We developed a method to detect Acidovorax avenae subsp. citrulli from cucurbit seeds that is more sensitive than conventional growing-out methods. Seeds were cultivated in test tubes for 2 weeks, and the bacteria grew semi-selectively on the seeds and seedlings. The plants were then vigorously shaken in water to suspend bacterial cells, and the suspension was spread on a selective medium for identifying the bacterium by colony appearance. The method is suitable for research purposes to evaluate the percentage of seeds infested with the bacteria, but not to detect bacterial contamination in commercial lots with thousands of seeds.  相似文献

In July 2006, black rot was observed on the leaves of 4-leaf-stage seedlings of salt-wort (Salsola komarovii) in Yamagata Prefecture, Japan. We isolated two single-conidial isolates from the diseased leaves. Although colony appearance of the isolates was different from that of each other, both isolates were identified as Colletotrichum truncatum by morphology and molecular similarity. After inoculation of healthy salt-wort plants with the isolates, the isolates were reisolated from symptomatic plants. We thus propose a new disease, anthracnose of salt-wort.  相似文献

A 12‐year‐old castrated male mixed breed dog was presented with anorexia, lethargy, intermittent vomiting, diarrhea, and weight loss. Clinicopathologic and imaging abnormalities included pancytopenia, icterus, and splenomegaly with multiple minute hypoechogenic nodules. Bone marrow (BM) smears revealed 2.5% hemophagocytic macrophages. In addition, an increased number of small to intermediate lymphocytes (16.3%) and plasma cells (3.2%) were recognized in the BM smears. More than 80% of the lymphocytes contained multiple small intracytoplasmic magenta granules. Histopathologic findings of the spleen revealed hemophagocytosis. Large granular lymphocytes (LGLs) were not found on the liver cytology or splenic histopathology at this time. PCR for antigen receptor rearrangement (PARR) analysis showed a clonal reaction in the T‐cell receptor ? (TCR?) gene in the BM sample. The dog was diagnosed with hemophagocytic syndrome (HPS). The dog was maintained in good condition with immunosuppressive therapy. However, the dog developed hepatic LGL lymphoma 7 months later. At this time, PARR analysis showed a clonal TCR? gene rearrangement in the hepatic LGL lymphoma samples. The BM and liver sample clonal rearrangements showed 100% homology, indicating that the small to intermediate granular lymphocytes in the BM at the HPS stage had progressed to hepatic LGL lymphoma. To our knowledge, this is the first report of canine secondary HPS caused by the occurrence of a BM LGL lymphoma clone that progressed to hepatic LGL lymphoma.  相似文献

The karyotypes of three shrew species (Mammalia, Soricomorpha, Soricidae) from Nepal were examined for the first time. Based on the karyotypes and overall differences in skull size, the current Episoriculus caudatus appeared to include two distinct species: the larger E. caudatus and the smaller Episoriculus sacratus. Episoriculus sacratus has three subspecies, E. s. soluensis in Nepal and Sikkim, E. s. umbrinus in Assam, Myanmar and the Yunnan Province of China, and E. s. sacratus in the Sichuan Province of China. Soriculus nigrescens had a diploid chromosome number (2n) and fundamental number (FN), including two X chromosomes, of 64 and 92, respectively, consisting of 11 metacentric or submetacentric, two subtelocentric and 18 acrocentric pairs of autosomes, a metacentric X and an acrocentric Y chromosome. Episoriculus sacratus soluensis had 2n = 74 and FN = 126, consisting of 12 metacentric or submetacentric, 13 subtelocentric and 11 acrocentric pairs of autosomes, a submetacentric X and an acrocentric Y chromosome. Episoriculus caudatus had 2n = 60 and FN = 118, consisting of 19 metacentric or submetacentric, nine subtelocentric and one acrocentric pair of autosomes, a subtelocentric X and an acrocentric Y chromosome. The karyotypes of these three species are characterized by their large 2n and FN values compared with other Soricidae.  相似文献

