Foliar late blight, caused by Phytophthora infestans, is an important disease problem worldwide. Foliar resistance to late blight was found in a hybrid population of the cultivated diploid species Solanum phureja-S. stenotomum (phu-stn). The objective of this study was to determine if resistance to late blight could be improved by recurrent maternal half-sib selection in the phu-stn population. Four clones from each of 72 maternal half-sib families plus the check cultivar ‘Atlantic’ were evaluated for late blight resistance in replicated field trials in Pennsylvania. The most late blight resistant clone from each of the 72 families was randomly intermated to advance the population. The cycle 1, 2, and 3 populations were evaluated in 1996–1997, 2003–2004, and 2009–2010, respectively. Percent infected foliage was recorded several times towards the end of the growing season and used to compute area under the disease progress curve (AUDPC). Mean AUDPC was 652 (cycle 1), 556 (cycle 2), and 276 (cycle 3), whereas the mean AUDPC of ‘Atlantic’ ranged from 775 to 863 each cycle. Narrow-sense heritability for resistance was estimated as 0.78, 0.77, and 0.80 in cycles 1, 2, and 3, respectively. Recurrent maternal half-sib selection continues to improve the levels of resistance to late blight in this diploid population with no adverse effect on the amount of additive genetic variance. Additional improvements for late blight resistance are likely to occur if this approach is continued. 相似文献

- Although the frequency of occurrence of plastic ingestion in the large-sized dolphinfish and tunas taken by the Hawai'i longline fishery is very low (frequency of occurrence < 5% of sampled individuals), the ingestion of plastic in smaller-sized specimens caught with pole-and-line gear by commercial and recreational fishers has not been investigated.
- This study examined ingestion of >0.25 mm marine plastic debris (MPD) by four predatory fish species caught by commercial fishers around the Main Hawaiian Islands, and documented ingestion in three species: 85.7% of albacore tuna (n = 7), 40.0% of skipjack tuna (n = 10) and 12.5% of dolphinfish (n = 8).
- Yellowfin tuna (n = 10) did not contain any MPD, probably owing to the high proportion of empty stomachs (60%).
- For skipjack tuna, the frequency of occurrence of MPD ingestion was significantly higher for the smaller-sized specimens caught with pole-and-line (40%), compared with the larger-sized specimens caught with longlines (0%).
- For dolphinfish, the frequency of occurrence of MPD ingestion was similar for the similar-sized specimens caught with pole-and-line and with longlines.
- The ingested MPD items were micro–meso plastics, between 1 and 25 mm. While most ingested items were fragments, albacore also ingested line and skipjack also ingested sheets.
- The predatory fishes ingested light MPD items that float in sea water, but there were species-specific differences in their polymer composition: albacore contained more polypropylene and polyethylene, and skipjack contained more elastomers, characterized by a high percentage of ester plasticizers.
- Altogether, these results suggest that albacore and skipjack tunas ingest plastic of different types and polymers. Yet more research is needed to understand how differences in vertical distribution, foraging ecology and diet influence the MPD sampled by these predatory fish species.

Summary Seedlings of Trifolium repens showed considerable variation with regard to the morphology and growth of their calli, and their ability for in vitro differentiation of shoots. One of the lines selected for regeneration in primary callus cultures also showed shoot formation from protoplasts. Somatic embryogenesis in callus cultures of T. pratense and T. arvense occurred only in selected seedling lines. This paper highlights the importance of screening a large number of plants within a cultivar of outbreeding species to achieve reproducible plant regeneration from tissue culture. 相似文献

