A metabolomics based approach has been used to study the infection of the Hwacheong rice cultivar (Oryza sativa L. cv. Hwacheong) with compatible (KJ201) and incompatible (KJ401) strains of the rice blast fungal pathogen Magnaporthe grisea. The metabolic response of the rice plants to each strain was assessed 0, 6, 12, 24, 36, and 48 h post inoculation. Nuclear
Magnetic Resonance (NMR) spectroscopy and Gas and Liquid Chromatography Tandem Mass spectrometry (GC/LC-MS/MS) were used to
study both aqueous and organic phase metabolites, collectively resulting in the identification of 93 compounds. Clear metabolic
profiles were observed at each time point but there were no significant differences in the metabolic response elicited by
each pathogen strain until 24 h post inoculation. The largest change was found to be in alanine, which was ~30% (±9%) higher
in the leaves from the compatible, compared to the resistant, plants. Together with several other metabolites (malate, glutamine,
proline, cinnamate and an unknown sugar) alanine exhibited a good correlation between time of fungal penetration into the
leaf and the divergence of metabolite profiles in each interaction. The results indicate both that a wide range of metabolites
can be identified in rice leaves and that metabolomics has potential for the study of biochemical changes in plant-pathogen

Spatial variability of humus layer (O layer) thicknesses can have important impacts upon soil water dynamics, nutrient storage and availability, as well as plant growth. The purpose of the present study was to elucidate the impact of forest‐transformation practices on the spatial variability of O layer thicknesses. The study focused on the Kahlenberg forest area (NE Germany) with stands of Scots pine (Pinus sylvestris) and European beech (Fagus sylvatica) of different age structures that form a transformation chronosequence from pure Scots pine stands towards pure European beech stands. Topsoil profiles including both, the O layer and the uppermost humic mineral soil horizon were excavated at intervals of 0.4 m along 15–20 m long transects, and spatial variability of O layer thicknesses was quantified by variogram analysis. The correlation lengths of total O layer thickness increased in the sequence consisting of pure pine stand (3.1 m) → older mixed stand (3.7 m) → pure beech stand (4.5 m), with the exception of the younger mixed stand, for which no correlation lengths of total O layer thickness could be determined. The degree of spatial correlation, i.e., the percentage of the total variance which can be described by variograms, was highest for the two monospecies stands, whereas this percentage was distinctly lower for the two mixed stands. A similar minimum for the two mixed stands was observed for the correlation lengths of the Oh horizon. These results suggest that the spatial structures of forest‐transformation stands may be interpreted in terms of a disturbance (in the form of the underplanting of beech trees). After this disturbance, the forest ecosystem requires at least 100 y to again reach relative equilibrium. These findings are in line with the results of other soil‐related investigations at these sites. 相似文献

Allergen detection is of increasing interest for food labeling purposes. A comparative study with a commercial hazelnut-specific PCR-ELISA and a sandwich-type ELISA detecting hazelnut protein was performed to investigate to what extent immunochemical and DNA-based techniques would correlate in the detection of trace amounts of potentially allergenic hazelnut residues. Both methods were highly sensitive and allowed the detection of even <10 ppm of hazelnut in complex food matrixes. The protein-ELISA was highly specific for hazelnut. However, some foods could lead to false-positive results at the 10 ppm level. The PCR-ELISA did not show any cross-reactions with non-hazelnut foods, thus reducing the probability of having false positives at the trace level. Forty-one commercial food products with and without hazelnut components on their labels were analyzed for the presence of hazelnut. Of the 27 products in which hazelnut components were detected, two samples were not identified by the protein-ELISA, and only one sample, namely one white chocolate having <1 ppm of hazelnut protein, was not detected by PCR-ELISA. The good correlation of the results of PCR-ELISA and protein-ELISA suggested that both PCR-based and immunochemical techniques are suitable for reliable detection of potentially allergenic hazelnut residues in foods at the trace level. 相似文献

The main results of the International Co-operative Programme on Effects on Materials, including Historic and Cultural Monuments (ICP Materials) within the United Nations Economic Commission for Europe (UN ECE) are summarised. The 8-year field exposure programme involves 39 test sites in 12 European countries and in the United States and Canada. Dose-response functions (DRF) expressing the effect of dry and wet deposition as individual terms have been obtained for a wide range of materials including bronze, copper, weathering steel, zinc, aluminium, nickel, tin, stone materials, paint coatings and glass materials. The DRF's includes parameters that are easily available on different geographical scales and can be used for mapping areas of increased corrosion rates and for calculation of costs. 相似文献

Forty-eight coal samples from the major coal mines in Guizhou were collected by means of subterranean channel sampling. The average mercury content in coal from this Chinese province is 0.53 mg kg-1, which is substantially higher than coal produced in any other province of China. A strong link is proposed to exist between atmospheric mercury emissions and the combustion of coal. During the last two decades, the amount of mercury emitted from coal combustion in Guizhou has increased invariably to 8.3 tons in 1998. Being concentrated to the pyrite fraction in the coal, the implementation of physical coal cleaning is likely to be an effective way to reduce the mercury content. Total and fractionated mercury in air and precipitation was measured in the provincial capital. During a measurement campaign in October 1999, the reactive gaseous mercury (RGM) concentration averaged at 450 pg m-3, which is much higher than that in pristine area (less than 20 pg m-3). The average total mercury in rainwater of 1996 was estimated to be 33 ng L-1, and the total annual wet deposition of mercury in the area was 39 g km-2 in 1996. 相似文献

