In an attempt to improve post‐harvest skin colour in cultured Australian snapper Pagrus auratus, a two‐factor experiment was carried out to investigate the effects of a short‐term change in cage colour before harvest, followed by immersion in K+‐enriched solutions of different concentrations. Snapper supplemented with 39 mg unesterified astaxanthin kg?1 for 50 days were transferred to black (for 1 day) or white cages (for 1 or 7 days) before euthanasia by immersing fish in seawater ice slurries supplemented with 0, 150, 300, 450 or 600 mmol L?1 K+ for 1 h. Each treatment was replicated with five snapper (mean weight=838 g) held individually within 0.2 m3 cages. L*, a* and b* skin colour values of all fish were measured after removal from K+ solutions at 0, 3, 6, 12, 24 and 48 h. After immersion in K+ solutions, fish were stored on ice. Both cage colour and K+ concentration significantly affected post‐harvest skin colour (P<0.05), and there was no interaction between these factors at any of the measurement times (P>0.05). Conditioning dark‐coloured snapper in white surroundings for 1 day was sufficient to significantly improve skin lightness (L*) after death. Although there was no difference between skin lightness values for fish held for either 1 or 7 days in white cages at measurement times up to 12 h, fish held in white cages for 7 days had significantly higher L* values (i.e. they were lighter) after 24 and 48 h of storage on ice than those held only in white cages for 1 day. K+ treatment also affected (improved) skin lightness post harvest although not until 24 and 48 h after removal of fish from solutions. Before this time, K+ treatment had no effect on skin lightness. Snapper killed by seawater ice slurry darkened (lower L*) markedly during the first 3 h of storage in contrast with all K+ treatments that prevented darkening. After 24 and 48 h of storage on ice, fish exposed to 450 and 600 mmol L?1 K+ were significantly lighter than fish from seawater ice slurries. In addition, skin redness (a*) and yellowness (b*) were strongly dependent on K+ concentration. The initial decline in response to K+ was overcome by a return of a* and b* values with time, most likely instigated by a redispersal of erythrosomes in skin erythrophores. Fish killed with 0 mmol L?1 K+ maintained the highest a* and b* values after death, but were associated with darker (lower L*) skin colouration. It is concluded that a combination of conditioning snapper in white surroundings for 1 day before harvest, followed by immersion in seawater ice slurries supplemented with 300–450 mmol L?1 K+ improves skin pigmentation after >24 h of storage on ice.  相似文献

This paper is based upon previous and ongoing research within a glacierized, alpine river system (Taillon‐Gabiétous basin, French Pyrenees), which adopts an interdisciplinary approach to investigate the climate–hydrology–ecology cascade. Data are used to advance hypotheses concerning impacts of climate change/variability upon alpine river system hydrology and ecology.
  • 3. The snowpacks and glaciers that are the source of Pyrenean streams are climatically sensitive, with glaciers in retreat for most of the historical period. Given anticipated changes in summer air‐mass frequencies, the volume of meltwater may decrease, the nature and rate of glacier drainage may alter, and the timing of peak snow‐ and ice‐melt may shift. However, rainfall‐runoff and groundwater may increase their relative contributions to stream flow.
  • 4. The influence of changing water source contributions on physico‐chemical habitat and, in turn, on benthic communities is assessed using an alternative alpine stream classification. This model predicts more rapid downstream change in benthic communities in the future as meltwater contributions decline and, at the basin‐scale, biodiversity may be reduced owing to less spatio‐temporal heterogeneity in water sources contributions and, thus, physico‐chemical habitat. However, predictions are complicated by potential changes in biotic interactions with altered species' distributions.

Integrated, long‐term research into the climate–hydrology–ecology cascade in other alpine river basins is vital because interdisciplinary science is fundamental to predicting stream hydrology and ecology under scenarios of future climate/variability, to assessing the utility of alpine river systems as indicators of global change, and to developing conservation strategies for these fragile ecosystems.

