The objective of this study was to use mouse embryos as a model system to investigate the effect of co-culture of cumulus cells in Sydney IVF sequential media (Cook) on embryo development, based on the hypothesis that feeder cells in co-culture with a sequential medium could work synergistically to further improve in vitro culture conditions for mammalian preimplantation embryos. The culture systems described here were evaluated by the ability to consistently produce high blastocyst formation rates and high cell number per blastocyst. The role of embryo-to-cell contact was assessed by using Transwell inserts with transparent microporous membranes. Pronuclear embryos of ICR mice were cultured to blastocysts in Cook sequential media, with and without mouse primary cultures of cumulus cells, and with or without inserts. Blastocyst formation rates and cell numbers of in vitro developing embryos in the different culture systems were compared to each other, and to in vivo derived blastocysts. Blastocyst formation rates for Cook medium only was 27.8% (without inserts) and 32.9% (with inserts), whereas Cook-Cumulus cells in identical culture systems was significantly higher at 45.8% (without inserts, P<0.05) and 55.6% (with inserts, P<0.05). When the embryos are suspended above the bottom of the well, for Cook medium significantly lower blastocyst formation rates were observed at 4.2% compared to Cook-Cumulus cells at 17.5% (P<0.05). Mean cell numbers of blastocysts obtained in all co-culture systems were significantly higher (P<0.05) compared to those developing in culture medium only. Although the putative mechanism is as yet unexplained, the improved blastocyst formation rates and cell numbers in co-culture when there is direct contact between the embryo and the cell monolayer suggest that the close proximity between the feeder cells and embryos is in part responsible for these effects.  相似文献

金钱鱼(Scatophagus argus)是我国东南沿海名优养殖鱼类, 具有 XY 性别决定系统, Dmrt1 是其性别决定候选基因。金钱鱼生长具有性别二态性, 雌鱼生长快于雄鱼。目前缺乏快速鉴定金钱鱼遗传性别的分子标记, 阻碍了其性别控制育种技术的建立。本研究以公布的金钱鱼基因组数据, 在 Dmrt1 附近设计多对标记引物, 并通过 PCR 扩增验证标记的性别特异性。其中, 标记引物 Dmrt1-Marker-4-F/R 在雌鱼中仅扩增出一条 593 bp X 染色体条带, 而在雄鱼中能扩增出 593 bp 和 693 bp 两条条带, 分别来自 X 和 Y 染色体, 表明该标记为共显性标记。利用该标记检测我国南海沿岸 3 个不同地理群体 213 尾金钱鱼的遗传性别与表型性别完全一致。此外, 快速 DNA 提取试剂盒提取的片段较短 DNA 样品也可用于该对标记引物准确鉴定遗传性别。本研究建立了一种快速、准确、经济可靠的金钱鱼遗传性别鉴定方法, 将旨为促进金钱鱼性别控制育种技术的建立, 并为金钱鱼性别决定与分化机制研究提供依据。  相似文献

Ten apparently healthy, adult laughing doves were used to document detailed histological, histochemical and surface ultrastructural features of the nasal cavity and to investigate the structure‐function relationship of the nasal cavity in this species. We observed that the nasal cavity of the laughing dove was composed of three main regions: nasal vestibule, respiratory and olfactory. Each region presented a characteristic epithelial lining. The epithelium varied along the nasal vestibule from keratinized stratified squamous rostrally to non‐keratinized stratified squamous in the middle and stratified cuboidal in the caudal region of the nasal vestibule. The respiratory region was lined with pseudostratified columnar epithelium and was initially devoid of both goblet cells and cilia, but cilia then appeared and increased gradually in number close to the olfactory region. The caudal part of the respiratory region presented a stratified cuboidal epithelium. Strong alcianophilic, intra‐epithelial mucous glands were identified, starting at the caudal region of the nasal vestibule and extended into the respiratory region. The olfactory region was lined with a pseudostratified epithelium that consisted of three different cell types: olfactory, support cells and basal cells. In conclusion, the current investigation presents new information concerning the histological, histochemical and ultrastructural features of the laughing dove's nasal cavity. Furthermore, the findings of this study may prove to be a valuable contribution to the avian histology and pathology literature.  相似文献

This paper presents the effect of cross-sectional morphologies and composite structures of glass fiber felts on their corresponding acoustic properties. Glass fiber felts with random and layered cross-sectional structure are produced by centrifugal-spinneret-blow system. Acoustic properties are determined by a B&K impedance tube. The results show that sound transmission loss (STL) of glass fiber felts with layered cross-sectional structure exhibit greater than that with random cross-sectional structure. However, there is a little difference between the absorption coefficient values for random and layered cross-section. It means that glass fiber felts with layered cross-section are better to improve the sound insulation. With the increase of thickness, STLs of glass fiber felts with the same areal density do not increase monotonically due to the changing of porosity and characteristic impedances. Different glass fiber felts with layered cross-sectional structure are combined to form a variety of composite structures. It finds that assembly order of glass fiber felts have an effect on the total sound insulation. The large mismatch between the acoustic impedances causes multiple reflections leading to the best STL.  相似文献

