Tuberculosis (TB) in deer is a serious zoonotic disease of worldwide distribution. Detection of infected animals is usually performed using single or comparative skin-testing (SST/CST), although false responses due to sensitization to other mycobacteria may occur, hampering diagnostic specificity. We describe the evolution of the responses to the SST, CST and to an in-house serological assay in a red deer farm subjected to regular TB testing in southern Spain in an attempt to understand the dynamics of possible non-specific reactions occurring under field conditions. We performed 2288 skin-tests and ELISAs in nine sampling periods between May 2009 and January 2011. In May 2010, a strong increase in skin fold thickness in response to avian purified protein derivative (PPD) (mean=4.0mm, 95% CI=3.5-4.5) and bovine PPD (mean=1.8mm, 95% CI=1.6-2.0) was observed in yearling deer hinds (n=150), compared to values recorded for the same individuals in November 2009 (avian PPD: mean=0.7 mm, 95% CI=0.6-0.8 and bovine PPD: mean=0.7 mm, 95% CI=0.6-0.7) and in January 2011 (avian PPD: mean=2.2mm, 95% CI=1.9-2.4 and bovine PPD: mean=1.1mm, 95% CI=1.0-1.2). Using SST, 54 animals (36%) of the yearlings tested in May 2010 would have been classified as positive reactors, while none of them was positive in the CST. The five animals with highest skin fold increases to mycobacterial antigens were culled and subjected to post-mortem analysis, which confirmed the absence of Mycobacterium tuberculosis complex (MTBC) infection but demonstrated the presence of environmental mycobacteria and closely related bacteria in four out of the five analyzed animals. Our results demonstrated how non-specific responses to mycobacterial antigens can adversely affect the specificity of TB diagnosis based on the SST. Thus, once TB infection has been ruled out using confirmatory techniques, application of comparative diagnostic tests is highly advisable to maximize test specificity and avoid the slaughter of false positive reactors.  相似文献

Objective-To determine the minimal electric threshold of neurostimulation dorsally and ventrally to the interarcuate ligament in the lumbosacral area necessary to cause muscle contraction of the hind limb or tail and determine whether a continuous electrical stimulation applied to an insulated needle during lumbosacral epidural needle placement could be used to distinguish the epidural from the intrathecal space in rabbits. Animals-24 New Zealand white rabbits. Procedures-Rabbits received iohexol (0.2 mL/kg) either dorsally (group 1) or ventrally to the interarcuate ligament in the lumbosacral area (groups 2 and 3). Correct placement of the needle was determined by use of the loss of resistance to injection technique (group 2) or a continuous electrical stimulation (group 3) and confirmed by examination of the iohexol distribution pattern on radiographs. Results-In all rabbits of group 1, iohexol was injected in the lumbosacral area, outside the epidural space. In groups 2 and 3, iohexol was injected intrathecally. No pure iohexol epidural migration of iohexol was observed. Mean ± SD minimal electric threshold to elicit a motor response was 1.2 ± 0.3 mA, 0.3 ± 0.1 mA, and 0.3 ± 0.1 mA in groups 1, 2, and 3, respectively. Conclusions and Clinical Relevance-Neurostimulation was a useful technique to determine correct intrathecal needle placement in rabbits but failed to detect the lumbosacral epidural space when the common technique, used in dogs and cats for the lumbosacral epidural approach, was used.  相似文献

Plant—animal interactions at the landscape level become particularly relevant when land use is diversified. Nothofagus forests in southern Patagonia have been used for timber and cattle grazing purposes during the last century, causing livestock to increase (Bos taurus and Ovis aries) and large native herbivores, such as guanaco (Lama guanicoe), either maintained or decreased their populations. Within this scenario, feeding interactions between guanaco and domestic herbivores were analysed in a mosaic of open habitats and different Nothofagus forest types and management histories, whereby a total of six habitat types were identified through satellite image analysis. A total of 205 floristic surveys were conducted to characterize the plant species composition at the landscape level. Diet composition of herbivores was assessed once each season during a year, using microhistological analysis of feces. Results showed higher plant richness in open lands and lower in closed, unmanaged forests. Overall, 43 plant taxa were detected in herbivore feces, which represent 56% of plant richness detected in the field. Both guanacos and domestic herbivores included mainly grasses in their diet, which were found predominantly in open lands. Tree seedlings and saplings were consumed by all herbivores, as well, except during winter. Differential forage use between guanaco and domestic herbivores in southern Patagonian Nothofagus forests did exist. However, competition for available resources among theses herbivores resulted in an alternation of feeding sites, which varied throughout the year. Management plans in southern Southern Patagonia (livestock, silvopastoral plans and timber harvesting) do not consider the direct or indirect consequences on guanaco populations. The challenge, therefore, is to generate management decisions to avoid either guanacos or domestic herbivores from becoming detrimental to the sustainability of managed forested ecosystems.  相似文献

