Two isoforms of the full-length cDNA of the growth hormone receptor (GHR) of the Atlantic salmon (Salmo salar; ss) were cloned by a PCR approach using RACE. Respectively, the cDNA sequences of ssGHR isoforms 1 and 2 are 2654 and 2608 nucleotides long, with 1782 and 1773 nucleotide ORFs. The resulting coded proteins
are 594 and 590 aa long, with 19 and 20 aa signal peptides. The two isoforms share 86% protein and 87% cDNA sequence similarity.
Isoform 1 is most similar to other salmonid GHR isoforms 1 while isoform 2 is most similar to salmonid GHR isoforms 2 (93–95%).
Similarity with other teleost species was lower (37–44%). The bioactivity of the cloned ssGHR was tested by transfecting the ssGHR isoform 1 cDNA into CHO-K1 hamster cells, incubating with recombinant salmon GH (sGH) or native ovine prolactin (oPRL),
and measuring cell proliferation by the MTT assay. The ssGHR-transfected cells significantly increased proliferation when stimulated by sGH at all concentrations. oPRL stimulated
ssGHR-transfected cells at higher concentrations due to receptor cross reaction. ssGHR isoforms 1 and 2 contain a single transmembrane domain and the typical conserved motifs found in other teleost GHRs, including
four paired cysteine residues and five potential N-glycosylation sites in the extracellular domain, Box I and Box II, as well as seven potential tyrosine phosphorylation sites
in the intracellular domain. However, in salmonids, these motifs differ from those of other teleosts, and could be responsible

This study quantifies the impact of four different grazing regimes (heavy, moderate, light and ungrazed) on the vegetation dynamics of rangeland ecosystems along the southern boundary area of the Mu Us Desert, China. As the grazing intensities decreased, the soil quality, canopy cover, height, density, above‐ and below‐ground biomass, litter, root/shoot ratio and native plant (Aneurolepidium dasystachys) and grass abundances significantly increased; the above‐ground biomass of grasses increased, but the above‐ground biomass of forbs decreased. Ungrazed grassland has significantly improved from grasslands experiencing three other levels of grazing pressure, especially in the grassland biomass. Species richness increased as the grazing intensity decreased in the grazing grasslands, but peak species richness appeared under moderate and light grazing against lower productivity. Grazing exclusion causes desirable transitions in plant communities of desert steppe rangelands. Therefore, appropriate and efficient grazing exclusion is an available way to counteract local grassland degradation and promote rangeland sustainability. 相似文献

Bronchoalveolar lavage was performed in 22 horses with pneumonia or pleuropneumonia. All horses had clinical evidence of pneumonia or pleuropneumonia on the basis of physical, radiographic, ultrasonographic, tracheobronchial aspirate or post-mortem findings. Results of lavage fluid analysis were normal in 9 horses, equivocal in 3 horses, and abnormal in 10 horses. Abnormal lavage fluid had increased total cell count, increased relative and absolute neutrophil counts, degenerative neutrophils, and decreased relative and absolute macrophage and lymphocyte counts. 相似文献

Nine laboratories participated in a collaborative study on determination of crude protein in animal feeds to compare a generically described combustion method with the AOAC mercury catalyst Kjeldahl method (7.015). The combustion method was written in general terms of method principle, apparatus specifications, and performance requirements. The sample set comprised closely matched pairs of feed ingredients and mixed products ranging from 10 to 90% protein. Ten pairs ground to 0.5 mm were the focus of the study; 4 pairs were ground to 1.0 mm for comparison. Nicotinic acid and lysine monohydrochloride were included as standards. Collaborators were instructed to report their results for performance checks using materials supplied. Only one laboratory failed to meet the proposed limits. Seven laboratories used the LECO Model FP-228 analyzer and 2 used the LECO CHN 600 analyzer. For the 0.5 mm pairs, repeatability standard deviations (Sr) ranged from 0.09 to 0.58 for the Kjeldahl method and from 0.14 to 0.33 for the combustion method, with a pooled Sr value of 0.28 and relative standard deviation (RSDr) of 0.59%. Reproducibility standard deviations (Sg) ranged from 0.23 to 0.86 (Kjeldahl) and from 0.30 to 0.61 (combustion), with a pooled Sg value of 0.52 and RSDg of 1.10%. Grand means for the samples ground to 0.5 mm were 47.65% protein by the combustion method and 47.41% protein by the Kjeldahl method. For samples ground to 1.0 mm, corresponding values were 31.82 and 31.50% protein. The generic combustion method has been approved interim official first action. 相似文献

