Study objectives were to determine the effects of continuously infusing glucose (GLC) or casein (CAS) into the terminal ileum on biomarkers of metabolism, inflammation, and intestinal morphology in growing pigs. Crossbred gilts (n = 19; 81 ± 3 kg body weight [BW]) previously fitted with T-cannulas at terminal ileum were used in the current experiment. Following 4 d of acclimation, pigs were enrolled in 2 experimental 4-d periods (P). During P1, pigs were housed in individual pens and fed ad libitum for collection of baseline parameters. At the beginning of P2, pigs were assigned to 1 of 3 infusion treatments: 1) control (CON; water; 3 liters/d; n = 7), 2) GLC (dextrose 50%; 500 g/d; n = 6;), or 3) CAS (casein sodium salt; 300 g/d; n = 6). Water, GLC, and CAS solutions were continuously infused at a rate of 125 mL/h for the entirety of P2. Animals were euthanized at the end of P2, and intestinal tissue was collected. During P2, average daily feed intake differed across treatments and was reduced in GLC compared with CON pigs (14%), while CAS pigs consumed an intermediate amount (P = 0.05). Average daily gain and final BW were similar across treatments. A treatment by time interaction was observed for blood urea nitrogen (BUN; P < 0.01), as it decreased in GLC (21%) while it gradually increased in CAS (76%) pigs relative to CON pigs. Mild hyperthermia occurred with both GLC and CAS infusions relative to CON (+0.3 and 0.2 °C, respectively; P < 0.01). Blood neutrophils increased in CAS relative to CON pigs (26%) but remained similar between CON and GLC treatments (P < 0.01). Blood monocytes decreased in GLC relative to CON pigs (24%) while CAS pigs had an intermediate value (P = 0.03). Circulating lipopolysaccharide binding protein tended to decrease in GLC (29%) relative to CON pigs but remained similar between CON and CAS pigs (P = 0.10). Plasma tumor necrosis factor-alpha was similar across treatments. Ileum villus height:crypt depth was increased in CAS compared with CON pigs (33%; P = 0.05) while GLC pigs had an intermediate value. Colon myeloperoxidase-stained area increased in CAS compared with CON pigs (45%; P = 0.03) but remained similar between GLC and CON pigs. In summary, continuously infusing GLC or CAS into the terminal ileum appeared to stimulate a mild immune response and differently altered BUN patterns but had little or no effects on blood inflammatory markers, intestinal morphology, or key production parameters.  相似文献

The objective of this experiment was to evaluate the growth performance and bone mineral content (BMC) of nursery pigs in response to increasing total calcium (Ca) to available phosphorus (aP) ratios in diets containing phytase (250 FTU/kg; Natuphos E, BASF, Florham Park, NJ). A total of 480 nursery pigs (body weight (BW) = 5.7 ± 0.6 kg) with 10 pigs per pen and 7 pens per treatment (6 pens fed 2.75:1 diet) were allotted to seven treatments consisting of increasing ratios of calcium to available phosphorus (Ca:aP): 1.25, 1.50, 1.75, 2.00, 2.25, 2.50, and 2.75. From day −7 to 0, pigs were fed a common diet. They were then fed the treatment diets during two experimental phases from day 1 to 14 and 15 to 28, respectively. Available P was formulated to 0.33% and 0.27% (approximately 90% of requirement) in dietary phases 1 and 2, respectively. BW, average daily gain (ADG), average daily feed intake (ADFI), and gain-to-feed ratio (G:F) were determined. BMC of the femur was measured on day 28 on one pig per pen using dual x-ray absorptiometry. Data were analyzed as a linear mixed model using PROC MIXED (SAS, 9.3). Orthogonal polynomial contrasts were used to determine the linear and quadratic effects of increasing the Ca:aP. Over the 28-d experimental period, increasing Ca:aP resulted in a linear decrease in ADG (353, 338, 328, 304, 317, 291, and 280 g/d; P < 0.01), ADFI (539, 528, 528, 500, 533, 512, and 489 g/d; P < 0.05), and G:F (0.68, 0.66, 0.64, 0.62, 0.61, 0.59, and 0.58; P < 0.01). Increasing Ca:aP also resulted in decreased BW on days 14 and 28 (P < 0.01). The BMC of the femur decreased with increasing Ca:aP (6.2, 6.3, 5.7, 5.9, 5.5, 5.6, and 5.3 g; P < 0.05). Regression analysis explained the impact of Ca:aP as follows on ADG (ADG [g/d] = 339 − 36x; r2 = 0.81), G:F (G:F = 0.61 – 0.03x; r2 = 0.72), and BMC (BMC [g] = 6.4 – 0.27x; r2 = 0.43), where x is the Ca:aP. In conclusion, all outcomes indicated that any level of calcium above the minimum used in this experiment impaired growth performance and skeletal development. Further research using even lower levels of dietary Ca is warranted.  相似文献