The bioavailability and interactive effects of calcium, iron and zinc fortified whole wheat flour chapattis on female Sprague–Dawley albino rats were determined. The whole wheat flour was fortified with CaCO3, FeSO4 and ZnSO4 at 1000, 40 and 20 mg/kg, respectively and fed to 64 female Sprague–Dawley rats for 4 weeks. Retention of Ca, Fe and Zn was determined in plasma, femur and liver tissues. Ca levels in plasma and liver did not alter significantly while the Ca retention in femur changed significantly with the change in fortificants. The calcium retention was higher in the femur of rats fed on the diet supplemented with Ca + Zn (221 ± 13.61 mg/g) than the rats fed on the diet supplemented with Ca + Fe (219 ± 18.92 mg/g). The iron in plasma, liver and femur ranged from 2.19 ± 0.11 to 2.77 ± 0.10 μg/ml, 203.5 ± 13.24 to 251.5 ± 17.19 μg/g and 43.5 ± 1.05 to 52 ± 2.07 μg/g while zinc content ranged from 1.31 ± 0.089 to 1.55 ± 0.114 μg/ml, 89.5 ± 3.41 to 132 ± 6.10 μg/g and 175 ± 8.29 to 191 ± 10.85 μg/g, respectively. The minerals interacted with each other resulting in decrease of bioavailability among each other. The absorption was higher in the rats fed on fortified diet than those fed on unfortified chapatti diet. Hence, it is concluded that fortification of whole wheat flour is feasible to overcome the Ca, Fe and Zn malnutrition in the vulnerable groups.  相似文献

In this work, water-extraction of Vitis Vinifera L. (Black Grenache) leaves and analysis of aqueous extracts for anthocyanins were investigated. Vegetable leaves were cultivated in North of Tunisia at different dates. Anthocyanins were identified as main constituents in these natural dyeing materials using high performance liquid chromatography (HPCL). Dyeability of wool and its fastness properties using Vitis Vinifera L. leaves extract were also determined. Aqueous extract of Vitis Vinifera L. leaves has markedly yielded shades with good fastness properties. Thus, affordability is a point favorable for Vitis Vinifera L. leaves. Effects of dye bath pH and temperature on dyeability and fastness proprieties of wool fabrics by aqueous extract of Vitis Vinifera L. leaves were evaluated. Experimental results showed that fastness properties of dyed fabrics ranged from average to very good. Vitis Vinifera L. leaves extracts showed potential to dyeing wool fibers under highly acidic conditions (pH ≤2). The results obtained here revealed that natural dyes extracted from Vitis Vinifera L. leaves can be cost-effective for dyeing wool fabrics.  相似文献

Climate change in Pakistan has a great impact on the spatial and temporal variation of precipitation and ultimately alters the frequency and duration of droughts. In this study, spatial and temporal trend analyses of precipitation and droughts were observed at 58 meteorological stations across Pakistan from 1981 to 2018. The existing trend analysis methods were evaluated to address the issue of serial correlation in the climatic data. Results of precipitation analysis showed significant decreasing trends in winter (November, December) and significant increasing trends were observed in summer (June and September) at a confidence level of 95 percent. The magnitude of the precipitation trends showed the highest variation during summer season and the least variation in winter season. Rotated Principal Component (RPC) analysis showed the severe droughts (high positive loading) in southeastern side (Sindh province) of Pakistan due to lack of summer rains. Furthermore, variance correction approaches are identified as the most suitable in coping with the effect of serial correlation. The highest drought frequencies were observed in the southern areas of Pakistan and the drought events are expected to occur more frequently in the late winter, early spring, and early autumn, while droughts were expected to occur least frequently in summer.

Objective-To compare the anesthetic and cardiorespiratory effects of total IV anesthesia with propofol (P-TIVA) or a ketamine-medetomidine-propofol combination (KMP-TIVA) in horses. Design-Randomized experimental trial. Animals-12 horses. Procedure-Horses received medetomidine (0.005 mg/kg [0.002 mg/lb], IV). Anesthesia was induced with midazolam (0.04 mg/kg [0.018 mg/lb], IV) and ketamine (2.5 mg/kg [1.14 mg/lb], IV). All horses received a loading dose of propofol (0.5 mg/kg [0.23 mg/lb], IV), and 6 horses underwent P-TIVA (propofol infusion). Six horses underwent KMP-TIVA (ketamine [1 mg/kg/h {0.45 mg/lb/h}] and medetomidine [0.00125 mg/kg/h {0.0006 mg/lb/h}] infusion; the rate of propofol infusion was adjusted to maintain anesthesia). Arterial blood pressure and heart rate were monitored. Qualities of anesthetic induction, transition to TIVA, and maintenance of and recovery from anesthesia were evaluated. Results-Administration of KMP IV provided satisfactory anesthesia in horses. Compared with the P-TIVA group, the propofol infusion rate was significantly less in horses undergoing KMP-TIVA (0.14 +/- 0.02 mg/kg/min [0.064 +/- 0.009 mg/lb/min] vs 0.22 +/- 0.03 mg/kg/min [0.1 +/- 0.014 mg/lb/min]). In the KMP-TIVA and P-TIVA groups, anesthesia time was 115 +/- 17 minutes and 112 +/- 11 minutes, respectively, and heart rate and arterial blood pressure were maintained within acceptable limits. There was no significant difference in time to standing after cessation of anesthesia between groups. Recovery from KMP-TIVA and P-TIVA was considered good and satisfactory, respectively. Conclusions and Clinical Relevance-In horses, KMP-TIVA and P-TIVA provided clinically useful anesthesia; the ketamine-medetomidine infusion provided a sparing effect on propofol requirement for maintaining anesthesia.  相似文献