Four cycles of modified recurrent full‐sib (FS) selection were conducted in an intermated F2 population of European flint maize. The objectives of our study were to monitor trends across selection cycles in the estimates of population mean, inbreeding coefficients and variance components, and to investigate the usefulness of best linear unbiased prediction (BLUP) of progeny performance under the recurrent FS selection scheme applied. We used a selection rate of 25% for a selection index, based on grain yield and dry matter content. A pseudo‐factorial mating scheme was used for recombination. In this scheme, the selected FS families were divided into an upper‐ranking group of parents mated to the lower‐ranking group. Variance components were estimated with restricted maximum likelihood (REML). Average grain yield increased 1.2 t/ha per cycle, average grain moisture decreased 20.1 g/kg per cycle, and the selection index relative to the F2 check entries decreased 0.3% per cycle. For a more precise calculation of selection response, the four cycles should be tested together in multi‐environmental trials. We observed a significant decrease in additive variance in the selection index, suggesting smaller future selection response. Predictions of FS family performance in Cn + 1 based on mean performance of parental FS families in Cn were of equal precision as those based on the mean additive genetic BLUP of their parents, and corresponding correlations were of moderate size for grain moisture and selection index. 相似文献

Activities of the antioxidant enzymes ascorbate peroxidase, catalase, dehydroascorbate reductase, glutathione reductase, guaiacol peroxidase, monodehydroascorbate reductase, and superoxide dismutase were assayed in honeydew (Cucumis melo L.) fruit and spinach (Spinacia oleracea L.) leaves either as fresh, frozen to -80 degrees C, frozen in liquid nitrogen, freeze-dried, or acetone powder, representing the various ways tissues are treated prior to enzyme extraction. Treated tissues were analyzed following treatment or stored for up to 8 weeks at -80 degrees C. Enzyme activities in fruit frozen with or without liquid nitrogen and leaves frozen with or without liquid nitrogen or freeze-dried were equal to those of fresh tissue. Enzyme activities in freeze-dried or acetone-powdered fruit and leaves and in acetone-powdered tissues were significantly higher or lower than those in fresh tissue. Enzyme activities in both tissues frozen with or without liquid nitrogen and stored for 8 weeks at -80 degrees C changed little; those in freeze-dried and acetone-powdered tissues, however, significantly increased/decreased over the same period. Fresh tissue should be used in antioxidant enzyme assays, but if storage is necessary, tissues should be placed directly into a -80 degrees C freezer. 相似文献

Many new in vitro methods have been developed to evaluate antioxidant activity. Unfortunately, these in vitro methods often correlate poorly with the ability of compounds to inhibit oxidative deterioration of foods because the in vitro assays do not account for factors such as the physical location of the antioxidant, its interaction with other food components, and environmental conditions. To accurately evaluate the potential of antioxidants in foods, models must be developed that have the chemical, physical, and environmental conditions expected in food products. This paper outlines model systems of the evaluation of antioxidants in three types of foods: bulk oil, oil-in-water emulsions, and muscle foods. These model systems are not intended to be inclusive of all possible methods to measure lipid oxidation and antioxidant activity. The mammalian target of rapamycin (mTOR) protein kinase is a master growth promoter that nucleates two complexes, mTORC1 and mTORC2. Despite the diverse processes controlled by mTOR, few substrates are known. We defined the mTOR-regulated phosphoproteome by quantitative mass spectrometry and characterized the primary sequence motif specificity of mTOR using positional scanning peptide libraries. We found that the phosphorylation response to insulin is largely mTOR dependent and that mTOR exhibits a unique preference for proline, hydrophobic, and aromatic residues at the +1 position. The adaptor protein Grb10 was identified as an mTORC1 substrate that mediates the inhibition of phosphoinositide 3-kinase typical of cells lacking tuberous sclerosis complex 2 (TSC2), a tumor suppressor and negative regulator of mTORC1. Our work clarifies how mTORC1 inhibits growth factor signaling and opens new areas of investigation in mTOR biology. 相似文献

The first encounter between the sun and the surrounding interstellar cloud appears to have occurred 2000 to 8000 years ago. The sun and cloud space motions are nearly perpendicular, an indication that the sun is skimming the cloud surface. The electron density derived for the surrounding cloud from the carbon component of the anomalous cosmic ray population in the solar system and from the interstellar ratio of Mg(+) to Mg degrees toward Sirius support an equilibrium model for cloud ionization (an electron density of 0.22 to 0.44 per cubic centimeter). The upwind magnetic field direction is nearly parallel to the cloud surface. The relative sun-cloud motion indicates that the solar system has a bow shock. 相似文献
