Imiquimod is an immune response modifier with potent antiviral and antitumour activity. The objective of this pilot study was to evaluate the efficacy of an imiquimod 5% cream (Aldaratrade mark: 3M, Saint Paul, MN, USA) as a topical treatment for equine sarcoids. Fifteen horses with a total of 19 tumours were enrolled, including mixed (7), fibroblastic (5), flat (3), verrucous (2), and nodular (2) types. Baseline data included history, physical examination, tumour location, measurement and digital photography. Imiquimod was applied by the owners three times a week until complete resolution of the tumour or 32 weeks, whichever occurred first. Tumours were measured and photographed every 4 weeks. Treatment efficacy was defined as 75% or greater reduction of tumour size by the end of the trial. Four sarcoids were withdrawn from the study. Twelve of the remaining 15 tumours (80%) showed more than 75% reduction in size and nine (60%) totally resolved between 8 and 32 weeks. The most common adverse effects of exudation, erythema, erosions, depigmentation and alopecia were limited to the tumour and adjacent areas. The results suggest that topical imiquimod is a therapeutic option for the treatment of equine sarcoids, although more detailed studies are required to corroborate these initial findings.  相似文献

OBJECTIVE: To evaluate the analgesic efficacy of epidural administration of morphine or a morphine-bupivacaine combination administered before orthopedic surgery in dogs that received opioid premedication. DESIGN: Prospective, randomized, blinded, clinical study. ANIMALS: 36 healthy adult dogs that underwent elective orthopedic surgery on a pelvic limb. PROCEDURES: Each dog received 1 of 3 epidural treatments before surgery. Anesthetic and supportive care protocols were standardized. Dogs under going different surgical procedures were randomly allocated among the 3 treatment groups. Respiratory and cardiovascular variables, end-tidal isoflurane concentration, and requirements for rescue analgesia were monitored. Postsurgical analgesia was evaluated with a multiparametric pain scoring system and by determination of rescue analgesia requirements and cortisolemia. RESULTS: The morphine-bupivacaine combination was associated with lower values than morphine or a saline solution for intraoperative arterial blood pressure; minimum and maximum isoflurane requirements; and postoperative pain scores, rescue analgesia requirements, and plasma cortisol concentrations. Values obtained after administration of morphine alone were not significantly different from those obtained after administration of saline solution for most variables. CONCLUSIONS AND CLINICAL RELEVANCE: The preoperative epidurally administered morphine-bupivacaine combination induced better analgesia than morphine alone and should be considered for use in clinical patients. The degree of hemodynamic depression associated with the combination was considered acceptable for healthy patients undergoing elective surgery.  相似文献

OBJECTIVE: To investigate the effects of preventive angiotensin converting enzyme inhibitor treatment with ramipril in dogs with progressively severe experimentally induced heart failure. ANIMALS: 20 dogs. PROCEDURES: Dogs were randomly allocated to receive no treatment (control) or ramipril (0.125 mg/kg, PO, daily) for 7 weeks. Physical examination, repetitive catheterization of the right side of the heart, and echocardiography were performed before the study (day 0) and weekly for 7 weeks. Renal plasma flow (RPF) as determined by para-aminohippuric acid clearance and glomerular filtration rate (GFR) as determined by creatinine and iohexol clearances were measured on day 0 and at weeks 4 and 7. RESULTS: Overpacing induced a progressive increase in right atrial pressure (RAP) and pulmonary artery pressure, occluded (PAPO), with a decrease in systemic arterial pressure. There were progressive alterations of echocardiographic indices of diastolic and systolic ventricular function. The RPF and GFR decreased before cardiac output decreased, and filtration fraction increased. The logarithm of the urinary sodium-to-potassium concentration ratio (log(10)[Na(+)/K(+)]) decreased. Significant effects of ramipril included a delay in clinical signs of heart failure, a late decrease in RAP and PAPO, and increases in the sodium excretion fraction and log(10)(Na(+)/K(+)). There was a satisfactory agreement between the creatinine and iohexol clearance measurements. Conclusions AND CLINICAL RELEVANCE: Results suggest that, in this rapid-evolving, dilated cardiomyopathy, activation of the renin-angiotensin system contributes to the pathophysiology of heart failure late in the disease and essentially by an activation of renal salt and water retention.  相似文献