As part of preliminary work aimed at the development of a formulated diet for the mud crab, Scylla serrata, an experiment was conducted with juvenile mud crabs (95.65±2.17 g) to determine apparent digestibility coefficients (ADC) for cellulose, fish meal, shrimp meal, blood meal, soybean meal, wheat flour and cod liver oil. Apparent digestibility coefficients for dry matter (ADCdm), energy (ADCenergy) and protein (ADCprotein) were in the ranges 70.0–95.7%, 77.4–97.1% and 57.7–97.9% respectively. Soybean meal had the highest ADCdm and wheat flour had the lowest value (P<0.05), while the ADCdm for fish meal, blood meal and shrimp meal were not different (P≥0.05). Similarly, soybean meal had the same ADCenergy as that of fish meal, but higher than those of cod liver oil, blood meal and shrimp meal (P<0.05). Moreover, the ADCprotein for blood meal or shrimp meal were not significantly different from fish meal (P≥0.05); nevertheless, they were lower than that of soybean meal and higher than that of wheat flour (P<0.05). Of significant interest was the ADCdm (78.0%) and ADCenergy (77.4%) for cellulose, which indicates that plant‐based nutrient sources may well be a useful component of formulated diets for mud crabs.  相似文献

We have previously demonstrated that sulforaphane is a potent inducer for thioredoxin reductase in HepG2 and MCF-7 cells (Zhang et al. Carcinogenesis 2003, 24, 497-503; Wang et al. J. Agric. Food Chem. 2005, 53, 1417-1421). In this study, we have shown that sulforaphane is not only an inducer for thioredoxin reductase but also an inducer for its substrate, thioredoxin in HepG2, and undifferentiated Caco-2 cells. Sulforaphane acts at two levels in the regulation of thioredoxin reductase/thioredoxin system by the upregulation of the expression of both the enzyme and the substrate. In human hepatoma HepG2 cells, sulforaphane induced thioredoxin reductase mRNA and protein by 4- and 2-fold, respectively, whereas thioredoxin mRNA was induced 2.9-fold and thioredoxin protein was unchanged in whole cell extracts, but an increase in nuclear accumulation (1.8-fold) was observed. Moreover, the induction of thioredoxin reductase was found faster than that of thioredoxin. The effects of PI3K and MAPK kinase inhibitors, LY294002, PD98059, SP600125, and SB202190, have been investigated on the sulforaphane-induced expression of thioredoxin reductase and thioredoxin. PD98059 abrogates the sulforaphane-induced thioredoxin reductase at both mRNA and protein levels in HepG2 cells, although other inhibitors were found less effective. However, both PD98059 and LY294002 significantly decrease thioredoxin mRNA expression in HepG2 cells. None of the inhibitors tested were able to modulate the level of expression of either thioredoxin reductase mRNA or protein in Caco-2 cells suggesting that there are cell-specific responses to sulforaphane. In summary, the dietary isothiocyanate, sulforaphane, is important in the regulation of thioredoxin reductase/thioredoxin redox system in cells.  相似文献

The crystallization and recrystallization of fats have a significant impact on the properties and quality of many food products. While crystallization has been the subject of a number of studies using pure triacylglycerols (TAG), recrystallization in similarly pure systems is rarely studied. In this work, perdeuterated tripalmitoylglycerol ( (2)H-PPP) was dissolved in medium chain triacylglycerol oil (MCT) to yield a saturated solution. The solution was heated to cause partial melting of the solid and dissolution of the molten fraction of (2)H-PPP in MCT and was then cooled to the original temperature to induce recrystallization from the supersaturated solution. (2)H NMR was used to monitor the disappearance of (2)H-PPP from the solution and showed that recrystallization occurred in two steps. The first step was rapid, taking place over a few minutes, and accounted for more than two-thirds of the total recrystallization. The second step was much slower, taking place over a remarkably long timescale of hours to days. It is proposed that dissolution occurs from all parts of the crystals, leaving an etched and pitted surface. The first step of crystallization is the infilling of these pits, while the second step is the continued growth on the smoothed crystal faces.  相似文献

Prior to 1995, the Montserrat oriole (Icterus oberi) was confined to ca. 30 km2 of hill forest on the Lesser Antillean island of Montserrat, but was not listed as globally threatened. Since then, the eruption of the Soufriere Hills volcano has destroyed more than half of the species’ range. Recent intensive monitoring has indicated that the species has also declined dramatically within the remaining intact forest, and is now critically threatened. Different monitoring and analytical methods indicate a decline of 8-52% p.a., and a remaining global population of ca. 100-400 pairs. This case study justifies the use of the restricted range criterion in designating threatened species. Despite intensive monitoring and the use of several analytical methods, it has proved surprisingly difficult to estimate the magnitude of the oriole’s decrease, or to control for potential artefacts in the census method. We discuss the reasons for this. The cause(s) of population decline in the intact forest are unclear, though two hypotheses appear plausible: a decrease in arthropod food, a result of volcanic ash fall, and an increase in nest predation as a result of increases in populations of opportunistic omnivores.  相似文献