The effect of dietary electrolyte balance on pigs fed lysine- or tryptophan-adequate or tryptophan-deficient diets was investigated in four experiments using 8- to 12-wk-old pigs. Electrolyte balance, expressed as Na+K-Cl in meq/kg of diet, was varied by altering dietary levels of Na and Cl while holding all other minerals constant. In two experiments in which the basal diet contained a balance of 135 meq/kg, simple lysine or tryptophan deficiences caused depressed growth, feed intake and efficiency of feed utilization, but none of these responses was altered by dietary supplementation with NaHCO3. In one experiment in which the electrolyte balance of the basal diet was 61 meq/kg and in which both lysine and tryptophan were limiting. NaHCO3 supplementation significantly increased growth and feed intake. This did not occur if the diet was also supplemented with tryptophan. A final experiment was conducted to determine the response of pigs to a range of electrolyte balance (-85 to 341 meq/kg) in a practical corn-soy diet containing adequate levels of all amino acids. Growth and feed intake appeared to be maximal for balances of 0 to 341 meq/kg Na+K-Cl, but were decreased at -85 meq/kg (P less than .05). Acid-base balance was adversely affected at 0 meq/kg. The results suggest that the response of lysine-deficient pigs to sodium bicarbonate is dependent upon the electrolyte balance of the diet, and also is influenced by other dietary amino acids.  相似文献

Objective

To determine the minimum infusion rate (MIR) of alfaxalone required to prevent purposeful movement in response to standardized stimulation while co-administered with lidocaine at three different doses by constant infusion rate infusion (CRI) in goats.

Study design

Prospective, blinded, randomized crossover, experimental.

Animals

A total of eight healthy goats: four does and four wethers.

Methods

Anaesthetic induction was with lidocaine at 1 mg kg?1 [low dose of lidocaine (L-Lid)], 2 mg kg?1 [moderate dose (M-Lid)] or 4 mg kg?1 [high dose (H-Lid)] and alfaxalone at 2 mg kg?1. Anaesthetic maintenance was with alfaxalone initially at 9.6 mg kg?1 hour?1 combined with one of three lidocaine treatments: 3 mg kg?1 hour?1 (L-Lid), 6 mg kg?1 hour?1 (M-Lid) or 12 mg kg?1 hour?1 (H-Lid). The MIR of alfaxalone was determined by testing for responses to a stimulation in the form of clamping on a digit with a Vulsellum forceps every 30 minutes during lidocaine CRI. Basic cardiopulmonary parameters were measured.

Results

The alfaxalone MIRs were 8.64 (6.72–10.56), 6.72 (6.72–8.64) and 6.72 (6.72–6.72) mg kg?1 hour?1 during L-Lid, M-Lid and H-Lid, respectively, without any significant differences among treatments. Compared to the initial rate of 9.6 mg kg?1 hour?1, these reductions in MIR are equivalent to 10, 30 and 30%, respectively. Significant increases in heart rate (HR) and arterial carbon dioxide partial pressure (PaCO2) and decreases in arterial haemoglobin saturation (SaO2), arterial oxygen partial pressure (PaO2) and respiratory frequency (fR) immediately after induction were observed during all lidocaine treatments.

Conclusions and clinical relevance

Lidocaine reduces the alfaxalone MIR by up to 30% with a tendency towards a plateauing in this effect at high CRIs. Immediate oxygen supplementation might be required to prevent hypoxaemia.  相似文献

