The Amazon River prawn Macrobrachium amazonicum is endemic to tropical South America and is being intensively exploited by artisanal fisheries in Brazil. Limited information is available about the nutritional requirements of M. amazonicum, although the production of this species is now technically feasible. The digestive process in this species is still unclear and investigation into the digestive cells of its hepatopancreatic epithelium is required. Thus, the hepatopancreas from 15 specimens were fixed in Karnovsky Solution and processed for Transmission Electron Microscopy. Our results indicate that E cells are located at the distal portion of the hepatopancreatic tubule and are involved in mitotic activity. The cylindrical R cells are sparse and are only found in the proximal portion of the hepatopancreatic tubule. According to its ultrastructural characteristics, this cell is involved in pinocytosis. M cells are generally found near the R cells. The F cells are scattered throughout the length of the hepatopancreatic tubules, and B cells are observed mainly in the proximal and middle regions. F cells and B cells are likely related to, respectively, the synthesis of enzymes and the intracelular digestion. R‐ and M cells are probably related to material storage. Thus, these findings provide basic information on the cell types that perform protein digestion in M. amazonicum, and will be useful in further nutritional research. The identification and characterization of digestive cells is an important step towards understanding the digestive mechanisms.  相似文献

The authors report a rare case of bilateral Leydig cell tumor in a noncryptorchid stallion, describing the gross and microscopic findings. An 8-year-old Appaloosa stallion was examined for a unilateral enlargement of the testis associated with discomfort during palpation and signs of colic. General health conditions were good. Fine-needle aspiration cytology of the testis provided the diagnosis of bilateral Leydig cell tumor. Bilateral orchiectomy was performed and the testes were submitted to histopathologic examination that confirmed the diagnosis of Leydig cell tumor.  相似文献

Salmonellosis due to Salmonella enterica serovar Abortusovis (S. Abortusovis) is mainly characterized by abortion in sheep. Little is known about the immune response, which develops in the host as a result of infection. We evaluated the immune response of pregnant ewes vaccinated and successively exposed to full virulent S. Abortusovis. We found that vaccine constituted by inactivated S. Abortusovis induced both humoral and cellular-mediated immune response and that it provided protection against a challenge infection due to a fully virulent S. Abortusovis. Furthermore, we found an association between the lack of capability to produce IFN-gamma and abortion. This evidence suggests that protection against abortion can be associated to an IFN-gamma mediated mechanism. Our findings represent an interesting insight to better understand the interplay between host and S. Abortusovis and the effector mechanisms underpinning immune-based protection.  相似文献

Several sociological, health and conservation arguments request a correct labelling of seafood products. Nowadays, molecular genetics is a useful tool for food chain traceability, particularly in regards to species identification. Among the variety of PCR-based molecular markers, AFLPs (Amplified Fragment Length Polymorphisms) have recently been used to investigate genomes of different complexities. This paper assesses the potential use of the AFLP technology to determine fish and seafood species in processed commercial products and domestic stocks. In particular a species database of fish, molluscs and crustaceans has been created with the aim to identify species of origin of seafood products by previously defined AFLP patterns. Different EcoRI and TaqI primer combinations were selected from 20 screened combinations in relation to the total number of detected fragments and polymorphic ones. Most informative combinations were E32/T32, E32/T33, E33/T33, E33/T37, E33/T38, E40/T33, E40/T37, E42/T32, E42/T37. The comparison of informative markers between unknown frozen or fresh products and reference samples has enabled the accurate identification of 32 different species. The taxonomic characterization has been performed either at the species or at the population level depending on the number of available individuals. AFLP variation at the population level is particularly helpful for the stock traceability of domestic strains. Size homoplasy was also investigated in one species to assess the rate of non-homologous comigrating fragments and to detect additional polymorphic markers to be used in stock identification. Results of Band Sharing Index (BSI) and percentage of polymorphic fragments are presented and are discussed in relation to the wide applicability of AFLPs both for fish and seafood safety and authenticity testing in such fields as food traceability and restocking management. Cholesterol is a necessary element in the diet of prawns and it is involved in the metabolic processes of sexual maturation and synthesis of the molting hormone. Freshwater prawns are not able to synthesise cholesterol from specialised metabolic pathways. The aim of this study was to describe changes in structural and morphometric patterns of hepatopancreas associated with morphotypic differentiation of males Macrobrachium amazonicum when fed with different levels of cholesterol. Considering the four morphotypes of this species, Translucent Claw morphotype specimens were split into experimental units and divided into four groups, fed with different levels of cholesterol (0%, 0.5%, 1%, 2%) for 50 days. They were collected as they reached Green Claw (GC) morphotype, weighed to determine the hepatosomatic index, and the hepatopancreas was processed in histological routine for morphologic and morphometric analysis. In this study, it was possible to observe that the 2% cholesterol treatment exhibited more animals that reached the GC morphotype, and there was no significant weight gain in all treatments. Histologically, B‐cell abundance and hypervacuolized as well as F‐ and R‐cells shortages were observed as the percentage of cholesterol increased. This alteration in cellular profile correlates with the morphometric analyses that exhibited an increase in epithelial area and alterations in the lumen shape. This imbalance observed in the lumen‐epithelium relationship may be associated with a low digestive performance and possibly congested the functions of hepatopancreas. Thus, high doses of cholesterol promoted morphotypic differentiation without weight gain, due to alterations on hepatopancreas.  相似文献