A water-soluble peridinin-chlorophyll-protein complex from the dinoflagellate Gonyaulax polyedra was found to have a molecular weight of about 38,000. The complex could be disrupted by digestion with proteolytic enzymes. No electron transfer was observed when the complex was irradiated. 相似文献

Objective – To describe the clinical presentation, case management, and outcome in 2 foals with Rhodococcus equi infection associated with presumptive severe immune‐mediated hemolytic anemia. Series Summary – Two foals diagnosed with R. equi pneumonia on the basis of tracheal wash cultures, thoracic radiographs, and thoracic ultrasonography were concurrently diagnosed with hemolytic anemia. Both foals required whole blood transfusions, and were treated with the antimicrobial combination of rifampin and a macrolide (eg, clarithromycin, erythromycin, or azithromycin). Dexamethasone was used to prevent further hemolysis in both foals, and to treat acute lung injury/acute respiratory distress syndrome in 1 of the foals. Both foals survived, and required prolonged antimicrobial therapy. New or Unique Information Provided – Although extra‐pulmonary disorders are commonly diagnosed in foals infected with R. equi, hemolytic anemia is rarely described. Dexamethasone is considered the treatment of choice for immune‐mediated hemolytic anemia, but may be contra‐indicated in foals with severe bacterial infections. In these foals, a relatively low dose and short duration of dexamethasone was utilized in an attempt to minimize immune suppression, although early discontinuation in 1 foal precipitated a second hemolytic crisis. 相似文献

An analytical procedure using accelerated solvent extraction and capillary gas chromatography with electron capture and flame photometric detections was developed to simultaneously determine residues of different pesticides in fruits and vegetables. Single laboratory validation of the method was carried out for 28 compounds selected from eight pesticide classes, in blank and fortified samples of fresh pear, cantaloupe, white potato, and cabbage. The method had to meet specific established validation criteria for regulatory purposes applicable to our laboratory. At each of the two fortification levels studied, 24 of the 28 pesticides gave recoveries of more than 70% with a coefficient of variation of less than 10%. With respect to existing procedures, the method showed acceptable limits of detection (from 0.0019 to 0.14 microg/g depending on the pesticide and matrix) while minimizing environmental concerns, time, and labor. 相似文献

A mammary gland neoplasm was identified at necropsy in a 9-year-old ovariectomized Holstein cow with a 1-year history of an enlarged left hindquarter. A sterile serous secretion was expressed from the quarter. The cow developed Corynebacterium pyogenes infection in the quarter one year later and died of secondary complications. The left hindquarter was found to contain multiple cysts. The histologic diagnosis was multicentric papillary cystadenoma. 相似文献

A 15-year-old, Standardbred broodmare with an aortic sinus aneurysm developed rupture of the aneurysm with subsequent rupture of a tricuspid valve chorda tendinae, tricuspid regurgitation, acute right-sided congestive heart failure, and pulmonary thromboembolism. Shunting of blood from the aorta through the ruptured aneurysm into the right ventricle resulted in decreased renal perfusion and acute renal failure. Initially, treatment of the mare with analgesics, fluids, and digoxin resulted in clinical improvement, but the mare's condition deteriorated after 8 days and the mare was euthanatized due to unrelenting pain and a poor prognosis. Echocardiography was useful in diagnosis of the cardiac disease in the broodmare. 相似文献