Wheat by-products are feedstuffs that vary in nutritional value, partly because of arabinoxylans that limit nutrient digestibility. Millrun is a byproduct from dry milling wheat into flour and contains varying amounts of the bran, middlings, screening, and shorts fractions. The digestible nutrient content of mill-run is not well known. Effects of xylanase supplementation (0 or 4,000 units/kg of diet) on energy, AA, P, and Ca digestibilities were studied in a wheat control diet and 5 diets containing 30% of a by-product (mill-run, middlings, shorts, screening, or bran) in a 2 x 6 factorial arrangement of treatments. The wheat control diet was formulated to contain 3.34 Mcal of DE/kg and 3.0 g of standardized ileal digestible Lys/Mcal of DE. Diets contained 0.4% chromic oxide. Each of 12 ileal-cannulated pigs (32.5 +/- 2.5 kg) was fed 6 or 7 of 12 diets at 3 times the DE requirement for maintenance in successive 10-d periods for 6 or 7 observations per diet. Feces and ileal digesta were each collected for 2 d. Xylanase tended to increase (P < 0.10) ileal energy digestibility by 2.2 percentage units and the DE content by 0.10 Mcal/kg of DM and increased (P < 0.05) ileal DM digestibility by 2.8 percentage units; a diet x xylanase interaction was not observed. Xylanase increased (P < 0.05) total tract energy and DM digestibilities and the DE content. A diet x xylanase interaction was observed; xylanase increased (P < 0.05) total tract energy digestibility of the millrun diet from 72.1 to 78.9%, DE content from 3.19 to 3.51 Mcal/kg of DM, and DM digestibility from 71.5 to 78.6%. Diet affected (P < 0.05) and xylanase improved (P < 0.05) digestibility and digestible contents of some AA in diets and by-products, including Lys, Thr, and Val. Xylanase increased (P < 0.05) Lys digestibility by 13.8, 5.0, 5.2, 6.0, and 14.1 percentage units in millrun, middlings, shorts, screening, and bran, respectively. Diet affected (P < 0.01) total tract P and Ca digestibilities. Xylanase increased (P < 0.05) digestible P and Ca contents. In summary, nutrient digestibility varies among wheat by-products. Millrun contained 2.65 Mcal of DE/kg of DM, which xylanase increased to 3.56 Mcal of DE/kg of DM. Xylanase improved nutrient digestibility and DE content in wheat by-products; and the extent of improvement depended on the by-product. Xylanase supplementation may maximize opportunities to include wheat byproducts in swine diets and ameliorate reductions in nutrient digestibility that may be associated with arabinoxylans.  相似文献

The objectives of this experiment were 1) to determine the NE of soybean oil (SBO) and choice white grease (CWG) fed to growing and finishing pigs, 2) to evaluate the effects of inclusion rate of SBO on the NE by growing and finishing pigs, and 3) to determine if there is a difference in the NE of SBO and CWG between growing and finishing pigs. Forty-eight growing (initial BW: 22.13 ± 1.78 kg) and 48 finishing (initial BW: 84.17 ± 5.80 kg) barrows were used, and they were housed and fed individually. Within each stage of growth, pigs were allotted to 8 outcome groups of 6 barrows based on BW. Within each outcome group, pigs were randomly allotted to 1 of 6 groups. Two groups at each stage of growth served as an initial slaughter group. Pigs in the remaining groups were assigned to 4 dietary treatments and slaughtered at the conclusion of the experiment. The basal diet contained corn, soybean meal, and no supplemental lipids. Three additional diets were formulated by mixing 95% of the basal diet and 5% SBO, 90% of the basal diet and 10% SBO, or 90% of the basal diet and 10% CWG. Average daily gain and G:F for finishing pigs and apparent total tract digestibility of energy for growing and finishing pigs increased (linear, P < 0.05) with lipid content, but was not affected by lipid source. The lipid gain:protein gain ratio and the energy retention also increased (linear, P ≤ 0.05) with lipid content in growing and finishing pigs. There were no interactive effects between lipid content and stage of growth or between lipid source and stage of growth on the NE of diets and the NE of dietary lipids. The NE of diets increased (linear, P < 0.01) with increasing SBO (2,056, 2,206, and 2,318 kcal/kg for diets containing 0, 5, or 10% SBO). The NE of the diet containing 10% CWG (2,440 kcal/kg) was greater (P < 0.05) than the NE of the diet containing 10% SBO. The NE of diets was greater (P < 0.05) for finishing pigs than for growing pigs regardless of lipid content or source. The NE of SBO included at 5% (5,073 kcal/kg) was not different from the NE of SBO included at 10% (4,679 kcal/kg), but the NE of CWG (5,900 kcal/kg) was greater (P < 0.05) than the NE of SBO. The stage of growth had no impact on the NE of SBO or CWG. In conclusion, the NE of lipids is not affected by the content of dietary lipids, but the NE of CWG is greater than the NE of SBO.  相似文献

Feed efficiency represents the cumulative efficiency with which the pig utilizes dietary nutrients for maintenance, lean gain and lipid accretion. It is closely linked with energy metabolism, as the oxidation of carbon-containing components in the feed drive all metabolic processes. While much is known about nutrient utilization and tissue metabolism, blending these subjects into a discussion on feed efficiency has proven to be difficult. For example, while increasing dietary energy concentration will almost certainly increase feed efficiency, the correlation between dietary energy concentration and feed efficiency is surprisingly low. This is likely due to the plethora of non-dietary factors that impact feed efficiency, such as the environment and health as well as individual variation in maintenance requirements, body composition and body weight.Nonetheless, a deeper understanding of feed efficiency is critical at many levels. To individual farms, it impacts profitability. To the pork industry, it represents its competitive position against other protein sources. To food economists, it means less demand on global feed resources. There are environmental and other societal implications as well.Interestingly, feed efficiency is not always reported simply as a ratio of body weight gain to feed consumed. This review will explain why this arithmetic calculation, as simple as it initially seems, and as universally applied as it is in science and commerce, can often be misleading due to errors inherent in recording of both weight gain and feed intake.This review discusses the importance of feed efficiency, the manner in which it can be measured and reported, its basis in biology and approaches to its improvement. It concludes with a summary of findings and recommendations for future efforts.  相似文献