Porcine reproductive and respiratory syndrome virus (PRRSV) is endemic in most parts of Asia, where genotype I and II strains of diverse virulence may coexist. This study evaluated the outcome of infection with a highly virulent Asian genotype II PRRSV isolate in piglets vaccinated with a genotype I vaccine. Twenty-one 3-week-old piglets were divided in three groups: Pigs in group V (n=8) were vaccinated with an attenuated genotype I commercial PRRSV vaccine, while pigs in group U (n=8) and a control group (group C; n=5) were unvaccinated; 6 weeks later, pigs in groups V and U were challenged intranasally with a highly virulent strain of genotype II PRRSV (1×10(5) 50% tissue culture infectious doses/mL), while pigs in group C received a placebo. Over a period of 21 days after challenge, vaccinated pigs had significantly lower mortality (0/8 versus 2/8), fewer days of fever, a lower frequency of catarrhal bronchopneumonia, higher weight gains (13.4 versus 6.6 kg) and lower levels of viraemia compared to unvaccinated challenged pigs. Immunisation with a genotype I attenuated PRRSV vaccine provided partial protection against challenge with a highly virulent genotype II strain.  相似文献

The risk of zoonoses is a major obstacle to xenotransplantation. Porcine endogenous retrovirus (PERV) poses a potential risk of zoonotic infection, and its control is a prerequisite for the development of clinical xenotransplantation. The copy number of PERV varies among different breeds, and it has been suggested that the PERV integrations number is increased by inbreeding. The purpose of this study was (i) to examine the copy number of PERV in different Spanish pig breeds, Spanish wild boar and commercial cross-bred pigs from five different farms and genetic background (CCP1-CCP5) and (ii) to investigate the correlation between PERV copy number and the genetic background of the pigs in order to improve the selection of pigs for xenotransplantation. PERV copy number was determined by quantitative, real-time polymerase chain reactions. Thirty-four microsatellite markers were genotyped to describe the genetic diversity within populations (observed and expected heterozygosities, Ho and He, respectively) and the inbreeding coefficient (F). Pearson's correlation coefficient was used to determine the relationship between PERV copy number and Ho, He and F. The copy number of PERV among different pig breeds was estimated to range between three (CCP1) and 43 copies (Iberian Pig). Statistical differences were found among the studied populations concerning PERV copy number. No correlation was found between the PERV copy number and the heterozygosity (calculated at an individual level or at a population level) or the inbreeding coefficient of each population. Our data suggest that pigs inbreeding does not increase PERV copy number and support the idea that careful selection of pigs for organ donation with reduced PERV copy number will minimize the risk of retrovirus transmission to the human receptor.  相似文献

The dynamic removal of cadmium from aqueous solutions by natural and surfactant-modified Mexican zeolitic rocks (clinoptilolite–heulandite type) in fixed bed column systems was investigated. The performances of fixed bed columns were described through the breakthrough curves obtained from column experiments and the values of column parameters predicted as a function of bed height. The column adsorption data were evaluated in terms of the bed adsorption capacity and the efficiency of the process. The experimental results fitted well the bed depth service time model (BDST) for both adsorbents, and the empty bed residence time model (EBRT) was used to optimize column operating conditions. The surface modification of the zeolitic rock with surfactant affected the removal of cadmium in fixed bed systems. Moreover, a column experiment with surfactant modified zeolitic rock previously saturated with 4-chlorophenol was carried out and the results showed that this saturation had a negative effect on the performance of the column.  相似文献

The concentrations of As and Zn in 100 georeferenced soils uniformly distributed throughout the area affected by the spill from the Aznalcóllar mine (April 1998) were analysed at three depths (0–10, 10–30, and 30–50 cm) and on four dates (autumn–winter 1998, 1999, 2001, and 2004). For an estimate of the geochemical background, 30 unaffected soils near the edge of the spill were also analysed at the same depths. The soils were contaminated before the spill and, the accident seriously increased the concentration of As and Zn in the first 10 cm of almost all the affected soils. After the enormous efforts of cleaning up the tailings, around 45% of the soils had a concentration higher than 100 mg As kg?1 dry soil, and some 35% had a concentration higher than 1,000 mg Zn kg?1 dry soil. Both As and Zn penetrated between 10 and 30 cm in 25% and 45% of the soils, respectively, but reached 30 cm in only 12% of the soils. The remediation actions, especially the tilling and homogenisation of the uppermost 25 cm of the all soils, caused the As and Zn concentrations to decline in the soils, but this change was not very effective from the standpoint of pollution. Thus, 6 years after the spill, the uppermost 10 cm of 30% of the soils continued to have an As concentration higher than 100 mg As kg?1, while the Zn concentration diminished considerably on the surface due to its greater mobility, accumulating between 10 and 30 cm in depth, where 20% of the soils continued to register more than 1,000 mg Zn kg?1 dry soil.  相似文献

The beneficial effects of inoculating with Azospirillum brasilense on crop productivity have been widely described, but extensive use in typical agricultural field environments is scarcely documented. The objective of this study was to quantify the productivity of wheat (Triticum aestivum L.) whose seed was inoculated with a liquid formulation containing Azospirillum brasilense INTA Az-39 strain under typical dryland farming conditions. The study was performed in the 2002–2006 growing seasons, evaluating inoculated and non-inoculated seed at 297 experimental locations in the Pampas region of Argentina. The inoculated crops exhibited more vigorous vegetative growth, with both greater shoot and root dry matter accumulation (12.9 and 22.0%, respectively). The inoculation increased the number of harvested grains by 6.1%, and grain yield by 260 kg ha?1 (8.0%). Positive responses were determined in about 70% of the sites, depending mostly on the attainable yield and independently of fertilization and other crop and soil management practices. In general, more response to inoculation was observed in the absence of major crop growth limitations, suggesting the complementary contribution of the Azospirillum brasilense treatment to more efficiently developing higher yielding wheat.  相似文献