For the increase of the occupation ratio of inoculum strain in the competition with indigenous rhizobia, the relationship between Rj-genotypes of soybean and the preference of Rj-cultivars for various types of rhizobia for nodulation was investigated by using the Rj 2 Rj 4-genotype of soybean isolated from the cross between the Rj 2 Rj 3-cultivar IAC-2 and Rj 4-one Hill (Ishizuka et al. 1993: Soil Sci. Plant Nutr., 39, 79-86). Firstly, these Rj 2 Rj 4-genotypes were found to harbor the Rj 3-gene. The Rj 2Rj3Rj4-genotypes of soybean were considered to exhibit a more narrow microsymbiont range for nodulation than the Rj 2 Rj 3-and Rj4-cultivars. Therefore, rhizobia were isolated from the nodules of various Rj-genotypes of soybeans grown in soils, and the preference of the Rj 2 Rj 3 Rj 4-genotype for indigenous rhizobia was examined. The nodule occupancy of serotype 110 was significantly higher in the bacteroids of the nodules from the Rj 2 Rj 3 Rj 4-rgenotypes than in those from the other genotypes, non Rj-, Rj 2 Rj 3-, and Rj 4-cultivars. These results demonstrated that the Rj 2 Rj 3 Rj 4-genotype prefers more actively serogroup USDA110 to the others of rhizobia. Thus, Rj 2 Rj 3 Rj 4-genotype is superior to non- Rj-, Rj 2 Rj 3-, and Rj 4-genotypes for the formation of efficient nodules for nitrogen fixation.  相似文献

Leaf mold of tomato was found on cv. Momotaro-fight in 2003 in Ehime Prefecture. The symptoms were marginal indefinite yellowing on the upper leaf surface, and downy, gray to brown sporulation on the lower surface underneath the spots. The symptoms and morphology were the same as seen with Passalora fulva. The fungal isolates were identified as races 2.4, 2.4.11, 4, and 4.11 by inoculation tests. Races 4 and 4.11 have never before been found in Japan.  相似文献

We developed a one-step immunochromatography assay kit to measure high levels of canine trypsin-like immunoreactivity (cTLI) for bedside estimation of canine pancreatitis. The serum cTLI level can be determined within 10 min by visual comparison of color strengths in the test and reference zones. The serum cTLI levels determined by this method correlate well with canine TLI-ELISA and can be classified into 3 categories: cTLI levels higher than 60 ng/ml were considered positive; 20-60 ng/ml, weakly positive; and less than 20 ng/ml, negative. Twelve dogs suspected of pancreatitis were examined using this method; 4 dogs were positive, 2 were weakly positive, and 6 were negative. This test can detect a high level of serum cTLI and a positive result in the TLIH test will provide critical information for evaluation of pancreatitis in dogs.  相似文献

In tomato plants, salt stress has been induced to improve the quality of fruit. In general, plants under salt stress produce reactive oxygen species (ROS). Plants have their own ROS scavenging systems (antioxidant systems). In tomato plants, salt-induced changes in antioxidant systems have been examined extensively in leaves and roots; however, detailed information about salt-stressed fruits is not available. We examined the salt-induced changes in the antioxidant systems of the pericarp (containing epidermis) and pulp (containing seeds, placenta, and locule) during fruit ripening. Salt treatments were applied by adding 100 mM NaCl to the nutrient solution. In the pericarp and pulp, lipid peroxidation and hydrogen peroxide content were not increased by salt stress during ripening, indicating the absence of salt-induced oxidative stress. In the pericarp, the activities of superoxide dismutase (EC 1.15.1.1) and ascorbate (ASA)–glutathione (GSH) cycle-related enzymes increased with salt stress at the turning stage. Thus, at the turning stage, the antioxidant system may contribute to the enzymatic reaction involved ASA–GSH cycle. However, during the red and over-ripe fruit stages, salt stress produces little effect on antioxidant enzymes. In addition, the concentrations of antioxidants, such as the reduced form of ASA and GSH, increased during ripening in the control fruit, but those in the salt-stressed fruit remained unchanged. Therefore, the antioxidant system may contribute to the nonenzymatic reactions such as ASA and GSH taking place during the red and over-ripe fruit stages. In contrast, in the pulp, salt stress produces little effect on antioxidants and antioxidant enzymes. These results indicate that the salt-stressed fruit has protection mechanisms against salt-induced oxidative stress during ripening in both the pericarp and pulp. Salt-induced changes in antioxidant systems differed between the pericarp and pulp.  相似文献

Breadmaking properties (bread height, mm, and specific volume, cm3/g ) showed marked deterioration when bread dough was frozen and stored at ‐20°C for one day. However, these properties of bread dough baked after storage for three to six days were not further deteriorated as compared with that baked after one day of storage. A large amount of liquid was oozed from the frozen‐and‐thawed bread dough. The liquid was separated from the bread dough by centrifugation (38,900 × g for 120 min at 4°C), and collected by tilting the centrifuge tube at an angle of 45° for 30 min. There was a strong correlation between the amount of centrifuged liquid and breadmaking properties (bread height and specific volume). The mechanism responsible for the oozing of liquid in frozen‐and‐ thawed bread dough was studied. The presence of yeast and salt in bread dough was suggested to be closely related to the amount of centrifuged liquid, and fermented products particularly had a large effect on the amount of centrifuged liquid.  相似文献