ObjectiveTo determine the minimum infusion rate (MIR) of alfaxalone required to prevent purposeful movement of the extremities in response to noxious stimulation.Study DesignProspective, experimental.AnimalsEight healthy goats; four does and four wethers.MethodsAnaesthesia was induced with alfaxalone 3 mg kg−1 intravenously (IV). A continuous IV infusion of alfaxalone, initially at 0.2 mg kg−1 minute−1, was initiated. Following endotracheal intubation the goats breathed spontaneously via a circle breathing circuit delivering supplementary oxygen. The initial infusion rate was maintained for 30 minutes before testing for responses. The stimulus was clamping on the proximal (soft) part of one digit of the hoof with Vulsellum forceps for 60 seconds. In the absence or presence of purposeful movement of the extremities, the infusion rate was reduced or increased by 0.02 mg kg−1 minute−1 and held constant for 30 minutes before claw-clamping again. Alfaxalone MIR was calculated as the mean of the infusion rates that allowed and abolished movement. Cardio-respiratory parameters were measured. Recovery from general anaesthesia was timed and quality scored. Results are presented as median (range).ResultsThe MIR of alfaxalone was 0.16 (0.14–0.18) mg kg−1 minute−1 or 9.6 (8.4–10.8) mg kg−1 hour−1. Induction of and recovery from anaesthesia were excitement-free. Cardio-respiratory changes were minimal, although compared to baseline HR increased, and at 2 minutes post-induction, (prior to oxygen supplementation), PaO2 decreased significantly from 84 (80–88) to 70 (51–72) mmHg [11.2 (10.7–11.7) to 9.3 (6.8–9.6) kPa]. Sporadic muscle twitches, unrelated to depth of anaesthesia, were observed during the period of general anaesthesia. Time (minutes) to sternal recumbency and standing were 4.0 (3.0–10.0) and 41.5 (25.0–57.0) respectively.Conclusions and Clinical RelevanceAlfaxalone can be used for total intravenous anaesthesia (TIVA) in goats and is associated with minimal adverse effects. Oxygen supplementation is advised, especially when working at higher altitudes.  相似文献

Site-segregated early weaning (SSEW) refers to the practice of weaning pigs from the sow at an early age and placing them in a nursery that is physically isolated from the breeding herd. An experiment involving 369 pigs was conducted at the Prairie Swine Center to investigate the impact of SSEW on pig performance when the herd of origin has a high health status and when housing and management conditions are kept as similar as possible across weaning regime. Three treatments were compared: weaning at 21+/-3 d and keeping the pigs on-site in an all-in-all-out nursery room (Control), weaning at 12+/-2 d of age and keeping the pigs on-site in a separate but identical all-in-all-out nursery room (OSEW), or weaning at 12+/-2 d of age and moving the pigs off-site to an all-in-all-out nursery room located 16 km from the Center (SSEW). Ventilation, feed, penning, feeders, and drinkers were kept as similar as possible for all treatments. Off-site weaning improved 56-d body weight (P < .05) by 12.5 and 8.3% compared with OSEW and Control, respectively. The improvement appeared to be the result of improvements primarily in feed intake but also due to enhanced efficiency of nutrient utilization. This experiment confirms that SSEW results in significant improvements in 56-d weights, even when the herd of origin has a relatively high health status.  相似文献

Acid-base balance and amino acid metabolism are intimately related. Changes in acid-base balance influence the metabolic fate of many amino acids. Also, acid-base homeostasis is achieved in part by alteration of amino acid metabolism, not only in the kidney, but also in liver, muscle and splanchnic tissue. Glutamine is the primary amino acid involved in renal ammonia-genesis, a process intimately related to acid excretion. The metabolism of other amino acids, such a serine, glycine and the branched-chain amino acids, also appears to be influenced by acid-base balance. Conversely, the metabolic fate of various amino acids will influence the daily acid load experienced by the animal. Oxidation of amino acids contributes to the total acid and base load imposed on the pig. The basic (cationic) amino acids (lysine, arginine and histidine) yield neutral end-products plus a proton; sulfur (methionine and cysteine) amino acids are also acidogenic because they generate sulfuric acid when oxidized. The dicarboxylic (anionic) amino acids (aspartate and glutamate, but not asparagine and glutamine) consume acid when oxidized and thus reduce the acid load of the diet. Acid-base balance and related phenomena are discussed in the context of practical and metabolic aspects of amino acid nutrition.  相似文献
