Cytokines have been associated with the progression of congestive heart failure (CHF) in humans and may be implicated in the pathophysiology of myxomatous mitral valve disease (MMVD) in dogs. The aim of this study was to determine the serum concentrations of cytokines in dogs with MMVD. The study included 16 Cairn terriers with no or minimal mitral regurgitation (MR), 41 Cavalier King Charles Spaniels (CKCS) with different degrees of MR and 11 dogs of different breeds with CHF due to MMVD. Granulocyte-macrophage colony-stimulating factor, interferon-γ, interleukin (IL)-2, IL-6, IL-7, IL-8, IL-10, IL-15, IL-18, keratinocyte-derived chemokine, interferon-γ-induced protein and monocyte chemoattractant protein-1 (MCP-1) were measured using a canine-specific multiplex immunoassay. CHF dogs had significantly higher MCP-1 concentrations than dogs with no or minimal MR. Among the CKCS, IL-2 and IL-7 decreased with increasing left atrial size and IL-7 also decreased with increasing MR. IL-8 decreased with increasing left ventricular end-systolic internal dimensions. MCP-1 was increased in CHF dogs compared to healthy control dogs and IL-2, IL-7 and IL-8 decreased with increasing indices of disease severity. The results suggest a role for these cytokines in canine MMVD and CHF.  相似文献

The serological characteristics of a group of 32 Pseudomonas anguilliseptica strains isolated in Spain from seabream (Sparus aurata) and turbot (Scophthalmus maximus) were compared with a total of 18 collection strains of this bacterial species with different geographical and host origin. The employment of different techniques, including slide agglutination, microagglutination and dot blot, allowed us to establish two serological groups, one comprising practically all the eel isolates, and the other including the majority of isolates from other fish species. The study of the lipopolysaccharides (LPS) and outer membrane proteins (OMP) corroborated these results, indicating that the serological differences among strains are due to the somatic antigen and not to antigenic determinants of protein nature. Therefore, a serological scheme of two "O" serotypes is proposed for P. anguilliseptica. The results obtained will be of importance for epidemiological studies as well as for the design of adequate vaccine formulations.  相似文献

Guazuma ulmifolia was experimentally pruned to determine when pruning should begin during the rainy season in order to extend the production of green tree-forage during the dry season. Three prunings (P-1, P-2, and P-3) were performed (5 weeks apart) during the rainy season (August, September, and October) and four forage harvests (C-a, C-b, C-c, and C-d) (3 weeks apart) took place during the dry season (February, March, and April). Over 2 years, forage biomass production was evaluated as total biomass (g dry matter tree?1), biomass of the morphological components (leaves, stems, and dead matter), and nutritional quality (crude protein, fiber, lignin, and digestibility). Date of pruning affected the production of total biomass (P = 0.001) with the earliest pruning (P-1) yielding the greatest forage quantity, while stems (P = 0.022) and dead matter (P = 0.032) varied due to a year by pruning interaction. Total biomass, leaves, stems, and dead matter varied by the interaction between forage harvest and year for all four variables (P < 0.037). In both years, the largest forage harvest occurred in C-b (P < 0.05), leaf production was highest in C-a and C-b (P < 0.001), stem production was greatest in C-b (P = 0.013) and dead matter was highest in C-b and C-d (P = 0.002). Leaf crude protein ranged between 10 and 19 %, and the interaction of pruning by forage harvest by year was significant (P = 0.035). Digestibility, neutral and acid detergent fiber and lignin differed significantly because of the interaction between forage harvest and year (P < 0.005), with February showing the lowest values for fiber and the highest digestibility. The best time to prune G. ulmifolia is in August so that the young trees will produce more total biomass with a higher crude protein content. The most suitable moment for forage harvest is in February when the trees have more leaves with greater digestibility and less fiber.  相似文献

Stocking rate manipulation was examined as a means of improving plant diversity (as a measure of pasture sustainability and forage value) in a native grass pasture used for dairy production in the humid tropics of Veracruz, Mexico. Given that environmental impact reduces biotic diversity, plant phylogenetic and functional diversity should decline with increased stocking rate. Stocking rates of 2, 3, and 4 cows · ha?1 and a rotational grazing plan of 3 d of occupation and 27 d of rest per pasture were applied continuously over 5 yr. Across 200 quadrats in each of two replicate paddocks per treatment, observed species richness, phylogenetic diversity (average taxonomic distinctness based on species presence/absence), and functional diversity (life-cycle duration and growth habit) were assessed. Most species were forb/herbs and forb/herb-subshrubs. Perennial species declined with increased stocking rate (F = 16.36, 0.05 > P > 0.02) while annual-perennial species increased (F = 76.88, 0.01 > P > 0.005); the proportion of annual species was least prominent and did not differ significantly. Observed species richness and phylogenetic diversity did not differ significantly with stocking rate. The correlation between functional diversity for life-cycle duration and phylogenetic diversity was significant and positive, suggesting that plant communities were predominantly assembled randomly from the surrounding species pool rather than through interspecies interactions acting to naturally filter immigrant species, thus leading to more opportunistic and undesired species. Although grazing pressure was not sufficient to alter indices or production measures, they did reveal shifts that may precede further pasture decline, indicating pasture sustainability was not being achieved. These rapid assessment methods permit monitoring for early warnings of reductions in pasture sustainability and forage quality for cattle.  相似文献

