragments. Our study suggests species most confined to forest interiors to be considered as potential umbrella species for landscape-scale conservation planning.  相似文献

A basic understanding of the relationships between key phenotypic characters of the shea butter tree is considered a crucial step to its genetic improvement. As such fruit samples of the shea butter tree, Vitellaria paradoxa, were collected from nine locations in July, 2006, to determine the fruit and nut characteristics of the species. The choice of the locations was informed by the need to have as broad a picture of the trait diversity as possible. The nine locations covered were Akwanga, Ilorin, Lokoja, Makurdi, Minna (in the Southern Guinea Savanna), Jalingo, Kachia (in the Northern Guinea Savanna), Kano and Yola (in the Sudan Savanna), recognized as the main shea butter tree belt in Nigeria. Analysis of variance indicated significant (P < 0.05) variations in fruit and nut traits across agro-ecologies and specific locations (representing individual accessions). The Northern Guinea Savanna accessions had higher values for nut weight, nut length kernel weight and fruit weight. The Southern Guinea Savanna fruits were, however, superior in percent pulp weight. Nuts sourced from Akwanga were comparatively heavier and longer, had wider diameters, heavier kernels and higher percent testa weights which did not differ from those of Minna. Also fruits from Akwanga had significantly (P < 0.05) higher values for fruit length, fruit diameter, fruit weight and pulp weight. The Lokoja accession was superior to the rest in percent pulp weight, but it recorded least nut weight and nut length values. Nut shape index varied from 2.3 (Makurdi) to 1.5 (Jalingo). Nuts from Akwanga, Jalingo and Kachia had wide variation in nut weight in contrast to those from Lokoja, Makurdi and Yola (that were relatively similar). Fruit length had significantly (P < 0.05) positive relationship with pulp weight, fruit weight and fruit diameter. Nut diameter, however, had a negative relationship with percent pulp weight. A strong influence of some environmental variables on certain phenotypic traits was detected. Principal components analysis indicated nut weight, nut shape index, percent pulp weight and fruit shape index as the key discriminant variables for grouping shea butter fruit and nut in Nigeria. Results of this study indicated considerable diversity in fruit and nut traits of the shea butter tree in Nigeria, suggesting the possibility of selection for desirable traits.  相似文献

This study reports 3 cases of spontaneous papillomavirus infection in 1-week-old calves. Thickening of the omasum and abomasum wall, with acute inflammation, necrosis, ulceration, and neoplastic changes were seen in 1 calf. In the other 2, small papillomas were observed in the omasal mucosa, exhibiting proliferation of the parakeratinized epithelium. Papillomavirus antigens were detected by immunohistochemistry and virus-like particles were seen through electron microscopy.  相似文献

The intent to solve biological and biomedical questions in high-throughput led to an immense interest in microarray technologies. Nowadays, DNA microarrays are routinely used to screen for oligonucleotide interactions within a large variety of potential interaction partners. To study interactions on the protein level with the same efficiency, protein and peptide microarrays offer similar advantages, but their production is more demanding. A new technology to produce peptide microarrays with a laser printer provides access to affordable and highly complex peptide microarrays. Such a peptide microarray can contain up to 775 peptide spots per cm2, whereby the position of each peptide spot and, thus, the amino acid sequence of the corresponding peptide, is exactly known. Compared to other techniques, such as the SPOT synthesis, more features per cm2 at lower costs can be synthesized which paves the way for laser printed peptide microarrays to take on roles as efficient and affordable biomedical sensors. Here, we describe the laser printer-based synthesis of peptide microarrays and focus on an application involving the blood sera of tetanus immunized individuals, indicating the potential of peptide arrays to sense immune responses.  相似文献

Genetic variation in cucumber (Cucumis sativus L. var. sativus) accessions from China was assessed by examining variation at 21 polymorphic isozyme loci. One hundred and forty-six Chinese accessions acquired by the U.S. National Plant Germplasm System (NPGS) before 1994 were compared to 67 recently collected Chinese accessions. Of the 67, 39 were collected during a 1994 U.S.–China expedition and 28 were donated in 1996. Isozymic profiles of all of these Chinese accessions were also compared with 853 previously examined NPGS cucumber accessions. Principal component analysis of allele frequencies identified two distinct groups of Chinese accessions: one containing accessions collected in 1994 (Group 1) and the other containing accessions received by NPGS in 1996 (Group 2) (P<0.01). Variation at Gpi, Gr, Mdh-2, Mpi-2, Pep-gl and Pep-la was important in the detection of this difference. Isozymic variation in U.S. NPGS accessions of Chinese origin acquired before 1994 differed significantly (P<0.01) from those collected during 1994 and those received in 1996. Eleven loci (Fdp-1, Gpi, Gr, Idh, Mpi-2, Pep-gl, Pep-la, Pep-pap, Per, Pgm and Pgd-1), were important in the detection of this difference. When Chinese accessions taken collectively (i.e., those acquired before 1994, and those during 1994 and 1996) were compared with an array of 853 C. s. var. sativus accessions examined previously, relationships between accessions grouped by country or subcontinent differed from those found in the previous work. Accessions from China and India were distinct from each other and from all other groupings and the C. s. var. hardwickii (feral form of C. s. var. sativus) accessions examined. Thus, Chinese and Indian accessions represent the most diverse genetic variation present in the NPGS cucumber collection. Further collection in India and China would be strategically important for increasing genetic diversity of the U.S. NPGS cucumber collection.  相似文献

Different semi‐natural habitats occur on farmland, and it is the vegetation's traits and structure that subsequently determine their ability to support natural enemies and their associated contribution to conservation biocontrol. New habitats can be created and existing ones improved with agri‐environment scheme funding in all EU member states. Understanding the contribution of each habitat type can aid the development of conservation control strategies. Here we review the extent to which the predominant habitat types in Europe support natural enemies, whether this results in enhanced natural enemy densities in the adjacent crop and whether this leads to reduced pest densities. Considerable variation exists in the available information for the different habitat types and trophic levels. Natural enemies within each habitat were the most studied, with less information on whether they were enhanced in adjacent fields, while their impact on pests was rarely investigated. Most information was available for woody and herbaceous linear habitats, yet not for woodland which can be the most common semi‐natural habitat in many regions. While the management and design of habitats offer potential to stimulate conservation biocontrol, we also identified knowledge gaps. A better understanding of the relationship between resource availability and arthropod communities across habitat types, the spatiotemporal distribution of resources in the landscape and interactions with other factors that play a role in pest regulation could contribute to an informed management of semi‐natural habitats for biocontrol. © 2016 Society of Chemical Industry  相似文献

Breeding soybeans for higher latitudes requires cultivars with an increased chilling stress tolerance, especially when flowering occurs. Phenotyping in climate chambers to select for this trait is labour‐intensive and requires an optimal allocation of resources due to limited space. We screened a diversity panel of 35 early maturity cultivars and a biparental population of 103 RILs for their cold stress tolerance at flowering stage. Pod number under control and stress conditions is highly heritable and showed only a weak correlation between the two treatments. Based on different testing scenarios, we could show that testing more genotypes with less replicates yields much higher responses to selection and hence should be pursued in such climate‐controlled experiments. We identified quantitative trait loci (QTL) for pod number under both conditions (chromosomes 7 and 13) and a cold tolerance‐specific QTL (chromosome 11). Furthermore, we performed genomic predictions using different test set scenarios and prediction models, showing that genomic prediction is a promising tool to select for cold stress tolerance, particularly if known QTL can be used as fixed effects in the model.  相似文献