Successful control of livestock diseases requires an understanding of how they spread amongst animals and between premises. Mathematical models can offer important insight into the dynamics of disease, especially when built upon experimental and/or field data. Here the dynamics of a range of epidemiological models are explored in order to determine which models perform best in capturing real-world heterogeneities at sufficient resolution. Individual based network models are considered together with one- and two-class compartmental models, for which the final epidemic size is calculated as a function of the probability of disease transmission occurring during a given physical contact between two individuals. For numerical results the special cases of a viral disease with a fast recovery rate (foot-and-mouth disease) and a bacterial disease with a slow recovery rate (brucellosis) amongst sheep are considered. Quantitative results from observational studies of physical contact amongst domestic sheep are applied and results from the differently structured flocks (ewes with newborn lambs, ewes with nearly weaned lambs and ewes only) compared. These indicate that the breeding cycle leads to significant changes in the expected basic reproduction ratio of diseases. The observed heterogeneity of contacts amongst animals is best captured by full network simulations, although simple compartmental models describe the key features of an outbreak but, as expected, often overestimate the speed of an outbreak. Here the weights of contacts are heterogeneous, with many low weight links. However, due to the well-connected nature of the networks, this has little effect and differences between models remain small. These results indicate that simple compartmental models can be a useful tool for modelling real-world flocks; their applicability will be greater still for more homogeneously mixed livestock, which could be promoted by higher intensity farming practices.  相似文献

Purpose

Vertical transfer of solid matter in soils (bioturbation and translocation) is responsible for changes in soil properties over time through the redistribution of most of the soil constituents with depth. Such transfers are, however, still poorly quantified.

Materials and methods

In this study, we examine matter transfer in four eutric Luvisols through an isotopic approach based on 137Cs, 210Pb(xs), and meteoric 10Be. These isotopes differ with respect to chemical behavior, input histories, and half-lives, which allows us to explore a large time range. Their vertical distributions were modeled by a diffusion-advection equation with depth-dependent parameters. We estimated a set of advection and diffusion coefficients able to simulate all isotope depth distributions and validated the resulting model by comparing the depth distribution of organic carbon (including 12/13C and 14C isotopes) and of the 0–2-μm particles with the data.

Results and discussion

We showed that (i) the model satisfactorily reproduces the organic carbon, 13C, and 14C depth distributions, indicating that organic carbon content and age can be explained by transport without invoking depth-dependent decay rates; (ii) translocation partly explains the 0–2-μm particle accumulation in the Bt horizon; and (iii) estimates of diffusion coefficients that quantify the soil mixing rate by bioturbation are significantly higher for the studied plots than those obtained by ecological studies.

Conclusions

This study presents a model capable of satisfactorily reproducing the isotopic profiles of several tracers and simulating the distribution of organic carbon and the translocation of 0–2-μm particles.

BACKGROUND: Limiting the use of fungicides is due to become an important issue in managing Erysiphe necator (Schwein) Burrill infections in vineyards. The authors determined how three fungicides currently used by vine growers could be managed to control the early stages of an E. necator‐induced epidemic. RESULTS: Leaf‐disc bioassays and field experiments suggested that the protectant quinoxyfen induced minor disruption in E. necator development, but compounds with protectant and curative properties (tebuconazole and trifloxystrobin) caused significant, although different, disruption during E. necator‐induced epidemics. Bioassays showed that each of the antifungals were most effective at different stages of fungal development, tebuconazole before sporulation and trifloxystrobin after sporulation of the colonies. Results from the bioassay also highlighted likely occurrences in the field, where several stages of fungal development are encountered simultaneously. CONCLUSION: The present findings were complementary: leaf‐disc tests showed when the fungicides were most effective at inhibiting E. necator infection cycles; the field trial provided results in terms of incidence and severity of disease on bunches without reference to the pathogenic cycle development. The significance of mastocytaemia in cats is different from that in dogs because it appears exclusively associated with mast cell neoplasia. The prevalence of mastocytaemia was 0.05% of all feline submissions to a private laboratory and 43% in cats with mast cell neoplasia. None of 30 healthy cats had mastocytaemia. There was no sex bias or significant age difference between mastocytaemic and non-mastocytaemic cats with mast cell tumours (MCT). Buffy coat (BC) examination was the best screening method for detection of mastocytaemia but direct blood film examination was more accurate for quantifying degree of mastocytaemia. BC examination should be performed in all cases of suspected/known mast cell neoplasia as mastocytaemia was missed on nearly 30% of occasions when direct film examination only was used. Mastocytaemia was associated with decreased haematocrit (HCT) but not with other haematological parameters. Mastocytaemic cats can survive significant lengths of time (up to 27 months) even when splenectomy is not performed.  相似文献
