Echinococcus granulosus sensu stricto (G1) and Echinococcus ortleppi (G5) are haplotypes of the parasite formerly known as Echinococcus granulosus sensu lato, which in its larval stage causes cystic hydatid disease, endemic in Southern Brazil. Epidemiological and molecular knowledge about the haplotypes occurring in a region is essential to control the spread of the disease. The aim of this work was to analyze the haplotype frequency and fertility of hydatid cysts in cattle from the state of Rio Grande do Sul. Cysts were collected and classified according to their fertility status. DNA was extracted from protoscoleces and germinal layers and then used as template for the amplification of the cytochrome c oxidase subunit 1 gene by PCR. Amplicons were purified and sequenced, and the sequences were analyzed for haplotype identification. A total of 638 fertile cysts collected in the last ten years were genotyped. On average, G1 (56.6%) was more frequent than G5 (43.4%). In lungs, the G5 haplotype exhibited a higher parasite load (52.8%), whereas in the liver, G1 was more frequent (90.4%). The analysis revealed an increase in the frequency of G5 haplotype cysts during the period of sampling, and an increase in the abundance of fertile cysts has also been observed in the last several years. Most infertile cysts were genotyped as G1. The possible factors involved in the increase in the proportion of E. ortleppi (G5) and the consequences of this increase are discussed. This study suggests that the proportion of E. ortleppi (G5) loads in cattle may be increasing overtime.  相似文献

This paper compares lake chemistry in the Adirondack region of New York measured by the Temporally Integrated Monitoring of Ecosystems (TIME) and Adirondack Long-Term Monitoring (ALTM) programs by examining the data from six lakes common to both programs. Both programs were initiated in the early 1990s to track the efficacy of emission reduction policies and to assess the full impacts of acid deposition on surface water chemistry. They now serve to inform on the emerging chemical recovery of these waters. The Adirondack TIME program utilizes a probability-based approach to assess chronic acidification in a population of lakes using one summer sample per year. The ALTM attempts to track changes in both chronic and episodic acidification across a gradient of lake types using monthly samples. The ALTM project has two important attributes that contrast with the TIME program in the Adirondacks: higher temporal resolution (monthly versus once during the summer or fall) and speciation of aluminum. In particular, the ALTM program provides inorganic monomeric aluminum (AlIM), the fraction of Al that is most toxic. The monthly sampling of the ALTM program includes the spring snowmelt period when acid-neutralizing capacity and pH are near their lowest and Al levels are near their highest. We compare chemistry trends (1992?C2008) for sulfate, nitrate, base cations, dissolved organic carbon, hydrogen ion, acid neutralizing capacity, and Al for the six lakes common to both programs. We also compare relatively high springtime AlIM concentrations from the ALTM with relatively low summertime total Al concentrations from the TIME, showing that the ALTM program provides a more biologically relevant indicator of the effects of acid deposition, illustrating the value of the complementary monitoring efforts in the Adirondack region.  相似文献

The use of live algae and preserved algal products was examined as to their utility in supporting growth and survival of the feather duster worm, Sabellastarte spectabilis. Hatchery‐reared 4‐mo‐old worms were provided the same cell densities of live T‐Iso, preserved T‐Iso, live Nannochlropsis sp., and preserved Nannochloropsis sp. over the course of 132 d. A group of worms placed in a water table that received a continuous flow of raw seawater served as controls. Live and preserved T‐Iso resulted in the highest survival (86.7 ± 6.2% and 78.3 ± 16.5%, respectively) compared to all the other treatments. Live T‐Iso fed worms resulted in significantly larger worms than all other treatments with the preserved T‐Iso treatment supporting similar growth to those being fed live Nannochloropsis sp. and exposed to raw seawater. The preserved Nannochloropsis sp. resulted in significantly lower survival and growth when compared to all the other treatments. Fatty acid profiles of the algal diets and worm carcasses obtained during the current investigation suggest that T‐Iso outperformed Nannochloropsis because of its higher energy or fatty acid content.  相似文献

The objectives of this study were to examine the physicochemical and structural properties of peptide derivatives of dermaseptin S4, investigate their detrimental effects on red blood and sperm cells and ascertain their antibacterial potency to control bacterial contaminants in fresh bovine semen. The dermaseptin S4 peptide derivatives used in this study were K4S4, S4(5–28), S4(5–28)a, K20S4(5–28), K4S4(1–16)a, K4S4(1–15)a and K4S4(1–15). Peptides K4S4, S4(5–28)a, K20S4(5–28), K4S4(1–15)a and K4S4(1–16)a, with a higher positive charge, were the most potent against the bacterial strains tested, with the lowest minimum inhibitory concentration (MIC), whereas S4(5–28) and K4S4(1–15), with a lower positive charge, showed the highest MIC (p < .01). Haemolysis percentage depended on peptide concentration (p < .01). The K4S4 was the most powerful haemolytic peptide, showing the highest haemolysis percentage at all peptide concentrations (p < .01). In contrast, S4(5–28), S4(5–28)a, K20S4(5–28) and K4S4(1–15) were not able to produce 50% cell lysis up to 100 µM (p < .01). All peptides reduced sperm motility in a dose-dependent manner when used in concentrations from 16 to 64 μM (p < .01). The highest reduction was seen due to K4S4 activity, and the lowest reductions of sperm motility were observed due to K4S4(1–16)a and K4S4(1–15)a activity (p < .01). Hence, we can conclude that K4S4(1–16)a and K4S4(1–15)a at a concentration of approximately 15 µM are the most promising peptides as antibacterial agents in fresh bovine semen, because at this concentration, they showed the most potent antibacterial activity against evaluated strains without significant effects on haemolysis or a reduction in sperm motility.  相似文献

In Brazil, water buffaloes have been used to produce milk for mozzarella cheese production. Consequently, the main selection criterion applied for the buffalo genetic improvement is the estimated mozzarella yield as a function of milk, fat and protein production. However, given the importance of reproductive traits in production systems, this study aimed to use techniques for identifying genomic regions that affect the age at first calving (AFC) and first calving interval (FCI) in buffalo cows and to select candidate genes for the identification of QTL and gene expression studies. The single-step GBLUP method was used for the identification of genomic regions. Windows of 1 Mb containing single-nucleotide polymorphisms were constructed and the 10 windows that explained the greatest proportion of genetic variance were considered candidate regions for each trait. Genes present into the selected windows were identified using the UOA_WB_1 assembly as the reference, and their ontology was defined with the Panther tool. Candidate regions for both traits were identified on BBU 3, 12, 21 and 22; for AFC, candidates were detected on BBU 6, 7, 8, 9 and 15 and for first calving interval on BBU 4, 14 and 19. This study identified regions with great contribution to the additive genetic variance of age at first calving and first calving interval in the population of buffalo cows studied. The ROCK2, PMVK, ADCY2, MAP2K6, BMP10 and GFPT1 genes are main candidates for reproductive traits in water dairy buffaloes, and these results may have future applications in animal breeding programs or in gene expression studies of the species.  相似文献