Streptococcus equi subsp. zooepidemicus is the pathogen most commonly isolated from the uterus of mares. S. zooepidemicus is an opportunistic pathogen and part of the resident flora in the caudal reproductive tract. The aim of this study was to investigate whether a genotypically distinct subpopulation of S. zooepidemicus is associated with endometritis in the mare, by genotyping and comparing uterine S. zooepidemicus strains with isolates from the vagina and clitoral fossa. Mares with (n = 18) or without (n = 11) clinical symptoms of endometritis were included. Uterine samples were obtained using a guarded endometrial biopsy punch, whereas a swab was used to recover samples from the cranial vagina and the clitoral fossa. If S. zooepidemicus was present, up to three colonies were selected from each anatomical location (max. 9 isolates per mare). Bacterial isolates were characterized by pulsed-field gel electrophoresis (PFGE) and multilocus sequence typing (MLST). S. zooepidemicus was isolated from the endometrium of 12 mares. A total of 88 isolates were analyzed by PFGE: 31 from the endometrium, 26 from the cranial vagina and 31 isolates from the clitoral fossa. For MLST 21 isolates were chosen. Results demonstrated a higher genetic similarity of the isolates obtained from infectious endometritis compared to isolates obtained from the caudal reproductive tract. In conclusion, we demonstrate for the first time that a genetically distinct group of S. zooepidemicus is associated with infectious endometritis in the mare.  相似文献

Transplacental transmission of bluetongue virus has been shown previously for the North European strain of serotype 8 (BTV-8) and for tissue culture or chicken egg-adapted vaccine strains but not for field strains of other serotypes. In this study, pregnant ewes (6 per group) were inoculated with either field or rescued strains of BTV-2 and BTV-8 in order to determine the ability of these viruses to cross the placental barrier. The field BTV-2 and BTV-8 strains was passaged once in Culicoides KC cells and once in mammalian cells. All virus inoculated sheep became infected and seroconverted against the different BTV strains used in this study. BTV RNA was detectable in the blood of all but two ewes for over 28 days but infectious virus could only be detected in the blood for a much shorter period. Interestingly, transplacental transmission of BTV-2 (both field and rescued strains) was demonstrated at high efficiency (6 out of 13 lambs born to BTV-2 infected ewes) while only 1 lamb of 12 born to BTV-8 infected ewes showed evidence of in utero infection. In addition, evidence for horizontal transmission of BTV-2 between ewes was observed. As expected, the parental BTV-2 and BTV-8 viruses and the viruses rescued by reverse genetics showed very similar properties to each other. This study showed, for the first time, that transplacental transmission of BTV-2, which had been minimally passaged in cell culture, can occur; hence such transmission might be more frequent than previously thought.  相似文献

The green fraction of humic acid (Pg) and the chloroform-extractable green fraction (CEGF) are characteristic soil organic matter (SOM) components. These alkaline solutions are green-colored due to the presence of 4,9-dihydroxyperylene-3,10-quinone (DHPQ) chromophore. While both of which are potential indicators for the effect of land use and paleoclimatic environment in the fields of soil science as well as geochemistry, CEGF as well as its relationship with Pg in soils are not yet fully documented. In this study, we firstly investigated the chemical properties of soil CEGF fractions by ultraviolet–visible (UV–Vis) and infrared (IR) method. Two CEGF components were separated by sequential liquid-liquid extraction using aqueous ammonium hydroxide (NH4OH) followed by aqueous sodium hydroxide (NaOH). Results showed that the UV–Vis spectral shape of NH4OH-extractable component is very similar to that of DHPQ, except that it is red-shifted. The solubility and UV–Vis spectrum of the NaOH-extractable fraction were completely identical with those of synthesized DHPQ. Their IR spectral shapes were also almost the same. Subsequently, the distribution of CEGF in humic acid (HA), fulvic acid (FA) and humin (HN) from Japanese Andosols and Cambisol was quantitatively evaluated by sequential extraction. Most of CEGF was detected in the HA (60–78%) and HN (22–40%), but not in the FA. While the UV–Vis spectral shape of CEGF extracted from Andosols HAs showed a relatively higher proportion of DHPQ than its derivative, the opposite was observed in Cambisol HA, whose CEGF is similar to that of sclerotium grain (one of the possible origin of CEGF). These results suggest the diversity of CEGF-producing soil fungi. Quantitative data also indicated that 35–49% of Pg consisted of a chloroform-soluble fraction (i.e., CEGF) and the remaining 51–65% of Pg was chloroform-insoluble. Based on these results, we propose that CEGF is composed of DHPQ and DHPQ-derivatives and that CEGF is one of the major fractions of Pg.  相似文献