Methyl isothiocyanate (MITC, CH3NCS) is the primary breakdown product of metam-sodium, and a potential replacement fumigant pesticide for methyl bromide. Methyl isothiocyanate is toxic and has a high potential for volatilization, therefore, minimizing its atmospheric emission is of the utmost importance. One method to reduce fumigant emissions is to enhance their degradation by incorporating organic amendments into the soil surface. In this study we determined the combined effect of temperature and chicken manure application rate on the degradation of MITC. The degradation of MITC was significantly accelerated by both increasing temperature and amendment rate. Differences between sterile and nonsterile degradation kinetics in unamended and organically amended soil indicate that MITC degradation is equally controlled by chemical and biological processes. The amelioration of soil with organic amendments should be further considered when designing fumigation practices that allow for reduced emissions. 相似文献

Bacteria were isolated from root-nodules collected from indigenous legumes at 38 separate locations in the Gascoyne and Pilbara regions of Western Australia. Authentication of cultures resulted in 31 being ascribed status as root-nodule bacteria based upon their nodulation of at least one of eight indigenous legume species. The authenticated isolates originated from eight legume genera from 19 sites. Isolates were characterised on the basis of their growth and physiology; 20 isolates were fast-growing and 11 were slow-growing (visible growth within 3 and 7 d, respectively). Fast-growers were isolated from Acacia, Isotropis, Lotus and Swainsona, whilst slow-growers were from Muelleranthus, Rhynchosia and Tephrosia. Indigofera produced one fast-growing isolate and seven slow-growing isolates. Three indigenous legumes (Swainsona formosa, Swainsona maccullochiana and Swainsona pterostylis) nodulated with fast-growing isolates and four species (Acacia saligna, Indigofera brevidens, Kennedia coccinea and Kennedia prorepens) nodulated with both fast- and slow-growing isolates. Swainsona kingii did not form nodules with any isolates. Fast-growing isolates were predominantly acid-sensitive, alkaline- and salt-tolerant. All slow-growing isolates grew well at pH 9.0 whilst more than half grew at pH 5.0, but all were salt-sensitive. All isolates were able to grow at 37 °C. The fast-growing isolates utilised disaccharides, whereas the slow-growing isolates did not. Symbiotic interactions of the isolates were assessed on three annual, one biennial and nine perennial exotic legume species that have agricultural use, or potential use, in southern Australia. Argyrolobium uniflorum, Chamaecytisus proliferus, Macroptilium atropurpureum, Ononis natrix, Phaseolus vulgaris and Sutherlandia microphylla nodulated with one or more of the authenticated isolates. Hedysarum coronarium, Medicago sativa, Ornithopus sativus, Ornithopus compressus, Trifolium burchellianum, Trifolium polymorphum and Trifolium uniflorum did not form nodules. Investigation of the 31 authenticated isolates by polymerase chain reaction with three primers resulted in the RPO1 primer distinguishing 20 separate banding patterns, while ERIC and PucFor primers distinguished 26 separate banding patterns. Sequencing the 16S rRNA gene for four fast- and two slow-growing isolates produced the following phylogenetic associations; WSM1701 and WSM1715 (isolated from Lotus cruentus and S. pterostylis, respectively) displayed 99% homology with Sinorhizobium meliloti, WSM1707 and WSM1721 (isolated from Sinorhizobium leeana and Indigofera sp., respectively) displayed 99% homology with Sinorhizobium terangae, WSM1704 (isolated from Tephrosia gardneri) shared 99% sequence homology with Bradyrhizobium elkanii, and WSM1743 (isolated from Indigofera sp.) displayed 99% homology with Bradyrhizobium japonicum. 相似文献

Abstract. In general, agricultural management has focused on differences between fields or on the gross differences within them. Recent developments in agricultural technology, yield mapping, Global Positioning Systems and variable rate applications, have made it possible to consider managing the considerable variation in soil and other properties within fields. This system is known as precision agriculture. More precise management of fields depends on a better understanding of the factors that affect crop input decisions. This paper examines the spatial variation in crop yield, soil nutrient status and soil pH within two agricultural fields using geostatistics. The observed properties vary considerably within each field. The relation between yield and the measured soil properties appears to be weak in general. However, the range of spatial correlation for yield, shown by the variogram, is similar to that of the soil chemical properties. In addition the latter changed little over two years. This suggests that information on the scale of variation of soil chemical properties can be derived from yield maps, which can also be used as a guide to a suitable sampling interval for soil properties. 相似文献

The mechanical properties of metastable soils have been approached from different backgrounds by civil engineers and soil scientists, with the main focus being on water‐induced self‐compaction rates. This study was conducted to determine the rebound‐recompression properties of metastable soil before and after wetting and to study the effect of water‐induced compaction on the mechanical stability depending on the overburden pressure. Rebound and recompression properties were measured with oedometer tests using sieved sandy loam, taken from a nonweathered Saalian glacial till layer, as the metastable test soil. The mechanical stability (preconsolidation load) of the dry and wetted soil was derived mathematically from recompression data after the test soil was unloaded. Void‐ratio changes during unloading and reloading were on a low level (Δe = 0.03). Rebound and recompression rates of the dry samples increased with load, whereas the void‐ratio change was twofold higher during recompression. Rebound rates of the wetted samples tended to decrease with load, while the recompression rates remained constant. Preconsolidation load calculated for the dry samples overestimated the simulated overburden pressure, whereas the wetted samples showed a high congruence. In both cases, the point of greatest curvature estimated in the earlier step of deviation gave values that were more realistic to the loads from which the unloading started. Our results indicate that both dry compaction and water‐induced compaction induce primarily plastic deformation. Furthermore, we found that in both the dry and wetted state the mechanical stability of the soil reflecting the overburden pressure remained constant. 相似文献
